Evolutionarily distinct strategies for the acquisition of inorganic carbon from seawater in marine diatoms.

The acquisition of dissolved inorganic carbon (DIC) in CO2-limited seawater is a central issue to understand in marine primary production. We previously demonstrated the occurrence of direct HCO3- uptake by solute carrier (SLC) 4 transporters in a diatom, a major marine primary producer. Homologs of SLC are found in both centric and pennate marine diatoms, suggesting that SLC transporters are generally conserved. Here, the generality of SLC-mediated DIC uptake in diatoms was examined using an SLC inhibitor, diisothiocyano-2,2'-stilbenedisulfonic acid (DIDS), and an inhibitor of external carbon

Relaxation of the non-photochemical chlorophyll fluorescence quenching in diatoms: kinetics, components and mechanisms.

Diatoms are especially important microorganisms because they constitute the larger group of microalgae. To survive the constant variations of the light environment, diatoms have developed mechanisms aiming at the dissipation of excess energy, such as the xanthophyll cycle and the non-photochemical chlorophyll (Chl) fluorescence quenching. This contribution is dedicated to the relaxation of the latter process when the adverse conditions cease. An original nonlinear regression analysis of the relaxation of non-photochemical Chl fluorescence quenching, qN, in diatoms is presented. It was used to

Role of extracellular carbonic anhydrase in the polar diatom Fragilariopsis cylindrus.

Diatoms are major primary producers in productive polar oceans, where cold temperatures and high CO2 solubility raise questions as to whether they rely on the CO2-concentrating mechanism (CCM) to saturate photosynthesis. Knowledge of CCM function will help better predict the sensitivity of diatom primary production to warming and acidifying polar oceans. Here, we characterized the CCM in the polar diatom, Fragilariopsis cylindrus, using membrane inlet mass spectrometry (MIMS) and a numerical model. Quantification of the role of external carbonic anhydrase (eCA) was enabled through the addition

Interactions with bacteria shape diatom adaptation to carbon concentration changes.

Diatoms are key contributors to global primary production, and have developed intricate partnerships with bacteria through long-term co-evolution. Here, we uncover a syntrophic relationship between the model obligate photoautotroph diatom Phaeodactylum tricornutum and the rod-shaped bacterium Loktanella vestfoldensis, which enables the diatom to indirectly utilize glucose. To be specific, growth of the diatom depends on the support of L. vestfoldensis for the supply of necessary carbon source when glucose serves as the sole carbon source, while L. vestfoldensis shows dependence on P. tricornut

Overexpression of PtNRPS1 enhances diatom-mediated bioremediation of salicylate pollution.

The accumulation of the emerging pollutant salicylic acid (SA) in the environment has gained much attention. In this study, overexpression of the non-ribosomal peptide synthase (NRPS) gene, PtNRPS1 in the marine diatom Phaeodactylum tricornutum (PtNRPS1-OE) increased resistance to SA pollutants. It was assumed that the enhanced tolerance was due to the high binding affinity between recombinant PtNRPS1 (rNRPS1) and SA pollutants. Moreover, tandem mass spectrometry analysis determined the amino acids that participated in the covalently binding of SA. The removal efficiency of SA pollutants by Pt

Ab initio spectroscopy and thermochemistry of diatomic platinum hydride, PtH.

Rovibrational levels of low-lying electronic states of the diatomic molecule PtH are computed using non-relativistic wavefunction methods and a relativistic core pseudopotential. Dynamical electron correlation is treated at the coupled-cluster with single and double excitations and a perturbative estimate of triple excitations level, with basis-set extrapolation. Spin-orbit coupling is treated by configuration interaction in a basis of multireference configuration interaction states. The results compare favorably with available experimental data, especially for low-lying electronic states. For

Hydrophobic Biodegradable Hyperbranched Copolymers with Excellent Marine Diatom Resistance.

As the utilization of degradable polymer coatings increased, the accompanying trade-off between good degradability and high-efficiency antidiatom adhesion due to their hydrophobic nature remains unresolved. The study presents a new hydrophobic surface-fragmenting coating consisting of degradable hyperbranched polymers (hereafter denoted as h-LLAx) synthesized by reversible complexation-mediated copolymerization with isobornyl acrylate (IBOA) and divinyl-functional oligomeric poly(l-lactide) (OLLA-V2), both derived from biomass, that exhibited superior resistance (∼0 cell mm-2) to marine diatom

Diatom extraction: A new technique with heated H2O2. A technical note.

The best method of diatom identification in animal and human tissues is still an important discussion topic, in terms of effectiveness and reliability. In this technical note, authors propose a new method of extraction of diatoms using heated hydrogen peroxide from animal and human tissue samples. This method has been compared with the traditional method of digestion with acids. The results of the comparison show that heated hydrogen peroxide extraction is more efficient in terms of reduction of sediment, extraction of the material and preservation of diatoms proving to be a viable alternative

Diatom responses to sewage inputs and hydrological alteration in Mediterranean streams.

We analyzed the conjoint effects of sewage inputs and hydrological alteration on the occurrence of teratological forms and on the assemblage composition of stream benthic diatoms. The study was performed in 11 Mediterranean streams which received treated or untreated urban sewage (Impact sites, I), whose composition and morphological anomalies were compared to upstream unaffected (Control, C) sites. The impact sites had high concentrations of ammonium, phosphorus, and pharmaceutical compounds (antibiotics, analgesics, and anti-inflammatories), particularly in those receiving untreated sewage.

Differential acclimation of a marine diatom to inorganic mercury and methylmercury exposure.

Aquatic organisms originating from metal polluted water may exhibit differences in their sensitivity to metals, but the underlying physiological mechanisms resulting in such responses have not been well reported. In the present study, a marine diatom Thalassiosira weissflogii was chronically exposed to different inorganic mercury (Hg(II), 0.5 and 5 μg Hg/L) or methylmercury (MeHg, 0.02 and 0.4 μg Hg/L) concentrations for over 18 generations. We then quantified the changes in the Hg(II) or MeHg sensitivity, Hg accumulation, subcellular distribution, as well as thiol compound induction in the di

Diatom extraction with HCl from animal tissues: a technical note.

Identification of diatoms from human or animal tissue may be affected by factors concerning techniques of extraction and analysis which cause fragmentation or loss of diatoms. In this brief technical note a novel efficient method of extraction of diatoms from animal tissues is presented. The authors have tested an H(2)O(2) plus HCl and HCl method to detect diatoms in several tissues from pig immersed in water rich in diatoms; they have also compared this method to other methods of extraction including enzymatic ones. The experiment showed that all siliceous frustules of fresh water diatoms are

CO2 sensing at ocean surface mediated by cAMP in a marine diatom.

Marine diatoms are known to be responsible for about a quarter of global primary production and their photosynthesis is sustained by inorganic carbon-concentrating mechanisms and/or C(4) metabolism. Activities of the inorganic carbon-concentrating mechanism are attenuated under enriched [CO(2)]; however, impacts of this factor on primary productivity and the molecular mechanisms of CO(2) responses in marine diatoms are unknown. In this study, transgenic cells were generated of the marine diatom Phaeodactylum tricornutum by the introduction of a beta-glucuronidase reporter gene under the contro

The role of ultraviolet-adaptation of a marine diatom in photoenhanced toxicity of acridine.

Cultures of the marine diatom Phaeodactylum tricornutum were grown under laboratory light with a different fraction of ultraviolet radiation (UV) to study the potential role of photoadaptation in determining the sensitivity to photoenhanced toxicity of acridine. In short-term experiments, a higher acridine concentration was needed to inhibit the photosynthetic electron flux, monitored with chlorophyll a fluorescence, in algae exposed to fluorescent light (low UV) than to mercury light (high UV), consistent with the expected role of UV. The two types of light in long-term exposures led to chang

Diatom-lipid-copepod nexus under threat by global change.

The central role of lipids in the ecology of high-latitude and temperate seas is under threat by global change. This hinges on the tight ecological links between diatoms and copepods of the genus Calanus and similar (e.g. Neocalanus, Calanoides) that produce and accumulate energy-rich lipids. Strongly coupled through seasonal water-column stability, nutrient supply, spring-bloom succession, and overwintering life strategies, this nexus is foundational to the highly productive food webs of these regions. We experimentally tested how a change in phytoplankton type from diatoms to flagellates, al

Biogenic Phosphonate Utilization by Globally Distributed Diatom Thalassiosira pseudonana.

Phosphonates are a class of organic phosphorus (P) compounds that contribute ~25% of dissolved organic P. Recent studies reveal the important role of phosphonates mediated by prokaryotes in the marine P redox cycle. However, its bioavailability by eukaryotic phytoplankton is under debate. 2-Aminoethylphosphonic acid (2-AEP) and 2-amino-3-phosphonopropionic acid (2-AP3) are two biogenic phosphonates in the marine environment. Here, Thalassiosira pseudonana, a common diatom species in the ocean, is able to recover growth from P starvation when provided with 2-AEP and 2-AP3. Moreover, 2-AEP cultu

The utility of drowning site inference through metagenomic diatom analysis.

The diatom test is one of the methods used to diagnose drowning in forensic autopsies. Metagenomic diatom analysis may reveal where a drowning occurred. We evaluated whether metagenomic diatom analysis could be used to infer waters, watersheds, and geographic locations using 166 water samples from 64 locations (freshwater: 55; seawater: 9). Principal component analysis (PCA) in all samples revealed no specific clusters for waters or watersheds. In one river, the three samples at the same site generally tended to be in close clusters, but there were some cases where the three sites were far fro

First Evidence of the Toxin Domoic Acid in Antarctic Diatom Species.

The Southern Ocean is one of the most productive ecosystems in the world. It is an area heavily dependent on marine primary production and serving as a feeding ground for numerous seabirds and marine mammals. Therefore, the phytoplankton composition and presence of toxic species are of crucial importance. Fifteen monoclonal strains of Pseudo-nitzschia subcurvata, a diatom species endemic to the Southern Ocean, were established, which were characterized by morphological and molecular data and then analysed for toxin content. The neurotoxins domoic acid and iso-domoic acid C were present in thre

The effect of elevated pCO2 on cadmium resistance of a globally important diatom.

Cadmium (Cd) pollution is a widespread threat to marine life, and ongoing ocean acidification (OA) is predicted to impact bio-toxicity of Cd compounds. However, the cascading effects of changed Cd toxicity to marine primary producers are not well characterized. Here, we studied the impact of OA on Cd toxicity responses in a globally important diatom Phaeodactylum tricornutum under both ambient and elevated pCO2 conditions. We found that increased pCO2 alleviated the impact of additive Cd toxicity on P. tricornutum not only under controlled indoor experiments but also in outdoor mesocosm experi
