Conversion of trichloroacetic acid to dichloroacetic acid in biological samples.

Trichloroethylene (TCE) has been identified as an environmental contaminant in groundwater. Trichloroacetic acid (TCA), dichloroacetic acid (DCA), chloral hydrate (CH), trichloroethanol (TCOH), and trichloroethanol glucuronide (TCOG) have been identified as metabolites of TCE. Studies have shown that TCA and DCA are toxicologically significant metabolites that can induce liver tumors in B6C3F1 mice. Methods for the analysis of these metabolites are important for conducting pharmacokinetic studies. In this study, TCA and DCA were derivatized to their methyl esters by dimethyl sulfate under acid

Sulfidation of Nano Zerovalent Iron (nZVI) for Improved Selectivity During In-Situ Chemical Reduction (ISCR).

The high reactivity of nano zerovalent iron (nZVI) leads to inefficient treatment due to competition with various natural reductant demand (NRD) processes, especially the reduction of water to hydrogen. Here we show that this limitation can be alleviated by sulfidation (i.e., modification by reducing sulfur compounds). nZVI synthesized on carboxylmethylcelluose (CMC-nZVI) was sulfidated with either sulfide or dithionite. The reactivity of the resulting materials was examined with three complementary assays: (i) direct measurement of hydrogen production, (ii) reduction of a colorimetric redox p

Impact of the chemicals, essential for the purification process of strict Fe-hydrogenase, on the corrosion of mild steel.

The influence of additional chemical molecules, necessary for the purification process of [Fe]-hydrogenase from Clostridium acetobutylicum, was studied on the anaerobic corrosion of mild steel. At the end of the purification process, the pure [Fe-Fe]-hydrogenase was recovered in a Tris-HCl medium containing three other chemicals at low concentration: DTT, dithionite and desthiobiotin. Firstly, mild steel coupons were exposed in parallel to a 0.1 M pH7 Tris-HCl medium with or without pure hydrogenase. The results showed that hydrogenase and the additional molecules were in competition, and the

The binding of cyanide to ferroperoxidase.

1. The equilibrium and kinetics of cyanide binding to ferroperoxidase were investigated. At pH9.1 the equilibrium and kinetic measurements agree closely and disclose a single process with an affinity constant of 1.1x10(3)m(-1) and combination and dissociation velocity constants of 29m(-1).s(-1) and 2.5x10(-2)s(-1) respectively. 2. At pH values below 8 the affinity constant falls until at pH6.0 the ferroperoxidase.cyanide complex is no longer formed. This is shown to be associated with the formation of ferriperoxidase.cyanide complex in the mixture even in the presence of excess of sodium dithi

Comparative biodistribution studies of technetium-99 m radiolabeled amphiphilic nanoparticles using three different reducing agents during the labeling procedure.

Considering the confusing biodistribution data through the literature and few reported alerts as well as our preliminary biodistribution results, we decided to evaluate the interaction and interference of the commonly present (99m) Tc (technetium-99m)-stannic oxide colloid during the direct stannous chloride (99m) Tc-labeling procedure and to assess its influence on the biodistribution pattern of amphiphilic poly(lactic-co-glycolic acid) nanoparticles. In order to confirm our thesis, beside stannous chloride, we employed two different reducing agents that don't form colloidal particles. The us

Characterization of the partially reduced cyanide-inhibited derivative of cytochrome c oxidase by optical, electron-paramagnetic-resonance and magnetic-circular-dichroism spectroscopy.

Optical. e.p.r. and near-infrared low-temperature m.c.d. (magnetic-circular-dichroism) spectroscopy were used to characterize the partially reduced cyanide-inhibited derivative of cytochrome c oxidase produced by anaerobic reductive titration with dithionite. The reductions of cytochrome a3+ and Cu2+a were followed by observation of the e.p.r. signals at g = 3.03, 2.21 and 1.5 and at g = 2.18, 2.03 and 1.99. As reduction proceeds new e.p.r. signals (g = 3.58 and 1.56) appear that quantify to give one haem per enzyme unit when a small excess of dithionite has been titrated in. The e.p.r. signal

Novel heme ligation in a c-type cytochrome involved in thiosulfate oxidation: EPR and MCD of SoxAX from Rhodovulum sulfidophilum.

The SoxAX complex of the bacterium Rhodovulum sulfidophilum is a heterodimeric c-type cytochrome that plays an essential role in photosynthetic thiosulfate and sulfide oxidation. The three heme sites of SoxAX have been analyzed using electronic absorption, electron paramagnetic resonance, and magnetic circular dichroism spectroscopies. Heme-3 in the ferric state is characterized by a Large g(max) EPR signal and has histidine and methionine axial heme iron ligands which are retained on reduction to the ferrous state. Hemes-1 and -2 both have thiolate plus nitrogenous ligand sets in the ferric s

Iron-ascorbate complex formation and redox behavior in Tris buffer under aerobic and anaerobic conditions: relevance to iron-deficiency anemia.

Iron-ascorbate (Fe-Asc) is a clinically approved drug for iron-deficiency anemia (IDA), yet its formation and redox behavior under varying conditions remain poorly understood. Herein, we report the first comprehensive examination of Fe-Asc formation and redox reactivity in Tris buffer as a function of stoichiometry, pH, and oxygen availability. Oxidation of Fe(II) in air-saturated buffer accelerates with pH, yielding multiple species, primarily di- and tri-nuclear clusters. Notably, FeSO₄ with AscH produces a distinct purple complex (λmax = 510 nm), whose assembly depends critically on O₂, 1:3

Cyanide binding to human plasma heme-hemopexin: a comparative study.

Hemopexin (HPX) displays a pivotal role in heme scavenging and delivery to the liver. In turn, heme-Fe-hemopexin (HPX-heme-Fe) displays heme-based spectroscopic and reactivity properties. Here, kinetics and thermodynamics of cyanide binding to ferric and ferrous hexa-coordinate human plasma HPX-heme-Fe (HHPX-heme-Fe(III) and HHPX-heme-Fe(II), respectively), and for the dithionite-mediated reduction of the HHPX-heme-Fe(III)-cyanide complex, at pH 7.4 and 20.0°C, are reported. Values of thermodynamic and kinetic parameters for cyanide binding to HHPX-heme-Fe(III) and HHPX-heme-Fe(II) are K = (4.

Comparative Evaluation of Mediated Electrochemical Reduction and Chemical Redox Titration for Quantifying the Electron Accepting Capacities of Soils and Redox-Active Soil Constituents.

The electron accepting capacity (EAC) of soil plays a pivotal role in the biogeochemical cycling of nutrients and transformation of redox-labile contaminants. Prior EAC studies of soils and soil constituents utilized different methods, reductants, and mediators, making cross-study comparison difficult. This study was conducted to quantify and compare the EACs of two soil constituents (hematite and Leonardite humic acid) and 12 soils of diverse composition, using chemical redox titration (CRT) with dithionite as the reductant and mediated electrochemical reduction (MER) with diquat as the media

Evidence for the inorganic nature of the cyanolyzable sulfur of molybdenum hydroxylases.

Activation of the desulfo forms of milk xanthine oxidase, chicken liver xanthine dehydrogenase, and aldehyde oxidase with S2- is greatly facilitated in the presence of reducing agents. Upon anaerobic incubation with 1 mM S2- and 1 mM dithionite, desulfo xanthine oxidase and chicken liver xanthine dehydrogenase prepared by cyanide treatment of active enzymes, are activated to the specific activity predicted by their molybdenum content. Routine preparations containing desulfo molecules are also similarly activated to the extent predicted. Cyanide-inactivated chicken liver xanthine dehydrogenase

Sulfidation of Magnetite for Superior Dechlorination of Trichloroethene.

The reported contributions of magnetite to the abiotic natural attenuation of chlorinated ethenes have generated interest in its potential for soil and groundwater remediation. In this study, we investigated the impact of the two-step sulfidation method on the physicochemical properties and reactivity of magnetite with trichloroethene (TCE). We systematically evaluated the effect of different sulfur precursors (dithionite, thiosulfate, and sulfide) and sulfur-to-iron ([S/Fe]dosed) molar ratios on the reactivity. Results were compared to those of sulfidated nZVI (S-nZVI) as a benchmark for asse

High-resolution structures of cholesterol oxidase in the reduced state provide insights into redox stabilization.

Cholesterol oxidase (CO) is a flavoenzyme that catalyzes the oxidation and isomerization of cholesterol to cholest-4-en-3-one. The reductive half reaction occurs via a hydride transfer from the substrate to the FAD cofactor. The structures of CO reduced with dithionite under aerobic conditions and in the presence of the substrate 2-propanol under both aerobic and anaerobic conditions are presented. The 1.32 Å resolution structure of the dithionite-reduced enzyme reveals a sulfite molecule covalently bound to the FAD cofactor. The isoalloxazine ring system displays a bent structure relative to

Reduction Pathway-Dependent Formation of Reactive Fe(II) Sites in Clay Minerals.

Structural Fe in clay minerals is an important, potentially renewable source of electron equivalents for contaminant reduction, yet our knowledge of how clay mineral Fe reduction pathways and Fe reduction extent affect clay mineral Fe(II) reactivity is limited. Here, we used a nitroaromatic compound (NAC) as a reactive probe molecule to assess the reactivity of chemically reduced (dithionite) and Fe(II)-reduced nontronite across a range of reduction extents. We observed biphasic transformation kinetics for all nontronite reduction extents of ≥5% Fe(II)/Fe(total) regardless of the reduction pat

Iron-Promoted Sulfinative Cyclization of N-Bromoacetyl Cyanamide with Inorganic Sulfur Dioxide Surrogate via Radical-Polar Crossover Reaction.

An iron-promoted sulfinative cyclization for the preparation of amidine-imbedded sultines under the mild conditions was described, in which available N-bromoacetyl cyanamide and easy-to-handle sodium dithionite, acting as an easy-to-handle sulfur dioxide surrogate and a readily reductant, facilitated the assembly of the target sultines via a radical-polar crossover pathway in a sustainable manner. This protocol is highly amenable to exploring the chemical space relevant to the construction of sulfur heterocycles incorporating nitrogen functionalities.

Kinetic and ligand binding evidence for two heme A-based terminal oxidases in plasma membranes from Bacillus subtilis.

Detergent-solubilized plasma membranes from Bacillus subtilis have been characterized for their cytochrome oxidase content. Triton X-100-solubilized membranes show high O2 turnover with ascorbate plus TMPD. Reduced-oxidized difference spectroscopy of ascorbate-TMPD-reduced membranes reveals the presence of cytochrome c and cytochrome a. An additional, b-type cytochrome appears when the membranes are reduced with dithionite. Time-resolved difference spectra taken during reduction by ascorbate-TMPD reveal two kinetic forms of heme A-containing cytochromes. There is a high-turnover form that is r

Hole-scavenging in photo-driven N2 reduction catalyzed by a CdS-nitrogenase MoFe protein biohybrid system.

The light-driven reduction of dinitrogen (N2) to ammonia (NH3) catalyzed by a cadmium sulfide (CdS) nanocrystal‑nitrogenase MoFe protein biohybrid is dependent on a range of different factors, including an appropriate hole-scavenging sacrificial electron donor (SED). Here, the impact of different SEDs on the overall rate of N2 reduction catalyzed by a CdS quantum dot (QD)-MoFe protein system was determined. The selection of SED was guided by several goals: (i) molecules with standard reduction potentials sufficient to reduce the oxidized CdS QD, (ii) molecules that do not absorb the excitation

Redox chemistry of anthracycline antitumor drugs and use of captodative radicals as tools for its elucidation and control.

The oxomorpholinyl radicals are unique materials in organic and medicinal chemistry. Their closest parallel lies in inorganic chemistry with dithionite, which exists in equilibrium with sulfur dioxide radical anion, also a one-electron reducing agent. However, dithionite is a more powerful reducing agent and is probably more toxic. The rate of release of the oxomorpholinyl radical from its dimer is medium and is structure dependent, which provides for some level of control. The oxomorpholinyl radicals TM-3 and DHM-3 are selective one-electron reducing agents for the anthracyclines, generating

Characterisation of 'fast' and 'slow' forms of bovine heart cytochrome-c oxidase.

We have prepared cytochrome-c oxidase from bovine heart (using a modification of the method of Kuboyama et al. (1972) J. Biol. Chem. 247, 6375-6383) which binds cyanide rapidly, shows no kinetic distinction between the two haems on reduction by dithionite, has a Soret absorption maximum above 424 nm, and has a negligible 'g' = 12' EPR signal. On incubation at pH 6.5 this 'fast' oxidase reverts to the 'slow' ('resting') form characterised by slow cyanide binding, slow reduction of haem a3 by dithionite, a blue-shifted Soret maximum and a large 'g' = 12' signal. Incubation of 'fast' oxidase with
