Mechanism of Gzma-mediated GEF-H1 activation in intestinal epithelial cells leading to intestinal barrier dysfunction in sepsis.

BACKGROUND: Sepsis-induced intestinal injury is a severe complication associated with dysfunction affecting multiple organ systems and a significantly elevated risk of death. Intestinal barrier dysfunction plays a central role, but the underlying molecular pathways remain incompletely understood. The present study sought to explore how the Gzma/GEF-H1/RhoA signalling axis contributes to the disruption of the intestinal epithelial barrier in sepsis. METHODS: Transcriptomic data, clinical samples, and a murine caecal ligation and puncture (CLP) model was used to assess Gzma expression and its c

Nail disorders in a woman treated with ixabepilone for metastatic breast cancer.

Ixabepilone (Ix) (BMS-247550) is a potent member of a new class of microtubule-stabilizing cytotoxic agents known as epothilones. In pre-clinical studies, Ix has shown anticancer activity against several cancer types, including paclitaxel-resistant models, both in vitro and in vivo. The major toxicities associated with Ix are myelosuppression, sensory neuropathy and neutropenia. Other minor side-effects include asthenia/fatigue, stomatitis, anorexia, alopecia, skin reaction, hypersensitivity reactions and a fluid-retention syndrome. Although Ix is functionally correlated to taxanes, no previou

Low dose tubulin-binding drugs rescue peroxisome trafficking deficit in patient-derived stem cells in Hereditary Spastic Paraplegia.

Hereditary Spastic Paraplegia (HSP) is a genetically heterogeneous group of disorders, diagnosed by progressive gait disturbances with muscle weakness and spasticity, for which there are no treatments targeted at the underlying pathophysiology. Mutations in spastin are a common cause of HSP. Spastin is a microtubule-severing protein whose mutation in mouse causes defective axonal transport. In human patient-derived olfactory neurosphere-derived (ONS) cells, spastin mutations lead to lower levels of acetylated α-tubulin, a marker of stabilised microtubules, and to slower speed of peroxisome tra

Ixabepilone: a novel microtubule-stabilizing agent for the treatment of metastatic breast cancer.

PURPOSE: The pharmacology, pharmacokinetics, clinical efficacy, safety, dosage, and administration of ixabepilone in patients with metastatic breast cancer are examined. SUMMARY: The clinical utility of the three main classes of chemotherapeutic agents used in breast cancer (i.e., anthracyclines, taxanes, and fluorinated pyrimidines) is limited in some patients by the emergence of drug resistance which leads to disease progression. A recent addition to the available drugs for the treatment of advanced breast cancer is the epothilone B analog ixabepilone, which has demonstrated clinical activi

Targeted environmental enrichment is more effective than bipedal treadmill training after thoracic spinal cord injury.

Rehabilitation is widely recognized as an essential component in evaluating the effectiveness of a therapeutic intervention in animal models of translational spinal cord injury research. Ideally, the rehabilitation method used should be optimized for the specific injury model and severity to adequately compare the efficacy of the investigational therapy. For studies utilizing thoracic spinal cord injuries, rehabilitation primarily involves bipedal or quadrupedal treadmill training aimed at restoring hindlimb function. Previously, we reported modest improvements to hindlimb motor recovery follo

Natural-Products-Inspired Use of the gem-Dimethyl Group in Medicinal Chemistry.

The gem-dimethyl moiety is a structural feature frequently found in many natural products of clinical interest, including, but not limited to, taxanes, epothilones, statins, retinoids, di-/triterpenes, noviose deoxysugar, and antibiotics derived from β-lactams, macrolides, and aminocoumarins. Inspired by this time-tested moiety, medicinal chemists have widely explored its use in developing bioactive molecules because of the possibility to (1) increase target engagement, potency, and selectivity through van der Waals interactions and entropically favorable restriction to a bioactive conformatio

Selected hybrid natural products as tubulin modulators.

Modulators of microtubule dynamics have received increasing attention because of their potential to stop cancer growth. Although it belongs to the category of complex protein-protein interactions (PPIs), which are generally considered difficult to modulate through small molecules, the use of microtubule is considered a well-validated target. There are a number of bioactive natural products and related compounds that are currently in use as drugs or in clinical trials as next generation anti-cancer agents. The present review article is focused on two such bioactive natural products, epothilone

Activation of the steroid and xenobiotic receptor (human pregnane X receptor) by nontaxane microtubule-stabilizing agents.

PURPOSE: Because induction of drug efflux transporters is one of the major underlying mechanisms of drug resistance in cancer chemotherapy, and human pregnane X receptor (hPXR) is one of the principal "xenobiotic" receptors whose activation induces transporter and drug-metabolizing enzyme gene transcription, it would be ideal to develop chemotherapy drugs that do not activate hPXR. This report describes studies undertaken to explore the characteristics of hPXR stimulation and mechanisms of drug-receptor interactions in vitro with new anti-tubulin drugs. EXPERIMENTAL DESIGN: In vitro transient

Utidelone induces mechanical and cold allodynia in mice via TRPA1 activation.

OBJECTIVE: Utidelone (UTD1), a recently approved epothilones analog in China for metastatic breast cancer, is endorsed in combination with capecitabine for metastatic breast cancer patients who have encountered first-line therapy failures. Despite its clinically verified therapeutic efficacy, it is concurrently associated with peripheral neuropathic pain, particularly affecting extremities. However, the etiology of UTD1-induced peripheral neuropathic pain remains unclear. METHODS: The present investigation built a mouse pain model induced by UTD1, resulting in marked mechanical and cold allod

Chemotherapy-induced peripheral neuropathy in children and adolescent cancer patients.

Brain cancer and leukemia are the most common cancers diagnosed in the pediatric population and are often treated with lifesaving chemotherapy. However, chemotherapy causes severe adverse effects and chemotherapy-induced peripheral neuropathy (CIPN) is a major dose-limiting and debilitating side effect. CIPN can greatly impair quality of life and increases morbidity of pediatric patients with cancer, with the accompanying symptoms frequently remaining underdiagnosed. Little is known about the incidence of CIPN, its impact on the pediatric population, and the underlying pathophysiological mecha

Anticancer therapy with novel tubulin-interacting drugs.

Antimitotic agents that target tubulin, including the taxanes and vinca alkaloids, are important components of current anticancer therapy. Whilst these antimitotic drugs are highly effective in the treatment of a number of cancers, both acquired and intrinsic resistance to these agents is a major clinical problem. Furthermore, the systemic toxicity, and in some cases lack of oral availability, make these agents less than ideal. Recently much effort has been directed on the isolation and synthesis of new antimitotic drugs that target the tubulin/microtubule system and display efficacy against d

Identification of molecular signatures and pathways to identify novel therapeutic targets in Alzheimer's disease: Insights from a systems biomedicine perspective.

Alzheimer's disease (AD) is a progressive neurodegenerative disease characterized by the accumulation of amyloid plaques and neurofibrillary tangles in the brain. However, there are no peripheral biomarkers available that can detect AD onset. This study aimed to identify the molecular signatures in AD through an integrative analysis of blood gene expression data. We used two microarray datasets (GSE4226 and GSE4229) comparing peripheral blood transcriptomes of AD patients and controls to identify differentially expressed genes (DEGs). Gene set and protein overrepresentation analysis, protein-p

Expression profiling stratifies mesothelioma tumors and signifies deregulation of spindle checkpoint pathway and microtubule network with therapeutic implications.

BACKGROUND: Malignant pleural mesothelioma (MPM) is a lethal neoplasm exhibiting resistance to most treatment regimens and requires effective therapeutic options. Though an effective strategy in many cancer, targeted therapy is relatively unexplored in MPM because the therapeutically important oncogenic pathways and networks in MPM are largely unknown. MATERIALS AND METHODS: We carried out gene expression microarray profiling of 53 surgically resected MPMs tumors along with paired normal tissue. We also carried out whole transcriptomic sequence (RNA-seq) analysis on eight tumor specimens. Taq

Potential for Treatment of Neurodegenerative Diseases with Natural Products or Synthetic Compounds that Stabilize Microtubules.

No effective therapeutics to treat neurodegenerative diseases exist, despite significant attempts to find drugs that can reduce or rescue the debilitating symptoms of tauopathies such as Alzheimer's disease, Parkinson's disease, frontotemporal dementia, amyotrophic lateral sclerosis, or Pick's disease. A number of in vitro and in vivo models exist for studying neurodegenerative diseases, including cell models employing induced-pluripotent stem cells, cerebral organoids, and animal models of disease. Recent research has focused on microtubulestabilizing agents, either natural products or synthe

Current advancements in tubulin targeting agents for breast cancer: Design strategies, structure-activity relationships, and pharmacological insights.

Breast cancer remains one of the most common causes of cancer mortality worldwide and, despite improvements in early detection and targeted therapies, resistance, recurrence, and metastasis still impair therapeutic success. Microtubule-targeting agents (MTAs) constitute a central pillar of breast cancer therapy by interfering with tubulin homeostasis, arresting mitosis and ultimately leading to apoptosis. Clinically utilized MTAs, such as vinca alkaloids, colchicine-site binders, taxanes, and epothilones, have greatly benefited many patients, for whom taxanes remain the mainstay of chemotherap

Comparative analysis of physicochemical properties, functional activities, and metabolomics of Annona squamosa juice fermented by different functional microorganisms.

Annona squamosa is a tropical fruit with unique flavor but rapid postharvest spoilage. This study evaluated fermentation with Bacillus subtilis, Acetobacter tropicalis, Limosilactobacillus reuteri, and Lactobacillus buchneri on juice properties. Fermentation increased total phenolics, flavonoids, and free amino acids, and improved taste. Fermented juices showed enhanced antioxidant, antimicrobial, and anti-inflammatory activities, with increased inhibition of α-amylase, α-glucosidase, xanthine oxidase, and pancreatic lipase (p < 0.001). Metabolomics revealed differential metabolites linked to

Enzymology of assembly line synthesis by modular polyketide synthases.

Modular polyketide synthases (PKSs) run catalytic reactions over dozens of steps in a highly orchestrated manner. To accomplish this synthetic feat, they form megadalton multienzyme complexes that are among the most intricate proteins on earth. Polyketide products are of elaborate chemistry with molecular weights of usually several hundred daltons and include clinically important drugs such as erythromycin (antibiotic), rapamycin (immunosuppressant) and epothilone (anticancer drug). The term 'modular' refers to a hierarchical structuring of modules and domains within an overall assembly line a

Structural basis of microtubule stabilization by laulimalide and peloruside A.

Laulimalide and peloruside A are microtubule-stabilizing agents (MSAs), the mechanism of action on microtubules of which is poorly defined. Here, using X-ray crystallography it is shown that laulimalide and peloruside A bind to a unique non-taxane site on β-tubulin and use their respective macrolide core structures to interact with a second tubulin dimer across protofilaments. At the same time, they allosterically stabilize the taxane-site M-loop that establishes lateral tubulin contacts in microtubules. Structures of ternary complexes of tubulin with laulimalide/peloruside A and epothilone A

Microtubule-stabilizing drugs suppress convulsions in a C. elegans model of CAMSAP disorders.

Recessive, loss of function, genomic variants in CAMSAP1 and 2 (calmodulin-regulated spectrin-associated proteins 1 and 2) cause a neurodevelopmental seizure disorder in humans that currently lacks specific treatments. CAMSAP proteins stabilize dynamics of the minus-ends of microtubules and regulate regenerative signaling cascades. We explored microtubule-stabilizing drugs as potential therapeutic interventions using a convulsion assay in the model organism Caenorhabditis elegans. We found that animals with genetically disrupted ptrn-1 (the C. elegans homolog of CAMSAP1) exhibited elevated con
