Metagenomic and paleopathological analyses of a historic documented collection explore ancient dental calculus as a diagnostic tool.

Dental calculus is a microbial biofilm that contains biomolecules from oral commensals and pathogens, including those potentially related to cause of death (CoD). To assess the utility of calculus as a diagnostically informative substrate, in conjunction with paleopathological analysis, calculus samples from 39 individuals in the Smithsonian Institution's Robert J. Terry Collection with CoDs of either syphilis or tuberculosis were assessed via shotgun metagenomic sequencing for the presence of Treponema pallidum subsp. pallidum and Mycobacterium tuberculosis complex (MTBC) DNA. Paleopathologic

Calculus and survivorship in medieval London: The association between dental disease and a demographic measure of general health.

OBJECTIVES: Dental plaque is associated with a variety of systemic diseases and mortality risks in living populations. However, bioarchaeologists have not fully investigated the mortality risks associated with plaque (or its mineralized form, calculus) in the past. This study examines the relationship between survivorship and calculus in a medieval skeletal sample. MATERIALS AND METHODS: Our sample (n = 1,098) from four medieval London cemeteries, c. 1000-1540 CE, includes people who died under attritional (normal) and catastrophic (famine and plague) conditions. The associations between age

Entrapment of drugs in dental calculus - Detection validation based on test results from post-mortem investigations.

For prospective investigation of drugs and metabolites in archaeological and contemporary dental calculus, a sensitive, broadly applicable ultra-high-performance liquid chromatography-tandem mass spectrometry method using pneumatically assisted electrospray ionisation (UHPLC-ESI-MS/MS) was developed. The dental calculus was treated with citric acid and the dissolution extracts were cleaned using weak and strong polymeric cation-exchange sorbents. The method was validated on hydroxyapatite for the analysis of 67 drugs and metabolites. Typically, the lower limits of quantification were in the ra

Functional Oral Intervention Associated With Reduced Dental Calculus Formation and Secondary Facial Changes: A Case Report.

Dental calculus accumulation is traditionally attributed to inadequate oral hygiene; however, oral function may also influence its formation. This case report describes a patient who presented with a sudden increase in calculus formation on the lingual surface of mandibular anterior teeth, despite no changes in oral hygiene habits. Clinical assessment revealed lip incompetence and signs suggestive of mouth breathing following prolonged mask use. A functional oral intervention, focusing on tongue mobility, bilateral mastication, and lip competence, was implemented without modifying brushing ins

An engineered peptide-polymer conjugate mimics statherin to block dental calculus formation while preserving oral microbiota.

Dental plaque accumulates calcium and phosphorus ions from saliva and gradually mineralizes into calculus, a primary etiological factor of periodontal disease. The complex oral environment limits the efficacy of conventional antibacterial and anticalculus agents in preventing calculus formation. Inspired by statherin's mineral-binding properties, a peptide-polymer conjugate SNA6-PEG-TCS (DPT) was synthesized comprising a mineral-binding peptide (SNA6), antifouling polyethylene glycol (PEG) and antibacterial triclosan (TCS). DPT exhibits high water solubility and eradicates 88.71% of plaque whi

Paleo-proteomic analysis of Iron Age dental calculus provides direct evidence of Scythian reliance on ruminant dairy.

The Scythians, often described as mounted horse-back warriors of the Iron Age steppe with lavish burial goods, have attracted increasing scientific interest over the past years. Recent genetic and multi-isotopic studies have uncovered that the 'Scythians' were neither a homogenous political nor a cultural group, but rather diverse populations of heterogeneous origins with intricate socio-political systems. Although populational differences in agro-pastoral subsistence regimes of Northern Black Sea Region groups have previously been identified through stable isotope analysis, it remains unclear

Modifiable early life risk factors for dental calculus in dogs: a retrospective cross-sectional study in Finland.

BACKGROUND: Dental calculus (DC) in dogs is a common oral health issue that can result in discomfort and may lead to systemic complications, if not managed effectively. While dental care practices are often overlooked by many owners, it remains crucial to identify and address possible DC risk and protective factors. The present retrospective cross-sectional study sought to examine the early life factors contributing to the emergence of owner-reported DC in dogs later in life, emphasizing the impact of modifiable variables. The study examined exposure data across five pivotal early life stages,

Unlocking the past: Dental calculus as key to understanding ancient health and disease through a One Health framework.

OBJECTIVE: Ancient dental calculus (DC) serves as a critical biomolecular repository, preserving dietary and extra-dietary residues related to past health and disease. This review examines DC as a potential 'One Paleopathology' resource, highlighting the interconnectedness of past human-animal-environmental health. Through a One Health lens, it critically assesses current research and opportunities for the next 50 years of study. RESULTS: By synthesizing the available literature and the current state of the art, we critically evaluate how DC research bridges paleopathology, bioarchaeology, an

A Journey into the Evolution of Human Host-Oral Microbiome Relationship through Ancient Dental Calculus: A Scoping Review.

One of the most promising areas of research in palaeomicrobiology is the study of the human microbiome. In particular, ancient dental calculus helps to reconstruct a substantial share of oral microbiome composition by mapping together human evolution with its state of health/oral disease. This review aims to trace microbial characteristics in ancient dental calculus to describe the evolution of the human host-oral microbiome relationship in oral health or disease in children and adults. Following the PRISMA-Extension for Scoping Reviews guidelines, the main scientific databases (PubMed, Scopus

Tooth wear and dental calculus in a group of orca (Orcinus orca) stranded on the New Zealand southern coast.

In 2014, a group of nine orca stranded in Te Waewae Bay, Western Southland, and their remains were collected, processed and studied in partnership with Ōraka Aparima Rūnaka. A total of 436 teeth were analysed to determine the location, type and intensity scores of tooth wear. The presence of dental calculus and percentage surface coverage was assessed on 1744 surfaces. Tooth wear and calculus were investigated in relation to sex and total body length (as a proxy for age). All orca had some degree of tooth wear; however, most teeth were worn superficially. Only two individuals had severe wear a

Commensal and Pathogenic Members of the Dental Calculus Microbiome of Badia Pozzeveri Individuals from the 11th to 19th Centuries.

The concept of the human oral microbiome was applied to understand health and disease, lifestyles, and dietary habits throughout part of human history. In the present study, we augment the understanding of ancient oral microbiomes by characterizing human dental calculus samples recovered from the ancient Abbey of Badia Pozzeveri (central Italy), with differences in socioeconomic status, time period, burial type, and sex. Samples dating from the Middle Ages (11th century) to the Industrial Revolution era (19th century) were characterized using high-throughput sequencing of the 16S ribosomal RNA

Palaeoproteomic characterization of archaeological dental calculus reveals precarious periodontal health in pre-Roman Italy (7th-4th century BCE).

Periodontitis, a chronic inflammatory disease affecting the tooth-supporting structures, is a key indicator of oral health in palaeopathology. While poor oral hygiene, systemic diseases, and genetics are well-established contributors, the dietary impact has often been underestimated. Clinical studies, however, link diets high in fermentable carbohydrates and meat to inflammation. We investigated periodontal disease by analyzing interdental septa in 63 individuals from elite and non-elite groups in pre-Roman Italy (7th-4th centuries BCE), a period of social stratification, intensified agricultu

Dental calculus: recent insights into occurrence, formation, prevention, removal and oral health effects of supragingival and subgingival deposits.

Dental calculus, both supra- and subgingival occurs in the majority of adults worldwide. Dental calculus is calcified dental plaque, composed primarily of calcium phosphate mineral salts deposited between and within remnants of formerly viable microorganisms. A viable dental plaque covers mineralized calculus deposits. Levels of calculus and location of formation are population specific and are affected by oral hygiene habits, access to professional care, diet, age, ethnic origin, time since last dental cleaning, systemic disease and the use of prescription medications. In populations that pra

The comparative efficacy of two commercial tartar control dentifrices in preventing calculus development and facilitating easier dental cleanings.

Tartar control dentifrices have been proven effective in reducing the build-up of supragingival calculus deposits between professional tooth cleanings. In addition to providing consumers with a cleaner dentition, these formulations can, in principle, contribute to easier professional tooth cleaning. Until recently, clinical methods for evaluating the effects of tartar control dentifrices were limited to the assessment of area coverage of calculus on the tooth surface. The development of the Quanticalc dental scaler permits the quantitative clinical measurement of force and stroke number associ

A pilot study on the novel use of an ultrasonic toothbrush in dogs: limited effect on dental calculus following a single treatment.

BACKGROUND: Dental calculus is common in dogs and contributes to periodontal disease by promoting plaque accumulation. While daily tooth brushing is the gold standard for prevention, the gold standard for calculus removal is professional dental cleaning under anesthesia at a veterinary clinic. Ultrasonic toothbrushes are marketed as alternatives for single-session calculus removal while the dog is awake; however, there is currently no scientific evidence supporting their effectiveness for this purpose. The aim of the study was to evaluate the effectiveness of a single ultrasonic toothbrush ses

Relief food subsistence revealed by microparticle and proteomic analyses of dental calculus from victims of the Great Irish Famine.

Food and diet were class markers in 19th-century Ireland, which became evident as nearly 1 million people, primarily the poor and destitute, died as a consequence of the notorious Great Famine of 1845 to 1852. Famine took hold after a blight (Phytophthora infestans) destroyed virtually the only means of subsistence-the potato crop-for a significant proportion of the population. This study seeks to elucidate the variability of diet in mid-19th-century Ireland through microparticle and proteomic analysis of human dental calculus samples (n = 42) from victims of the famine. The samples derive fro
