Prolactin and growth hormone responses in the amenorrhea-galactorrhea syndrome.

Fifteen patients with hyperprolactinemia, amenorrhea, and galactorrhea were studied; 10 of these patients had apparent tumors. Nine of 12 patients had a suppression of prolactin with oral L-dopa, and all of 4 patients undergoing dopamine infusion had suppression of prolactin. Eleven and 8 patients underwent chlorpromazine stimulation and insulin-induced hypoglycemia, respectively; none responded. Three of the 4 patients did not show a growth hormone response with dopamine infusion, and L-dopa testing failed to achieve a growth hormone response in 7 of 10 patients. The data suggest an intact do

Acute dyskinetic reaction in a healthy toddler following methylphenidate ingestion.

BACKGROUND: Acute dyskinetic or dystonic reactions are a long-recognized complication of medications that alter dopamine signaling. Most reactions occur following exposure to agents that block dopamine receptors (e.g., neuroleptics). However, agents that increase dopaminergic transmission (such as methylphenidate) can also trigger acute dyskinesias. This has been previously reported only in patients also taking dopamine antagonists or, less commonly, in children with developmental abnormalities. CASE DESCRIPTION: The present report describes a previously healthy toddler who developed transien

Psychopharmacological treatment for cognitive impairment in survivors of traumatic brain injury: a critical review.

Traumatic brain injury (TBI) is a growing national health issue that commonly results in clinically significant cognitive impairments. This article reviews and evaluates the many proposed psychopharmacological treatments for TBI-related cognitive impairment. A literature review was utilized to focus on stimulant and nonstimulant dopamine enhancing agents, acetylcholinesterase inhibitors, antidepressant agents, mood stabilizers, antipsychotics, and benzodiazepines. The most consistent evidence supports the use of dopamine enhancing medications. However, other medications such as acetylcholinest

Clozapine fails to prevent the development of haloperidol-induced behavioral hypersensitivity in a cotreatment paradigm.

We have previously established that chronic cotreatments involving antimuscarinic agents and haloperidol attenuate the development of behavioral hypersensitivity without affecting dopamine receptor proliferation. The antipsychotic agent clozapine also has significant antimuscarinic activity and was coadministered with haloperidol in rats for 2 months to determine if it would similarly attenuate the development of hypersensitivity. Clozapine or chlorpromazine cotreatment, unlike thioridazine cotreatment, did not attenuate the development of haloperidol-induced behavioral hypersensitivity. Cloza

Cardiovascular dopamine receptors: physiological, pharmacological and therapeutic implications.

Dopamine receptor activation can lead to pronounced changes in cardiovascular function. The myriad of effects produced by dopamine receptor agonists results from the activation of dopamine receptors located at different anatomical sites in the cardiovascular system. Further basic research is required to better characterize these dopamine receptors so as to allow the development of more specific dopamine receptor agonists. Endogenous dopamine may be involved in the physiological control of fluid and electrolyte balance and continuing research efforts in this area should provide for a better und

Effect of clozapine on dopamine-induced inhibition of prolactin release from cultured rat pituitary cells.

Although the atypical antipsychotic agent clozapine has little propensity to induce hyperprolactinemia in humans it increases serum prolactin levels in the rat. In this study, the effects of clozapine and of some typical antipsychotic drugs on basal and dopamine-mediated prolactin secretion from cultured rat pituitary cells were compared. Despite being less potent than the other antipsychotic agents tested, clozapine reverted the effect of dopamine on prolactin secretion in vitro. This finding suggests that clozapine interferes with dopamine receptors in the pituitary gland.

Antiparkinsonian drugs: inhibition of dopamine uptake in the corpus striatum as a possible mechanism of action.

A variety of antiparkinsonian drugs are potent, noncompetitive inhibitors of dopamine uptake into synaptosomes in homogenates of rat corpus striatum. Inhibition of dopamine uptake may potentiate the synaptic actions of dopamine in the striatum and could explain the antiparkinsonian effects of these drugs. This hypothesis accounts for several clinical features of Parkinson's disease and predicts compounds which may be new therapeutic agents.

Targeting Dopaminergic Pathways: Current and Emerging Dopamine Inhibitors in Neurological and Psychiatric Disorders.

Dopaminergic signaling plays a central role in regulating motor control, reward processing, and emotional balance. Dysregulation of dopamine pathways is implicated in multiple neuropsychiatric disorders. This review primarily focuses on current and emerging dopaminergic inhibitors, including dopamine receptor antagonists, dopamine transporter blockers, and indirect modulators, while also comparatively addressing pro-dopaminergic approaches. Emphasis is placed on their mechanisms, clinical applications, adverse effects, and future therapeutic directions in neurological and psychiatric disorders

Coenzyme Q10 and nicotinamide and a free radical spin trap protect against MPTP neurotoxicity.

1-Methyl-4-phenyl-1,2,5,6-tetrahydropyridine (MPTP) produces Parkinsonism in both experimental animals and in man. MPTP is metabolized to 1-methyl-4-phenylpridinium, an inhibitor of mitochondrial complex I. MPTP administration produces ATP depletions in vivo, which may lead to secondary excitotoxicity and free radical generation. If this is the case then agents which improve mitochondrial function or free radical scavengers should attenuate MPTP neurotoxicity. In the present experiments three regimens of MPTP administration produced varying degrees of striatal dopamine depletion. A combination

Role of oxidative stress and the glutathione system in loss of dopamine neurons due to impairment of energy metabolism.

Alterations in the glutathione system and impairment in energy metabolism have both been implicated in the loss of dopamine neurons in Parkinson's disease. This study examined the importance of cellular glutathione and the involvement of oxidative stress in the loss of mesencephalic dopamine and GABA neurons due to inhibition of energy metabolism with malonate, the reversible, competitive inhibitor of succinate dehydrogenase. Consistent with previous findings, exposure to malonate for 24 h followed by 48 h of recovery caused a dose-dependent loss of the dopamine population with little effect o

The physiology, signaling, and pharmacology of dopamine receptors.

G protein-coupled dopamine receptors (D1, D2, D3, D4, and D5) mediate all of the physiological functions of the catecholaminergic neurotransmitter dopamine, ranging from voluntary movement and reward to hormonal regulation and hypertension. Pharmacological agents targeting dopaminergic neurotransmission have been clinically used in the management of several neurological and psychiatric disorders, including Parkinson's disease, schizophrenia, bipolar disorder, Huntington's disease, attention deficit hyperactivity disorder (ADHD(1)), and Tourette's syndrome. Numerous advances have occurred in un

Fungal-derived semiochemical 1-octen-3-ol disrupts dopamine packaging and causes neurodegeneration.

Parkinson disease (PD) is the most common movement disorder and, although the exact causes are unknown, recent epidemiological and experimental studies indicate that several environmental agents may be significant risk factors. To date, these suspected environmental risk factors have been man-made chemicals. In this report, we demonstrate via genetic, biochemical, and immunological studies that the common volatile fungal semiochemical 1-octen-3-ol reduces dopamine levels and causes dopamine neuron degeneration in Drosophila melanogaster. Overexpression of the vesicular monoamine transporter (V

Electrochemically Grown Hole-Rich NiO(OH) Thin Films toward Hole-Mediated Very Fast and Selective Enzyme-Free Electrochemical Sensing of Dopamine under Simulated Environment.

This work aimed to develop an enzyme-free semiconductor-assisted electrochemical technique for the selective detection of the neurotransmitter dopamine. In this case, electrochemically grown nickel oxyhydroxide [NiO(OH)] thin films were chosen to fabricate the sensing platform, i.e., the electrodes. Chronoamperometry was used to deposit the films on indium tin oxide (ITO) coated glass substrates. The films were thoroughly characterized to establish their structure, composition, phase purity, and electrochemical attributes. Electrochemical sensing characteristics were investigated by means of c

Smaller effect of propofol than sevoflurane anesthesia on dopamine turnover induced by methamphetamine and nomifensine in the rat striatum: an in vivo microdialysis study.

Volatile anesthetics accelerate dopamine turnover in the brain, especially when used in conjunction with psychotropic agents such as methamphetamine and nomifensine. The effect of intravenous propofol anesthesia on the extracellular dopamine concentrations is unclear. The aim of this study was to compare the effect of two anesthetics on the extracellular concentrations of dopamine and metabolites using an in vivo microdialysis model. Male Sprague Dawley rats were implanted with a microdialysis probe into the right striatum. The probe was perfused with modified Ringer's solution, and the dialys

Detection of Dopamine Receptors Using Nanoscale Dendrimer for Potential Application in Targeted Delivery and Whole-Body Imaging: Synthesis and In Vivo Organ Distribution.

Dopamine is one of the most important neurotransmitters released by neurons in the central nervous system, and a variety of neurological illnesses and mental disorders are associated with impairments in the secretion and functionality of dopamine. Dopamine, depending on the type of receptors, can act as a stimulant or an inhibitor. In this study, dendrimer-conjugated dopamine was utilized as a chelating agent for Technetium-99m to investigate the organ distribution of this compound in vivo using the single-photon emission computed tomography (SPECT) technique. For this purpose, dendrimers were

An outline of anti-emetic treatment.

With the advent of new chemotherapeutic agents, and their well-known side effect of emesis, the need for a greater facility with anti-emetics has emerged. As well as the common problem of chemotherapy-induced emesis, other problems have become apparent such as delayed emesis and anticipatory emesis. The control of such emetic problems may be affected by certain patient characteristics, such as a history of chronic high alcohol intake, and age. Blockade of the different types of neuroreceptors can lead to effective emetic control. Dopamine receptor blockers such as the phenothiazines, butyrophe

Comparison of dopamine and norepinephrine in the treatment of shock.

BACKGROUND: Both dopamine and norepinephrine are recommended as first-line vasopressor agents in the treatment of shock. There is a continuing controversy about whether one agent is superior to the other. METHODS: In this multicenter, randomized trial, we assigned patients with shock to receive either dopamine or norepinephrine as first-line vasopressor therapy to restore and maintain blood pressure. When blood pressure could not be maintained with a dose of 20 microg per kilogram of body weight per minute for dopamine or a dose of 0.19 microg per kilogram per minute for norepinephrine, open-
