IDMME and IDMDE: inducible CRISPR-dCasRx platforms for spatiotemporal RNA m5C editing.

RNA 5-methylcytosine (m5C) is a dynamic epigenetic mark implicated in tumorigenesis, however, existing editors lack spatiotemporal regulation and reversibility. Here, we develop abscisic acid (ABA)-inducible CRISPR-dCasRx systems for programmable m5C methylation (IDMME) and demethylation (IDMDE). These editors enable site-specific, low off-target m5C modification through ligand-dependent assembly of split effector domains. We further integrate photocaged ABA to achieve light-controlled activation. Application in renal carcinoma models shows that targeted m5C editing modulates transcript functi

Chromatin-mediated alternative splicing regulates cocaine-reward behavior.

Neuronal alternative splicing is a key gene regulatory mechanism in the brain. However, the spliceosome machinery is insufficient to fully specify splicing complexity. In considering the role of the epigenome in activity-dependent alternative splicing, we and others find the histone modification H3K36me3 to be a putative splicing regulator. In this study, we found that mouse cocaine self-administration caused widespread differential alternative splicing, concomitant with the enrichment of H3K36me3 at differentially spliced junctions. Importantly, only targeted epigenetic editing can distinguis

Epigenetic control of gene regulation during development and disease: A view from the retina.

Complex biological processes, such as organogenesis and homeostasis, are stringently regulated by genetic programs that are fine-tuned by epigenetic factors to establish cell fates and/or to respond to the microenvironment. Gene regulatory networks that guide cell differentiation and function are modulated and stabilized by modifications to DNA, RNA and proteins. In this review, we focus on two key epigenetic changes - DNA methylation and histone modifications - and discuss their contribution to retinal development, aging and disease, especially in the context of age-related macular degenerati

Generating mRNA Encoding Anti-HBV Designer Epigenome Modifiers.

Chronic hepatitis B virus (HBV) infection poses significant global health challenges, with an estimated 257.5 million individuals affected, leading to approximately 880,000 deaths annually from complications such as cirrhosis and hepatocellular carcinoma. Although antiviral therapies are available, they often fail to eliminate the stable viral reservoir of covalently closed circular DNA within infected cells. Recent advancements in epigenome editing offer a promising alternative to traditional mutagenic approaches, providing a strategy to silence HBV replication and transcription directly. Thi

Fertile androgenetic mice generated by targeted epigenetic editing of imprinting control regions.

Each new mammalian life begins with the fusion of an oocyte and a sperm to produce a fertilized egg containing two sets of genomes, one from the mother and one from the father. Androgenesis, a way for producing offspring solely from male genetic material, is limited in mammals, presumably due to barriers arising from genomic imprinting, an epigenetic mechanism leading to monoallelic gene expression. Here, we report adult mammalian offspring derived from the genetic material of two sperm cells. These mice, which we refer to as androgenetic mice, were produced via targeted DNA methylation editin

Key to Delivery: The (Epi-)genome Editing Vector Toolbox.

Curing a genetic disease by repairing the underlying genetic defect is a fascinating concept that has been addressed so far by gene compensation therapy. For this, a functional copy of the gene in question together with elements controlling its expression is produced as a vector and introduced ex vivo into the patient's own cells that subsequently are reinfused. Alternatively, vectors are administered directly in vivo. Although this strategy resulted in impressive therapeutic benefits for patients, the ultimate goal of gene therapy, i.e., a cure by repairing the actual genetic or epigenetic de

Unlocking the epigenetic code: future Frontiers in predictive biomarkers for intellectual disability in fragile X syndrome.

Fragile X Syndrome (FXS) remains the leading inherited cause of intellectual disability (ID), yet the classic CGG repeat expansion on the Fragile X Messenger Ribonucleoprotein 1 (FMR1) gene often fails to provide a precise prognostic map for cognitive outcomes. While the expansion is the primary trigger, the resulting epigenetic landscape, characterized by DNA hypermethylation and chromatin remodelling, is the true driver of gene silencing and phenotypic severity. This review critically evaluates the shift from traditional genetic screening to advanced epigenetic profiling as a robust framewor

Epigenetics of scleroderma: Integrating genetic, ethnic, age, and environmental effects.

Scleroderma or systemic sclerosis is thought to result from the interplay between environmental or non-genetic factors in a genetically susceptible individual. Epigenetic modifications are influenced by genetic variation and environmental exposures, and change with chronological age and between populations. Despite progress in identifying genetic, epigenetic, and environmental risk factors, the underlying mechanism of systemic sclerosis remains unclear. Since epigenetics provides the regulatory mechanism linking genetic and non-genetic factors to gene expression, understanding the role of epig

DNMT1-Mediated Epigenetic Reprogramming Drives KMT2A Amplifications and Rearrangements.

UNLABELLED: MLL/KMT2A amplifications and rearrangements are prevalent in infant, adult, and therapy-induced leukemia; however, the molecular contributors controlling these alterations remain elusive. Here, we reveal that rapid CTCF degradation generates local genome structure alterations at KMT2A and copy gains and rearrangements. We then established a conserved, coordinated interplay between DNA and histone methylation pathways that control CTCF occupancy, and in turn, KMT2A locus stability. For example, DNMT1 overexpression promotes increased H3K9 methylation, reduced CTCF occupancy, and cau

Endothelial p300 Promotes Portal Hypertension and Hepatic Fibrosis Through C-C Motif Chemokine Ligand 2-Mediated Angiocrine Signaling.

BACKGROUND AND AIMS: During liver fibrosis, liver sinusoidal endothelial cells (LSECs) release angiocrine signals to recruit inflammatory cells into the liver. p300, a master regulator of gene transcription, is associated with pathological inflammatory response. Therefore, we examined how endothelial p300 regulates angiocrine signaling and inflammation related to portal hypertension and fibrogenesis. APPROACH AND RESULTS: CCl4 or partial inferior vena cava ligation (pIVCL) was used to induce liver injury. Mice with LSEC-specific p300 deletion (p300LSECΔ/Δ ) or C-C motif chemokine ligand 2 (Cc
