Tizanidine-induced acute severe cystitis in a female taking famotidine.

Cystitis is a possible adverse drug reaction associated with the use of tizanidine. Such cases have been rarely reported in literature because of the difficulty in establishing the causality. However, from a pharmacovigilance point of view, it is better to report such cases of a possible association between a drug's use and potential adverse drug reactions. We report a case of a 26-year-old Asian female on famotidine who presented with acute severe urinary burning after taking tizanidine.

Effect of famotidine on oesophageal sensitivity in gastro-oesophageal reflux disease.

Gastric acid suppression could improve heartburn by healing oesophagitis or by reduction of oesophageal sensitivity to acid. To independently assess changes in oesophageal sensitivity, it would be necessary to study patients with reflux disease but no oesophagitis. The aim of this study was to investigate the effect of acid suppression on oesophageal sensitivity and to assess the time course of any measured effect. Twenty seven patients were recruited, of whom 25 completed the study (14 men and 11 women, mean (SD) age 50 (15) years). All had classic symptoms of gastro-oesophageal reflux but no

[Treatment of mild to moderate reflux esophagitis with the H2-blocker famotidine].

The efficacy of a single daily dose of 40 mg famotidine was compared with that of the same dose administered twice daily for 4 and 8 weeks respectively, in the treatment of reflux oesophagitis of varying severity (oesophagitis stage I to III). A total of 20 patients took part in the trial; the rate of total recovery was 50%. A significant improvement was recorded in a further 10% of the patients, and no change was observed in 30%. In 5% of cases, a deterioration was seen, that is to say that the treatment was discontinued for 1 patient due to adverse reactions. Following a 4 week course of tre

Clinical efficacy of famotidine in the treatment of acid-related diseases: an overview.

The clinical efficacy of famotidine has been clearly shown in the short- and long-term treatment of a number of acid-related diseases such as duodenal and gastric ulcer as well as in the acute management of reflux esophagitis. In addition, famotidine effectively treats anastomotic ulcer and nonulcer dyspepsia. Based on a number of considerations--chemical structure, pharmacokinetics, pharmacodynamics, clinical efficacy, tolerability, and dosage regimen--famotidine represents an end-point in the development of the H2-antagonist group of anti-ulcer agents.

Dose--response effect of famotidine on patterns of gastro-oesophageal reflux.

The present study attempts to assess the alteration in patterns of gastro-oesophageal reflux as assessed by 24-h oesophageal pH monitoring by varying degrees of H2-receptor blockade with famotidine. Subjects were 12 patients with complaints of daily heartburn who demonstrated at least 6% of acid mucosal contact time by 24-h ambulatory oesophageal pH monitoring. All subjects had a positive Bernstein test, and nine of the 12 subjects had erosive oesophagitis. The study was conducted as a double-blind crossover design utilizing 40 mg nocte, 20 mg b.d., and 40 mg b.d. and placebo treatments. Resul

Famotidine in the treatment of gastric and duodenal ulceration: overview of clinical experience.

The new H2 receptor antagonist, famotidine, has been tested in open and double-blind studies in over 2,000 acute duodenal or gastric ulcer patients as well as in the maintenance of chronic duodenal ulcer patients. In early studies, dosages of 20 mg b.i.d. were found to achieve better results than 10 mg b.i.d. and equivalent to 20 mg t.i.d. Trials comparing famotidine to cimetidine, ranitidine, and gefarnate found the new agent to be approximately equivalent to cimetidine and ranitidine and superior to gefarnate in the treatment of acute duodenal ulcer. Similar trials were conducted to compare

Comparative study of nizatidine and famotidine for maintenance therapy of erosive esophagitis.

BACKGROUND: The therapeutic effect of combined administration of prokinetics and histamine H2 receptor antagonists (H2RA) in gastroesophageal reflux disease is reported to be superior to that of monotherapy with H2RA alone. In addition to its acid-suppressing effect, the H2RA nizatidine also has a prokinetic action by suppressing acetylcholine esterase. The present multicenter, randomized controlled study was performed to investigate whether nizatidine is superior to famotidine, which does not suppress acetylcholine esterase activity, in maintenance therapy for erosive esophagitis. In addition

Control of intragastric acidity with over-the-counter doses of ranitidine or famotidine.

BACKGROUND: Histamine H2-receptor antagonists are available over the counter for the treatment of heartburn. AIM: To compare the effects of low doses of ranitidine and famotidine on intragastric acidity in a three-way crossover study. METHODS: Healthy subjects (12 male, 12 female) were dosed on three occasions with single oral doses of placebo, ranitidine, 75 mg, and famotidine, 10 mg, 1 h after lunch. The pH of gastric aspirates was then measured for 20 h. Subjects ate standard meals and snacks. Analysis of variance was used to determine the statistical significance of differences in acidit

Interaction of diazepam with famotidine and cimetidine, two H2-receptor antagonists.

Famotidine is currently under investigation as an H2-receptor antagonist. Eleven healthy male volunteers received a single 10 mg intravenous dose of diazepam on three occasions: once during coadministration of famotidine 40 mg bid, once during coadministration of cimetidine 300 mg qid, and once without other drug treatment (control). Multiple blood samples were drawn during the seven days after each diazepam dose. Diazepam and desmethyldiazepam plasma concentrations were measured by electron capture gas chromatography. There were no significant differences among the three treatment conditions

Interaction of ranitidine and famotidine with guinea pig-isolated parietal cells.

The inhibitory effects of the histamine H2-receptor antagonists, ranitidine and famotidine on histamine-stimulated gastric acid secretion have been studied in guinea pig isolated, enriched parietal cells using the 14C-aminopyrine accumulation technique. The 14C-aminopyrine accumulation curves in response to histamine were shifted towards the right in a parallel fashion by ranitidine, and in a nonparallel fashion by famotidine. The inhibitory effect of ranitidine, but not that of famotidine, was readily reversed by washing the parietal cells. It is concluded that the histamine H2-receptors in g

Famotidine activates the vagus nerve inflammatory reflex to attenuate cytokine storm.

Background. Severe COVID-19 is characterized by pro-inflammatory cytokine release syndrome (cytokine storm) which causes high morbidity and mortality. Recent observational and clinical studies suggest famotidine, a histamine 2 receptor (H2R) antagonist widely used to treat gastroesophageal reflux disease , attenuates the clinical course of COVID-19. Because evidence is lacking for a direct antiviral activity of famotidine, a proposed mechanism of action is blocking the effects of histamine released by mast cells. Here we hypothesized that famotidine activates the inflammatory reflex, a brain-i

Optimization studies on floating multiparticulate gastroretentive drug delivery system of famotidine.

The objective of this study was to optimize floating microballoons of famotidine by the emulsion solvent diffusion technique using central composite design. Formulations F1-F15 were prepared using three independent variables (pH of medium, drug: Eudragit S100 ratio and ethanol : dichloromethane ratio) and evaluated for dependent variables (shape, percentage buoyancy, and encapsulation). The optimized formulation F9 was fractionated and a polymer combination of (Eudragit S100 : Eudragit L100-55, 9.5:0.5) resulted in microballoons that exhibited zero order release (94.73%) with 84.20% buoyancy a

Suppression of nocturnal acid secretion with famotidine accelerates gastric ulcer healing.

Although nocturnal acid secretion has been emphasized in the pathophysiology and treatment of duodenal ulcer, its importance in gastric ulcer disease has been questioned. To explore this area, this multicenter U.S. trial compared the effect of a once-daily nighttime dose of H2-receptor antagonist with placebo on the healing of gastric ulcer and relief of associated symptoms. One hundred fifty-seven patients with endoscopically verified benign gastric ulcers were randomized in a double-blind fashion to either famotidine (40 mg at bedtime) or placebo. Antacid tablets were allowed as needed. The

Physiologically based Pharmacokinetic/Pharmacodynamic Modeling (PBPK/PD) of Famotidine in Pregnancy.

Famotidine, a H2-receptor antagonist, is commonly used to treat heartburn and gastroesophageal reflux disease during pregnancy. However, information on the pharmacokinetics (PK) of famotidine in pregnant patients is limited since pregnant patients are usually excluded from clinical trials. This study aimed to develop and evaluate a physiologically based pharmacokinetic (PBPK) model for famotidine in non-pregnant and pregnant populations, and to combine it with a pharmacodynamic (PD) model to predict the effect of famotidine on intragastric pH. Clinical data for model evaluation were taken from

Famotidine promotes inflammation by triggering cell pyroptosis in gastric cancer cells.

BACKGROUND: Cell pyroptosis has been characterized by cell swelling and pro-inflammatory factors release to aggravate inflammatory reaction., such as interlukin-1 beta (IL-1β) and interlukin18 (IL-18). However, the function of famotidine, an antagonist of histamine H2-receptor antagonists, in cell pyroptosis remained unknown. METHODS: Real-time quantitative PCR (qPCR), western blotting (WB), LDH release assay and enzyme linked immunosorbent assay (Elisa) combined with inhibitor were performed to analyze the effect of famotidine on cell pyroptosis-related gene expression. Dispersive Liquid-Liquid Microextraction of Famotidine and Nizatidine from Water Samples.

A new extraction method was developed for isolation of the antihistaminic drugs (histamine H2 receptor antagonists) famotidine (FMT) and nizatidine (NZT) from aqueous samples. A low solvent consumption method using dispersive liquid-liquid microextraction (DLLME) was applied for this purpose. The parameters of the isolation, namely, volume of extraction and disperser solvents, pH of sample and shaking time were optimized. The influence of foreign substances on the extraction process was also studied. After separation of FMT and NZT using DLLME, the analytes in organic extracts were determinate
