Fos-related antigens (Fra) were detected in the nuclei of neurones in young adult Acheta domesticus female crickets by immunohistochemical analysis, using an antibody that recognizes the amino-acid sequence 127-152 of c-Fos protein. Specificity of Fra immunoreactivity was confirmed by Western blot analysis of nuclear extracts from neural tissues. A major immunoreactive doublet with an apparent molecular mass of 52,000/54,000 Da was detected in nuclear extracts. Immunostaining of the 52,000/54,000 Da doublet showed variations in intensity during the first 5 days following the imaginal molt. Sta

Metamorphosis in insects includes a series of programmed tissue histolysis and remolding processes that are controlled by two major classes of hormones, juvenile hormones and ecdysteroids. Precise pulses of ecdysteroids (the most active ecdysteroid is 20-hydroxyecdysone, 20E), are regulated by both biosynthesis and metabolism. In this study, we show that ecdysone oxidase (EO), a 20E inactivation enzyme, expresses predominantly in the midgut during the early pupal stage in the lepidopteran model insect, Bombyx mori. Depletion of BmEO using the transgenic CRISPR/Cas9 (clustered regularly intersp

A comprehensive metabolite profiling of the medicinal plant Silene viridiflora using an UHPLC-ESI-MS/MS method is described for the first time. A total of 71 compounds were identified and annotated, the most common of which were flavonoids, triterpene glycosides, and ecdysteroids. The three major compounds schaftoside, 26-hydroxyecdysone, and silviridoside can be chosen as the markers for the assessment of the quality of S. viridiflora preparations. The methanol extract and a variety of metabolites identified in S. viridiflora were screened for their cytotoxic and Wnt pathway-inhibiting activi

Mutations in either of the two human Niemann-Pick type C (NPC) genes, NPC1 and NPC2, cause a fatal neurodegenerative disease associated with abnormal cholesterol accumulation in cells. npc1a, the Drosophila NPC1 ortholog, regulates sterol homeostasis and is essential for molting hormone (20-hydroxyecdysone; 20E) biosynthesis. While only one npc2 gene is present in yeast, worm, mouse and human genomes, a family of eight npc2 genes (npc2a-h) exists in Drosophila. Among the encoded proteins, Npc2a has the broadest expression pattern and is most similar in sequence to vertebrate Npc2. Mutation of

Paired Y-organs secrete ecdysteroid hormones that control cycles of growth and molting in crustaceans. Y-Organs are regulated, at least in part, by molt-inhibiting hormone (MIH), a polypeptide produced and released by the X-organ/sinus gland complex of the eyestalks. In the present studies, crab (Callinectes sapidus) Y-organs were incubated in vitro in the presence of [(35)S]methionine, and cyclic nucleotide analogs or experimental agents that influence the cAMP signaling pathway. In 4-hr incubations, 8-Br-cAMP and db-cAMP (but not 8-Br-cGMP) suppressed incorporation of [(35)S]methionine into

BACKGROUND: Filarial nematodes, including Brugia malayi, the causative agent of lymphatic filariasis, undergo molting in both arthropod and mammalian hosts to complete their life cycles. An understanding of how these parasites cross developmental checkpoints may reveal potential targets for intervention. Pharmacological evidence suggests that ecdysteroids play a role in parasitic nematode molting and fertility although their specific function remains unknown. In insects, ecdysone triggers molting through the activation of the ecdysone receptor: a heterodimer of EcR (ecdysone receptor) and USP

The induction of vitellogenin in oviparous vertebrates has become the gold standard biomarker of exposure to estrogenic chemicals in the environment. This biomarker of estrogen exposure also has been used in arthropods, however, little is known of the factors that regulate the expression of vitellogenin in these organisms. We investigated changes in accumulation of mRNA products of the vitellogenin gene Vtg2 in daphnids (Daphnia magna) exposed to a diverse array of chemicals. We further evaluated the involvement of hormonal factors in the regulation of vitellogenin expression that may be targe

BACKGROUND: The brown planthopper (BPH), Nilaparvata lugens, is a destructive rice pest across Asia. Suppressing insect development is enduring goals in insect pest management, underpinning the action of existing insecticides and guiding the design of new ones. Insect molting, which is tightly regulated by ecdysteroid signaling, is essential for insect growth and development. Cytochrome P450 (CYP) enzymes play critical roles in ecdysteroid biosynthesis, and identifying P450 genes involved in this process may provide new insights into insect developmental regulation and potential targets for pe

Changes in ecdysteroid levels that lead to commitment of pupal and adult development were studied in the yellow fever mosquito, Aedes aegypti. Application of juvenile hormone at the time of pupal commitment usually results in delay or blockage of pupal and adult development. With methoprene, a juvenile hormone mimic, the JH sensitive period was found to be within 19 h after ecdysis to the fourth instar, at which time methoprene treatment caused a delay in pupation. Consistent with this observation, there was a small peak of ecdysteroid levels between 14 and 28 h after ecdysis to the fourth ins

The Y-organs (YOs) synthesize and secrete molting hormones, and thus regulate crustacean growth and reproduction. However, the YOs of the orange mud crab Scylla olivacea is yet to be described due to its minute size and ambiguous feature. This study describes the location, morphological characteristics, histology, and the changes of YOs at different molt stages, and examines in vitro ecdysteroid secreted by the YOs of S. olivacea. Mature male crabs (> 90 mm carapace width, CW) were used to identify the location of the YOs while male juveniles (50-65 mm CW) at three molt stages (intermoult, pre

To further understand the function of morphogenetic hormones in honeybee eye differentiation, the alterations in ommatidial patterning induced by pyriproxyfen, a juvenile hormone (JH) analogue, were studied by scanning and transmission electron microscopy. Prepupae of prospective honeybee workers were treated with pyriproxyfen and the effects on ommatidial differentiation were described at the end of the pupal development. The results show that the entire ommatidia, i.e., the dioptric as well as the receptor systems, were affected by the JH analogue. The wave of ommatidial differentiation, whi

Wolbachia pipientis is a widespread endosymbiont of filarial nematodes and arthropods. While in worms the symbiosis is obligate, in arthropods Wolbachia induces several reproductive manipulations (i.e., cytoplasmic incompatibility, parthenogenesis, feminization of genetic males, and male-killing) in order to increase the number of infected females. These various phenotypic effects may be linked to differences in host physiology, and in particular to endocrine-related processes governing growth, development, and reproduction. Indeed, a number of evidences links Wolbachia symbiosis to insulin an

BACKGROUND: Searching for new effective drugs against human and animal toxoplasmosis we decided to test the anti-Toxoplasma potential of phytoecdysteroids (α-ecdysone and 20-hydroxyecdysone) characterized by the pleiotropic activity on mammalian organisms including the enhancement of host's anti-parasitic defence. This objective was accomplished by the in vitro evaluation of T. gondii growth in phytoecdysteroid-treated immunocompetent cells of selected hosts: humans and two strains of inbred mice with genetically determined different susceptibility to toxoplasmosis.

Ecdysteroids secreted by the prothoracic gland (PG) cells of insects control the developmental timing of their immature life stages. These cells have been historically considered as carrying out a single function in insects, namely the biochemical conversion of cholesterol to ecdysteroids and their secretion. A growing body of evidence shows that PG cells receive multiple cues during insect development so we tested the hypothesis that they carry out more than just one function in insects. We characterised the molecular nature and developmental profiles of cell membrane receptors in PG cells of

For many insects, the ecdysteroid biosynthetic pathway that converts cholesterol to ecdysone includes multiple cytochrome P450 monooxygenases, a Rieske oxygenase, and a short-chain dehydrogenase. Collectively, the genes encoding these enzymes are referred to as Halloween genes. While the importance of these enzymes in oogenesis is well-documented in holometabolous insects, less is known about their oogenic role in hemipterans. To address this, we used RNA interference-mediated knockdown to determine the role that the Halloween transcripts neverland, spookiest, shroud, phantom, disembodied, sha

The transition from larval to pupal stage in holometabolous insects is a critical developmental decision regulated by hormonal and nutritional cues. The studies reported here revealed a central role for the TGF-β ligand Myoglianin (Myo) in integrating these signals in a major lepidopteran pest, the fall armyworm, Spodoptera frugiperda. We found that CRISPR/Cas9-mediated Myo knockout (KO) resulted in supernumerary larval molts and an impairment of the larval-pupal transition. Myo-KO larvae exhibited a disrupted hormone milieu, characterized by elevated juvenile hormone (JH) levels and reduced e

The function of two autophagy genes, an activating molecule BECN1 regulated autophagy (AMBRA1) and autophagy-related gene 8 (ATG8) in the midgut remodeling of Aedes aegypti was investigated. Real-time quantitative polymerase chain reaction (RT-qPCR) analysis of RNA samples collected from the last instar larvae and pupae showed that these two genes are predominantly expressed during the last 12 h and first 24 h of the last larval and pupal stages, respectively. Stable ecdysteroid analog induced and juvenile hormone (JH) analog suppressed these genes. RNA interference (RNAi) studies showed that

In modern life, the use of plant stress-protectors has taken on particular significance due to the wide distribution of neurosis-like and neurotic diseases caused by neuroendocrine-immune system imbalance. Special attention has been paid to the plants containing ecdysteroids, i.e., hormone-like bioactive substances with high adaptogenic activity. The article deals with the study of bioactivity of two plant extracts as Rhaponticum uniflorum (L.) DC. and Serratula centauroides L. with a high content of ecdysteroids and phenolic compounds. The models of acute and chronic emotional stress in white

Metamorphosis in class Insecta is defined as the drastic transition of an organism from a larval stage to a morphologically very different adult form. Insects such as Drosophila melanogaster undergo metamorphosis, which requires precise temporal control, exerted by ecdysteroids, the primary regulators of these developmental transitions. The active form of ecdysone is 20-hydroxyecdysone (20E), which acts in pulses that trigger the onset of different developmental stages. A massive pulse of 20E occurs between the prepupal and pupal stages, triggering pupariation. Precise quantification of 20E le

Wolbachia pipientis is known to infect only arthropods and nematodes (mainly filarial worms). A unique feature shared by the two Phyla is the ability to replace the exoskeleton, a process known as ecdysis. This shared characteristic is thought to reflect a common ancestry. Arthropod moulting is induced by the steroid hormone 20-hydroxyecdysone (20E) and a role for ecdysteroids in nematode ecdysis has also been suggested. Removing Wolbachia from filarial worms impairs the host's development. From analyses of the genome of Wolbachia harbored by the filarial nematode Brugia malayi and that of its
