E2F1 is a novel fibrogenic gene that regulates cholestatic liver fibrosis through the Egr-1/SHP/EID1 network.

UNLABELLED: E2F transcription factor 1 (E2F1) is an important regulator of metabolic diseases; however, its role in liver function remains elusive. This study unraveled a regulatory cascade involving E2F1, early growth response-1 (Egr-1), nuclear receptor small heterodimer partner (SHP, NR0B2), and EIA-like inhibitor of differentiation 1 (EID1) in cholestatic liver fibrosis. Liver E2F1 messenger RNA (mRNA) and protein expression was strongly up-regulated in human nonalcoholic steatohepatitis (NASH) and alcohol cirrhosis; the latter was inversely correlated with diminished SHP expression. E2F1

Phosphorylation at Ser10 triggered p27 degradation and promoted gallbladder carcinoma cell migration and invasion by regulating stathmin1 under glucose deficiency.

Gallbladder carcinoma (GBC) is a considerable challenge because of its high metastatic potential. The tumor microenvironment is characterized by nutrient starvation, which promotes tumor metastasis. Stathmin1, an important microtubuleregulating protein, is overexpressed and promotes metastasis in GBC. It remains unclear how the harsh tumor microenvironment regulates stathmin1 expression to affect GBC metastasis. We employed glucose deficiency to mimic nutrient starvation and found that glucose deficiency upregulated stathmin1 transcription. However, glucose deficiency also promoted p27 degrada

Long noncoding RNA CASC11 promotes hepatocarcinogenesis and HCC progression through EIF4A3-mediated E2F1 activation.

BACKGROUND: Growing evidences have been revealing that long noncoding RNAs are vital factors in oncogenesis and tumor development. Among them, cancer susceptibility candidate 11 (CASC11) has displayed an impressively essential role in various kinds of cancers including hepatocellular carcinoma (HCC). Nevertheless, its role and potential mechanism in HCC still remain to be fully investigated. METHODS: CASC11 expression level was evaluated by real-time polymerase chain reaction, western blotting, and in situ hybridization staining in HCC patients, and its prognostic effect was analyzed. The rol

USP5-Laden ApoEVs and E2F1 Stabilization in Disc Repair: Unresolved Questions on Senescence, Specificity and Endosomal Escape.

The seminal work by Shi et al. unveils a novel cytoprotective mechanism in intervertebral disc degeneration, whereby USP5-rich apoptotic extracellular vesicles (ApoEVs) stabilize the transcription factor E2F1 to suppress apoptosis and enhance DNA repair in nucleus pulposus cells. While this study elegantly reframes the reparative potential of ApoEVs, its conceptual and translational premises invite critical scrutiny. This letter highlights several overlooked challenges. We argue that the contextual duality of E2F1-a well-established regulator of both cell survival and senescence-poses a consid

RBM15 affects mitophagy in intracerebral hemorrhage by promoting E2F1 nuclear translocation via NXF1.

Nuclear mRNA transport is essential for maintaining mRNA homeostasis and normal cellular functions; however, the mechanisms regulating the nuclear transport of E2F transcription factor 1 (E2F1) mRNA remain poorly understood. In this study, we show that the nuclear RNA transport factor 1 (NXF1) is upregulated in Hemin-treated HT22 cells and intracerebral hemorrhage (ICH) mice, suggesting its involvement in ICH pathogenesis. NXF1 knockdown exerted a protective effect against Hemin-induced inflammatory responses in HT22 cells. Consistently, NXF1 downregulation ameliorated neurological deficits, r

Current concepts of the crosstalk between lncRNA and E2F1: shedding light on the cancer therapy.

Long noncoding RNAs (lncRNAs) constitute a distinctive subset of RNA molecules with limited protein-coding potential, which exert crucial impacts on various biological activities. In the context of cancer, dysregulated lncRNAs function as essential regulators that affect tumor initiation and malignant progression. These lncRNAs serve as competitive endogenous RNAs (ceRNAs) through sponging microRNAs and regulating the expression of targeted genes. Moreover, they also directly bind to RNA-binding proteins, which can be integrated into a complex mechanistic network. E2F1, an extensively studied

Exploring the Research Progress of Vascular Dementia and Key Regulatory Molecules: E2F1.

Vascular dementia (VaD) is a type of dementia caused by cerebrovascular factors, which can arise from both ischemic cerebrovascular disease and hemorrhagic cerebrovascular disease. The incidence rate of VaD is second only to Alzheimer's disease and dementia with Lewy bodies. Currently, there are no effective drugs specifically targeting VaD, making the discovery of new therapeutic targets of great significance. This article provides an overview of the research progress on VaD, with a focus on elucidating its pathogenesis, aiming to identify targets that play a regulatory role in the mechanism.

Nuclear Import of E2F1 Regulated by KPNB1 is Essential for Proliferation and Invasion of Cervical Cancer Cells.

As a nuclear transport receptor, KPNB1 transport proteins from cytoplasm into nucleus, performing a vital function in intracellular traffic. The aberrant upregulation of KPNB1 has been associated with various malignancies and transformed cells, where it is indispensable for cellular proliferation and survival. Transcription factors like E2F1 possess a nuclear localization motif that triggers the expression of a range of crucial factors during tumorigenesis. This investigation explores the influence of KPNB1 in cervical cancer (CC) cell metastasis and proliferation through the modulation of E2F

Co-inhibition of ATM and ROCK synergistically improves cell proliferation in replicative senescence by activating FOXM1 and E2F1.

The multifaceted nature of senescent cell cycle arrest necessitates the targeting of multiple factors arresting or promoting the cell cycle. We report that co-inhibition of ATM and ROCK by KU-60019 and Y-27632, respectively, synergistically increases the proliferation of human diploid fibroblasts undergoing replicative senescence through activation of the transcription factors E2F1 and FOXM1. Time-course transcriptome analysis identified FOXM1 and E2F1 as crucial factors promoting proliferation. Co-inhibition of the kinases ATM and ROCK first promotes the G2/M transition via FOXM1 activation,

Identification of key genes in the liver of offspring from obesity maternal mice by bioinformatics analysis.

Murine maternal obesity predisposes offspring to obesity and other non-communicable diseases. Fetal programming enables researchers to trace this detrimental effect in the early life of the offspring. The aim of the current study was to explore the molecular impact of maternal obesity on the livers of murine offspring at weaning age. C57BL/6 female mice were exposed to a high-fat diet to induce obesity, after which they were mated to produce offspring. At weaning age, the metabolic health of female offspring was assessed and hepatic mRNAs were investigated via mRNA high throughput sequencing.

The downregulation of the RNA-binding protein Staufen2 in response to DNA damage promotes apoptosis.

Staufen2 (Stau2) is an RNA-binding protein involved in cell fate decision by controlling several facets of mRNA processing including localization, splicing, translation and stability. Herein we report that exposure to DNA-damaging agents that generate replicative stress such as camptothecin (CPT), 5-fluoro-uracil (5FU) and ultraviolet radiation (UVC) causes downregulation of Stau2 in HCT116 colorectal cancer cells. In contrast, other agents such as doxorubicin and ionizing radiation had no effect on Stau2 expression. Consistently, Stau2 expression is regulated by the ataxia telangiectasia and

Hypoxia-Sensitive COMMD1 Integrates Signaling and Cellular Metabolism in Human Macrophages and Suppresses Osteoclastogenesis.

Hypoxia augments inflammatory responses and osteoclastogenesis by incompletely understood mechanisms. We identified COMMD1 as a cell-intrinsic negative regulator of osteoclastogenesis that is suppressed by hypoxia. In human macrophages, COMMD1 restrained induction of NF-κB signaling and a transcription factor E2F1-dependent metabolic pathway by the cytokine RANKL. Downregulation of COMMD1 protein expression by hypoxia augmented RANKL-induced expression of inflammatory and E2F1 target genes and downstream osteoclastogenesis. E2F1 targets included glycolysis and metabolic genes including CKB tha

An investigation of targeted inhibition of transcription factor activity with pyrrole imidazole polyamide (PA) in chronic myeloid leukemia (CML) blast crisis cells.

Tyrosine kinase inhibitor (TKI) therapy is the standard treatment for chronic phase (CP)-chronic myeloid leukemia (CML), yet patients in blast crisis (BC) phase of CML are unlikely to respond to TKI therapy. The transcription factor E2F1 is a down-stream target of the tyrosine kinase BCR-ABL1 and is up-regulated in TKI-resistant leukemia stem cells (LSC). Pyrrole imidazole polyamides (PA) are minor groove binders which can be programmed to target DNA sequences in a gene-selective manner. This manuscript describes such an approach with a PA designed to down-regulate E2F1 controlled gene express

E2F1 copy number variations in germline and breast cancer: a retrospective study of 222 Italian women.

BACKGROUND: Breast cancer is the most common neoplasia among women in developed countries. The risk factors of breast cancer can be distinguished in modifiable and unmodifiable factors and, among the latter, genetic factors play a key role. Copy number variations (CNVs) are genetic variants that are classified as rare when present in less than 1% of the healthy population. Since rare CNVs are often cause of diseases, over the last years, their contribution in carcinogenesis has become a relevant matter of study. E2F1 is a transcriptional factor that plays an important role in regulating cell c

E2F1-mediated transcriptional regulation of TMEM33 promotes colorectal cancer proliferation, metastasis, and EMT.

Colorectal cancer (CRC) remains one of the most lethal malignancies worldwide, with tumor metastasis representing the primary driver of treatment failure and poor prognosis. Transmembrane Protein 33 (TMEM33) has been implicated in tumor progression across several cancer types, yet its expression pattern, biological function, and upstream transcriptional regulation in CRC remain undefined. In this study, TMEM33 expression in CRC and matched adjacent normal tissues was assessed by quantitative reverse transcription polymerase chain reaction (qRT-PCR), Western blot, and immunohistochemistry (IHC)

Transcriptional regulation of fibrillin-2 gene by E2F family members in chondrocyte differentiation.

Mutation in fibrillin-2, a major structural component of extracellular microfibrils in connective tissue, results in the autosomal dominant disease congenital contractural arachnodactyly. This genetic disease is characterized by dolichostenomelia and arachnodactyly, in addition to contractures of the large joints and abnormal pinnae formation, thus indicating the significance of fibrillin-2 in chondrogenesis. In this study, we investigated the transcriptional regulation of fibrillin-2 in chondrogenic differentiation. Although mRNA expression of fibrillin-1, a highly homologous protein to fibri

Genome-wide analysis of the oxyntic proliferative isthmus zone reveals ASPM as a possible gastric stem/progenitor cell marker over-expressed in cancer.

The oxyntic proliferative isthmus zone contains the main stem/progenitor cells that provide for physiological renewal of the distinct mature cell lineages in the oxyntic epithelium of the stomach. These cells are also proposed to be the potential cells-of-origin of gastric cancer, although little is known about their molecular characteristics and specific biological markers are lacking. In this study, we developed a method for serial section-navigated laser microdissection to isolate cells from the proliferative isthmus zone of rat gastric oxyntic mucosa for genome-wide microarray gene express

In human salivary gland cells, overexpression of E2F1 overcomes an interferon-gamma- and tumor necrosis factor-alpha-induced growth arrest but does not result in complete mitosis.

Increased levels of cytokines in the salivary glands have been associated with the loss of secretory cells and reduced salivary function. It has been demonstrated that interferon-gamma (IFN-gamma) and tumor necrosis factor-alpha (TNF-alpha) treatment of a human submandibular gland (HSG) cell line causes growth arrest in the G0/G1 phase of the cell cycle, followed by apoptosis. To stimulate DNA synthesis and reverse this growth arrest, we used an adenovirus vector to overexpress the transcription factor E2F1 in HSG cells. Initially, cells were synchronized by a double thymidine block and then i
