Echinostoma paraensei (Digenea: Echinostomatidae) lives in the duodenum and bile duct of rodents and is reported as a useful model for studies on the biology of flatworms. Here, we compared the growth and development of pre and post ovigerous worms collected 3, 7, 14 and 21 days post infection from experimentally infected hamster (permissive host) and Wistar rat (less permissive hosts). Linear measurements and ratios were examined by light (morphology and morphometry) and confocal laser scanning microscopy. At day 3, either worm from hamsters or rats were small with poorly developed gonads. At

The rapid spread of triclabendazole resistance in veterinary medicine is an important motivation for the discovery and development of novel fasciocidal drugs. The aim of this study was to characterize the fasciocidal properties of 1,2,4,5-tetraoxane (MT04 and MT14) and 1,2,4-trioxane (ST16 and ST28) analogues of the fasciocidal drug candidate OZ78, a 1,2,4-trioxolane. Dose response relationships were determined against juvenile and adult Fasciola hepatica in rats and Echinostoma caproni in mice. The temporal effects of MT04, MT14, ST16, and ST28 compared to OZ78 on the viability of F. hepatica

Invertebrates lack adaptive immune systems homologous to those of vertebrates, yet it is becoming increasingly clear that they can produce diversified antigen recognition molecules. We have previously noted that the snail Biomphalaria glabrata produces a secreted lectin, fibrinogen-related protein 3 (FREP3), unusual among invertebrate defense molecules because it is somatically diversified by gene conversion and point mutation. Here we implicate FREP3 in playing a central role in resistance to a major group of snail pathogens, digenetic trematodes. FREP3 is up-regulated in three models of resi

INTRODUCTION: Trematodiases cause significant financial losses to livestock worldwide and some of which are zoonotic, raising public health concerns. In Rwanda, information on the prevalence of bovine trematodiases is scanty, and this hampers efforts to control and prevent them in the country.
METHODS: This cross-sectional study aimed to determine the prevalence of bovine trematodiases in Nyagatare district and associated risk factors. One hundred cattle were randomly selected for this study and faecal samples were collected directly from the rectum to identify trematode eggs using a simple s

Echinostomiasis is a food-borne, intestinal, zoonotic, snail-mediated helminthiasis caused by digenean trematodes of the family Echinostomatidae with seven species of the genus Echinostoma infecting humans or domestic and wildlife animals. Echinostoma paraensei is a peristomic 37-collar-spined echinostome belonging to the "revolutum group". Praziquantel (PZQ) is the drug of choice for treatment and control of human schistosomiasis and food-borne trematodiasis. In the present study we used scanning and transmission electron microscopy to further elucidate the trematocidal effect of PZQ on adult

OBJECTIVE: To investigate the morphological characteristics of Echinostoma miyagawai in domestic ducks in Wuhu area, and to explore the feasibility of the cytochrome oxidase subunit-1 (Cox1) gene as a molecular marker for the identification of E. miyagawai.
METHODS: E. miyagawai was isolated from free-ranged domestic ducks in Wuhu area, and the parasites were stained and identified. In addition, the mitochondrial Cox1 gene of E. miyagawai was amplified using a PCR assay, and the amplification product was sequenced and aligned with the GenBank database to yield the homology for the identificat

This study examined survival and distribution of Echinostoma caproni in the small intestine of ICR mice at various times up to 36 hr following the death of the host. Adult worms were obtained at 2-wk postinfection of 21 ICR mice each infected with 50 metacercarial cysts. Mice were killed with light ether anesthetization and cervical dislocation and maintained at room temperature (22 +/- 1C) until examination at 0 (controls), 2, 4, 6, 12, 24, and 36 hr postmortem. Survival was based on worm activity and distribution was assessed on the basis of worm location in 1 of 5 equal intestinal segments

Intestinal histopathological changes due to infection with Echinostoma hortense (Trematoda) were studied in rats after experimental infection with the metacercariae. The metacercariae were obtained from the tadpoles of Rana nigromaculata, a second intermediate host infected in the laboratory. Total 18 albino rats (Sprague-Dawley) were given 200 matacercariae each and sacrificed on the day 1, 3, 7, 11, 22 or 44 post-infection (PI). Segments of the small intestine at 1, 3, 5, 8 and 30 cm posterior to the pylorus (PTP) were resected and studied histopathologically. 1. The flukes were seen to have

Experiments were designed to study the effect of Trypanosoma brucei on the expulsion of Echinostoma revolutum and on the development and maintenance of homologous E. revolutum resistance in the mouse. T. brucei given 3, 2, and 1 wk before and 1 wk after infection with E. revolutum completely inhibited the expulsion of the E. revolutum worm burden for a period of at least 6 wk following infection, and T. brucei given 2 or 3 wk after infection with E. revolutum conferred a significant delay in the expulsion of the E. revolutum worm burden. T. brucei given 1 wk before and 1 wk after a primary E.

Precise discrimination of Echinostoma species within the 'revolutum' group is quite difficult because of their morphological similarities. The objective of this study was to precisely characterize the echinostomes of ducks from Bangladesh based on both morphological and molecular characteristics. Two Echinostoma species were identified: E. revolutum and E. robustum. In the phylogenetic trees (ITS2 and nad1), E. revolutum and E. robustum belonged to their respective Eurasian clade, which is distinct from the American clade. These results suggest that both species have two distinct and geographi

Amphibian populations around the world are facing threats that include disease and pollution. Although the effect of environmental contaminants on susceptibility to infection has been demonstrated for several amphibian species, to our knowledge, the opposite interaction, infection status affecting contaminant susceptibility, has not been studied. We conducted standard 48-h toxicity tests to compare susceptibility to malathion, a widely used organophosphate insecticide, of uninfected pickerel frog (Rana palustris) tadpoles and tadpoles infected with two levels (10 or 30 cercariae) of the tremat

Parasites can influence host population dynamics, community composition and evolution. Prediction of these effects, however, requires an understanding of the influence of ecological context on parasite distributions and the consequences of infection for host fitness. We address these issues with an amphibian - trematode (Digenea: Echinostomatidae) host-parasite system. We initially performed a field survey of trematode infection in first (snail) and second (larval green frog, Rana clamitans) intermediate hosts over 5 years across a landscape of 23 ponds in southeastern Michigan. We then combin

A comparative morphometric analysis was conducted on two European species of Echinostoma in order to examine the degree of the variability in the metrical characteristics of the adults and to assess their value in discriminating species. Adult E. miyagawai and E. revolutum, obtained experimentally, were compared using univariate and multivariate statistical analyses of 35 and 25 metrical characters, respectively. All subsets of worms of different ages represented homogeneous samples with respect to their morphometric characteristics; however, univariate analyses revealed significant difference

BACKGROUND: Echinostoma caproni (Trematoda: Echinostomatidae) is an intestinal trematode, which has been extensively used to investigate the factors that determine the rejection of intestinal helminths. In this sense, several studies have shown that IL-25 is critical for the development of resistance against E. caproni in mice. In fact, treatment of mice with recombinant IL-25 generates resistance against primary E. caproni infection. However, the mechanisms by which IL-25 induces resistance remain unknown.
METHODS: To study the mechanisms responsible for resistance elicited by IL-25, we anal

Anthropogenic activities are promoting the proliferation of aquatic primary producers in freshwater habitats, including cyanobacteria. Among various problems stemming from eutrophication, cyanobacterial blooms can be toxic due to the production of secondary compounds, including microcystins such as microcystin-LR (MC-LR); however, it is unknown whether cyanotoxins can affect the susceptibility of aquatic vertebrates such as fish and larval amphibians to parasites or pathogens even though infectious diseases can significantly affect natural populations. Here, we examined how exposure to environ

Maternal plasma levels of pregnancy-associated murine protein-1 (PAMP-1) were measured by rocket immunoelectrophoresis in BALB/cABom mice experimentally infected with the two intestinal parasites Schistosoma mansoni and Echinostoma caproni. In the first half of pregnancy the PAMP-1 level was significantly lower in the infected females compared with the healthy controls, indicating that PAMP-1 is not essential for implantation and early gestation. In the latter half of pregnancy a rapid increase in the plasma levels of PAMP-1 in the infected females was recorded. This might suggest that pituita

Conventional mice and congenitally athymic, nude mice were infected with 20 metacercariae of the intestinal trematode Echinostoma revolutum. The sequential events in the pathological changes in the intestine were studied at different intervals post-infection. By day 11 onwards the conventional mice displayed dilatation of the region of the intestine which harboured the parasites. The mucosa in the dilated region showed marked crypt hyperplasia, villous atrophy and subepithelial fibrosis as the most conspicuous features which, together with a hypertrophy of the muscular layers, made the wall of

UNLABELLED: Echinostoma caproni (Trematoda: Echinostomatidae) is an intestinal trematode, which has been widely employed to investigate the factors determining the rejection of intestinal helminths. Protein production patterns of intestinal epithelial cells are related to the infection-induced changes that determine the course of E. caproni infections. Herein, we compare the protein production profiles in the ileum of four experimental groups of mice: control; infected; dewormed and reinfected. Worm burdens were significantly lower in secondary infections, confirming the generation of partial

In the present paper, we analyse the effect of a primary infection of ICR mice with Echinostoma caproni (Trematoda: Echinostomatidae) on the generation of resistance against homologous challenge infections. In ICR mice, E. caproni induces chronic infections concomitantly with strong responses characterized by the development of T-helper 1 (Th1)-type local immune responses with elevated levels of local interferon-gamma (IFN-γ) and inflammatory and antibody responses. Here, the effect of the response generated against a primary infection with E. caproni in the generation of resistance against su

Mucosal mast cell (MMC) responses and worm recovery rates in rats infected with Echinostoma hortense were investigated from day 3 to day 56 post-infection (p.i.). Experimental infected group showed apparently higher number of MMC in each part of the small intestine than that of the control group. The number of MMC in the duodenum increased gradually after the infection and reached a peak on day 35 p.i. Thereafter, the number of MMC continued to decrease at a slow pace. The kinetics of MMC responses in the upper and lower jejunum were similar to that of the duodenum, but the number of MMC in th
