Spatiotemporal expression patterns and RNA interference efficiency of key diapause genes in Lymantria dispar-Investigating the heritability of RNA interference effects for pest biocontrol.

The spongy moth (Lymantria dispar Linnaeus), a major quarantine pest, relies on diapause as a key survival strategy. This study examined the temporal and spatial expression of four diapause-associated genes, LdGCLC, LdGLUD1_2, LdIDH1, and LdIDH2. Using fluorescence in situ hybridization and qPCR, their expression was analyzed across developmental stages and tissues. Additionally, RNA interference (RNAi) via microinjection and immersion was employed to systematically silence these genes. The results revealed that (1) all four genes were expressed in the brain; (2) dsRNA microinjection reduced t

CO2 narcosis induces a metabolic shift mediated via juvenile hormone in Bombus impatiens gynes.

Carbon dioxide (CO2) has pleiotropic effects on insect physiology and behavior. Although diverse, many impacts are related to changes in metabolism and reallocation of macronutrients. Here we examined the metabolic shift induced by CO2 and its regulation using Bombus impatiens. CO2 applied to bumble bee gynes induces bypass of diapause and transition into reproduction. We analyzed ovary activation and macronutrient amounts in four tissues/body parts (fat body, thorax, ovaries, and crop) at three timepoints following CO2 administration. To tease apart the effects of CO2 on reproduction and meta

Ecdysteroid signalling components in metamorphosis and development of the desert locust, Schistocerca gregaria.

The arthropod-specific hormone family of ecdysteroids plays an important role in regulating diverse physiological processes, such as moulting and metamorphosis, reproduction, diapause and innate immunity. Ecdysteroids mediate their response by binding to a heterodimeric complex of two nuclear receptors, the ecdysone receptor (EcR) and the retinoid-X-receptor/ultraspiracle (RXR/USP). In this study we investigated the role of EcR and RXR in metamorphosis and development of the desert locust, Schistocerca gregaria. The desert locust is a voracious, phytophagous, swarming pest that can ruin crops

Quantitative parameters and ecological implications of a specialized tritrophic interaction involving a seed-feeding tortricid, Pseudargyrotoza conwagana, a braconid parasitoid, Bracon otiosus, and the wild privet, Ligustrum vulgare.

Little is known about tritrophic interactions involving seed-feeding insects, parasitoid wasps, and wild fleshy fruits. Here, we examine relationships between Pseudargyrotoza conwagana (F.) (Lepidoptera: Tortricidae), Bracon otiosus Marshall (Hymenoptera: Braconidae), and the wild privet, Ligustrum vulgare L. (Lamiales: Oleaceae), after collecting fruits in a hedgerow habitat in northwest Spain and rearing insects indoors. No other insect species was detected in this trophic system. Each fruit contained one to four seeds, each infested fruit contained only one seed-feeding tortricid caterpilla

Understanding geographic and host range expansion of stored product insects to improve quarantine and pest management programs.

Many species of stored product insects have been spread by commerce, a few recently enough to have a written record. The bean weevil, Acanthoscelides obtectus (Say), Mediterranean flour moth, Ephestia kuehniella (Zeller), and Angoumois grain moth, Sitotroga cerealella (Olivier), have some of the most complete early records. The larger black flour beetle, Cynaeus angustus (LeConte) may have the most complete record of both geographic and host range expansion. The introduction of heated buildings and storage sites has allowed tropical species to establish in temperate climates. The adoption of c

Understanding the regulation of overwintering diapause molecular mechanisms in Culex pipiens pallens through comparative proteomics.

To reveal overwintering dormancy (diapause) mechanisms of Culex pipiens pallens (L.), global protein expression differences at three separate time points represent nondiapause, diapause preparation and overwintering diapause phases of Cx. pipiens pallens were compared using iTRAQ. Cx. pipiens pallens females accumulate more lipid droplets during diapause preparation and overwintering diapause maintenance than during the nondiapause phase. A total of 1030 proteins were identified, among which 1020 were quantified and compared. Gene Ontology, Kyoto Encyclopedia of Genes and Genomes (KEGG), Domai

Experimental host preference of diapause and non-diapause induced Culex pipiens pipiens (Diptera: Culicidae).

BACKGROUND: Culex pipiens pipiens plays an important role in the transmission of several vector-borne pathogens such as West Nile virus (WNV) in North America. Laboratory and field studies suggest that this species is ornithophilic but because of genetic hybridization with sibling species during the active mosquito season, it may occasionally feed on mammals. Adult female Cx. p. pipiens undergo a facultative diapause and may serve as an overwintering mechanism for WNV. To determine the effect of diapause on the innate host preference of Cx. p. pipiens emerging from winter hibernation, we condu

Parental thermal environment affects offspring resilience to winter and spring weather anomalies.

Recent increases in the frequency and intensity of thermal anomalies can result in recurrent stressful weather events. Among annual insects, this phenomenon may expose parents and offspring to a succession of different weather anomalies (e.g. summer heatwaves and late spring frosts). Parental stress can alter offspring physiology and performance, potentially affecting their tolerance of environmental variation. We investigated the transgenerational effects of thermal anomalies on a univoltine wetland butterfly. We induced parental thermal stress by exposing pupae to a constant temperature, ran

The adipokinetic hormone system in Culicinae (Diptera: Culicidae): molecular identification and characterization of two adipokinetic hormone (AKH) precursors from Aedes aegypti and Culex pipiens and two putative AKH receptor variants from A. aegypti.

Insect neuropeptides of the adipokinetic hormone (AKH) family induce the mobilization of energy stores to fuel flight, but also affect the nutritional balance during diapause and oogenesis. They are therefore important regulators for flight, hibernation, and reproduction in mosquitoes including those that transmit human pathogens. In this study, we identified and analyzed the genes encoding two AKH preprohormones in the Yellow fever mosquito, Aedes aegypti: Aedae-AKH-I encodes the octapeptide pELFTPSWa and Aedae-AKH-II the decapeptide pEVTFSRDWNAa. Identical AKHs were identified in the West Ni

Embryonic dormancy in Aedes aegypti and Aedes albopictus (Diptera: Culicidae): A survival and dispersal mechanism.

Aedes aegypti and Aedes albopictus are the main vectors of arboviruses such as dengue, Zika virus, and chikungunya. Ae. aegypti is a widely spread mosquito in tropical and subtropical regions, whereas Ae. albopictus is a culicid of Asian origin that shows exophilic behavior and can be found in subtropical and temperate areas. Climatic factors could influence the distribution of both species, making them use genetic and environmental resources to adapt to the environment, activating survival mechanisms (embryonic dormancy) that increase the developmental period and keep their offspring in the e

Functional expression and characterization of CAPA receptor in the digestive tract and life stages of Drosophila suzukii, and differential activities with insect PRXamide peptides.

Spotted-wing drosophila, Drosophila suzukii (Matsumura), is an invasive vinegar fly that is a major threat to the small fruits industries globally. Insect capa genes encode multiple neuropeptides, including CAPA-periviscerokinin (CAPA-PVK) peptides, that are specifically known to cause diuresis or anti-diuresis in various organisms. Here we identified and characterized a corresponding G protein-coupled receptor (GPCR) of the D. suzukii CAPA-PVK peptides: CAPA receptor (CAPA-R). To better characterize the behavior of D. suzukii CAPA-R, we used insect cell-based functional expression assays to e

Effect of Climate Warming on Culex Mosquito Population Dynamics in Newfoundland.

Mosquitoes are important vectors of infectious diseases affecting humans and animals. In North America, Culex mosquitoes transmit West Nile virus and other arboviruses, while their persistence in temperate regions depends strongly on seasonal processes such as diapause. Temperature, rainfall, and photoperiod jointly regulate mosquito development, survival, reproduction, and seasonal activity, yet many climate-driven models do not explicitly represent diapause or interacting environmental drivers. We develop a mechanistic, stage-structured model for temperate Culex populations that includes egg

Toxicity of the venom from Nasonia vitripennis (Hymenoptera: Pteromalidae) toward fly hosts, nontarget insects, different developmental stages, and cultured insect cells.

A venom preparation from Nasonia vitripennis, a wasp ectoparasitoid of fly pupae, was assayed for lethality in different stages of insects representing ten different orders and in cultured insect cells. In most cases, the motor activity of the injected insects remained completely normal for 1-2 days after the injection and displayed none of the symptoms of paralysis commonly reported for venoms of the Hymenoptera. A natural host, the flesh fly Sarcophaga bullata, was highly sensitive in the pupal stage (LD50 = 5.4 and 5.5 VRE/g for nondiapausing and diapausing pupae, respectively), the stage t

An analytical study of the neurosecretory cells of the brain of the mature larvae and diapause pupae of Amsacta collaris Hampson (Lepidoptera: Arctiidae) under different temperature regimes.

The neurosecretory cells (NSCs) of the brain of the mature larvae and 3 months old diapause pupae of Amsacta collaris Hampson (Lepidopera: arctiidae) have been studied under different temperature regimes (0, 7, 18, 37, and 42 degrees C), using paraldehyde fuchsin and performic acid resorcin fuchsin staining techniques. The 0 degree C is fatal to the larvae, and at 7 degrees C, within 24 h the quantity of NSM reduced considerably in all the subtypes of A-cells; activity started 3rd day onwards, and on 5th day, the A1-cells appeared loaded with NSM, the A2-cells moderately filled, but no change

Cadmium exposure induces acute and delayed physiological effects in bumblebee (Bombus terrestris) gynes.

Cadmium (Cd), a potent environmental contaminant, persists widely in the environment and threatens global ecosystems. However, the underlying mechanisms of its acute and delayed toxicity effects on pollinating insects still remains elusive. Here, we systematically investigated whether and how environmentally relevant concentrations of Cd exposure impose acute and delayed toxic effects on bumblebees. Seven day exposure of environmentally relevant Cd concentration to gynes exhibited acute toxic effects, impairing their learning and memory capacity. Nutrient reserves were impaired and their growt

QTL identification and characterization of the recombination landscape of the mountain pine beetle (Dendroctonus ponderosae).

Insect pests can rapidly accumulate in number and thrive in diverse environments, making them valuable models for studying phenotypic plasticity and the genetic basis of local adaptation. The mountain pine beetle (Dendroctonus ponderosae) is a major forest pest, and adult body size and generation time are 2 traits that vary among populations and directly influence reproductive success and outbreak dynamics. To identify regions of the genome linked to these 2 traits, we generated double-digest RAD sequencing data from an F2 intercross, using populations from 2 Y haplogroups with phenotypic and

Drug resistance against bacteria and fungi has become common in recent years, and it is urgent to discover novel antimicrobial peptides to manage this problem. Many antimicrobial peptides from insects have been reported to have antifungal activity and are candidate molecules in the treatment of human diseases. In the present study, we characterized an antifungal peptide named blapstin that was isolated from the Chinese medicinal beetle Blaps rhynchopetera used in folk medicine. The complete coding sequence was cloned from the cDNA library prepared from the midgut of B. rhynchopetera. It is a 4

Functional Characterization of Trypsin Genes in Spodoptera litura and Their RNA Silencing in Combination with Insecticides.

Trypsin is one of the major digestive proteinases secreted by the insect midgut. It plays important roles in digestion, molting, diapause, zymogen activation, innate immunity, reproduction and insecticide resistance. However, the functions of trypsin and its potential as a target for the control of Spodoptera litura, a polyphagous agricultural pest, remain poorly understood. In this study, seven trypsin genes related to S. litura development were characterized. Temporal-spatial expression results showed that the seven trypsin genes were predominantly expressed during the larval stage and mainl

[Ecology of flea groups of the species conformis (Siphonaptera: Pulicidae: Xenopsylla) of the fauna of Russia and adjacent countries (review)].

Within the boundaries of the former USSR, the northern part of the conformis group distribution is located. It spreads over the arid regions of the Trans-Cacucasus, Pricaspijckaja lowland, Kazakhstan and Middle Asia. In this area 10 species and subspecies occur. They are mainly parasites of gerbillins. Unlike many other Siphonaptera the conformis fleas, when in the host's home, do not concentrate in the nest but inhabit the passages of burrow and food chambers throughout the year. On this reason the preimaginal development and existence of the adults take place not at the temperature of the ha
