Extruded Nucleoli of Human Dental Pulp Cells.

Background and Objectives: The dental pulp stem cells are highly proliferative and can differentiate into various cell types, including endothelial cells. We aimed to evaluate the ultrastructural characteristics of the human dental pulp cells of the permanent frontal teeth. Materials and Methods: Human adult bioptic dental pulp was collected from n = 10 healthy frontal teeth of five adult patients, prior to prosthetic treatments for aesthetic purposes. Tissues were examined under transmission electron microscopy. Results: We identified cells with a peculiar trait: giant nucleoli resembling int

Dental Pulp Cell Behavior in Biomimetic Environments.

There is emerging recognition of the importance of a physiologically relevant in vitro cell culture environment to promote maintenance of stem cells for tissue engineering and regenerative medicine purposes. In vivo, appropriate cellular cues are provided by local tissue extracellular matrix (ECM), and these are not currently recapitulated well in vitro using traditional cultureware. We therefore hypothesized that better replication of the in vivo environment for cell culture and differentiation could be achieved by culturing dental pulp cells with their associated ECM. Primary dental pulp cel

Bacterial invasion of non-exposed dental pulp.

Anaerobic procedures were adopted to demonstrate the early bacterial invasion of non-exposed dental pulps, and to isolate and identify the bacteria. Of 19 freshly extracted teeth which originally exhibited deep dentinal lesions, clinical examination and electric pulp testing showed that nine of them had no pulpal exposure. Thus the pulps of these teeth were covered by clinically sound dentine beneath the carious lesion. Bacteria were found to have invaded the pulps of six of these nine teeth. The predominant bacteria were obligate anaerobes belonging to the genera Eubacterium, Propionibacteriu

Lymphatic vessels in inflamed human dental pulp.

Investigation has been performed on both the light and electron microscopic characteristics of the lymphatic vessels present in the dental pulp of human teeth which have been affected by serious carious lesions. These conditions provoke a severe inflammatory response resulting in structural and functional modifications of the tissue; increase of the tissue pressure is followed by the need for a more intensive lymphatic drainage. In the inflamed pulps, dilated lymphatic vessels with distended walls and "open junctions" between endothelial cells are detectable. On the other hand they lack certai

Dental pulp lymphatic vessel dynamics during tooth development and pulp stimulation in rodents.

AIM: The anatomy and functions of lymphatic vessels (LV) in mammals remain poorly understood compared to the blood vascular system. In particular, whether or not LV exist in the dental pulp is still controversial. This study aims to identify the existence of LV in the mouse dental pulp using a Prox1 (Prospero homeobox 1)-eGFP transgenic mouse model combined with a tissue-clearing technique. METHODOLOGY: Mandible or mandibular first molars of Prox1-eGFP mice were extracted, cleared for whole-mount observation and imaged using confocal microscopy. Dylight 594-lectin was injected intracardially

Defence reaction in dental pulp after pulp capping and partial pulpectomy in dogs.

The dental pulp was capped indirectly or directly, or partial vital pulpectomy was performed on the 12 functionally most important teeth of 24 beagle dogs. For pulp capping, calcium hydroxide was used, followed by zinc phosphate as a lining, and the preparation was restored with amalgam or composite material. Histological sections were prepared and examined for degree and type of pulp inflammation (hyperaemia, pulpitis, necrosis or gangrene). Degenerative changes of dental pulp (vacuolation, calcification, amyloid or hyaline changes) were also determined. For indirect pulp capping the width of

FOXE1 promotes the progression of pulp inflammation by activating PANoptosis in dental pulp cells.

PANoptosis is a type of programmed cell death (PCD) that is characterized by the simultaneous activation of pyroptosis, necroptosis, and apoptosis. This process triggers amplified inflammatory responses, which may compromise the success of vital pulp therapy (VPT). In this study, a PANoptotic dental pulp cell (DPC) cluster was identified in single-cell RNA sequencing data from inflamed dental pulp tissue. The occurrence of PANoptosis was further confirmed in human clinical pulpitis samples, primary cultured DPCs, and a mouse experimental pulpitis model. We subsequently identified forkhead box

Understanding dental pulp inflammation: from signaling to structure.

The pulp is a unique tissue within each tooth that is susceptible to painful inflammation, known as pulpitis, triggered by microbial invasion from carious lesions or trauma that affect many individuals. The host response involves complex immunological processes for pathogen defense and dentin apposition at the site of infection. The interplay of signaling between the immune and non-immune cells via cytokines, chemokines, neuropeptides, proteases, and reactive nitrogen and oxygen species leads to tissue reactions and structural changes in the pulp that escalate beyond a certain threshold to irr

Using Dental Pulp Stem Cells for Stroke Therapy.

Stroke is a leading cause of permanent disability world-wide, but aside from rehabilitation, there is currently no clinically-proven pharmaceutical or biological agent to improve neurological disability. Cell-based therapies using stem cells, such as dental pulp stem cells, are a promising alternative for treatment of neurological diseases, including stroke. The ischaemic environment in stroke affects multiple cell populations, thus stem cells, which act through cellular and molecular mechanisms, are promising candidates. The most common stem cell population studied in the neurological setting

Hypoxia promotes mineralization of human dental pulp cells.

INTRODUCTION: Dental pulp can be exposed to hypoxic conditions in case of trauma or inflammation. Dental pulp cells (DPCs) have mineralization potential, which plays a key role in pulp repair and reparative dentinogenesis process. Little information is available about DPC mineralization in hypoxic condition. The purpose of this study was to assess the influence of hypoxia on DPC mineralization to pave the way for a better understanding of dental pulp regeneration and reparative dentin formation. METHODS: Human DPCs were obtained by using tissue explant technique in vitro and cultured in normo

Vascular reactions in the dental pulp during inflammation.

This article deals with blood flow and tissue pressure changes in the inflamed dental pulp. While detailed patho-physiological studies of the circulation in the inflamed pulp are still partly lacking, available data from studies of the normal pulp tissue are also discussed. Knowledge of the normal physiological adjustments may indicate which circulatory changes would take place during inflammation. One of the focal points in the present article is how a localized increased tissue pressure may persist in the inflamed area without a circumferential spread to the rest of the pulp. The article is

Advanced Scaffolds for Dental Pulp and Periodontal Regeneration.

No current therapy promotes root canal disinfection and regeneration of the pulp-dentin complex in cases of pulp necrosis. Antibiotic pastes used to eradicate canal infection negatively affect stem cell survival. Three-dimensional easy-to-fit antibiotic-eluting nanofibers, combined with injectable scaffolds, enriched or not with stem cells and/or growth factors, may increase the likelihood of achieving predictable dental pulp regeneration. Periodontitis is an aggressive disease that impairs the integrity of tooth-supporting structures and may lead to tooth loss. The latest advances in membrane
