A New Day 4 Grading System to Assess Embryo Quality in Frozen Embryo Transfer Cycles.

To present a new day 4 (D4) embryo grading system to assess embryos in frozen-thawed embryo transfer (FET) cycles. A new grading system (grades A-E) was developed from the 2011 ESHRE Istanbul Consensus for D4 embryos in FET cycles. Embryos with complete compaction were classified as grade A; those with partial compaction were assigned as grade B; and those without compaction were classified as grades C, D, and E according to their different blastomere number ratio (BNR; number of embryo blastomeres on D4/number of embryo blastomeres on D3, D4/D3). Embryos with a BNR of ≥ 1.5 were defined as gr

A comparison of culture and cooling for the short term preservation of in vivo derived dromedary camel embryos of varying morphological quality.

Despite recent advancements in the cryopreservation of dromedary camel embryos, widespread application of the technique is still limited by the need for specialised vitrification equipment and supplies. Temporary, liquid-phase embryo storage methods provide a useful tool for short-term preservation of camel embryos. In the current study, we compared the use of in vitro embryo culture with cold liquid storage in order to maintain both high- (Grade 1- Excellent and 2-Good) and low- (Grade 3- Moderate and 4-Poor) morphological grade Day-7 dromedary camel embryos in vitro for up to 3 days. Embryos

Effects of the Sex Ratio Organism on IN VITRO Differentiation of Drosophila Embryonic Cells.

Effects of Sex Ratio Organism (SRO) on the development of embryonic cells of Drosophila melanogaster were studied in vitro. The single embryo culture technique and a histochemical staining method were employed to distinguish male embryo cultures from female ones. SRO-infected Zw(n) females were crossed to wild-type males, and their embryos were separately dissociated and cultured in vitro. Cell growth and differentiation in each culture were examined by phase contrast microscopy and described both qualitatively and quantitatively. Neurons, imaginal disk cells and plasmatocyte-like cells barely

Early embryonic development, assisted reproductive technologies, and pluripotent stem cell biology in domestic mammals.

Over many decades assisted reproductive technologies, including artificial insemination, embryo transfer, in vitro production (IVP) of embryos, cloning by somatic cell nuclear transfer (SCNT), and stem cell culture, have been developed with the aim of refining breeding strategies for improved production and health in animal husbandry. More recently, biomedical applications of these technologies, in particular, SCNT and stem cell culture, have been pursued in domestic mammals in order to create models for human disease and therapy. The following review focuses on presenting important aspects of

A study on methods for preimplantation genetic testing (PGT) on in vivo- and in vitro-produced equine embryos, with emphasis on embryonic sex determination.

Two methods for preimplantation genetic testing (PGT) have been described for equine embryos: trophoblast cell biopsy (TCB) or blastocoele fluid aspiration (BFA). While TCB is widely applied for both in vivo- and in vitro-produced embryos, BFA has been mostly utilized for in vivo-produced embryos. Alternative methods for PGT, including analysis of cell-free DNA (CFD) in the medium where in vitro-produced embryos are cultured, have been reported in humans but not for equine embryos. In Experiment 1, in vivo- (n = 10) and in vitro-produced (n = 13) equine embryos were subjected to BFA, cultured

Engineering the human endometrium at the intersection of development and reproduction.

The human endometrium plays a central role in embryo implantation, pregnancy, and reproductive and systemic health, making it of great biological and clinical interest. Animal models of the endometrium often fail to mimic many intrinsic characteristics at the base of human physiology and pathology. Although two-dimensional cell-culture systems have yielded foundational insights, recent advances in three-dimensional modelling techniques have unlocked long-term culture of hormone-responsive, multicellular systems that more closely reproduce endometrial physiology. Here we trace the evolution of

Purification and Characterization of Extracellular Vesicles from Embryo-Conditioned Medium.

Extracellular vesicles (EVs) serve as key communication signals among embryos. They are secreted into the embryo culture medium and hold potential as biomarkers for assessing embryo quality. However, the limited volume of embryo-conditioned medium presents challenges for EV enrichment. This chapter details an effective method for purifying and characterizing EVs from in vitro embryo cultures by using the OptiPrep™ density gradient ultracentrifugation (ODG) method. Additionally, we also provide methods for preparing EV-free embryo culture media.

Designing 3-Dimensional In Vitro Oviduct Culture Systems to Study Mammalian Fertilization and Embryo Production.

The oviduct was long considered a largely passive conduit for gametes and embryos. However, an increasing number of studies into oviduct physiology have demonstrated that it specifically and significantly influences gamete interaction, fertilization and early embryo development. While oviduct epithelial cell (OEC) function has been examined during maintenance in conventional tissue culture dishes, cells seeded into these two-dimensional (2-D) conditions suffer a rapid loss of differentiated OEC characteristics, such as ciliation and secretory activity. Recently, three-dimensional (3-D) cell cu

A 3D "sandwich" co-culture system with vascular niche supports mouse embryo development from E3.5 to E7.5 in vitro.

BACKGROUND: Various methods for ex utero culture systems have been explored. However, limitations remain regarding the in vitro culture platforms used before implanting mouse embryos and the normal development of mouse blastocysts in vitro. Furthermore, vascular niche support during mouse embryo development from embryonic day (E) 3.5 to E7.5 is unknown in vitro. METHODS: This study established a three-dimensional (3D) "sandwich" vascular niche culture system with in vitro culture medium (IVCM) using human placenta perivascular stem cells (hPPSCs) and human umbilical vein endothelial cells (hU

Third party assisted conception: an African perspective.

The central importance of reproduction in all human cultures has given rise to many methods and techniques of assisting reproduction or overcoming infertility. Such methods and techniques have achieved spectacular successes in the Western world, where processes like in vitro fertilization (IVF) constitute a remarkable breakthrough. In this paper, the author attempts to reflect critically on assisted reproduction technologies (ART) from the background and perspective of African culture, a culture within which human reproduction is given the highest priority but which also exhibits a highly ambi

[Optimization of the in vitro culture system for chicken small intestinal organoids].

In order to establish a stable in vitro culture platform for chicken small intestine three-dimensional (3D) organoids, in this study, crypt cells were collected from the small intestine of 18-day-old embryos of AA broilers. On the basis of the L-WRN conditioned medium, we optimized the culture conditions of chicken small intestinal organoids by adjusting the proportions of nicotinamide, N-acetylcysteine, LY2157299, CHIR99021, Jagged-1, FGF, and other cytokines to select the medium suitable for the long-term stable growth of the organoids. The optimization results showed that the addition of 1.

Development of external morphological malformations induced by hyperthermia exposure during the blastula stage in an ex-ovo (shell-less) culture of Gallus gallus domesticus embryos.

The chicken is a valuable model to study embryonic development, genetic manipulation, regenerative medicine, tumorigenesis, vaccine development, toxicology, and teratology research. Different methods have been described for incubating chicken embryos outside the eggshell (ex-ovo or shell-less incubation). To date, no studies have focused on using ex-ovo incubation as a model to study the effects of hyperthermia as a teratogen. In this work, a total of 350 fertile chicken eggs were used to study the development of congenital malformations in an ex-ovo and in-ovo incubation system exposed to nor

Advances in pig embryo production for cryopreservation, transfer and genome editing.

Pig embryo production has emerged as a cornerstone for advancing both agricultural efficiency and biomedical innovation. In vivo-derived embryos remain the gold standard for cryopreservation and embryo transfer due to their superior developmental competence; however, the need for surgical recovery continues to limit their applicability on a large scale. Recent advances in nonsurgical transfer techniques and refined donor-management protocols are bringing widespread implementation closer to feasibility. Concurrently, in vitro production (IVP) systems provide a scalable alternative by generating

Highly efficient vitrification for cryopreservation of human oocytes and embryos: the Cryotop method.

Vitrification is frequently referred to as a novel technology of cryopreservation in embryology, although some young embryologists were born after its first successful application. Unfortunately, in spite of the accumulated evidence regarding its enormous potential value, most domestic animal and human laboratories use exclusively the traditional slow-rate freezing with its compromised efficiency and inconsistency. The purpose of this paper is to clarify terms and conditions, to summarize arguments supporting or disapproving the use of vitrification, and to outline its role among assisted repr

NMR studies of preimplantation embryo metabolism in human assisted reproductive techniques: a new biomarker for assessment of embryo implantation potential.

There has been growing interest in understanding energy metabolism in human embryos generated using assisted reproductive techniques (ART) for improving the overall success rate of the method. Using NMR spectroscopy as a noninvasive tool, we studied human embryo metabolism to identify specific biomarkers to assess the quality of embryos for their implantation potential. The study was based on estimation of pyruvate, lactate and alanine levels in the growth medium, ISM1, used in the culture of embryos. An NMR study involving 127 embryos from 48 couples revealed that embryos transferred on Day 3

Artificial intelligence in time-lapse system: advances, applications, and future perspectives in reproductive medicine.

With the rising demand for in vitro fertilization (IVF) cycles, there is a growing need for innovative techniques to optimize procedure outcomes. One such technique is time-lapse system (TLS) for embryo incubation, which minimizes environmental changes in the embryo culture process. TLS also significantly advances predicting embryo quality, a crucial determinant of IVF cycle success. However, the current subjective nature of embryo assessments is due to inter- and intra-observer subjectivity, resulting in highly variable results. To address this challenge, reproductive medicine has gradually t

Artificial Uterus and Artificial Embryos: Unsolved Tasks.

Ex utero embryo culture is one of the important tasks that has not been completely solved. In the first part of the current review, we focus on potential roles ex utero mammalian embryo culture could play, such as in infertility treatment or ex utero embryo treatment. Particularly, we discuss ex utero genetic therapy of embryos for correction and prevention of congenital pathologies. We also discuss the benefits of ex utero embryo culture in comparison to the traditional surrogacy programs and review its potential influence on social aspects. In the second part, we discuss key methods for ex u

Ovine Trophoblast Cells: Cell Isolation and Culturing from the Placenta at the Early Stage of Pregnancy.

Embryo development is dependent upon the exchange of oxygen and nutrients through the placenta, mainly composed of peculiar epithelioid cells, known as trophoblast cells. Normal trophoblast functionality plays a key role during the whole pregnancy, especially in the first stage of placentation. This chapter explains the techniques to obtain sheep primary trophoblast cells from the early placenta. Overall, procedures for cell isolation, culture, characterization, and cryopreservation are described.
