Abstract We immobilized 47 white rhinoceroses (Ceratotherium simum) for dehorning with 1-4 mg of etorphine HCl, 10-40 mg of azaperone, and 7,500 IU of hyaluronidase, at a game ranch in South Africa in November 2012. Forty-four received butorphanol intravenously 5 min after recumbency, at the rate of 10 mg of butorphanol per 1 mg of etorphine, and three animals did not. When possible, blood gas and physiologic parameters were measured immediately before butorphanol administration and 10 min later. Statistically significant improvements were observed, with a reduction in pH, partial pressure of

Nineteen white rhinoceroses ( Ceratotherium simum ) were anesthetized with 4 mg of etorphine hydrochloride; 35-40 mg of midazolam; and 7,500 international units of hyaluronidase for dehorning purposes at a game ranch in South Africa, to investigate this anesthetic combination. Median time to recumbency was 548 sec (range 361-787 sec). Good muscle relaxation and no muscle rigidity or tremors were observed in 18 animals, and only 1 individual showed slight tremors. In addition, all animals received butorphanol i.v. 5 min after recumbency at the ratio of 10 mg of butorphanol per 1 mg of etorphine

Etorphine is an non-selective opioid receptor agonist with very potent analgesic effect. Low concentrations (< nM) of most opioid receptor agonists decrease the K+ conductance (gK) in cultures of dissociated mouse dorsal root ganglion neurons regardless of the presence of Ba2+ However, low concentrations of etorphine, in contrast to all other opioids tested, decreased gK only in the absence of Ba2+. In the presence of Ba2+, pM-nM etorphine elicited dose-dependent increases, instead of decreases in gK. Higher concentrations of etorphine (> nM) not only increased gK but, in addition, appreciably

Buprenorphine is an opiate drug with a mixed agonist-antagonist profile and has therapeutic efficacy in attenuating drug craving and addiction. Because the adenylyl cyclase system has been implicated in the biochemical basis of opiate withdrawal phenomena, we have compared the acute and chronic effects of buprenorphine with the full opiate agonist etorphine on cyclic AMP (cAMP) synthesis in the human neuroblastoma cell SK-N-SH. Both drugs acutely inhibited prostaglandin (PG)E1-stimulated cAMP accumulation; the inhibition caused by either drug was prevented by pretreatment with the opiate antag

Chemical immobilization of captive European bison (Bison bonasus) is often required for veterinary care, transportation, or husbandry practices playing an important role in conservation breeding and reintroduction of the species. We evaluated the efficiency and physiological effects of an etorphine-acepromazine-xylazine combination with supplemental oxygen in 39 captive European bison. Animals were darted with a combination of 1.4 mg of etorphine, 4.5 mg of acepromazine, and 20 mg of xylazine per 100 kg based on estimated body mass. Arterial blood was sampled on average 20 min after recumbency

Historically, free-ranging musk oxen (Ovibos moschatus) were anesthetized with potent opioids such as etorphine, which are often associated with marked respiratory depression. The goal of this study was to investigate alternatives to the etorphine-xylazine combination (EtXy) traditionally used in this species. First, the potential benefit of postinduction butorphanol administrations to musk oxen anesthetized with EtXy was assessed. Then, the use of butorphanol-azaperone-medetomidine (BAM-II) as an alternative to the use of potent opioids for the immobilization of free-ranging musk oxen was eva

Reversal of opioid-related respiratory depression is often accompanied by an "acute abstinence like syndrome" with hypertension, tachycardia, and pain. This overshoot was used to investigate the extent at which opioids of high potency but different structure are involved in naloxone-induced abstinence. In 10 awake and trained mongrel dogs two highly mu-selective compounds, alfentanil and fentanyl, were given in cumulative doses and at different occasions (30-60-120-240 micrograms/kg, and 6-12-24-48 micrograms/kg, respectively). Subsequently, a high dose of naloxone (100 micrograms/kg) was give

The effects of etorphine on the pulmonary vascular system of white rhinoceros ( Ceratotherium simum) have not been described and could play a role in the severe hypoxemia that develops after immobilization with etorphine-based drug combinations. Characterization of these effects requires measurement of pulmonary vascular pressures and cardiac output (CO). To refine a technique for pulmonary arterial catheterization, five boma-habituated white rhinoceros (three females and two males weighing 1,012-1,572 kg) were immobilized by remote injection with etorphine plus azaperone followed by butorphan

BACKGROUND: Etorphine, a highly potent opioid widely used in wildlife immobilisation, is known to cause cardiorespiratory compromise. This study aimed to investigate the effects of a serotonergic agonist, BIMU-8 and an alpha-2 adrenoreceptor antagonist, vatinoxan on pulmonary hypertension and cardiovascular function in etorphine-immobilised sheep, as a model for wild ungulates. Six sheep were immobilised three times in a randomised, prospective, controlled crossover design using intramuscular etorphine (0.05 mg·kg−1). Seven minutes later, sheep received intravenous BIMU-8 (1.5 mg·kg−1), vatino

BACKGROUND: Opioid-induced respiratory compromise remains a significant challenge in etorphine-immobilised wildlife. Serotonergic agonists offer a potential avenue for preventing or treating opioid-induced respiratory compromise. We therefore aimed to determine whether the selective 5-hydroxytryptamine receptor 4 (5-HT4) agonist, BIMU-8, reverses opioid-induced respiratory compromise in etorphine-immobilised goats.
METHODS: Seven healthy adult goats were immobilised with etorphine, then treated with BIMU-8 or sterile water 5 minutes later in a randomised, prospective cross-over study. Cardior
