In-Stent Re-Endothelialization Strategies: Cells, Extracellular Matrix, and Extracellular Vesicles.

Arterial stenosis caused by atherosclerosis often requires stent implantation to increase the patency of target artery. However, such external devices often lead to in-stent restenosis due to inadequate re-endothelialization and subsequent inflammatory responses. Therefore, re-endothelialization strategies after stent implantation have been developed to enhance endothelial cell recruitment or to capture circulating endothelial progenitor cells. Notably, recent research indicates that coating stent surfaces with biogenic materials enhances the long-term safety of implantation, markedly diminish

Spatiotemporally Resolved Tracking of Extracellular Vesicle Cargo via Photocatalytic Extracellular Vesicles Protein Tracking (PhoEVT).

Extracellular vesicle (EV)-mediated protein transport has been recognized as a crucial mechanism for intercellular communication and interaction. Proximity labeling has emerged as a promising approach to study the dynamic protein interactions in cellular activity, but tracking extracellular protein transport pathways remains to be further investigated. Here, we employ Ru(bpy)3 2+ (RU) to construct a spatiotemporally resolved Photocatalytic Extracellular Vesicles protein Tracking (PhoEVT) system. Upon 450 nm irradiation, RU generates singlet oxygen to trigger the biotinylation of EV-associated

Mitochondrial extracellular vesicles between pathology and regeneration.

Mitochondria are central regulators of cellular energy production, metabolic homeostasis, and stress responses, and their dysfunction represents a critical hallmark of neurodegenerative and neuroinflammatory diseases. To preserve mitochondrial integrity, cells rely on an intricate mitochondrial quality control system encompassing mitochondrial dynamics, mitophagy, biogenesis, and vesicle-mediated pathways. Emerging evidence highlights the pivotal role of mitochondria-derived vesicles as vehicles for trafficking mitochondrial components within cells, thereby contributing significantly to intrac

Extracellular Vesicles Drive Coagulopathy Across Hematologic Cancers.

Coagulopathies are common in hematological malignancies and can cause life-threatening bleeding or thrombosis. Extracellular vesicles (EVs), including exosomes, microvesicles, and apoptotic bodies, carry cargo that reflects their cellular origin. They frequently express tissue factor (TF) and expose phosphatidylserine (PS), which initiate and amplify coagulation and can also transport fibrinolytic mediators that modulate plasmin generation. Procoagulant EVs are commonly assessed using TF- and PS-dependent functional assays, global hemostasis tests such as overall hemostatic potential, and flow

Extracellular Vesicles in Migraine: Biomarkers and Therapeutics.

The diagnosis and therapeutic evaluation of migraine have long relied on subjective clinical descriptions, lacking objective, stable, and quantifiable biological markers. Extracellular vesicles (EVs), which carry specific bioactive molecules, have been recognized in recent years as a potential breakthrough in research due to their role in intercellular communication, the stability of their contents, and their reflection of pathological states. This article comprehensively evaluates studies on changes in EVs and their contents in migraine patients, discusses methods for isolating and characteri

Kidney-heart crosstalk: the extracellular vesicles connection.

Chronic kidney disease (CKD) is a major public health concern, closely linked to an increased risk of cardiovascular disease (CVD), which remains the leading cause of morbidity and mortality in this population. While traditional risk factors such as hypertension and diabetes are prevalent in CKD, disease-specific mechanisms-including chronic inflammation, oxidative stress, mineral disturbances and the accumulation of uraemic toxins-further amplify cardiovascular vulnerability. In CKD, both the abundance and molecular cargo of circulating extracellular vesicles (EVs) are altered, reflecting the

Prevotella intermedia-Derived Extracellular Vesicles Promote Pulpitis.

Although bacterial infection is the primary etiological factor of pulpitis, the underlying mechanisms remain unclear. Bacterial extracellular vesicles (BEVs) play significant roles in the pathogenesis of infectious diseases independent of bacteria. However, the involvement of BEVs in pulpitis has yet to be explored. In this study, we investigated for the first time the specific mechanisms of action of EVs derived from Prevotella intermedia (Pi) in pulpitis. Our results revealed that Pi-EVs were taken up by macrophages, which amplified their pathogenic effects, thereby inducing inflammation in

Extracellular vesicles and their translational therapeutic potential.

BACKGROUND AND PURPOSE: Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have emerged as a promising cell-free therapy for osteoarthritis (OA). However, their transition into clinical therapeutics is constrained by unstandardized pharmacokinetics and heterogeneous preclinical reporting. EXPERIMENTAL APPROACH: A meta-analysis of preclinical in vivo OA models evaluating MSC-EV therapy was conducted. Study quality was rigorously assessed using MISEV 2023 (Minimal Information for Studies of Extracellular Vesicles, 2023) criteria and the SYRCLE (Systematic Review Centre for Laborator

Gas-powered extracellular vesicles promote bone regeneration.

The signaling gas hydrogen sulfide (H2S) has recently been implicated in the regulation of bone remodeling and the maintenance of periodontal health. Exploring the underlying mechanisms for this regulation holds promise for the development of new treatment strategies to block bone resorption and stimulate bone regeneration. A recent study by Zhou et al. (Bioactive Materials, 2024) showed that treatment with H2S stimulated changes in the extracellular vesicles (EVs) released by M2 macrophages, enhancing their capacity to promote the osteogenic differentiation of mesenchymal stem cells in vitro.

Small Extracellular Vesicles Promote Stiffness-mediated Metastasis.

UNLABELLED: Tissue stiffness is a critical prognostic factor in breast cancer and is associated with metastatic progression. Here we show an alternative and complementary hypothesis of tumor progression whereby physiologic matrix stiffness affects the quantity and protein cargo of small extracellular vesicles (EV) produced by cancer cells, which in turn aid cancer cell dissemination. Primary patient breast tissue released by cancer cells on matrices that model human breast tumors (25 kPa; stiff EVs) feature increased adhesion molecule presentation (ITGα2β1, ITGα6β4, ITGα6β1, CD44) compared wit

Polyelectrolytes Are Effective Cryoprotectants for Extracellular Vesicles.

Extracellular vesicles (EVs) have been widely recognized as a heterogeneous group of membrane-coated submicrometer particles released by different types of cells, including stem cells (SCs). Due to their ability to harbor and transfer bioactive cargo into the recipient cells, EVs have been reported as important paracrine factors involved in the regulation of a variety of biological processes. Growing data demonstrate that EVs may serve as potential next-generation cell-free therapeutic factors. However, clinical application of EVs in tissue regeneration requires the development of standardized

Application of extracellular vesicles in diabetic osteoporosis.

As the population ages, the occurrence of osteoporosis is becoming more common. Diabetes mellitus is one of the factors in the development of osteoporosis. Compared with the general population, the incidence of osteoporosis is significantly higher in diabetic patients. Diabetic osteoporosis (DOP) is a metabolic bone disease characterized by abnormal bone tissue structure due to hyperglycemia and insulin resistance, reduced bone strength and increased risk of fractures. This is a complex mechanism that occurs at the cellular level due to factors such as blood vessels, inflammation, and hypergly

Monocyte derived large extracellular vesicles in polytrauma.

Despite significant progress in the medical field, there is still a pressing need for minimal-invasive tools to assist with decision-making, especially in cases of polytrauma. Our team explored the potential of serum-derived large extracellular vesicles, so called microparticles/microvesicles/ectosomes, to serve as a supportive tool in decision-making in polytrauma situations. We focused on whether monocyte derived large EVs may differentiate between polytrauma patients with internal organ injury (ISS > 15) and those without. Thus, we compared our EV data to soluble biomarkers such as tumour n

Enterococcus faecalis Extracellular Vesicles Promote Apical Periodontitis.

Enterococcus faecalis is an important contributor to the persistence of chronic apical periodontitis. However, the mechanism by which E. faecalis infection in the root canals and dentinal tubules affects periapical tissue remains unclear. Bacterial extracellular vesicles (EVs) act as natural carriers of microbe-associated molecular patterns (MAMPs) and have recently attracted considerable attention. In this study, we investigated the role of EVs derived from E. faecalis in the pathogenesis of apical periodontitis. We observed that E. faecalis EVs can induce inflammatory bone destruction in the

Investigating serum extracellular vesicles in Cystic Fibrosis.

BACKGROUND: Extracellular vesicles (EVs) are emerging as biomarkers of disease with diagnostic potential in CF. With the advent of highly effective modulator therapy, sputum production is less common and there is a need to identify novel markers of CF disease progression, exacerbation and response to therapies in accessible fluids such as serum. METHODS: We used size exclusion chromatography (SEC) to isolate and characterise EVs from the blood of PWCF of different ages and compared to ultracentrifugation (UC). We used nanoparticle tracking analysis to measure the number of EVs present in seru

Extracellular vesicles in cardiomyopathies: A narrative review.

Extracellular vesicles (EVs) are membrane-bound particles released by all cells under physiological and pathological conditions. EVs constitute a potential tool to unravel cell-specific pathophysiological mechanisms at the root of disease states and retain the potential to act as biomarkers for cardiac diseases. By being able to carry bioactive cargo (such as proteins and miRNAs), EVs harness great potential as accessible "liquid biopsies", given their ability to reflect the state of their cell of origin. Cardiomyopathies encompass a variety of myocardial disorders associated with mechanical,
