Altered splenic T-cell subpopulation dynamics in a murine model of bovine digital dermatitis-associated Treponema spp. infection.

Bovine digital dermatitis (BDD) is an infectious hoof disease associated with multiple Treponema species, causing significant economic and welfare concerns in the cattle industry. Studies regarding T-cell mediated immune responses are limited in BDD. Using a mouse model, this study analyzed different T-cell subsets involved in immune responses against BDD-associated treponemes. Additionally, this study measured Treponema-specific IgG antibody titers to elucidate humoral immune responses. Mice were experimentally infected with 2 × 109 cells/mL of live T. phagedenis HNW1, T. pedis GNW45, T. pedi

Investigation on the effectiveness of a new hoof care product to sustainably reduce and prevent digital dermatitis in dairy cow herds.

The regular application of hoof care and cleaning products is an important part of protocols designed to reduce burden of disease caused by the ubiquitous and multifactorial hoof disease digital dermatitis (DD) in dairy cows. Commonly used hoof care products such as formalin or copper sulfate applied through foot baths or by spraying hindfeet are often irritant to the skin as well as harmful to the environment or human health, and scientifically proven evidence of their efficacy is scarce. Thus, in a clinical controlled trial, we investigated whether the use of a hoof care product based on a m

Effect of footbath protocols for prevention and treatment of digital dermatitis in dairy cattle: A systematic review and network meta-analysis.

Digital dermatitis (DD) is an infectious bacterial disease affecting cattle feet. Footbaths are a common herd-level control method for DD; however, variations in product, concentration, and frequency of use complicate comparisons between protocols. The objective of this systematic review was to evaluate all walk-through footbath protocols reported that determined efficacy for prevention and treatment of DD lesions in dairy cattle. An extensive literature search was conducted, including electronic databases and gray literature updated until March 2018. Studies identified included all liquid wal

Transcriptomic analysis of bovine peripheral blood mononuclear cells stimulated with whole cell lysates from bovine digital dermatitis associated treponemes.

Bovine digital dermatitis (BDD) is an infectious disease in cattle associated with multiple Treponema species yet information on the immune responses of circulating immune cells to these pathogens are limited. In this study, we characterized the transcriptomic responses of bovine peripheral blood mononuclear cells (PBMCs) following stimulation with combined whole cell lysates derived from three BDD-associated Treponema species. PBMCs were stimulated for 12 h and differentially expressed genes between non-stimulated control and stimulated groups were identified. Functional enrichment of DEGs wa

Digital dermatitis in dairy cattle in Egypt: Herd-level risk factors and Treponema spp. prevalence across lesion M-scores.

Digital dermatitis (DD) is an infectious disease of the digital skin of dairy cows that is associated with compromised animal welfare and significant economic losses. The hind feet of 16,098 dairy cows from 55 herds were examined in the milking parlor, and DD lesions identified were classified using the M-score system and swabbed for PCR testing. Swabs were also collected from hind feet with normal digital skin for comparison. Data on farm construction and lameness prevention measures were collected through direct observation and in-person interviews with farm managers. Herd-level risk factors

In vitro susceptibility of bovine digital dermatitis treponemes to conventional and novel antimicrobial agents, and evidence of heavy metal resistance.

OBJECTIVES: Bovine digital dermatitis (BDD) is a globally endemic, painful infectious foot disease of cattle; however, gaps in knowledge regarding treatment efficacy limit our ability to control it. Our objectives were to collect data on minimum inhibitory concentrations and minimum bactericidal concentrations (MICs and MBCs) for conventional and naturally derived therapeutic agents against specific Treponema strains important in BDD pathogenesis in the context of the need for responsible use of antimicrobials. Since antibacterial metals are widely used to control infectious lameness in rumina

Proteolytic dysfunction in bovine digital dermatitis caused by Treponema spp. and the therapeutic use of non-antimicrobial matrix metalloproteinase inhibitors.

Digital dermatitis (DD) is a painful hoof disease in cattle, closely linked to Treponema spp., for which the pathogenesis remains poorly understood and effective treatments are lacking. This study identifies a persistent pro-inflammatory proteomic signature in untreated, non-healing active DD lesions in the foot skin of cattle, characterized by increased immune cell infiltration and elevated matrix metalloproteinase (MMP) activity. Treatment of active DD lesions with CMC2.24, a non-antibiotic MMP inhibitor, maintains the lesions in an M2 stage in most cases; however, it reduces histological de

Metabolic Footprint of Treponema phagedenis and Treponema pedis Reveals Potential Interaction Towards Community Succession and Pathogenesis in Bovine Digital Dermatitis.

Bovine digital dermatitis (BDD) is a cattle infection causing hoof lesions and lameness, with treponemes as key pathogens. We analyzed the metabolic activity of Treponema phagedenis and Treponema pedis using gas chromatography-mass spectrometry for organic acids (OAs), amino acids (AAs), and fatty acids (FAs), and high-performance liquid chromatography for short-chain fatty acids (SCFAs). Key findings include a 61.5% reduction in pyruvic acid in T. pedis and 81.0% in T. phagedenis. 2-hydroxybutyric acid increased by 493.8% in T. pedis, while succinic acid increased by 31.3%, potentially suppor

Multilocus sequence typing of pathogenic treponemes isolated from contagious ovine digital dermatitis stage five lesions: Implications for disease transmission dynamics.

Contagious ovine digital dermatitis (CODD) causes a severe, infectious foot disease and lameness of sheep, is common within the UK and is now also emerging in other countries. As well as causing severe animal welfare issues, huge economic losses emerge from the disease due to weight loss/lack of weight gain, and veterinary treatments. CODD lesion progress is measured, with a scoring system from 1 (early lesions) to 5 (healed). Here, using samples from an experimental flock infected by natural means, samples were taken from CODD stage 5 lesions, post treatment, and subjected to bacterial isolat

M-score and wound healing assessment of 2 nonantibiotic topical gel treatments of active digital dermatitis lesions in dairy cattle.

This open-label, randomized clinical trial with positive control compared the treatment of active digital dermatitis (DD) lesions (stages M1, M2, and M4.1) on dairy cattle hind feet with an enzyme alginogel or a copper and zinc chelate gel (coppergel). Upon recruitment (d 0), active DD lesions were cleaned, photographed, treated, and bandaged. This procedure was repeated on d 3 and d 7, with treatment and bandaging discontinued for those lesions that had transitioned to the M0, M3, or M4 stage on d 7. Day 10 was considered the end of the treatment trial, and all recruited feet were cleaned and

A filter-assisted culture method for isolation of Treponema spp. from bovine digital dermatitis and their identification by MALDI-TOF MS.

Digital dermatitis (DD) is a major infectious foot disease of cattle worldwide. Some DD stages are associated with lameness, and the disease has significant economic and animal welfare consequences. The pathogenesis of the disease is not yet fully understood, but Treponema spp. have been associated consistently with clinical cases. Isolation of these fastidious bacteria is difficult and cumbersome. We describe an improved method enabling the culturing of the 3 Treponema spp. (T. pedis, T. phagedenis, and T. medium) from bovine foot specimens derived from DD lesions, using a combination of memb

At-Home Evaluation of Both Wearable and Touchless Digital Health Technologies for Measuring Nocturnal Scratching in Atopic Dermatitis: Analytical Validation Study.

BACKGROUND: The most common symptom of atopic dermatitis (AD) is pruritus, which is often exacerbated at night and leads to nocturnal scratching and sleep disturbance. The quantification of nocturnal scratching provides an objective measure, which could be used as a clinical trial endpoint tracking this AD-related behavior. However, it is not clear how digital health technologies (DHTs) intended to measure scratching perform in the real-world environment of patient homes. OBJECTIVE: In this study, we present the analytical validation of 2 DHTs: the GENEActiv wristband with Philips sleep and s

Proof of an optimized salicylic acid paste-based treatment concept of ulcerative M2-stage digital dermatitis lesions in 21 dairy cows.

The efficacy of salicylic acid paste (SA) in the treatment of ulcerative bovine digital dermatitis (BDD) was assessed by combining clinical and histopathological analyses with molecular biological techniques. The latter were conducted in a blinded manner to reach maximum objectivity. Prior to treatment, M2-stage BDD lesions (n = 26, diagnosed in 21 dairy cows) exhibited ulceration, with severe perivascular, chronic, lymphoplasmacytic dermatitis and extensive keratinolysis being noted in most cases. Pretreatment biopsy samples (n = 12) followed by povidone-iodine ointment under bandage for one

Surveying bovine digital dermatitis and non-healing bovine foot lesions for the presence of Fusobacterium necrophorum, Porphyromonas endodontalis and Treponema pallidum.

BACKGROUND: Non-healing bovine foot lesions, including non-healing white line disease, non-healing sole ulcer and toe necrosis, are an increasingly important cause of chronic lameness that are poorly responsive to treatment. Recent studies have demonstrated a high-level association between these non-healing lesions and the Treponema phylogroups implicated in bovine digital dermatitis (BDD). However, a polymicrobial aetiology involving other gram-stain-negative anaerobes is suspected. METHODS: A PCR-based bacteriological survey of uncomplicated BDD lesions (n=10) and non-healing bovine foot le

Tracking Reservoirs of Antimicrobial Resistance Genes in a Complex Microbial Community Using Metagenomic Hi-C: The Case of Bovine Digital Dermatitis.

Bovine digital dermatitis (DD) is a contagious infectious cause of lameness in cattle with unknown definitive etiologies. Many of the bacterial species detected in metagenomic analyses of DD lesions are difficult to culture, and their antimicrobial resistance status is largely unknown. Recently, a novel proximity ligation-guided metagenomic approach (Hi-C ProxiMeta) has been used to identify bacterial reservoirs of antimicrobial resistance genes (ARGs) directly from microbial communities, without the need to culture individual bacteria. The objective of this study was to track tetracycline res

Detection of spirochetes by polymerase chain reaction and its relation to the course of digital dermatitis after local antibiotic treatment in dairy cattle.

The aim of the study was to monitor the course of digital dermatitis after local antibiotic treatment in an experimental group (treated on diagnosis) and a control group (treated 5 days later). The present study was carried out on 2 farms involving 18 animals. Monitoring was performed by means of clinical findings and detection of spirochetes on the surface of the lesions, using a polymerase chain reaction. Superficial wound smears were taken before and after treatment. Twelve animals on both farms followed the classical healing process, but six animals responded poorly to treatment. We observ

Short communication: Informative value of an ELISA applied to bulk tank milk to assess within-herd prevalence of digital dermatitis in dairy herds.

The objective of this study was to assess whether an antibody ELISA applied to bulk tank milk (BTM) could be used to accurately estimate within-herd prevalence of digital dermatitis (DD). The ELISA was designed for the detection of antibodies against Treponema phagedenis-like strain V1 (PrrA antigen). The hind feet of all lactating cows from 40 commercial French dairy herds with a history of DD were scored by an observer in the milking parlor, using the 4 M-stage system. After milking, a BTM sample was collected and tested for anti-Treponema phagedenis-like antibodies using the antibody ELISA.
