A novel cytochrome P450 gene, CYP12A2, regulates reproduction of Mythimna separata by modulating 20E biosynthesis.

BACKGROUND: Several P450 genes in the 20-hydroxyecdysone (20E) signaling pathway are known to critically regulate reproductive development in insects. However, the contribution of other P450 family members to insect reproduction remains poorly characterized. RESULTS: Here, we identified a novel P450 gene, CYP12A2, from the highly polyphagous pest Mythimna separata. Quantitative real-time polymerase chain reaction analysis revealed that CYP12A2 was upregulated during sexual maturation and predominantly enriched in the ovaries of 3-day-old female adults. RNA interference (RNAi)-mediated knockdo

Nutritional Resources Regulate the Reproduction or Migration of Spodoptera frugiperda through Juvenile Hormones and 20-Hydroxyecdysone.

Spodoptera frugiperda is a significant agricultural pest that migrates annually based on the growth stages of its host plants. However, the role of host plant growth stages in the residency or migration of S. frugiperda remains unclear. We allowed S. frugiperda to feed on maize leaves and seeds, and results showed that maize seeds had higher amino acids, carbohydrates, and triglyceride (TG). S. frugiperda that fed on seeds had longer adult stages; larger wings; higher TG, juvenile hormone II (JH II), and JH III; and more developed flight muscles, indicating tendencies toward migration. Developmental regulation of Dirofilaria immitis microfilariae and evaluation of ecdysone signaling pathway transcript level using droplet digital PCR.

BACKGROUND: Current measures for the prevention of dirofilariasis, caused by the dog heartworm, Dirofilaria immitis, rely on macrocyclic lactones, but evidence of drug-resistant isolates has called for alternative approaches to disease intervention. As microfilariae are known to be in a state of developmental arrest in their mammalian host and then undergo two molts once inside the arthropod, the aim of this study was to look at the developmental regulation of D. immitis microfilariae that occurs in their arthropod host using in vitro approaches and to investigate the role of the ecdysone sign

Evaluation of ecdysteroid antisera for a competitive enzyme immunoassay and extraction procedures for the measurement of mosquito ecdysteroids.

Ecdysteroid hormones regulate several aspects of insect development and reproduction. The predominant ecdysteroids produced by insects including mosquitoes are ecdysone (E) and 20-hydroxyecdysone (20E). The ability to measure E and 20E titers is essential for many studies, but few sensitive, low cost options are currently available for doing so. To address this deficiency, we developed a new enzyme-linked immunoassay (EIA). In the first part of the study, we compared the affinity of two new antisera named EAB25 and EAB27 to other available ecdysteroid antisera. EAB25 had a 27-fold higher affin

Changes in the Gene Expression Profiles of the Hypopharyngeal Gland of Worker Honeybees in Association with Worker Behavior and Hormonal Factors.

The hypopharyngeal glands (HPGs) of worker honeybees undergo physiological changes along with the age-dependent role change from nursing to foraging: nurse bee HPGs secrete mainly major royal jelly proteins, whereas forager HPGs secrete mainly α-glucosidase III, which converts the sucrose in the nectar into glucose and fructose. We previously identified two other genes, Apis mellifera buffy (Ambuffy) and Apis mellifera matrix metalloproteinase 1 (AmMMP1), with enriched expression in nurse bee and forager HPGs, respectively. In the present study, to clarify the molecular mechanisms that coordin

Control of follicular epithelium development and vitelline envelope formation in the mosquito; role of juvenile hormone and 20-hydroxyecdysone.

Using microsurgical manipulations, hormone applications, and transmission electron microscopy we have investigated the regulation of differentiation of the follicular epithelium and formation of the vitelline envelope (VE) in primary follicles in the ovary of the mosquito, Aedes aegypti. During the first 3 days after eclosion, the primary follicle grows, and cells of the follicular epithelium differentiate, their content of mitochondria, rough endoplasmic reticulum, and Golgi complexes increases significantly. Growth and differentiation of the follicular epithelium appear to be under the contr

Lipases are differentially regulated by hormones to maintain free fatty acid homeostasis for insect brain development.

BACKGROUND: Free fatty acids (FFAs) play vital roles as energy sources and substrates in organisms; however, the molecular mechanism regulating the homeostasis of FFA levels in various circumstances, such as feeding and nonfeeding stages, is not fully clarified. Holometabolous insects digest dietary triglycerides (TAGs) during larval feeding stages and degrade stored TAGs in the fat body during metamorphosis after feeding cessation, which presents a suitable model for this study. RESULTS: This study reported that two lipases are differentially regulated by hormones to maintain the homeostasis

Effect of sublethal doses of cadmium, inorganic mercury and methylmercury on the cell morphology of an insect cell line (Aedes albopictus, C6/36).

The effect of CdCl2 (44 microM), HgCl2 (3.7 microM), and MeHgCl (2 microM) on the morphology of Aedes albopictus C6/36 cells was studied at the light microscopical level. Treatment times and metal concentrations were in the sublethal range as determined by a fluorometric dye exclusion test. The three metal species had profound effects on the cell morphology. MeHgCl treatment induced the development of a large number of short, actin-supported, tangled filopodia. Both CdCl2 and HgCl2 induced long extensions. Pretreatment with colchicine but not with cytochalasin B prevented formation of these ex

Ecdysteroid-like compounds in the serum and urine of African patients infected with Loa loa and Mansonella perstans microfilariae.

Ecdysteroids are compounds related to 20-hydroxyecdysone, the insect moulting hormone. Surprisingly, they have been found in serum and urine of patients infected with helminths. In these cases, the substances are assumed to be produced by the parasites and, therefore, might be used as a marker of parasitic infection. Thus, we need to know exactly which species, at which developmental stage, can release ecdysteroids in such large quantities that they could be detected in the biological fluids of the host. Large-scale investigations must, accordingly, be devoted to the major species of helminths

Tinosporide promotes melanogenesis via activation of cAMP/PKA/CREB-MITF signaling in B16F10 and SK-MEL-2 cells.

Hypopigmentation disorders are characterized by impaired melanogenesis and remain challenging to manage due to limited efficacy of current interventions and insufficient mechanistic insights. Natural products represent a valuable source of bioactive phytoconstituents capable of regulating key pathways involved in melanin synthesis. Despite its extensive traditional use and diverse pharmacological activities, Tinospora cordifolia and its major bioactive constituent tinosporide remain unexplored for their role in melanogenesis. The present study aimed to evaluate the melanogenesis-promoting pote

SOX12 and DELTA mediate ecdysone-driven adult neural cell proliferation during metamorphosis.

During metamorphic development from larva to adult, insect brains exhibit cell proliferation; however, the molecular mechanisms governing this process remain poorly understood. Employing the lepidopteran insect Helicoverpa armigera (cotton bollworm), an agricultural pest, as a model system, this study demonstrates that the steroid hormone 20-hydroxyecdysone (20E) regulates neural cell proliferation during imaginal brain development by upregulating the expression of SOX12 and DELTA via nuclear receptor EcRA. SOX12 exhibits significantly elevated expression and is localized in the brain during m

Regulation and dysregulation of vitellogenin mRNA accumulation in daphnids (Daphnia magna).

The induction of vitellogenin in oviparous vertebrates has become the gold standard biomarker of exposure to estrogenic chemicals in the environment. This biomarker of estrogen exposure also has been used in arthropods, however, little is known of the factors that regulate the expression of vitellogenin in these organisms. We investigated changes in accumulation of mRNA products of the vitellogenin gene Vtg2 in daphnids (Daphnia magna) exposed to a diverse array of chemicals. We further evaluated the involvement of hormonal factors in the regulation of vitellogenin expression that may be targe

Homeostatic and developmental control of cell size and shape in an insect epithelium, the epidermis of Manduca sexta.

In contrast to previously described transformations of insect epidermal cells, morphological changes in the larval epidermis of Manduca sexta involve large changes in cell volume: a threefold increase at the beginning and a twofold decrease at the end of the last feeding phase. The volume changes are determined, in part, or entirely, by changes in cell height (cell elongation and cell shortening). Initial cell elongation occurs in a region-specific manner, whereas subsequent cell shortening affects all of the epidermis equally. As shown by ligation experiments and hormone treatments in tissue

20-hydroxyecdysone (20E) signaling regulates amnioserosa morphogenesis during Drosophila dorsal closure: EcR modulates gene expression in a complex with the AP-1 subunit, Jun.

BACKGROUND: Engaging young people (10-24 years old) in research has been shown to improve the uptake and impact of HIV interventions in youth. However, how youth are engaged in Youth Advisory Boards (YABs) in low- and middle-income countries (LMICs) remains under-explored. Therefore, we conducted a scoping review and reflected on YAB members' experiences to examine YABs in youth HIV research. METHODS: We conducted this study using a two-phase process. In phase one, we conducted a scoping review searching seven literature databases for peer-reviewed articles published in English between 2014 a

Molecular action of pyriproxyfen: Role of the Methoprene-tolerant protein in the pyriproxyfen-induced sterilization of adult female mosquitoes.

Exposure of adult mosquitoes to pyriproxyfen (PPF), an analog of insect juvenile hormone (JH), has shown promise to effectively sterilize female mosquitoes. However, the underlying mechanisms of the PPF-induced decrease in mosquito fecundity are largely unknown. We performed a comprehensive study to dissect the mode of PPF action in Aedes aegypti mosquitoes. Exposure to PPF prompted the overgrowth of primary follicles in sugar-fed Ae. aegypti females but blocked the development of primary follicles at Christopher's Stage III after blood feeding. Secondary follicles were precociously activated

β-ecdysone promotes osteogenic differentiation of bone marrow mesenchymal stem cells.

BACKGROUND: β-ecdysone (βEcd) has numerous pharmacological effects, although its role in the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) has not yet been explored. METHODS: In cell experiments, BMSCs were induced to differentiate by osteogenic induction medium (OIM) or βEcd. In animal experiments, an osteonecrosis of the femoral head (ONFH) rat model was established using lipopolysaccharide plus methylprednisolone and treating the rats with βEcd. Postembryonic hematopoiesis in Drosophila.

We have investigated the blood cell types present in Drosophila at postembryonic stages and have analysed their modifications during development and under immune conditions. The anterior lobes of the larval hematopoietic organ or lymph gland contain numerous active secretory cells, plasmatocytes, few crystal cells, and a number of undifferentiated prohemocytes. The posterior lobes contain essentially prohemocytes. The blood cell population in larval hemolymph differs and consists mainly of plasmatocytes which are phagocytes, and of a low percentage of crystal cells which reportedly play a role

High PPC impairs female reproduction by inhibiting the 20E-Vg/VgR pathway.

Hyperproteinemia is characterized by an unusually high plasma protein concentration (PPC). It affects both humans and animals. In this study, we used a silkworm model of hyperproteinemia to explore the mechanisms by which high PPC impairs female reproduction. Analysis of ovarian transcriptomes and qRT-PCR revealed that high PPC reduced the expression of vitellogenin (Vg) in the fat body and vitellogenin receptor (VgR) in the ovary. Further biochemical and gene expression analyses showed that high PPC decreased the titer of 20-hydroxyecdysone (20E) and the expression of key genes in the 20E-Vg/
