Transmission from pregnanglionic fibres in the hypogastric nerve to peripheral ganglia of male guinea-pigs.

1. Intracellular records were obtained from ganglion cells of the pelvic plexus of male guinea-pigs.2. The input resistance of cells which responded to intracellular stimulation varied from 40 to 150 MOmega. Slope resistance decreased when the membrane was hyperpolarized. Time constants varied from 5 to 200 msec. Resting membrane potentials ranged from 40 to 70 mV.3. Action potentials in response to direct stimulation were followed by a prolonged phase of after-hyperpolarization.4. A second type of cell was also impaled which did not respond to electrical stimulation. These cells had resting m

Involvement of alpha7- and alpha4beta2-type postsynaptic nicotinic acetylcholine receptors in nicotine-induced excitation of dopaminergic neurons in the substantia nigra: a patch clamp and single-cell PCR study using acutely dissociated nigral neurons.

The receptor subtypes, which mediate nicotine-induced excitation of dopaminergic neurons in the substantia nigra, were investigated by whole-cell patch clamp studies and single-cell RT-PCR using acutely dissociated nigral neurons. Three types of current were observed when acetylcholine (1 mM) was applied to the neurons in the presence of atropine (1 microM) by the U-tube system, which allowed the rapid application of drugs. In 50% of neurons examined, acetylcholine (1 mM) plus atropine (1 microM) evoked a current with a rapidly desensitizing decay phase (designated as type Ia current). In 14%

A study of the inferior mesenteric and pelvic ganglia of guinea-pigs with intracellular electrodes.

1. Ganglion cells in the inferior mesenteric ganglion (IMG) and the pelvic plexus of the guinea-pig were studied using intracellular micro-electrodes.2. Ganglion cells had resting membrane potentials of 55-65 mV. Threshold for initiation of an action potential ranged from 10 to 20 mV depolarization. Action potentials often exceeded 100 mV in amplitude and were followed by an after-hyperpolarization of up to 20 mV.3. Synaptic responses were recorded from cells in the IMG in response to stimulation of the right and left hypogastric nerves, ascending mesenteric, inferior splanchnic and colonic ne

Inhibition of Nigrostriatal Dopamine Release by Striatal GABAA and GABAB Receptors.

Nigrostriatal dopamine (DA) is critical to action selection and learning. Axonal DA release is locally influenced by striatal neurotransmitters. Striatal neurons are principally GABAergic projection neurons and interneurons, and a small minority of other neurons are cholinergic interneurons (ChIs). ChIs strongly gate striatal DA release via nicotinic receptors (nAChRs) identified on DA axons. Striatal GABA is thought to modulate DA, but GABA receptors have not been documented conclusively on DA axons. However, ChIs express GABA receptors and are therefore candidates for potential mediators of

Three acetylcholine receptors in Aplysia neurones.

1. In the pleural ganglion of Aplysia californica, groups of cells were identified that respond differently to an iontophoretic injection of ACh. The anterior neurones are excited by ACh, whereas the medial cells are inhibited. The inhibitory response is biphasic, consisting of a short-latency, rapid component and a longer-latency, slow component. Homologous responses (e.p.s.p.s in the anterior cells and a two-component inhibitory response in the medial cells) are evoked in these same cell groups by stimulation of an identifiable presynaptic neurone.2. The e.p.s.p. and the corresponding ACh po

Multiple nicotine training doses in mice as a basis for differentiating the effects of smoking cessation aids.

RATIONALE: Receptor mechanisms underlying the behavioral effects of clinically used nicotinic acetylcholine receptor agonists have not been fully established. OBJECTIVE: Drug discrimination was used to compare receptor mechanisms underlying the effects of smoking cessation aids. METHODS: Separate groups of male C57BL/6J mice discriminated 0.56, 1, or 1.78 mg/kg of nicotine base. Nicotine, varenicline, and cytisine were administered alone, in combination with each other, and in combination with mecamylamine and dihydro-β-erythroidine (DHβE). Midazolam and morphine were tested to examine sensi

(-)-nicotine ameliorates corticosterone's potentiation of N-methyl-d-aspartate receptor-mediated cornu ammonis 1 toxicity.

Hypercortisolemia, long-term exposure of the brain to high concentrations of stress hormones (i.e. cortisol), may occur in patients suffering from depression, alcoholism, and other disorders. This has been suggested to produce neuropathological effects, in part, via increased function or sensitivity of N-methyl-d-aspartate (NMDA)-type glutamate receptors. Given that cigarette smoking is highly prevalent in some of these patient groups and nicotine has been shown to reduce toxic consequences of NMDA receptor function, it may be suggested that nicotine intake may attenuate the neurotoxic effects

Selective antagonism of amino acid-induced and synaptic excitation in the cat spinal cord.

1. The effects of D-alpha-aminoadipate (DalphaAA), D-alpha-aminosuberate (DalphaAS) and other excitatory amino acid antagonists have been compared on the excitatory responses of neurones of the cat spinal cord to acetylcholine, a range of glutamate-related amino acids and stimulation of appropriate excitatory synaptic pathways. The ionophoretic technique was used for administration of excitants and antagonists. 2. DalphaAA and DalphaAS had little or no effect on acetylcholine-induced excitation of Renshaw cells. Responses of either Renshaw cells or dorsal horn neurones in the spinal cord to ex

Ni(2+) affects dopamine uptake which limits suitability as inhibitor of T-type voltage-gated Ca(2+) channels.

Neuronal T-type voltage-gated Ca(2+) channels are reported to have physiological roles that include regulation of burst firing, Ca(2+) oscillations, and neurotransmitter release. These roles are often exposed experimentally by blocking T-type channels with micromolar Ni(2+). We used Ni(2+) to explore the role of axonal T-type channels in dopamine (DA) release in mouse striatum, but identified significant off-target effects on DA uptake. Ni(2+) (100 μM) reversibly increased electrically evoked DA release and markedly extended its extracellular lifetime, detected using fast-scan cyclic voltammet

Augmented responses to intrathecal nicotinic agonists in spontaneous hypertension.

Abnormal central cholinergic activity has been reported to be responsible in part for the pathogenesis of high blood pressure in spontaneously hypertensive rats (SHR). Administration of cholinergic agonists in brain and spinal cord results in exaggerated pressor responses in SHR. Studies to date have focused largely on the muscarinic cholinergic system. Recently, we demonstrated that intrathecal administration of nicotinic agonists results in pressor, tachycardic, and irritation responses. In the present study we examine the cardiovascular and behavioral responses to nicotine and cytisine admi

Inhibition of peripheral macrophages by nicotinic acetylcholine receptor agonists suppresses spinal microglial activation and neuropathic pain in mice with peripheral nerve injury.

BACKGROUND: Neuro-immune interaction underlies chronic neuroinflammation and aberrant sensory processing resulting in neuropathic pain. Despite the pathological significance of both neuroinflammation-driven peripheral sensitization and spinal sensitization, the functional relationship between these two distinct events has not been understood. METHODS: In this study, we determined whether inhibition of inflammatory macrophages by administration of α4β2 nicotinic acetylcholine receptor (nAChR) agonists improves neuropathic pain and affects microglial activation in the spinal dorsal horn (SDH) i

Contribution of α4β2 nAChR in nicotine-induced intracellular calcium response and excitability of MSDB neurons.

The neurons of medial septal diagonal band of broca (MSDB) project to hippocampus and play an important role in MSDB-hippocampal synaptic transmission, plasticity and network oscillation. Nicotinic acetylcholine receptor (nAChR) subunits, α4β2 and α7 nAChRs, are expressed in MSDB neurons and permeable to calcium ions, which may modulate the function of MSDB neurons. The aims of this study are to determine the roles of selective nAChR activation on the calcium responses and membrane currents in MSDB neurons. Our results showed that nicotine increased calcium responses in the majority of MSDB ne

(S)-3-methyl-5-(1-methyl-2-pyrrolidinyl) isoxazole (ABT 418): a novel cholinergic ligand with cognition-enhancing and anxiolytic activities: I. In vitro characterization.

A diversity of nicotinic acetylcholine receptor (nAChR) subtypes has been identified in mammalian brain using recombinant DNA technology. Alterations in the activity of these acetylcholinegated ion channels have been implicated in a number of central nervous system disorders including Alzheimer's disease (AD). The potential therapeutic usefulness of (-)-nicotine [(S)-3-(1-methyl-2-pyrrolidinyl) pyridine], the prototypic agonist at nAChRs, is severely limited by side effects that are the result of activation of both cholinergic and noncholinergic pathways in the central and peripheral nervous s

Kappa 2-bungarotoxin and kappa 3-bungarotoxin: two new neuronal nicotinic receptor antagonists isolated from the venom of Bungarus multicinctus.

Neuronal nicotinic acetylcholine receptors are recognized with high affinity by two snake venom kappa-neurotoxins, kappa-bungarotoxin and kappa-flavitoxin. Native and radiolabeled kappa-neurotoxins have been used to localize and quantitate neuronal nicotinic receptors in a variety of species. We now report the identification of two new kappa-neurotoxins. kappa 2-Bungarotoxin and kappa 3-bungarotoxin were purified from the venom of Bungarus multicinctus collected in the province of Guangdong, China. kappa-Bungarotoxin has as yet not been found in this venom, although it is the only kappa-neurot

A study of the interaction between motoneurones in the frog spinal cord.

1. A short-latency interaction between motoneurones has been studied with intracellular and root potential recordings from the isolated spinal cord of the frog. Antidromic stimulation of one ventral root causes brief depolarization (VR-EPSP) of the motoneurones of adjacent, non-excited motoneurones. The summed activity of many such VR-EPSPs can be seen as a brief depolarization (VR-VRP) passing out an adjacent ventral root.2. Both intracellular and root-recorded signs of this interaction are graded in amplitude.3. It was found that this interaction decreased with increasing temperature. This i

Brainstem cholinergic pathways diminish cardiovascular and neuroinflammatory actions of endotoxemia in rats: Role of NFκB/α7/α4β2AChRs signaling.

Nicotine improves endotoxic manifestations of hypotension and cardiac autonomic dysfunction in rats. Here, we test the hypothesis that brainstem antiinflammatory pathways of α7/α4β2 nicotinic acetylcholine receptors (nAChRs) modulate endotoxic cardiovascular derangements. Pharmacologic and molecular studies were performed to determine the influence of nicotine or selective α7/α4β2-nAChR ligands on cardiovascular derangements and brainstem neuroinflammation caused by endotoxemia in conscious rats. The i.v. administration of nicotine (50 μg/kg) abolished the lipopolysaccharide (LPS, 10 mg/kg i.v

Genetically encoded nAChR upregulation is neuroprotective in female parkinsonian mice.

Parkinson's disease is projected to rise to pandemic proportions by 2050, which has resulted in an urgent need for disease-modifying treatments. In this regard, we previously showed that in a mouse model of parkinsonism with unilateral 6-hydroxydopamine (6-OHDA) injection into the dorsolateral striatum (DLS), low doses of the neuronal nicotinic acetylcholine receptor (nAChR) partial agonist and smoking cessation drug, cytisine exerts sex-specific neuroprotection in substantia nigra pars compacta (SNc) dopaminergic (DA) neurons of only female mice by reducing apoptotic endoplasmic reticulum (ER

In vivo PET imaging of the α4β2 nicotinic acetylcholine receptor as a marker for brain inflammation after cerebral ischemia.

PET imaging of nicotinic acetylcholine receptors (nAChRs) could become an effective tool for the diagnosis and therapy evaluation of neurologic diseases. Despite this, the role of nAChRs α4β2 receptors after brain diseases such as cerebral ischemia and its involvement in inflammatory reaction is still largely unknown. To investigate this, we performed in parallel in vivo magnetic resonance imaging (MRI) and positron emission tomography (PET) with 2[(18)F]-fluoro-A85380 and [(11)C]PK11195 at 1, 3, 7, 14, 21, and 28 d after middle cerebral artery occlusion (MCAO) in rats. Presynaptic α4β2 nicotinic acetylcholine receptors increase glutamate release and serotonin neuron excitability in the dorsal raphe nucleus.

Several behavioral effects of nicotine are mediated by changes in serotonin (5-HT) release in brain areas that receive serotonergic afferents from the dorsal raphe nucleus (DRN). In vitro experiments have demonstrated that nicotine increases the firing activity in the majority of DRN 5-HT neurons and that DRN contains nicotinic acetylcholine receptors (nAChRs) located at both somata and presynaptic elements. One of the most common presynaptic effects of nicotine is to increase glutamate release. Although DRN receives profuse glutamatergic afferents, the effect of nicotine on glutamate release
