Metal Probe Microextraction Coupled to Dielectric Barrier Discharge Ionization-Mass Spectrometry for Detecting Drug Residues in Organisms.

In this study, a novel method for the direct coupling of metal probe microextraction (MPME) and a dielectric barrier discharge ionization (DBDI) source with mass spectrometry (MS) is reported. Analytes adsorbed on a tungsten needle were directly transferred to the DBDI source via rapid thermal desorption, which resulted in a limit of detection as low as 8 pg/mL. This is in part due to the "active capillary" configuration of the plasma ion source, where the efficiency of ion transfer to the MS is ∼100%. Specialty gases to maintain the plasma and carry analytes to the MS are not required. In con

Multi-class, multi-residue liquid chromatography/tandem mass spectrometry screening and confirmation methods for drug residues in milk.

This paper describes the development and optimization of a multi-residue veterinary drug screening method for whole milk. The drug residues of regulatory interest in milk include beta-lactams, sulfonamides, tetracyclines, fluoroquinolones, and macrolides. Milk samples were extracted with acetonitrile and the samples were then subjected to a clean-up procedure using a bonded solid-phase extraction cartridge and a molecular weight cut-off filter. Liquid chromatography/tandem mass spectrometry (LC/MS/MS) triple quadrupole electrospray methods were developed to monitor for the drugs in milk. Since

Atomic modelling and systematic mutagenesis identify residues in multiple drug binding sites that are essential for drug resistance in the major Candida transporter Cdr1.

The ABC (ATP-Binding Cassette) transporter Cdr1 (Candida drug resistance 1) protein (Cdr1p) of Candida albicans, shows promiscuity towards the substrate it exports and plays a major role in antifungal resistance. It has two transmembrane domains (TMDs) comprising of six transmembrane helices (TMH) that envisage and confer the substrate specificity and two nucleotide binding domains (NBDs), interconnected by extracellular loops (ECLs) and intracellular loops (ICLs) Cdr1p. This study explores the diverse substrate specificity spectrum to get a deeper insight into the structural and functional fe

Recognition of Potential COVID-19 Drug Treatments through the Study of Existing Protein-Drug and Protein-Protein Structures: An Analysis of Kinetically Active Residues.

We report the results of our in silico study of approved drugs as potential treatments for COVID-19. The study is based on the analysis of normal modes of proteins. The drugs studied include chloroquine, ivermectin, remdesivir, sofosbuvir, boceprevir, and α-difluoromethylornithine (DMFO). We applied the tools we developed and standard tools used in the structural biology community. Our results indicate that small molecules selectively bind to stable, kinetically active residues and residues adjoining them on the surface of proteins and inside protein pockets, and that some prefer hydrophobic s

Residues of carcinogenic animal drugs in food: difficulties in evaluation of human safety.

The indisputable need to intensify animal production in order to provide an adequate food supply for the world population involves the use of substances that are highly potent pharmacologically and toxicologically. The history of regulatory action with regard to such additives is similar to that for other substances: first, no regulation; next, an over-reaction; and now decisions based on judicious evaluation of scientific facts. One factor that differentiates the chemicals used in animal production from other food additives is that both the parent compounds and their metabolites appear in edi

[Do pharmaceutical waste and drug residue pose a risk to public health?].

Recently, awareness has developed of the environmental consequences of drug waste and disposal. These residues are identified as coming from either diffuse sources, the most significant of which is via the discharge of these residues in urine and feces, and thus the sewage system and water contains these drug remnants and their metabolites, or from point sources, sometimes with very high levels of concentration in waste from chemical and pharmaceutical industries, health care settings, but also from intensive livestock farming and aquaculture. Depending on their physical chemistry properties,

Pressure-Sensitive Adhesive Combined with Paper Spray Mass Spectrometry for Low-Cost Collection and Analysis of Drug Residues.

Illicit drug use causes over half a million deaths worldwide every year. Drugs of abuse are commonly smuggled through customs and border checkpoints and, increasingly, through parcel delivery services. Improved methods for detection of trace drug residues from surfaces are needed. Such methods should be robust, fieldable, sensitive, and capable of detecting a wide range of drugs. In this work, commercially produced paper with a pressure-sensitive adhesive coating was utilized for the collection and analysis of trace drug residues by paper spray mass spectrometry (MS). This modified substrate w

Chromatographic analysis of bromhexine and oxytetracycline residues in milk as a drug analysis medium with greenness profile appraisal.

Tetracyclines are frequently employed in animal husbandry. Bromhexine is a mucolytic drug that improves the efficacy of tetracyclines. It has been reported that residues of tetracyclines in milk may have negative effects on humans. Two versatile and accurate methods were developed for concurrent analysis of oxytetracycline (OTC) and bromhexine (BR) residues in spiked milk samples. Sample preparation was carefully considered for extraction and cleanup using the ecofriendly chemicals acetic acid, 0.1 N EDTA and ethanol or methanol. The first method was a TLC-densitometric method in which TLC pla

Development of in vitro antimicrobial resistance in bacteria exposed to residue level exposures of antimicrobial drugs, pesticides and veterinary drugs.

Ten ppb or less of antibacterial drugs, pesticides and veterinary drugs could increase antibacterial resistance in bacteria. The minimum inhibitory concentration (MIC) was the indicator of resistance and Staphylococcus aureus ATCC 9144 the indicator organism. Seventeen compounds used in human, veterinary medicine, crop production, and found in the environment, were studied singly, and as combinations. Single compounds decreased MIC values in 5.3% of possibilities; showed no changes in 74.4% of values; increased values in 21.8%; 16.5% of MICs increased 4- to 8-fold; 4.7% of changes were >8-fold

Deciphering Transporter-Mediated Drug Interactions Affecting Marbofloxacin Residues in Sheep Milk: From In Silico Predictions to In Vivo Validation.

Veterinary drug residues in milk represent an important food safety concern, particularly under conditions of co-administration where transporter-mediated drug-drug interactions may alter residue profiles. Marbofloxacin, a fluoroquinolone antimicrobial, is actively secreted into milk through the ATP-binding cassette transporter ABCG2 (BCRP), whereas ivermectin has been reported to interact with ABC transporters. The present study integrated in silico and in vivo approaches to investigate whether ivermectin may influence marbofloxacin secretion into sheep milk through transporter-mediated mecha

Monitoring of veterinary drug residues in mutton based on hyperspectral combined with explainable AI: A case study of OFX.

Veterinary drug residues in meat seriously harm human health. Rapid and accurate detection of veterinary drug residues is necessary to minimize contamination. Taking ofloxacin (OFX) residues in mutton as an example, the near-infrared hyperspectral imaging combined with explainable AI was used to evaluate the importance of feature wavelengths in the convolutional neural network-stacked sparse auto-encoder (CNN-SSAE) model for chemical properties. Based on this, the qualitative (residue identification-residue level identification) and quantitative detection of OFX residues in mutton was realized

Unveiling of phosphodiesterase-5 hot residues binding to xanthine derivatives for erectile dysfunction therapy: A computational drug repurposing approach.

Overexpression of phosphodiesterase 5 (PDE-5) presents a compelling target for the therapy of erectile dysfunction. Sildenafil and other conventional PDE-5 inhibitors may lead to adverse effects, including visual disturbances and migraines. Therefore, the investigation of novel inhibitors with enhanced safety profiles is imperative. This research employed a computational drug repurposing approach to assess US-FDA-approved xanthine derivatives (XDs) for their efficacy in targeting PDE-5. XDs exhibit a favorable affinity for the active site of the PDE-5 receptor, with binding scores between -10.
