The heartworm Dirofilaria immitis is the causative agent of cardiopulmonary dirofilariosis in dogs and cats, which also infects a wide range of wild mammals and humans. The complex life cycle of D. immitis with several developmental stages in its invertebrate mosquito vectors and its vertebrate hosts indicates the importance of miRNA in growth and development, and their ability to regulate infection of mammalian hosts. This study identified the miRNA profiles of D. immitis of zoonotic significance by deep sequencing. A total of 1063 conserved miRNA candidates, including 68 anti-sense miRNA (mi

Reports of loss-of-efficacy (LOE) events in dogs infected with Dirofilaria immitis despite adherence to accepted prophylaxis regimens with a macrocyclic lactone anthelmintic are attracting considerable attention. It is crucially important to distinguish among several possible causes for these LOE reports, one of which is the evolution of resistance to these drugs in heartworms. We review here recent evidence at the molecular level that supports the hypothesis that parasites derived from LOE cases have experienced a strong selection event and that these populations are characterized by very hig

Blood samples from 260 dogs over 1 year of age from eight districts of the western part of Albania (20 to 50 dogs per district) were collected in 1995 and 1996 and tested for circulating Dirofilaria immitis antigen using the PetChek HTWM PF (IDEXX) ELISA test kit. Overall seroprevalence of D. immitis infection among these dogs was 13.5% (95% confidence interval = 9.3-17.6%), ranging from 5% to 30% between the districts of origin of the dogs. There was no difference for the seroprevalence of heartworm infection between the sexes and age classes or between pure and mixed breed dogs; however, D.

Three dogs were experimentally infected with Dirofilaria immitis. All dogs were euthanised at 30, 36 and 37 weeks after inoculation of D. immitis for the recovery of adult worms. Three cases accounted to 42.91 % recovery of inoculated worms. Serum samples from dogs experimentally inoculated with D. immitis were analyzed by ELISA and immunoblotting methods. Antibody titers of dogs detected by ELISA peaked between 7 and 14 weeks then decreased between weeks 15 to 24 followed by another increase during weeks 25 to 30 and persisted throughout the remainder of the experiment period. Analysis of adu

Canine heartworm or Dirofilaria immitis can occasionally infect man. We present the case of a 36-year-old Balkan woman referred to us for a thoracoscopic biopsy of a well defined pulmonary mass. The latter was thoracoscopically resected and proved to be due to Dirofilaria immitis on histopathological examination. To our knowledge this is the second reported case in Western Australia, the first being that reported by Brine et al. The prevalence of the disease in Australia was recognized in 1969 in one study in the state of Queensland, where 12% of 761 dogs at one veterinary clinic and 20% of 29

One worm recovered from the orbit of a patient in Indiana, and two from the peritoneal cavity of another patient in California are described and classified as Dirofilaria. Both patients were elderly men who have lived all their lives in the United States. The main morphologic feature of these three immature worms is the presence of a smooth cuticle devoid of longitudinal ridges, similar to the cuticle of D. immitis. However, because other dirofilariid with similar cuticular characteristics have been found in animals in the Western Hemisphere, and they cannot be conclusively ruled out as the ca

Superoxide dismutase (SOD) may not only perform a housekeeping role in filarial worms but also assist in defense against oxidants generated by host immune cells. Both Dirofilaria and Onchocerca adult filariae and microfilariae contain relatively high activities of the antioxidant enzyme SOD; adult Dirofilaria worms also secrete SOD in vitro. In addition, superoxide radicals are relatively impotent against Dirofilaria and Onchocerca microfilariae in vitro. In assessing the role of SOD, we determined the anatomic localization of SOD in D. immitis adult worms by immunolocalization at the light-mi

To help to develop better diagnostic tests for zoonotic pulmonary dirofilariasis, seven patients with histologically confirmed pulmonary dirofilariasis were evaluated using the immunoblot technique. Six patients exhibited a response to the excretory-secretory (AS) antigen proteins with molecular weights of 20-19.5, 17.5-17 and 14 derived from Dirofilaria immitis adult works. However, when adult worm extracts were used as the antigen, there was no qualitative difference in antigen recognition between the Dirofilaria patients, other patients with non-filarial parasitic infections or with lung ca

A method for the identification of circulating parasite antigens in filarial nematode infections was developed using canine infections with Dirofilaria immitis as a model. Filarial antigens ranging in molecular weight from 211 to 13 kDa were extracted from the sera of microfilaremic dogs by a solid phase immunobinding procedure and identified by immunostaining of Western blots. A major antigen of 104 kDa was selected for further characterization. The 104 kDa circulating antigen showed antigenic and biochemical identity with 104 kDa peptides found in extracts of adult male and microfilarial sta

The immunopathogenesis of the anaphylactoid Mazzotti reactions has been studied by comparing physiologic and immunologic aspects of diethylcarbamazine-induced shock in Dirofilaria immitis infected dogs with antigen induced anaphylaxis in infected and uninfected controls. Filarial antigen, specific host IgG antibody, and C1 and C3 complement levels were quantitatively measured over time in relation to the levels of histamine and prostaglandin D2 in the blood and changes in mean blood pressure. Fourteen beagles infected with larvae (microfilariae) of Dirofilaria immitis, were randomly selected from another study in which the toxic effects of subfilaricidal doses of diethylcarbamazine were being evaluated. This group of 14 dogs, together with 4 uninfected control animals, were variably sacrificed between 14 and 25 months after larval inoculations, and the ensuing renal lesions were studied by light and ultrastructural microscopy and by immunofluorescence and antibody elution techniques. On the basis of these studies, two groups of animals were distinguished. The first group was charac

Canine dirofilarioses are mosquito-borne zoonotic diseases with a continuous expansion of their geographical distribution, as a consequence of different climatic and ecological factors. Dirofilaria immitis, the aetiological agent of heartworm disease, has gained a major veterinary interest, mainly due to its severe clinical implication. In the last decades, D. repens, despite of being regarded as a less pathogenic species, regained attention due to its recognized zoonotic potential. Romania has been traditionally regarded as a non-endemic country, but recent epidemiological surveys are highlig

This survey was conducted to investigate the presence of Dirofilaria immitis and Dirofilaria repens in native dogs and cats living in an area of Italy where animal and human dirofilarioses have been sporadically reported in the past few years. From May 2008 to March 2009 individual blood samples were collected from 300 native cats and 300 native dogs in the Abruzzo region of central Italy. All 600 samples were subjected to a Knott modified method, to a test for the detection of circulating D. immitis antigen and to two species-specific PCR-coupled sequencing protocols. Epidemiological data wer

BACKGROUND: Dirofilariasis is a vector-borne disease caused by parasitic nematodes of the genus Dirofilaria spp., considered an emerging concern in both veterinary and human medicine. Climate changes and human activities, such as pet travel, contribute to the spread of diseases to new non-endemic regions. Poland is dominated by subcutaneous dirofilariasis caused by D. repens infections. Cardiopulmonary dirofilariasis, also known as a heartworm disease is much more rare with only single autochthonous cases reported so far. Also, imported infections are observed sporadically in dogs traveling to

We report a case of Dirofilaria immitis nematode infection in a dog imported from Venezuela that had been living for 2 years in Santiago, Chile, where this parasite had not been reported before. Our findings warrant surveillance for all dogs imported to Chile, given that suitable conditions exist for establishing this parasite.

Microfilariae (Mfs) of filarial nematode parasites exhibit nocturnal periodicity, with their numbers in peripheral blood peaking at night and decreasing during the day. However, the reason for their appearance at night remains unknown. In this study, in vitro photobiostimulation experiments showed that Mfs exhibited positive phototaxis toward infrared light with lower photon flux densities of infrared light at wavelengths of 890 and 700 nm, in particular, mediating paradoxically higher velocity than intense ones. Microarray analysis revealed that infrared light stimulation influenced gene expr

BACKGROUND: Heartworm (Dirofilaria immitis) in dogs is considered endemic in Australia, but the clinical heartworm disease caused by the heartworm is rare and prevalence is low. The mainstream prevention of the heartworm is based on macrocyclic lactone (ML) administration. The aim of this study was to confirm endemism of the heartworm under current Australian conditions using a cohort of recent microfilaria-positive dogs which were on variable heartworm prevention.
METHODS: A hotspot of canine heartworm antigen-positive and microfilaria-positive dogs has been detected recently in Queensland,

BACKGROUND: This paper forecasts next year's canine heartworm prevalence in the United States from 16 climate, geographic and societal factors. The forecast's construction and an assessment of its performance are described.
METHODS: The forecast is based on a spatial-temporal conditional autoregressive model fitted to over 31 million antigen heartworm tests conducted in the 48 contiguous United States during 2011-2015. The forecast uses county-level data on 16 predictive factors, including temperature, precipitation, median household income, local forest and surface water coverage, and presen

Dirofilaria immitis is the causal agent of cardiopulmonary dirofilariosis (heartworm disease). Adult worms lodge in the pulmonary arteries and right ventricle, thus vascular endothelium is exposed to high concentrations of Dirofilaria antigenic products. Heartworm disease habitually develops as a chronic foreseeable pathology. Moreover, the simultaneous death of many adult worms, naturally or induced by a filaricide treatment, can cause acute thromboembolisms and endarteritis. To better understand the effects of the massive death of D. immitis adult worms on the blood vessel endothelium, we cu
