Changes in filterability during 28-day bloodbank storage of normal, sickle trait, and sickle cell anaemia red cells were investigated. The technique used involved constant positive-pressure filtration of red cell-saline suspensions through cellulose nitrate membrane filters of 8-microns pore size for 2 min. Erythrocyte filtration rate was expressed as the number of red cells filtered per minute; calculated from the volume and red cell count of the filtrates. This was a departure from the commonly used filtrate volume measurements alone, and seemed to permit a clearer definition of changes in f

The effects of two local anaesthetics, procain and tetracaine, on erythrocyte aggregation and deformability under in vitro conditions are analysed. Fresh blood samples from healthy volunteers are obtained by venepuncture in test tubes containing heparin as anticoagulant. After centrifugation, plasma and cells are separated and buffy coat is discarded. The erythrocyte suspensions for aggregation and deformability measurement are prepared at hematocrit 34 and at 8% in buffered medium containing glucose and albumin, respectively. A part of the suspending medium in these samples is replaced by an

The deformability of human erythrocytes was studied at pH values ranging from 6.4 to 8.5 using micropipette aspiration of whole cells. Also the mean cell volume was measured in dependence on pH. The observed high increase of the transit pressure pt with lowering of the pH in a fairly narrow interval suggests that within the range of pH values under consideration pi is mainly controlled by the effect of the critical volume of the cells.

Erythrocyte deformability was determined in more than 500 clinical samples, and was found to be elevated in conditions in which fetal-like red cells are produced: aplastic anemia (3/3 cases), myelodysplastic syndromes, polycythemias, sickle cell anemia during treatment with hydroxyurea, paroxysmal nocturnal hemoglobinuria, and recovery from B12 deficiency. Elevated deformability was observed in neonatal erythrocytes, and during recovery from transient erythroblastopenia of childhood, when fetal-like red cells are known to be produced. Increased deformability appears to be a feature of fetal an

This study compared erythrocyte changes between a group of subjects with sickle cell trait (SCT) and controls (subjects without hemoglobinopathy) during a soccer game in two conditions: with and without hydration. Erythrocyte deformability of subjects was assessed by the coefficient of erythrocyte rigidity (Tk) which was calculated before and after football match. Our results showed a significant increase in erythrocyte rigidity (Tk) in SCT at the end of physical activities without hydration; however when water was provided ad libitum their Tk decreased significantly, reaching values of contro

Oxidative stress from ischemia-reperfusion (IR) injury severely compromises erythrocyte deformability, a critical factor that disrupts microvascular flow and thereby exacerbates reperfusion injury. This study provides a novel perspective by directly investigating the protective effects of 6-Gingerol, known for its potent antioxidant properties, on erythrocyte function in a rat model of lower-extremity skeletal muscle IR. Twenty-four Wistar albino rats were divided into four groups: Sham (n = 6), DMSO (n = 6), IR (n = 6), and 6-Gingerol-IR (6G-IR, n = 6, 6 mg/kg i.p., 1 h before ischemia). Nine

The alpha- and beta-thalassaemias are characterized by decreased erythrocyte deformability. To determine what effects excess alpha- and beta-haemoglobin (globin) chains have on cellular and membrane deformability, purified haem-containing alpha- and beta-chains were entrapped within normal erythrocytes. Entrapment of purified alpha-chains in normal erythrocytes resulted in a significant decrease in cellular and membrane deformability similar to that observed in beta-thalassaemia intermedia. The decreased deformability was correlated with alpha-chain membrane deposition, an alteration in membra

We present an application of wavelet-based Information Theory quantifiers (Normalized Total Shannon Entropy, MPR-Statistical Complexity and Entropy-Complexity plane) on red blood cells membrane viscoelasticity characterization. These quantifiers exhibit important localization advantages provided by the Wavelet Theory. The present approach produces a clear characterization of this dynamical system, finding out an evident manifestation of a random process on the red cell samples of healthy individuals, and its sharp reduction of randomness on analyzing a human haematological disease, such as β-t

Cellular deformability has been proposed in the past as a major determinant of lectin-mediated agglutination of cells. In this paper we have evaluated the correlation between deformability and Con A-agglutinability of human erythrocytes by subjecting them to agents that alter either one of the properties and evaluating the effect on the other property. The following results have been obtained: (i) Treatment with pronase or trypsin, which makes the Con A-nonagglutinable normal red cells highly agglutinable, has practically no effect on deformability; while neuraminidase treatment, with a simila

Impairment of red cell deformability (and decrease of survival time) is the common trait of congenital haemolytic anemias with the exception of the enzyme defect glucose-6-phosphate-dehydrogenase (GPDH) deficiency. Causes for increased red cell rigidity may be: spherocytosis (familial hemolytic anemia), instable less fluid Hb (instable hemoglobinopathy), abnormal HbS-formation (sickle cell disease), genetic synthesis defect of Hb chains (homozygous beta-thalassemia), enzyme defects (autosomal recessive pyruvate-kinase deficiency). With GPDH red cell deformability remains unchanged. Splenectomy

We studied 45 full-term newborns divided into 3 groups. Group 1: 17 newborns with bilirubin <10 mg/dL; Group 2: 18 newborns with hemolytic ictericia (bilirubin 11-20 mg/dL) and Group 3: 10 newborns with moderate hemolytic ictericia needing exchange transfusion. The following were studied: erythrocytic deformability, plasma viscosity, plasmatic osmolarity, seric bilirubin, bilirubin/albumin ratio, free fatty acids and corpuscular volume of the erythrocytes. In full-term newborns, the following are risk factors for increased erythrocytic rigidity: neonatal hemolytic illness (p = 0.004, odds rati

Blood from patients with homozygous sickle cell anemia gives a "cross shaped" diffraction pattern, when submitted to shear stresses in the Ektacytometer. This pattern, which seems to be specific to these cells, is due to the existence of two populations of elliptical cells: one population (normal, deformable cells) aligned parallel to the direction of the flow, and the other (abnormal, rigid cells) perpendicular to the direction of the flow.

BACKGROUND: Plasmodium vivax accounts for the majority of human malaria infections outside Africa and is being increasingly associated in fatal outcomes with anaemia as one of the major complications. One of the causes of malarial anaemia is the augmented removal of circulating non-infected red blood cells (nRBCs), an issue not yet fully understood. High levels of auto-antibodies against RBCs have been associated with severe anaemia and reduced survival of nRBCs in patients with falciparum malaria. Since there are no substantial data about the role of those antibodies in vivax malaria, this st

Relationship between erythrocyte deformability and: a) soluble cell adhesion molecules concentration, b) glomerular filtration rate (eGFR) has been investigated in three study groups: a group of 20 patients with diagnosed arterial hypertension, a group of 20 individuals with exclusively hypercholesterolemia and a group of 22 healthy persons. The individuals with hypertension or hypercholesterolemia were free of any other cardiovascular disease risk factor and were not on any therapy prior to entering the study. Clinical and laboratory data included systolic and diastolic blood pressure (obtain

Deformability of erythrocytes of a patient with chronic nonspherocytic hemolytic anemia caused by a G-6-PD variant (G-6-PD Hamburg) in red cells was studied. The flow rate of erythrocytes from this patient through 5 mum polycarbonate sieves was higher than that of red cells from healthy adults. Even under oxidative stress in vitro, the deformability of erythrocytes of the patient was only slightly decreased. The residual hemoglobin content of red cell membranes (ghosts) from this patient was lower than that of ghosts from healthy adults and of ghosts prepared from comparable reticulocyte-rich

The rheological properties of human erythrocytes were investigated after incubation in ethanol-containing suspensions. Ethanol concentrations were adjusted to the blood ethanol level reached in severe alcohol intoxication. Red cell deformability was measured by the filterability and by viscosimetry of suspended erythrocytes. Red cell deformability becomes significantly decreased by ethanol addition to the suspending media in close connection with the increased osmolarity of the solution. The influence of these alterations on blood viscosity and microcirculation in shocked and non-shocked patie

A new gravity filtration technique for the study of sickle cell deformability has shown a significant loss of filterability of oxygenated erythrocytes on day 2 of painful crisis in eight patients with homozygous sickle cell anaemia; there was no increase in irreversibly sickled cell (ISC) count. Serial study (mean of 5.2 occasions) in six out-patients when asymptomatic also showed considerable fluctuation in sickle cell filterability with time; this was again independent of ISC count. Erythrocyte filtration methods demonstrate fluctuations in the rheology of sickle cells that are not detected
