The p38/CYLD Pathway is Involved in Necroptosis Induced by Oxygen-glucose Deprivation Combined with ZVAD in Primary Cortical Neurons.

Recently, necroptosis, a form of programmed necrosis, has been widely studied. It has previously been shown that knockout of lysine 63 deubiquitinase CYLD significantly inhibits necroptosis in other cell lines, and serum response factor (SRF) could regulate CYLD gene expression through p38 mitogen-activated protein kinase (p38 MAPK). In the following study, we show oxygen-glucose deprivation (OGD) combined with a caspase inhibitor, ZVAD (OGD/ZVAD), induced CYLD protein expression in a time-dependent manner. Immunofluorescence studies showed that CYLD was localized strongly to the nucleus and w

Genistein alleviates Osteoporosis by inhibiting the differentiation and autophagy of osteoclasts via regulating the CYLD/p62/RANKL axis.

Osteoporosis is a highly prevalent metabolic bone disorder characterized by an imbalance in bone remodeling, primarily due to excessive osteoclast-mediated bone resorption. The present study systematically investigates the protective effects of genistein, a naturally occurring soy isoflavone, against osteoporosis by targeting osteoclast differentiation and autophagy, as well we the underlying mechanism. Using RANKL-stimulated RAW264.7 cells as an in vitro model, we found that genistein treatment significantly and dose-dependently inhibited osteoclast formation and autophagic activity, which wa

Osteoclastic miR-301-b knockout reduces ovariectomy (OVX)-induced bone loss by regulating CYDR/NF-κB signaling pathway.

Postmenopausal osteoporosis (PMOP) is a frequent bone disorder responsible for an increased risk of disability to millions of individuals in the world. For identifying novel and effective targets to treat this disease, it is essential to explore the underlying molecular mechanisms. MicroRNAs (miRNAs) have been widely investigated due to their involvement in the pathophysiology of bone loss. In this study, we attempted to elucidate the role of miR-301-b in murine osteoclastogenesis. We found that miR-301-b expression was increased in the bone tissues from PMOP patients, along with up-regulated

FGFR4 suppresses antitumor immunity and cuproptosis by disrupting CYLD-mediated stabilization of STING and FDX1.

The fibroblast growth factor receptor (FGFR) pathway is frequently dysregulated in cancer and contributes to tumor progression and immune evasion; however, the underlying immunosuppressive mechanisms remain incompletely understood. Here, we identify FGFR4 as a key FGFR family member that suppresses antitumor immunity. Mechanistically, FGFR4 promotes GRB2-dependent phosphorylation and inactivation of the deubiquitinase CYLD, leading to ubiquitin-proteasome-mediated degradation of STING and FDX1. Loss of STING attenuates type I interferon signaling and impairs cytotoxic immune responses, while d

Beneficial effects of chlorogenic acid treatment on neuroinflammation after deep hypothermic circulatory arrest may be mediated through CYLD/NF-κB signaling.

Deep hypothermic circulatory arrest (DHCA) during heart surgery may induce neuroinflammation leading to neurocognitive dysfunction. Chlorogenic acid (CA) is a common phytochemical, which can attenuate neuroinflammation. Nevertheless, the underlying mechanism involved in the anti-inflammatory effect of CA after DHCA is unknown. The present study therefore characterized the anti-inflammatory functions of CA after DHCA using in vivo and in vitro DHCA models. The activation of microglia, inflammatory cytokine levels, and the NF-κB pathway were measured. The results showed that CA treatment amelior

OTUD4 stabilizes CYLD and counteracts BCL3 K63-linked polyubiquitination to suppress melanoma progression.

Melanoma is a highly aggressive malignancy in which dysregulated ubiquitin-dependent signaling contributes to tumor progression. OTUD4 is frequently downregulated in melanoma, but the functional significance of this alteration remains unclear. Here, we identify OTUD4 as a dual-mode deubiquitinase that suppresses melanoma progression by coordinating K48- and K63-linked polyubiquitination. OTUD4 directly binds and stabilizes CYLD by counteracting K48-linked polyubiquitination, whereas it independently binds BCL3 and counteracts BCL3 K63-linked polyubiquitination. OTUD4 depletion promotes BCL3 ub

SPATA2 Attenuates Inflammatory Injury Through CYLD-Related Regulation of Microglial Inflammatory Responses After Focal Cerebral Ischemia/Reperfusion in Rats.

BACKGROUND: Post-ischemic inflammation is strongly influenced by interactions between neurons and microglia. We previously observed that spermatogenesis-associated protein 2 (SPATA2) alleviates inflammatory injury following cerebral ischemia/reperfusion; however, the relationship between SPATA2, cylindromatosis (CYLD)-associated inflammatory signaling, and microglial inflammatory responses remains incompletely understood. METHODS: Adult Sprague-Dawley (SD) rats were subjected to transient middle cerebral artery occlusion followed by reperfusion. SPATA2 expression was knocked down in vivo via

Baicalein attenuates atherogenesis by suppressing endothelial pyroptosis via cylindromatosis-mediated tumor necrosis factor receptor-associated factor 6 deubiquitination.

BACKGROUND: Endothelial pyroptosis is a critical contributor to the development of atherosclerosis. Baicalein (BAI) exhibits anti-atherosclerotic potential; however, whether it alleviates atherosclerosis by modulating endothelial pyroptosis and the underlying molecular mechanisms remain unclear. PURPOSE: This study aimed to elucidate the mechanism by which baicalein attenuates atherosclerosis, with a particular focus on Cylindromatosis (CYLD)-mediated deubiquitination of tumor necrosis factor receptor-associated factor 6 (TRAF6). METHODS: Atherosclerosis was induced in ApoE⁻/⁻ mice, and ox-L

lncRNA XIST suppresses acute myeloid leukemia progression by promoting necroptosis through the miR-92a-3p/CYLD axis.

Acute myeloid leukemia (AML) remains a highly lethal hematologic malignancy, with relapse and therapeutic resistance largely driven by apoptosis evasion. Necroptosis, a regulated form of non-apoptotic cell death, has emerged as a potential strategy to eliminate apoptosis-resistant cancer cells; however, its upstream regulatory mechanisms in AML remain incompletely understood. Here, we identify the long non-coding RNA XIST as a previously underappreciated regulator of necroptosis-associated signaling in AML. XIST and CYLD were consistently downregulated in AML cell lines and primary patient sam

Metastatic malignant cylindroma arising on a background of digenic inheritance of BRCA2 and CYLD pathogenic variants targeted with PARP inhibition.

BACKGROUND: CYLD cutaneous syndrome (CCS) is caused by germline heterozygous pathogenic variants in CYLD and results in the progressive formation of cylindromas, spiradenomas or trichoepitheliomas. Malignant cylindroma is a rare skin adnexal tumour occurring in CCS, which can metastasize with lethal outcomes and has limited genomic characterization. BRCA2 loss in CCS is not described and may modulate the cutaneous cancer risk of CCS. OBJECTIVES: To establish whether BRCA deficiency drives metastatic malignant cylindroma and to report the phenotype of three siblings with digenic inheritance of

Immune dysregulation in SHARPIN-deficient mice is dependent on CYLD-mediated cell death.

SHARPIN, together with RNF31/HOIP and RBCK1/HOIL1, form the linear ubiquitin chain assembly complex (LUBAC) E3 ligase that catalyzes M1-linked polyubiquitination. Mutations in RNF31/HOIP and RBCK/HOIL1 in humans and Sharpin in mice lead to autoinflammation and immunodeficiency, but the mechanism underlying the immune dysregulation remains unclear. We now show that the phenotype of the Sharpincpdm/cpdm mice is dependent on CYLD, a deubiquitinase previously shown to mediate removal of K63-linked polyubiquitin chains. Dermatitis, disrupted splenic architecture, and loss of Peyer's patches in the

Inherited pulmonary cylindromas: extending the phenotype of CYLD mutation carriers.

BACKGROUND: Germline mutations in the tumour suppressor gene CYLD are recognized to be associated with the development of multiple cutaneous cylindromas. We encountered such a patient who presented with breathlessness because of multiple pulmonary cylindromas. OBJECTIVES: To search for clinical and radiological features of multiple pulmonary cylindromas in a cohort of 16 patients with CYLD mutations. METHODS: A retrospective case-note review was carried out in a tertiary dermatogenetics clinic where CYLD mutation carriers are reviewed on an annual basis. In-depth investigation was carried ou

The deubiquitinase CYLD inhibits thrombin-induced p38 MAPK-p65 NF-κB signaling and inflammatory cytokine production to suppress triple-negative breast cancer progression.

Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by chronic inflammation with limited therapeutic options. Although thrombin activation of protease-activated receptor-1 (PAR1) promotes ubiquitin-dependent signaling and inflammatory responses in endothelial cells, its contribution to inflammatory effects in TNBC remain poorly understood. Here, we show that loss of the deubiquitinase cylindromatosis (CYLD) markedly increases basal phosphorylation of p38 mitogen-activated protein kinase and p65 nuclear factor kappa B subunit to levels comparable to thrombin stimulation,

Associations of CYLD, JAK2 and TLR4 Genotypes with PSA Levels and Immunophenotype in Benign Prostatic Hyperplasia and Prostate Cancer.

BACKGROUND: Prostate cancer (PCa) is one of the most common malignant tumors of the male urinary system, and its incidence and mortality rates have been increasing worldwide. Benign prostatic hyperplasia (BPH) represents stromal and epithelial cell proliferation in the prostate in elderly males. Abnormal activation of inflammation-related signalling molecules, such as toll-like receptor 4 (TLR4) and Janus kinase/signal transducers and activators of transcription (JAK/STAT) has been linked to the initiation and progression of various human diseases including PCa and BPH. Cylindromatosis (CYLD)

Targeting noncanonical nuclear factor kappa B signalling in CYLD cutaneous syndrome by selective inhibition of IκB kinase alpha.

BACKGROUND: CYLD cutaneous syndrome (CCS) skin tumours develop from puberty onwards, can number in the hundreds and progressively grow over time. Patients with CCS lack medical therapies and require repeated surgery to control tumour burden. CYLD loss of heterozygosity drives tumour growth, and CCS tumours have previously been shown to demonstrate increased canonical nuclear factor kappa B (NF-κB) and Wnt signalling. OBJECTIVES: To investigate NF-κB signalling in CCS tumours and CCS tumour keratinocytes, with the aim of identifying druggable targets. METHODS: We used complementary bulk trans

Up-regulation of CYLD enhances Listeria monocytogenes induced apoptosis in THP-1 cells.

Listeria monocytogenes (Lm), a facultative anaerobic gram-positive bacterium, causes listeriosis. Immune cell apoptosis is considered to be one pathogenic factor for listeriosis. As a deubiquitinase, CYLD is an important regulator both in innate immune response and apoptosis by negatively modulating NF-κB pathway. However the role of CYLD in Lm induced apoptosis remains unclear. Here we found that CYLD is significantly up-regulated in macrophages upon its infection. There is a moderate decrease in Lm proliferation and apoptotic cells in siRNA-induced CYLD knockdown THP-1 cells. Thereby CYLD ma

Brooke-Spiegler syndrome tumor spectrum beyond the skin: a patient carrying germline R936X CYLD mutation and a somatic CYLD mutation in Brenner tumor.

Brooke-Spiegler syndrome is a hereditary disorder characterized by a predisposition to the development of skin appendage neoplasms and the major and minor salivary glands neoplasms. The role of the CYLD mutation in visceral neoplasms is still unclear, except for the parathyroid tumor. We report the case of a 46-year-old patient with multiple cylindromas and trichoepitheliomas, a Brenner tumor of the ovary and a negative family history for Brooke-Spiegler phenotype. Genetic analysis revealed R936X germline mutation in the proband, but not in the patient's relatives. The same somatic mutation wa

Cylindromatosis gene CYLD regulates hepatocyte growth factor expression in hepatic stellate cells through interaction with histone deacetylase 7.

UNLABELLED: Hepatic fibrosis is considered as a physiological wound-healing response to liver injury. The process involves several factors, such as hepatocyte growth factor (HGF), which restrains hepatic injury and facilitates reversibility of fibrotic reaction in response to an acute insult. Chronic liver injury and sustained inflammation cause progressive fibrosis and, ultimately, organ dysfunction. The mechanisms tipping the balance from restoration to progressive liver tissue scarring are not well understood. In the present study, we identify a mechanism in which the tumor-suppressor gene,

Influenza A virus infection activates caspase-8 to enhance innate antiviral immunity by cleaving CYLD and blocking TAK1 and RIG-I deubiquitination.

Caspase-8, an aspartate-specific cysteine protease that primarily functions as an initiator caspase to induce apoptosis, can downregulate innate immunity in part by cleaving RIPK1 and IRF3. However, patients with caspase-8 mutations or deficiency develop immunodeficiency and are prone to viral infections. The molecular mechanism underlying this controversy remains unknown. Whether caspase-8 enhances or suppresses antiviral responses against influenza A virus (IAV) infection remains to be determined. Here, we report that caspase-8 is readily activated in A549 and NL20 cells infected with the H5
