Correction by pentoxifylline of the abnormal fluorescence polarization of erythrocyte membranes from diabetic patients.

Erythrocytes from diabetic patients show abnormal rheology. Pentoxifylline, a methylxanthine, improves the abnormal deformability of diabetic erythrocytes, but its mechanism of action remains unclear. We have studied the effect of pentoxifylline on the lipid order of erythrocyte membranes from controls and patients with Type I diabetes. We studied the structural organization of membrane lipids in individual erythrocyte ghosts by fluorescence polarization using a cell sorter. Fluorescence polarization values (P) for 17 controls (P = 0.244) and 20 diabetic patients (P = 0.215) were significantly

Superoxide dismutase and glutathione peroxidase abnormalities in erythrocytes and lymphoid cells in Down syndrome.

After validating a superoxide dismutase (SOD) assay, this enzyme was measured together with a large series of cell age sensitive marker enzymes, in the erythrocytes of 24 persons with the 24 persons without Down syndrome. In addition to the expected elevation of SOD-1, significant elevations of erythrocyte glutathione peroxidase (GSHPxase) and phosphofructokinase activities were found in Down syndrome. None of these changes could be accounted for by a decreased mean red cell age. The elevation of GSHPxase was also found in lymphoid cells but does not represent a gene dose effect at this locus.

Abnormal erythrocyte-related parameters in children with Pb, Cr, Cu and Zn exposure.

The link between exposure to a particular heavy metal or metalloid and the development of anemia is well established. However, the association between combined exposure to multiple heavy metal(loid)s and anemia in children is still lacking in evidence. In this study, a total of 266 children aged 3 to 7 were recruited from Guiyu, China. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure blood heavy metal(loid) concentrations. Blood cell count, hemoglobin (HGB), mean corpuscular hemoglobin (MCH), mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCH

Acetaldehyde adducts in blood and bone marrow of patients with ethanol-induced erythrocyte abnormalities.

BACKGROUND: Although alcohol abuse is known to cause a wide array of adverse effects on blood cell formation, the molecular mechanisms by which alcohol exerts its toxic actions remain poorly defined. We examine here the formation of acetaldehyde-derived protein modifications in erythrocytes and in their bone marrow precursors using antibodies specifically recognizing acetaldehyde-modified epitopes in proteins independently of the nature of the carrier protein. MATERIALS AND METHODS: We studied 138 consecutive adult patients undergoing bone marrow aspiration due to macrocytosis (MCV values abo

Comparative Assessment of Short- and Long-Term Effects of Triadimenol Fungicide on Danio rerio Erythrocytes Using the Micronucleus and Erythrocyte Nuclear Abnormality Assays.

Triadimenol is a systemic fungicide widely used in agriculture to manage plant diseases, especially fungal infections. This study aims to evaluate the short-term (24, 48, 72 and 96 h) and long-term (10, 20, and 30 days) genotoxic effects of different concentrations of triadimenol on zebrafish (Danio rerio) erythrocytes using micronucleus (MN) and erythrocyte nuclear abnormal (ENA) assay. Fish were treated with 1.5, 3, and 6 mg/L concentrations of triadimenol for short and long-term periods. After the treatment period, blood was collected with heparin syringe, smears were prepared, the preparat

Abnormality in membrane fatty acid compositions of cells measured on erythrocyte in alcoholic liver disease.

It has been proven that the fatty acids of esterified phospholipids in the cell membrane play an important role in membrane fluidity. Our previous in vitro experiment indicated that the impairment of erythrocyte membrane fluidity might be largely because of the change in fatty acids. The aim of this study is to clarify changes of cell membrane fatty acids in more detail in relation to various stages and pathology of alcoholic liver disease. For the analysis, erythrocyte membranes were exploited on the assumption that their fatty acid compositions may be similar to those of other organs. In alc

Absence of abnormal erythrocyte superoxide dismutase, copper, or zinc levels in patients with retinitis pigmentosa.

Blood samples obtained from 22 patients with retinitis pigmentosa, 6 unaffected family members, and 8 unrelated controls showed serum copper and zinc to be in the normal range, contrary to the results in earlier reports. Likewise no significant variation of erythrocyte superoxide dismutase (SOD) and catalase levels was found between the 3 groups or when the patients were grouped by sex, age, or genetic distribution. The SOD proteins from controls and retinitis pigmentosa patients had identical mobilities on gel electrophoresis, isoelectric points, and heat stabilities. Our studies do not suppo

Intercellular adhesion molecule-4 and CD36 are implicated in the abnormal adhesiveness of sickle cell SAD mouse erythrocytes to endothelium.

BACKGROUND: Abnormal adhesiveness of red blood cells to endothelium has been implicated in vaso-occlusive crisis of sickle cell disease. The present study examined whether the SAD mouse model exhibits the same abnormalities of red blood cell adhesion as those found in human sickle cell disease. DESIGN AND METHODS: The repertoire of adhesive molecules on murine erythrocytes and bEnd.3 microvascular endothelial cells was determined by flow cytometry using monoclonal antibodies or by western blotting. Adhesion was investigated in dynamic conditions and measured at different shear stresses. RESU

Nuclear and Cellular Abnormalities of Erythrocytes in Response to Thermal Stress in Common Carp Cyprinus carpio.

As a consequence of global warming, increase of water temperature is likely to alter physiological functions of fish. Hence, we examined the effects of high temperature on blood glucose, hematological parameters [hemoglobin (Hb), red blood cell (RBC), and white blood cell (WBC)], and nuclear and cellular structure of blood cells of common carp (Cyprinus carpio) after exposure to three temperature regimes (27, 31, and 35°C) for 14 days. Fish were sacrificed on 3, 7, and 14 days of exposure. The blood glucose level increased significantly in the fish exposed to 35°C compared to 27 and 31°C. The

Micronuclei and other erythrocyte nuclear abnormalities in fishes from the Great Lakes Basin, USA.

Biological markers (biomarkers) sensitive to genotoxic and mutagenic contamination in fishes are widely used to identify exposure effects in aquatic environments. The micronucleus assay was incorporated into a suite of indicators to assess exposure to genotoxic and mutagenic contamination at five Great Lakes Areas of Concern (AOCs), as well as one non-AOC (reference) site. The assay allowed enumeration of micronuclei as well as other nuclear abnormalities for both site and species comparisons. Erythrocyte abnormality data was also compared to skin and liver tumor prevalence and hepatic transcr

Nutritional dwarfing: a growth abnormality associated with reduced erythrocyte Na+,K(+)-ATPase activity.

To assess whether nonorganic nutritional dwarfing (ND) is accompanied by a biochemical adaptation to reduced nutrient intake, Na+,K(+)-ATPase activity of erythrocytes was examined. The study included 27 children with ND who demonstrated deteriorating linear growth and poor weight gain (caused by self-imposed dietary restrictions), 20 patients who initially exhibited ND growth patterns and subsequently experienced catch-up weight gain and growth after nutritional rehabilitation (NDR), and 32 normally growing children who had familial short stature and/or constitutional growth delay (F/CSS). Abnormal pattern of erythrocyte ageing in hereditary spherocytosis as shown by Percoll density gradient centrifugation.

Erythrocytes from individuals with Hereditary Spherocytosis (HS) were fractionated on Percoll density gradients. The osmotic fragility, mean cell volume, mean cell haemoglobin concentration, reticulocyte count and cell morphology of the fractions showed that the gradients separate the erythrocytes according to age, the older small spherocytes appearing in the dense fractions. Comparison of the well established parameters of erythrocyte ageing, pyruvate kinase activity, acetylcholinesterase activity and cell potassium content in normal and HS erythrocyte fractions of similar age revealed abnorm

[Abnormal cation transport in erythrocyte membranes during congenital hemolytic anemia (author's transl)].

In this paper we review our knowledge on: 1. The physiological mechanisms of constant low Na+, high K+ concentration in human red blood cells (cation content, cation movements through the erythrocyte membrane, membrane ATPases). 2. The pathologic findings observed in congenital hemolytic anemia. These alterations may explain hyperhemolysis in numerous diseases. In no case a primary genetically transmitted abnormal erythrocyte membrane protein can be formely demonstrated.
