Echinococcus multilocularis: mouse strain difference in hydatid development.

Female mice of nine inbred strains (A, AKR, BALB/c, C3H, C57BL/6, C57BL/10, CBA, DBA/1 and DBA/2) and H-2 congenic B10.D2, at 9-10 weeks of age, were infected with larval Echinococcus multilocularis by trans-portal injection of hydatid homogenate. Parasitized livers were histologically examined 9 or 13 weeks after infection. Hydatid development was quite different among mouse strains. Multivesiculation was prominent in C57BL/10, DBA/1, C57BL/6 and BALB/c. Protoscoleces were well developed in DBA/2, AKR, DBA/1 and CBA. H-2 congenic B10.D2, which has the background genes of C57BL/10 except for t

Dual inhibition of the Echinococcus multilocularis energy metabolism.

Alveolar echinococcosis is caused by the metacestode stage of the zoonotic parasite Echinococcus multilocularis. Current chemotherapeutic treatment options rely on benzimidazoles, which have limited curative capabilities and can cause severe side effects. Thus, novel treatment options are urgently needed. In search for novel targetable pathways we focused on the mitochondrial energy metabolism of E. multilocularis. The parasite relies hereby on two pathways: The classical oxidative phosphorylation including the electron transfer chain (ETC), and the anaerobic malate dismutation (MD). We screen

First detection of Echinococcus multilocularis in Bosnia and Herzegovina.

Echinococcus multilocularis has been spreading through Central Eastern Europe but has not yet been reported in Bosnia and Herzegovina (B&H). Recently, this parasite is confirmed in Croatia suggesting the movement of the parasite's distribution limit further south. Given that there is no surveillance or monitoring system for echinococcosis in B&H, our study was designed as a pilot study of E. multilocularis. A total of 57 red foxes originating from 24 localities all over the country were collected during the routine rabies monitoring, autopsied and examined for the presence of echinococcosis. B

Limited fertility of the subcutaneous cysts of Echinococcus multilocularis.

Echinococcus multilocularis is a tiny devastating worm that causes alveolar echinococcosis in humans. This disease mainly occurs in the liver but rarely in other organs. We report the subcutaneous encystment of E. multilocularis metacestodes in experimentally infected mice. Subcutaneous cysts had remarkably fewer protoscoleces (2.05 ± 1.47, n = 20) and small irregular-shape vesicles (ISVs) in the lumen as compared to liver cysts (69.6 ± 55.65, n = 10). Moreover, abnormal development of a protoscolex was also observed in a subcutaneous cyst. The results suggest that subcutaneous encystment may

Surveillance and management of Echinococcus multilocularis in a wildlife park.

The fox tapeworm Echinococcus multilocularis is the causative agent of alveolar echinococcosis, a severe zoonotic disease that may be fatal if untreated. A broad spectrum of mammalian species may be accidentally infected even in captivity. In April 2011, liver lesions due to E. multilocularis were observed during the necropsy of a captive-born nutria (Myocastor coypus) in a French wildlife park, leading to initiation of a study to survey the parasite's presence in the park. A comparable environmental contamination with fox's feces infected by E. multilocularis was reported inside (17.8%) and o

Survival strategy of Echinococcus multilocularis in the human host.

As exemplified by "aborted" calcified liver lesions commonly found in patients from endemic areas, Echinococcus multilocularis metacestodes develop only in a minority of individuals exposed to infection with the papasite. Clinical research has disclosed some aspects of the survival strategy of E. multilocularis in human hosts. Clinical observations in liver transplantation and AIDS suggest that suppression of cellular/Th1-related immunity increases disease severity. Most of the studies have stressed a role for CD8+ T cells and for Interleukin-10 in the development of tolerance. A spontaneous s

Diagnosis of the Echinococcus multilocularis infection in final hosts.

In view of the considerable public health significance of Echinococcus multilocularis, the causative agent of the highly lethal human alveolar echinococcosis, there is an urgent need for reliable and simple techniques for the diagnosis of the infection in populations of final hosts (foxes, dogs, cats) and also in individual dogs and cats. The standard technique presently used is parsitological examination of the small intestine at necropsy. This reliable technique requires high expenditure and special safety precautions. An alternative approach is coproantigen detection. Recently, in our labor

Echinococcus multilocularis in a European beaver from Switzerland.

Infection with the larval stage of the cestode parasite Echinococcus multilocularis was diagnosed in a European beaver (Castor fiber) in central Switzerland. The animal was hit, run over by a car, and died of trauma. It was in normal body condition and no signs of disease were seen. At necropsy, multiple cystic structures up to 1 cm in diameter were found in the liver adjacent to the hilus. Within the parasite vesicles, multiple protoscolices were visible. The species was determined to be Echinococcus multilocularis by upon polymerase chain reaction and direct immunofluorescence with MAbG11-FI

Echinococcus multilocularis (Cestoda, Cyclophyllidea, Taeniidae): oncospheral hook morphogenesis.

Ultrastructural characteristics of the oncospheral hook morphogenesis in the taeniid cestode Echinococcus multilocularis Leuckart, 1863, a parasite of medical and veterinary importance, are described. Oncospheral hook primordia appear at the preoncospheral phase of the embryonic development. Within six specialised cells of the so-called oncoblasts, high concentration of mitochondria, numerous ribosomes and extended Golgi regions are involved in hook development. During hook growth, the blade and base gradually protrude outside the oncoblast plasma membrane. The nucleated oncoblast persists aro

Spinal cord compression due to costal Echinococcus multilocularis.

We present computerized tomography (CT) and magnetic resonance imaging (MRI) findings of a costal hydatid cyst (Echinococcus multilocularis) causing spinal cord compression. The hydatid disease was proved histologically. MRI was not only very useful for determining the spinal extension of the disease by its multiplanar imaging capability, but also gave important information about the texture of the cyst, thus aiding the preoperative diagnosis.

[Experimental imaginal Echinococcus multilocularis infestation of golden hamsters].

The imago stage of Echinococcus multilocularis development (prior to maturation of parasite oocysts) has been described in the intestine of golden hamsters infected by parenteral penetration of an Alveococcus agent and given hydrocortisone subcutaneously for 3 days before the infection and on the day of the infection, mean total drug dose being about 250 mg/kg. The number of helminths developed on days 8-35 of postinvasion ranged from 432 to 1863, their length was 1.0 to 4.0 mm and the number of segments did not exceed 2. Oocyst maturation was noted on day 20 of postinvasion. High oocyst invas

[Echinococcus multilocularis causing extensive biliary obstruction (author's transl)].

Infection of man by Echinococcus multilocularis occurs relatively seldom, most commonly localizing in the liver in the manner of a malignant tumour. A case of alveolar hydatid disease is reported presenting with extensive biliary obstruction, and a large cavitation in the liver due to necrosis. A review of epidemiology, clinical and roentgenological findings is given.

Differential activation of hepatic stellate cells by Echinococcus multilocularis and Echinococcus granulosus protoscoleces.

Echinococcosis, caused by Echinococcus multilocularis (Em) and Echinococcus granulosus (Eg), induces distinct patterns of liver fibrosis. Understanding how protoscoleces (PSCs) from these species interact with hepatic stellate cells (HSCs) is crucial to elucidating the mechanisms of fibrogenesis. In this study, HSC-LX2 cells were co-cultured with Em and Eg PSCs. ELISA was used to quantify collagen-I, α-smooth muscle actin (α-SMA), osteopontin (OPN), and TNF-α levels, while transmission electron microscopy (TEM) examined ultrastructural changes. Both E. multilocularis- and E. granulosus-derived

Macroscopic and Microscopic Aspects of Echinococcus multilocularis and Echinococcus granulosus in Humans: A Guideline for Pathologists.

BACKGROUND: Echinococcosis is a parasitic disease, caused by the larval stage of the genus Echinococcus. Echinococcus multilocularis and Echinococcus granulosus cause alveolar echinococcosis (AE) and cystic echinococcosis (CE) in humans. Pathologists may be confronted with the larval stage of both species. Diagnosis may be difficult due to the low incidence in daily routine. However, right diagnosis of the pathologist is still the gold standard and crucial for further treatment of the patient. SUMMARY: Macroscopically, specimens of AE show multiple small cysts and a bread-like appearance. Ins

Genetic diversity of Echinococcus multilocularis and Echinococcus granulosus sensu lato in Kyrgyzstan: The A2 haplotype of E. multilocularis is the predominant variant infecting humans.

Alveolar and cystic echinococcosis (AE, CE) caused by E. multilocularis and E. granulosus s.l., respectively, are considered emerging zoonotic diseases in Kyrgyzstan with some of the world highest regional incidences. Little is known regarding the molecular variability of both species in Kyrgyzstan. In this study we provide molecular data from a total of 72 parasite isolates derived from humans (52 AE and 20 CE patients) and 43 samples from dogs (23 infected with E. multilocularis and 20 with E. granulosus s.l.).Genetic variability in E. multilocularis was studied using the concatenated comple

Efficient delivery of Echinococcus multilocularis miRNAs using chitosan nanoparticles.

Alveolar echinococcosis caused by Echinococcus multilocularis is an important zoonotic disease, a great threat to human health due to limited interventions. microRNAs are a type of small non-coding RNA that plays a key role in many diseases and is considered as a potential therapeutic target for control of parasitic diseases. However, naked miRNAs are difficult to enter into cells and are easily degraded in both external and internal environments. Chitosan (CS) has recently been used as a promising vehicle for delivery of nucleic acids. Therefore, we prepared miRNA-bearing CS nanoparticles and

Long-term (35 years) cryopreservation of Echinococcus multilocularis metacestodes.

The metacestode of Echinococcus multilocularis is the etiological agent of alveolar echinococcosis. The metacestode stage used for research is maintained in rodents by serial passages. In order to determine whether cryopreservation of E. multilocularis metacestodes would be suitable for long-term maintenance and replace serial passages, isolates of different geographic origin were cryopreserved in 1984-1986. The aim of the current study was to test the viability of cryopreserved isolates following long-term cryopreservation (up to 35 years) and to determine the phylogenetic clades these isolat

Activity of mefloquine and mefloquine derivatives against Echinococcus multilocularis.

The cestode E. multilocularis causes the disease alveolar echinococcosis (AE) in humans. The continuously proliferating metacestode (larval stage) of the parasite infects mostly the liver and exhibits tumor-like growth. Current chemotherapeutical treatment options rely on benzimidazoles, which are rarely curative and have to be applied daily and life-long. This can result in considerable hepatotoxicity and thus treatment discontinuation. Therefore, novel drugs against AE are urgently needed. The anti-malarial mefloquine was previously shown to be active against E. multilocularis metacestodes i
