Biomarker responses and metabolism in Lumbricus terrestris exposed to drugs of environmental concern, an in vivo and in vitro approach.

The earthworm Lumbricus terrestris is an anecic species living in natural soils but it is also a sentinel in pollution monitoring. Specimens of L.terrestris were exposed for 48 h though the filter paper contact test at 1 mg/mL of the chemicals: Lamotrigine (LMG), Cocaine (COC), Fipronil (FIP) and the pesticide bis-4-nitrophenyl phosphate (BNPP). After that period, the activities of Acetylcholinesterase, Glutathione S-transferase, Carboxylesterase (CE) using different substrates, and lipid peroxidation levels were evaluated in the exposed whole tissue earthworms. The results revealed difference

Biomarkers of Environmental Enteropathy are Positively Associated with Immune Responses to an Oral Cholera Vaccine in Bangladeshi Children.

Environmental enteropathy (EE) is a poorly understood condition that refers to chronic alterations in intestinal permeability, absorption, and inflammation, which mainly affects young children in resource-limited settings. Recently, EE has been linked to suboptimal oral vaccine responses in children, although immunological mechanisms are poorly defined. The objective of this study was to determine host factors associated with immune responses to an oral cholera vaccine (OCV). We measured antibody and memory T cell immune responses to cholera antigens, micronutrient markers in blood, and EE mar

Explainable machine learning integrating environmental chemical biomarkers and maternal clinical factors for prediction of hypertensive disorder of pregnancy.

Hypertensive disorders of pregnancy (HDP) represent a leading cause of maternal morbidity, yet environmental chemical contributors to HDP risk remain poorly characterized within predictive frameworks. We developed and evaluated an explainable machine-learning model integrating environmental biomarkers with maternal clinical characteristics for HDP prediction using data from 4,260 pregnant women enrolled in the Korean Children's Environmental Health Study (Ko-CHENS) prospective birth cohort. An Extreme Gradient Boosting (XGBoost) classifier incorporating 18 environmental biomarkers-including bl

Urinary oxidative stress biomarkers are associated with preterm birth: an Environmental Influences on Child Health Outcomes program study.

BACKGROUND: Preterm birth is the leading cause of infant morbidity and mortality worldwide. Elevated levels of oxidative stress have been associated with an increased risk of delivering before term. However, most studies testing this hypothesis have been conducted in racially and demographically homogenous study populations, which do not reflect the diversity within the United States. OBJECTIVE: We leveraged 4 cohorts participating in the Environmental Influences on Child Health Outcomes Program to conduct the largest study to date examining biomarkers of oxidative stress and preterm birth (N

A survey of liquid chromatographic-mass spectrometric analysis of mercapturic acid biomarkers in occupational and environmental exposure monitoring.

High-performance liquid chromatography/mass spectrometry (HPLC/MS) is sensitive and specific for targeted quantitative analysis and is readily utilized for small molecules from biological matrices. This brief review describes recent selected HPLC/MS methods for the determination of urinary mercapturic acids (mercapturates) which are useful as biomarkers in characterizing human exposure to electrophilic industrial chemicals in occupational and environmental studies. Electrophilic compounds owing to their reactivity are used in chemical and industrial processes. They are present in industrial em

Influence of systems biology response and environmental exposure level on between-subject variability in breath and blood biomarkers.

To explain the underlying causes of apparently stochastic disease, current research is focusing on systems biology approaches wherein individual genetic makeup and specific 'gene-environment' interactions are considered. This is an extraordinarily complex task because both the environmental exposure profiles and the specific genetic susceptibilities presumably have large variance components. In this article, the focus is on the initial steps along the path to disease outcome namely environmental uptake, biologically available dose, and preclinical effect. The general approach is to articulate

Urinary 1-hydroxypyrene and other PAH metabolites as biomarkers of exposure to environmental PAH in air particulate matter.

Humans are exposed to polycyclic aromatic hydrocarbons (PAHs) from occupational, environmental, medicinal and dietary sources. PAH metabolites in human urine can be used as biomarkers of internal dose to assess recent exposure to PAHs. The most widely used urinary PAH metabolites are 1-hydroxypyrene (1-OHP) or 1-hydroxypyrene-O-glucuronide (1-OHP-gluc), the major form of 1-OHP in human urine, because of their relatively high concentration and prevalence in urine and their relative ease of measurement. Elevated levels of 1-OHP or 1-OHP-gluc have been demonstrated in smokers, in patients receivi

Glutathione Transferase as a Potential First-Tier Biomarker of Environmental Pollution in Workers and Residents of Umbria Region, Italy.

Erythrocyte glutathione transferase (e-GST) and oxidized human serum albumin (HSAox) were used as biomarkers to assess the environmental health of a foundry and a ceramics factory located in the Umbria region (Italy). While HSAox level indicates that no relevant oxidative stress is present in workers of either company, a distinct overexpression of e-GST, also present in non-workers residents, suggests exposure to broad environmental stressors that may be attributed to common sources. A more careful analysis of different job sectors and the effect of filtering facepieces indicates that the work

Effects of benzo[a]pyrene as an environmental pollutant and two natural antioxidants on biomarkers of reproductive dysfunction in male rats.

Benzo[a]pyrene (B[a]P) is an environmental toxicant and endocrine disruptor. Therefore, the aim of the present study was to investigate the toxicity of B[a]P in testis of rats and also to study the role of silymarin and thymoquinone (TQ) as natural antioxidants in the alleviation of such toxicity. Data of the present study showed that levels of testosterone, estrogen and progesterone were significantly decreased after treatment of rats with B[a]P. In addition, B[a]P caused downregulation of the expressions of steroidogenic enzymes including CYP17A1 and CP19A1, and decreased the activity of 17-

Urinary mercury and biomarkers of early renal dysfunction in environmentally and occupationally exposed adults: a three-country study.

We conducted a cross-sectional study in Sweden, Italy and Poland to assess environmental and occupational exposure to mercury from chloralkali (CA) plants and the potential association with biomarkers of early renal dysfunction. Questionnaire data and first-morning urine samples were collected from 757 eligible subjects. Urine samples were analysed for mercury corrected for creatinine (U-HgC), alpha-1-microglobulin (A1M), N-acetyl-beta-glucosaminidase (NAG) and albumin. Determinants of urinary mercury excretion were examined. Levels of kidney markers were compared in three U-HgC categories, an

Thyroid disruption in walleye (Sander vitreus) exposed to environmental contaminants: cloning and use of iodothyronine deiodinases as molecular biomarkers.

Thyroid hormones play a role in the initiation of ovarian maturation in fish. Thus, reports of delayed sexual maturation in female walleye (Sander vitreus) exposed to contaminants in the Ottawa River suggest the presence of endocrine disrupting chemicals. The objectives of this study were to assess the effects of environmental contaminants in the Ottawa River on thyroid hormones of immature walleye and to develop a molecular biomarker of thyroid status. Walleye were sampled in the Ottawa River at Deep River (reference site), at Rivière Blanche (downstream from the Ottawa and Gatineau municipal

DNA Damage in Bat Blood Leukocytes Using a Chromatin Dispersion Test (CDT): Biomarker of Environmental Genotoxicity.

Environmental pollutants produce adverse effects on organisms and ecosystems. Biomonitoring and biomarkers offer a reasonable approach to make these assessments. Induced genetic changes can be using as a biomarker in organisms that react to a given compound in the ecosystem. Monitoring environmental genotoxicity necessitates the choice of model animals known as "sentinels or biological monitors" and the suitability of validated tests for DNA damage evaluation. We aimed to estimate the DNA damage produced by thermal stress in the leukocytes of the Mexican free-tailed bat (Tadarida brasiliensis)

Urinary Trans, Trans-Muconic Acid is Not a Reliable Biomarker for Low-level Environmental and Occupational Benzene Exposures.

BACKGROUND: Benzene is a known occupational and environmental pollutant. Its urinary metabolite trans, trans-muconic acid (tt-MA) has been introduced by some environmental and occupational health regulatory associations as a biological index for the assessment of benzene exposure; however, recently, doubts have been raised about the specificity of tt-MA for low-level benzene exposures. In the present study, we investigated the association between urinary levels of tt-MA and inhalational exposure to benzene in different exposure groups. METHODS: Benzene exposure was assessed by personal air sa
