Environmental risk factors of type 2 diabetes-an exposome approach.

Type 2 diabetes is one of the major chronic diseases accounting for a substantial proportion of disease burden in Western countries. The majority of the burden of type 2 diabetes is attributed to environmental risks and modifiable risk factors such as lifestyle. The environment we live in, and changes to it, can thus contribute substantially to the prevention of type 2 diabetes at a population level. The 'exposome' represents the (measurable) totality of environmental, i.e. nongenetic, drivers of health and disease. The external exposome comprises aspects of the built environment, the social e

Hidden mysteries behind genome, epigenome, and exposome of lupus erythematosus.

As an extremely heterogeneous disease, lupus erythematosus (LE) occurs when a genetically susceptible individual encounters specific environmental triggers. Epigenetics bridges genetics and environment and helps explain their interactions. Comprehensively understanding the hidden mysteries behind the genome, epigenome, and exposome of lupus could contribute to precision prevention and treatment of lupus.

Exposome contribution to the brain metabolome: importance of body brain connection.

INTRODUCTION: Mounting evidence support exposome influences on brain function and health, complementing genome influences. Understanding the molecular imprint of exposome on brain metabolism and the biochemical communication between the body and brain can impact our fundamental understanding and treatment of neuropsychiatric diseases. METHODS: Leveraging two complementary metabolomics platforms, we classified 1400 features in 514 brains from the ROSMAP collection. We evaluated the origin of these compounds using literature and databases. We correlated those metabolites with cognitive function

Linking the exposome to frailty: pathways, mechanisms, clinical implications, and prevention.

BACKGROUND: Frailty is a state of vulnerability that emerges from cumulative, non-genetic influences acting across the life course. This review applies an exposome lens, integrating general external exposures, specific external exposures, and internal biologic processes. We aimed to synthesize how social, environmental, behavioural, clinical, and biological exposures contribute to frailty and identify clinically actionable prevention targets. METHODS: This structured narrative review synthesized epidemiological, clinical, environmental, and mechanistic evidence linking exposome-related exposu

An integrated workflow for combined organic and inorganic chemical exposome analysis.

Humans are exposed to a multitude of non-genetic factors throughout their lives, all of which are encompassed in the concept of the exposome. To assess its chemical component, combinations of complementary analytical techniques are needed for broad chemical space coverage. However, approaches for combined organic and inorganic analysis remain scarce and are further constrained by limited sample availability (e.g., in historical biobanks or preterm infants), as individual techniques require different sample preparation workflows. Therefore, we developed and evaluated an approach to combine samp

The fetal exposome: cellular and epigenetic programming of neonatal immune function.

Fetal life represents a critical period in the development of the neonatal immune system. During this period of high epigenomic plasticity, environmental exposures, maternal health conditions and perinatal factors can have a lifelong impact on gene expression and function, influencing neonatal immune function and contributing to the later development of immune-mediated disease. We conducted a narrative review of human and translational evidence linking antenatal exposures with cellular and epigenetic regulation of neonatal immune function. Evidence was synthesised across maternal health, envir

Adverse Social Exposome During the Life Course and Vascular Brain Injury.

IMPORTANCE: The adverse social exposome, the entirety of harmful lifetime social-environmental exposures, is associated with Alzheimer disease and related dementias (ADRD) risk and may be key to informing modifiable systemic and behavioral risk interventions. With increasing emphasis to understand lifetime risk, methods are needed to measure and study social exposome relative to ADRD-related biological processes, including cerebrovascular disease. OBJECTIVE: To evaluate the association between adverse social exposome and the burden of vascular brain injury (VBI) post mortem. DESIGN, SETTING,

[Translated article] Multicenter, Prospective, Case-Control Study of Exposome in Melanoma.

UV radiation is the main etiological agent of skin cancer. Other factors such as pollution, diet and lifestyle are also contributing factors. Our aim was to investigate the association between melanoma and exposome factors in the Spanish population with a prospective multicenter case-control study. Case group included patients with melanoma while the control group included people who attended the consultations as companions without a past medical history of skin cancer. A total of 73 melanoma patients and 126 controls were included. The former group included more outdoor workers, a history of

The exposome and liver disease - how environmental factors affect liver health.

Since the initial development of the exposome concept, much effort has been devoted to the characterisation of the exposome through analytical, epidemiological, and toxicological/mechanistic studies. There is now an urgent need to link the exposome to human diseases and to include exposomics in the characterisation of environment-linked pathologies together with genomics and other omics. Liver diseases are particularly well suited for such studies since major functions of the liver include the detection, detoxification, and elimination of xenobiotics, as well as inflammatory responses. It is w

Exposome-wide ranking of modifiable risk factors for cardiometabolic disease traits.

The present study assessed the temporal associations of ~ 300 lifestyle exposures with nine cardiometabolic traits  to identify exposures/exposure groups that might inform lifestyle interventions for the reduction of cardiometabolic disease risk. The analyses were undertaken in a longitudinal sample comprising > 31,000 adults living in northern Sweden. Linear mixed models were used to assess the average associations of lifestyle exposures and linear regression models were used to test associations with 10-year change in the cardiometabolic traits. 'Physical activity' and 'General Health' were

Early-Life Environmental Exposures and Childhood Obesity: An Exposome-Wide Approach.

BACKGROUND: Chemical and nonchemical environmental exposures are increasingly suspected to influence the development of obesity, especially during early life, but studies mostly consider single exposure groups. OBJECTIVES: Our study aimed to systematically assess the association between a wide array of early-life environmental exposures and childhood obesity, using an exposome-wide approach. METHODS: The HELIX (Human Early Life Exposome) study measured child body mass index (BMI), waist circumference, skinfold thickness, and body fat mass in 1,301 children from six European birth cohorts age

Large language models in Hidradenitis suppurativa: strong empathy, weak exposome awareness.

BACKGROUND: Hidradenitis suppurativa (HS) imposes a substantial biopsychosocial burden. As patients increasingly consult large language models (LLMs) for health information, their capacity to provide clinically appropriate, empathic, readable and exposome-aware counseling requires evaluation. OBJECTIVES: To benchmark LLM-generated responses to simulated HS consultations for clinical performance, exposome awareness, quality-of-life (QoL) coverage and readability. Metabolome informs about the chemical exposome and links to brain health.

The metabolome is an intermediate phenotype, summarizing the profile of all small molecules (<1.5 kDa) in biospecimens. The metabolome provides a readout for the net influence of the chemical exposome, diet, gut microbiome, and genome on human health. Metabolic changes observed in exposome studies may thus provide clues about adverse outcome pathways related to cancer, diabetes, heart disease, cognitive impairment and other neurological conditions such as Alzheimer's disease (AD). Whilst the number of human cohort studies including both metabolomic and exposomic profiles is increasing, they ar

The exposome impact on hair health: non-pharmacological management. Part II⋆.

Hair holds a significance that surpasses mere aesthetics, as it plays a pivotal role in our social interactions and contributes significantly to the definition of our self-esteem. Central to this understanding is the concept of the exposome, which encompasses intrinsic elements like genetics and physiological changes, as well as extrinsic factors such as UV radiation, pollution, lifestyle choices, and chemical treatments. These factors may significantly impact hair health and hair aging. Expanding upon the groundwork laid by the first part of this research (Cedirian et al., 2024), this study a

Exposome Signatures of Prescription Medication and Mild Traumatic Brain Injury Sequelae.

INTRODUCTION: Mild traumatic brain injury (mTBI) has been termed the signature wound of active duty service members (ADSM) returning from Operation Iraqi Freedom and Operation Enduring Freedom with estimated incidence rates between 19.5% and 23%, respectively. Locally, mTBI accounted for 9% of all diagnoses at Fort Bragg, NC, between 2016 and 2022. Our study examines long-term effects of medications prescribed to reduce or prevent mTBI complications that may lead to postconcussive syndrome (PCS). METHODS: The Medical Assessment and Readiness System (MARS) housed at Womack Army Medical Center,

Characterization of Chemical Exposome in A Paired Human Preconception Pilot Study.

Parental preconception exposure to synthetic chemicals may have critical influences on fertility and reproduction. Here, we present a robust LC-MS/MS method covering up to 95 diverse xenobiotics in human urine, serum, seminal and follicular fluids to support exposome-wide assessment in reproductive health outcomes. Extraction recoveries of validated analytes ranged from 62% to 137% and limits of quantification from 0.01 to 6.0 ng/mL in all biofluids. We applied the validated method to a preconception cohort of Australian couples (n = 30) receiving fertility treatment. In total, 36 and 38 xenob
