Extremophilic Fungi from Marine Environments: Underexplored Sources of Antitumor, Anti-Infective and Other Biologically Active Agents.

Marine environments are underexplored terrains containing fungi that produce a diversity of natural products given unique environmental pressures and nutrients. While bacteria are commonly the most studied microorganism for natural products in the marine world, marine fungi are also abundant but remain an untapped source of bioactive metabolites. Given that their terrestrial counterparts have been a source of many blockbuster antitumor agents and anti-infectives, including camptothecin, the penicillins, and cyclosporin A, marine fungi also have the potential to produce new chemical scaffolds a

Microbial ecology of acidic, biogenic gypsum: community structure and distribution of extremophiles on freshly formed and relict sulfate deposits in a hydrogen sulfide-rich cave.

UNLABELLED: Sulfate minerals are abundant on the Martian surface, and many of these evaporite deposits are thought to have precipitated from acidic fluids. On Earth, gypsum (CaSO4•2H2O) and other sulfates sometimes form under acidic conditions, so exploring the extremophilic life that occurs in these mineral environments can help evaluate the astrobiological potential of acid sulfate depositional settings. Here, we characterized the microbial communities associated with acidic gypsum deposits in a sulfuric acid cave, where sulfate precipitation is driven by sulfide-oxidizing bacteria and archa

Genome-scale insights into metabolic streamlining and photosynthetic energy balance in the extremophile green alga Picocystis salinarum (Picocystophyceae, Chlorophyta).

Picocystis salinarum is an early-diverging chlorophyte and the sole described member of the Picocystophyceae, frequently dominating hypersaline and alkaline lakes despite extreme physicochemical constraints. To elucidate the genomic foundations of its ecological success, we generated a fully annotated, chromosome-scale nuclear genome assembly of the type strain originally isolated from a saline pond in San Francisco Bay. The 18.5-Mb genome comprises 30 chromosomal assemblies, exhibits clear diploidy, and contains multiple copies of intact Ty3/Gypsy and Ty1/Copia long terminal repeat retrotrans

Extremophilic Bacteria Improve Germination, Early Vigor, and Oxidative-Stress Status in Phaseolus vulgaris Under Simulated Water Deficit.

Water deficit constrains common bean (Phaseolus vulgaris L.) production in the acidic Andisols of south-central Chile. We evaluated Bacillus haynesii CamB6 (thermotolerant, Chilean hydrothermal spring) and Microbacterium sp. Se63 (psychrotolerant, Antarctic soil), singly and as a two-strain consortium, for in vitro plant-growth-promoting traits and, via seed bacterization, on P. vulgaris cvs. Manteca and Señorita under simulated water deficit (PEG-8000, 0-15% w/v). The strains showed complementary in vitro profiles: Se63 solubilized more phosphate and produced siderophores and lipase, whereas

Studying The Role of Aprotic and Protic Solvents on Extremophilic Laccase From Thermus thermophilus for Solvent-Mediated One-Pot Biocatalysis.

In present times, globally, solvent-mediated biocatalysis is gaining a pivotal role in sustainable biotechnological solutions for organic synthesis. The versatile green catalyst, laccase, a multicopper oxidoreductase, has gained significant importance in aromatic bioconversion in solvent systems. Laccases portray varied behavioral patterns and functionality in solvents. With this interest, the influence of aprotic polar organic solvents and a benchmark out-of-the-ordinary protic solvent, an ionic liquid, i.e., 1-Ethyl-3-methylimidazolium acetate (EmimOAc), on extremophilic laccase from Thermus

Manganese-Induced Production of Antioxidant Polyene Steroids in the Extremophile Fungus Talaromyces fuscoviridis Isolated From Copper-Mine Soil.

The fungus Talaromyces fuscoviridis was isolated from soil collected in a copper-mine tailings basin in Pará State, Brazil, an extreme environment that can induce the production of bioactive secondary metabolites. Extracts and fractions obtained after cultivation were analyzed by liquid chromatography coupled to high-resolution mass spectrometry (LC-HRMS) and compared using Global Natural Products Social molecular networking (GNPS), which revealed a family of seven putative polyunsaturated steroids. Isolation led to the characterization of two major steroids: the known Ergosta-3(4),5(6),7(8),1

Bioinspired Antimicrobial Strategy: An Extremophile Deep Sea Peptide to Combat Cystic Fibrosis Infections Caused by Pseudomonas aeruginosa and Staphylococcus aureus.

Cystic fibrosis (CF)-associated lung infections caused by Pseudomonas aeruginosa (P. aeruginosa) and Staphylococcus aureus (S. aureus) remain difficult to treat due to multidrug resistance and the redox instability of the pulmonary environment, which can impair antibiotic efficacy. In this study, we investigated alvinellacin (ALV), a disulfide-stabilized β-hairpin antimicrobial peptide (AMP) derived from the deep-sea polychaete Alvinella pompejana (A. pompejana), as a potential therapeutic agent naturally adapted to redox-fluctuating conditions. The antibacterial and antibiofilm activities of

Eco-morphological differentiation in Lake Magadi tilapia, an extremophile cichlid fish living in hot, alkaline and hypersaline lakes in East Africa.

Ecological diversification through divergent selection is thought to be a major force during the process of adaptive radiations. However, the large sizes and complexity of most radiations such as those of the cichlids in the African Great Lakes make it impossible to infer the exact evolutionary history of any population divergence event. The genus Alcolapia, a small cichlid lineage endemic to Lakes Magadi and Natron in East Africa, exhibits phenotypes similar to some of those found in cichlids of the radiations of the African Great Lakes. The simplicity within Alcolapia makes it an excellent m

Antioxidant-producing extremophile regulates gut microbiota of black soldier fly larvae to inhibit conjugative transfer mediated by plasmids in municipal sludge.

Oxidative stress induced by heavy metals and antibiotics in municipal sludge inhibits the gut development of black soldier fly larvae (BSFL), compromising pollutant-processing capacity of their gut microbiota. Herein, Deinococcus radiodurans was colonized into the BSFL gut and established an antioxidant cascade regulatory system in concert with the gut microbiota, synergistically reducing the enrichment of antibiotic resistance genes (ARGs) in municipal sludge. Results showed that bioaugmentation with Deinococcus radiodurans scavenges reactive oxygen species and upregulates the expression of G

Extremophilic Acinetobacter strains from high-altitude lakes in Argentinean Puna: remarkable UV-B resistance and efficient DNA damage repair.

High-Altitude Andean Lakes (HAAL) of the South American Andes are almost unexplored ecosystems of shallow lakes. The HAAL are recognized by a remarkably high UV exposure, strong changes in temperature and salinity, and a high content of toxic elements, especially arsenic. Being exposed to remarkably extreme conditions, they have been classified as model systems for the study of life on other planets. Particularly, Acinetobacter strains isolated from the HAAL were studied for their survival competence under strong UV-B irradiation. Clinical isolates, Acinetobacter baumannii and Acinetobacter jo

Mechanisms of microbial hyper-resistance to heavy metals: Cellular metal accumulation, metabolic reorganization, and GroEL chaperonin in extremophilic bacterium Sulfobacillus thermotolerans in response to zinc.

Mine waste disposal in dumps and stockpiles causes environmental pollution, particularly through microbe-assisted acid mine drainage (AMD) generation and groundwater contamination with hazardous heavy metal(loid)s. Metal hyper-resistance in acidophilic microorganisms remains an underexplored intriguing phenomenon. Using a multi-level approach, we provide the first data on extreme zinc resistance mechanisms in Sulfobacillus thermotolerans, recognized as one of the most metal-resistant organisms known. Under high zinc levels, Sb. thermotolerans cells exhibited efficient zinc sorption and low int

Optimized Chemical Extraction Methods of Antimicrobial Peptides from Roots and Leaves of Extremophilic Plants: Anthyllis sericea and Astragalus armatus Collected from the Tunisian Desert.

Extraction methods depend mainly on the chemical nature of the extracted molecule. For these reasons, the selection of the extraction medium is a vital part of obtaining these molecules. The extraction of antimicrobial peptides (AMPs) from extremophile plants is important because of its potential pharmaceutical applications. This work focused on the evaluation of several solvents for the extraction of AMPs from the following two extremophile plants: Astragalus armatus and Anthyllis sericea from southern Tunisia. In order to identify the most efficient solvents and extraction solutions, we used

Protein Intrinsic Disorder and Adaptation to Extreme Environments: Resilience of Chaos.

Extremophiles are organisms adapted not only to survive in harsh conditions but thrive in environments considered extreme or uninhabitable by most life forms. They serve as a living reflection of a popular motto "What doesn't kill you makes you stronger" and show that life on Earth is spread well outside the human-centric comfort zone. In fact, extremophiles, being found in deep crustal and oceanic depths, outer space, highly acidic and basic conditions, extreme temperatures (+122 °C to -20 °C), and in the presence of toxins and high radiation, demonstrate life's remarkable adaptability. Life in blue: copper resistance mechanisms of bacteria and archaea used in industrial biomining of minerals.

Industrial biomining processes to extract copper, gold and other metals involve the use of extremophiles such as the acidophilic Acidithiobacillus ferrooxidans (Bacteria), and the thermoacidophilic Sulfolobus metallicus (Archaea). Together with other extremophiles these microorganisms subsist in habitats where they are exposed to copper concentrations higher than 100mM. Herein we review the current knowledge on the Cu-resistance mechanisms found in these microorganisms. Recent information suggests that biomining extremophiles respond to extremely high Cu concentrations by using simultaneously

Fungi from the extremes of life: an untapped treasure for bioactive compounds.

More than 80% of the Earth surface is consisted of hostile and harsh environments, classified as extreme from an anthropogenic perspective. Microorganisms with acclimatized nature dominate these extreme ecosystems of the biosphere. Survivals in such environments initiate an inductive force leading to the production of noteworthy metabolites having peculiar biochemistry. Recent investigations on extremophilic fungi for unprecedented bioactive compounds emphasize their remarkable potential as sources of new therapeutics. The present review covers the literature published in the last 15 years and

Nonsterile microbial production of chemicals based on Halomonas spp.

The use of extremophile organisms such as Halomomas spp. can eliminate the need for fermentation sterilization, significantly reducing process costs. Microbial fermentation is considered a pivotal strategy to reduce reliance on fossil fuel resources; however, sustainable processes continue to incur higher costs than their chemical industry counterparts. Most organisms require equipment sterilization to prevent contamination, a practice that introduces complexity and financial strain. Fermentations involving extremophile organisms can eliminate the sterilization process, relying instead on cond

Toward Sustainable Polyhydroxyalkanoates: A Next-Gen Biotechnology Approach.

Polyhydroxyalkanoates (PHAs) are biodegradable biopolymers synthesized by microorganisms and serve as sustainable alternatives to petroleum-based plastics. While traditional PHA production relies on refined carbon sources and pure cultures, high costs and scalability challenges limit commercial viability. Extremophiles, particularly halophiles, have emerged as promising candidates for cost-effective, large-scale production of PHAs. Their ability to thrive in extreme environments reduces contamination risks, minimizes the need for sterilization, and lowers operational costs. Advancements in met
