Biological Matrices Promote Odontoblastic Markers Expression in Stem Cells from Apical Papilla: An In Vitro Study.

INTRODUCTION: Odontoblastic differentiation is a cellular process that requires signaling molecules and extracellular matrix components provided by the dentin-pulp microenvironment. Several scaffolds have been proposed to induce odontoblast differentiation for pulp regeneration. This study evaluated the ability of collagen sponge and decellularized dental pulp, along with dentin, to promote the expression of odontoblastic markers in stem cells from the apical papilla (SCAPs). METHODS: Experimental groups consisted of 2 different biological matrices: (1) human decellularized dental pulp seeded

Fluoride or/and aluminum induced toxicity in guinea pig teeth with the low expression of dentine phosphoprotein.

This study investigated the damage and expression of dentine phosphoprotein (DPP) in guinea pig teeth by the administration of fluoride (F) or/and aluminum (Al). Fifty-two guinea pigs were divided randomly into four groups (control, F, Al, and F+Al). F (150 mg NaF/L) or/and Al (300 mg AlCl3 /L) were added in their drinking water for 90 days. The levels of F ion, dentine sialophosphoprotein (DSPP) gene, and DPP protein in incisor and molar were determined, respectively. The results showed that the concentrations of F ion in F and F+Al groups were increased significantly. F induced the mottled e

[Mineralized dental enamel matrix proteins].

The organic matrix of the developing enamel is highly heterogeneous, comprising proteins derived from a number of different genes, including amelogenin, enamelin, ameloblastin, tuftelin, dentine sialophosphoprotein, enzymes and serum proteins. Each of these classes appears to undergo post-secretory sequential degradation which contributes towards further matrix heterogeneity. Possible functions of these proteins are discussed. Proteins of the organic matrix are presumed to play an important role in the modulation of mineral deposition and growth during tooth morphogenesis but the precise funct

DSPP Is Essential for Normal Development of the Dental-Craniofacial Complex.

The craniofacial skeleton is derived from both neural crest cells and mesodermal cells; however, the majority of the bone, cartilage, and connective tissue is derived from the neural crest. Dentin sialophosphoprotein (DSPP) is a precursor protein that is expressed by the connective tissues of the craniofacial skeleton, namely, bone and dentin with high expression levels in the dentin matrix. Gene ablation studies have shown severe dental defects in DSPP-null mutant mice. Therefore, to elucidate the role of DSPP on the developing dental-craniofacial complex, we evaluated phenotypic changes in t

Effects of Red LED Irradiation in Enhancing the Mineralization of Human Dental Pulp Cells In Vitro.

Dentin regeneration is the preferred method used to preserve dental pulp vitality after pulp exposure due to caries. Red light-emitting diode irradiation (LEDI), which is based on photobiomodulation (PBM), has been used to promote hard-tissue regeneration. However, the underlying mechanism still needs elucidation. This study aimed to explore the mechanism involved in red LEDI affecting dentin regeneration. Alizarin red S (ARS) staining revealed that red LEDI induced mineralization of human dental pulp cells (HDPCs) in vitro. We further distinguished the cell proliferation (0-6 d), differentiat

Association of bovine dentine phosphophoryn with collagen fragments.

Bovine dentine phosphophoryn (BDP), a protein rich in aspartyl (Asp) and O-phosphoseryl (Ser(P)) residues, is synthesized by odontoblasts and believed to be involved in matrix-mediated biomineralization of dentine. Phosphophoryn was purified from bovine dentine using EDTA extraction, Ca(2+) precipitation, anion exchange and size exclusion chromatography. The purified protein migrated on SDS-PAGGE as a single band. The protein was dephosphorylated using a chelex alkaline dialysis procedure, repurified using anion exchange and size exclusion chromatography and then subjected to cleavage with try

The Role of Transient Receptor Potential Cation Channel, Subfamily C, Member 1 in the Odontoblast-like Differentiation of Human Dental Pulp Cells.

INTRODUCTION: Calcium ions (Ca2+) actively participate in reparative dentin formation by promoting cellular proliferation and differentiation of human dental pulp cells (hDPCs). Transient receptor potential cation channel, subfamily C, member 1 (TRPC1) activates Ca2+ entry upon store depletion in a variety of cell types. However, the function of TRPC1 in hDPCs has not been reported. Therefore, we aimed to analyze the role of TRPC1 in hDPCs undergoing odontoblast-like differentiation. METHODS: Immunohistochemical staining was used to determine the distribution of TRPC1 in pulp tissues. Western

Exploring vitamin K2 and B12 as bioactive micronutrients for dentin conditioning in regenerative endodontics procedures.

This study assessed the potential of vitamins K2 and B12 as dentin surface conditioners to modulate the regenerative behavior of human periodontal ligament stem cells (hPDLSCs). hPDLSCs were isolated and characterized by flow cytometry, which confirmed the mesenchymal stem cell phenotype. Cell viability assays showed no cytotoxicity at the tested concentrations; therefore, 16 µg/mL was selected for further analysis. Both vitamins significantly enhanced cell migration, attachment to dentin disks, and mineralization potential, as demonstrated by Alizarin Red staining and alkaline phosphatase act

Optogenetic control of odontoblastic differentiation in dental pulp stem cells (DPSC).

BACKGROUND/PURPOSE: Dental Pulp Stem Cells (DPSC) are sensitive to ionic and electrical cues governing mineralization. We developed a photo-responsive in vitro model using a synthetic optogenetic construct to test whether light-gated ion flux can modulate odontoblastic differentiation without biochemical induction. MATERIALS AND METHODS: A commercially available, non-clinical-grade DPSC line was transfected with a modular light-gated cation channel (Opto-Ca2+Switch) driven by a mineralization-associated synthetic promoter. Cells were embedded in gelatin methacrylate (GelMA)-carbon-dot composi

Performance of electrospun poly(ε-caprolactone) fiber meshes used with mineral trioxide aggregates in a pulp capping procedure.

Living dental pulp tissue exposed to the oral environment should be protected with an appropriate pulp capping material to support the dentinogenesis potential of the pulp cells. Mineral trioxide aggregate (MTA) is the material of choice for the treatment of pulp. However, due to cytotoxicity during the initial setting phase of MTA, a new material is required that can act as a barrier to direct contact but facilitate the favorable effect of MTA. This study examined the feasibility of using electrospun poly(ε-caprolactone) fiber (PCL-F) meshes in the MTA-based pulp capping procedures. An experi

Osteopontin Is Essential for Type I Collagen Secretion in Reparative Dentin.

Osteopontin (OPN) is a highly phosphorylated glycoprotein that is a prominent component of the mineralized extracellular matrix of bone. The secretion of OPN by immunocompetent cells plays a role in the differentiation of odontoblast-like cells during pulpal healing following tooth transplantation. This study aimed to clarify the role of OPN during reparative dentinogenesis. A groove-shaped cavity was prepared on the mesial surface of the upper first molars of wild-type (WT) and Opn knockout (KO) mice, and the samples were collected at intervals of 1 to 14 d. The demineralized sections were pr

Icariin negatively regulated lipopolysaccharide-induced inflammation and ameliorated the odontogenic activity of human dental pulp cells in vitro.

Alleviating inflammation and promoting dentine regeneration is critical for the healing of pulpitis. In this study, we investigated the anti-inflammatory, angiogenesis and odontogenesis function of icariin on Human dental pulp cells (HDPCs) under inflammatory state. Furthermore, the underlying mechanisms was also evaluated. Icariin attenuated the LPS-induced pro-inflammatory marker expression, such as interleukin-1β (IL-1β), IL-6 and IL-8. The immunoblotting and immunofluorescence staining results showed that icariin suppressed the inflammatory responses mediated by the protein kinase B (Akt)

Delivery of affordable and scalable encapsulated allogenic/autologous mesenchymal stem cells in coagulated platelet poor plasma for dental pulp regeneration.

The main goal of regenerative endodontics procedures (REPs) is to revitalize teeth by the regeneration of healthy dental pulp. In this study, we evaluated the potential of combining a natural and accessible biomaterial based on Platelet Poor Plasma (PPP) as a support for dental pulp stem cells (DPSC) and umbilical cord mesenchymal stem cells (UC-MSC). A comparison study between the two cell sources revealed compatibility with the PPP based scaffold with differences noted in the proliferation and angiogenic properties in vitro. Additionally, the release of growth factors including VEGF, HGF and

Hypoxia promotes mineralization of human dental pulp cells.

INTRODUCTION: Dental pulp can be exposed to hypoxic conditions in case of trauma or inflammation. Dental pulp cells (DPCs) have mineralization potential, which plays a key role in pulp repair and reparative dentinogenesis process. Little information is available about DPC mineralization in hypoxic condition. The purpose of this study was to assess the influence of hypoxia on DPC mineralization to pave the way for a better understanding of dental pulp regeneration and reparative dentin formation. METHODS: Human DPCs were obtained by using tissue explant technique in vitro and cultured in normo

Cellular Dynamics and Dentine Bridge Formation Following Direct Pulp Capping With MTA and Biodentine in a Murine Model.

AIM: Formation of a mineralized dentine bridge is the hallmark of successful direct pulp capping (DPC). Mineral trioxide aggregate (MTA) and Biodentine are widely used capping materials, but in vivo evidence comparing their regenerative potential remains inconclusive. This study used a controlled murine model to compare dentine bridge formation and pulp response to MTA and Biodentine. METHODS: DPC was performed on maxillary first molars of mice using either MTA or Biodentine. Reparative dentine bridge formation was assessed by micro-computed tomography (bridge volume, mineral content, density

DSPP dosage affects tooth development and dentin mineralization.

Dentin Sialoprotein (DSP) and phosphophoryn (PP) are two most dominant non-collagenous proteins in dentin, which are the cleavage products of the DSPP (dentin sialophosphoprotein) precursor protein. The absence of the DSPP gene in DSPP knock-out (KO) mice results in characteristics that are consistent with dentinogenesis imperfecta type III in humans. Symptoms include thin dentin, bigger pulp chamber with frequent pulp exposure as well as abnormal epithelial-mesenchymal interactions, and the appearance of chondrocyte-like cells in dental pulp. To better understand how DSPP influences tooth dev

Hypophosphatasia-associated deficiencies in mineralization and gene expression in cultured dental pulp cells obtained from human teeth.

INTRODUCTION: Mutations in the gene ALPL in hypophosphatasia (HPP) reduce the function of tissue nonspecific alkaline phosphatase, and the resulting increase in pyrophosphate (PP(i)) contributes to bone and tooth mineralization defects by inhibiting physiologic calcium-phosphate (P(i)) precipitation. Although periodontal phenotypes are well documented, pulp/dentin abnormalities have been suggested in the clinical literature although reports are variable and underlying mechanisms remains unclear. In vitro analyses were used to identify mechanisms involved in HPP-associated pulp/dentin phenotype

Ca-binding domains in the odontoblast layer of rat molars and incisors under normal and pathological conditions.

We recently reported the presence of high concentrations of a Ca-binding matrix in the circumpulpal dentin of rat incisors which had been prevented from mineralization by a systemic administration of 1-hydroxyethylidene-1,1-bisphosphonate (HEBP), a type of bisphosphonates, thus suggesting the role of the putative Ca-binding matrix in the appositional mineralization of circumpulpal dentin (TAKANO et al., 1998, 2000; OHMA et al., 2000). In this study, we examined the distribution of Ca-binding domains in the pulp tissue of normal rat teeth and its changes under the influence of HEBP, in order to

Histone acetylation promotes FKBP5 to increase reparative dentin formation in pulp repair.

IMPORTANCE: A well-formed dentin bridge isolates irritants and prevents bacterial penetration into the remaining pulp tissue, which is essential for pulp regeneration. The existing literature on promoting reparative dentin formation mostly focuses on the effects of pulp-capping materials but lacks investigations into the underlying mechanisms. OBJECTIVE: To identify potential targets for enhancing reparative dentin formation as well as the epigenetic mechanisms underlying these effects. DESIGN: Laboratory-based study combining multi-omics sequencing, epigenetic assays, in vitro FK506-binding
