Proteomic and miRNA profiling of radon-induced skin damage in mice: FASN regulated by miRNAs.

Radon is a naturally occurring radioactive gas and considered as a serious carcinogen to humans. Continuous radioactive decay of this gas emits high-energy alpha particles. Long-term radon exposure induces oxidative stress and inflammatory response, which results in chronic lung diseases. However, biological effects after radon exposure in other organs have been rarely reported. As the outermost organ of the human body, the skin suffers from environmental damage to agents such as air pollution. Epidemiological studies indicated that areas with high level of radon had a high incidence of skin c

Targeting Fatty Acid Synthase With Berberine Promotes IL-10 Production in Macrophages and Prevents Relapse in a Murine Colitis Model.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder characterized by alternating periods of remission and relapse. Current therapeutic options for maintaining long-term remission are limited, and no established animal model exists for evaluating relapse. We hypothesized that enhancing interleukin-10 (IL-10) production in intestinal macrophages could serve as a novel therapeutic strategy. Through the screening of a library of natural compounds derived from medicinal herbs, we identified berberine as a promising IL-10 enhancer. This study aimed to develop an experimental relapse

Acetylation of fatty acid synthase regulates microglial lipid droplets accumulation and pro-inflammatory activity following traumatic brain injury.

Lipid droplet accumulation in microglia has been implicated in inflammatory functions associated with aging and demyelinating diseases. However, the molecular mechanisms driving lipid droplet formation under pathological conditions remain unrevealed. It is demonstrated herein that the acetylation of fatty acid synthase (FASN) plays a key regulatory role in the accumulation of lipid droplets in microglia following traumatic brain injury (TBI). Through mass spectrometry analysis, we identified hyperacetylation at lysine K673 of FASN as a critical driver of lipid droplet formation in microglia. N

Ovarian multi-omics analysis reveals key rate-limiting enzymes FASN, SCD5, FADS1, 3BHSD, and STAR as potential targets for regulating kidding traits in goats.

The kidding traits of goats are an important index of production. However, the molecular regulatory mechanisms of kidding traits in goats have not been fully elucidated. This study aimed to investigate the molecular regulatory network of kidding traits in goats. Multi-omics revealed the enrichment of 10 signaling pathways, with fatty acid biosynthesis, biosynthesis of unsaturated fatty acids, and steroid hormone biosynthesis pathways being closely related to reproduction. Interestingly, the key rate-limiting enzymes, fatty acid synthase (FASN), stearoyl-CoA desaturase 5 (SCD5), fatty acid desa

Morphing Natural Product Platensimycin via Heck, Sonogashira, and One-Pot Sonogashira/Cycloaddition Reactions to Produce Antibiotics with In Vivo Activity.

Type II fatty acid synthases are promising drug targets against major bacterial pathogens. Platensimycin (PTM) is a potent inhibitor against β-ketoacyl-[acyl carrier protein] synthase II (FabF) and β-ketoacyl-[acyl carrier protein] synthase I (FabB), while the poor pharmacokinetics has prevented its further development. In this work, thirty-two PTM derivatives were rapidly prepared via Heck, Sonogashira, and one-pot Sonogashira/cycloaddition cascade reactions based on the Gram-scale synthesis of 6-iodo PTM (4). About half of the synthesized compounds were approximately equipotent to PTM agains

Fermented black radish (Raphanus sativus L. var. niger) attenuates methionine and choline deficient diet-induced nonalcoholic fatty liver disease in mice.

As one of the wide-ranging form of chronic liver disease, there are only limited therapeutic options for nonalcoholic fatty liver disease (NAFLD). We evaluated whether fermented black radish (Raphanus sativus L. var. niger; FBR) ameliorates lipid accumulation, inflammation, and hepatic fibrosis, which are characteristics of the pathogenesis of NAFLD. Fermented black radish treatment reduced lipid accumulation in 3T3-L1 adipocytes, which appeared to be associated with the downregulation of adipogenic transcription factors, including sterol regulatory element-binding protein 1c, CCAAT/enhancer-b

A missense mutation in the fabB (beta-ketoacyl-acyl carrier protein synthase I) gene confers tiolactomycin resistance to Escherichia coli.

Thiolactomycin (TLM) is an antibiotic that inhibits bacterial type II fatty acid synthesis at the condensing enzyme step, and beta-ketoacyl-acyl carrier protein synthase I (FabB) is the relevant target in Escherichia coli. TLM resistance is associated with the upregulation of efflux pumps. Therefore, a tolC knockout mutant (strain ANS1) was constructed to eliminate the contribution of type I secretion systems to TLM resistance. Six independent TLM-resistant clones of strain ANS1 were isolated, and all possessed the same missense mutation in the fabB gene (T1168G) that directed the expression o

Fatty acid synthase promotes Hantaan virus infection through lipogenic regulation in the tree shrew model.

Hantaan virus (HTNV) is the principal etiological agent of hemorrhagic fever with renal syndrome (HFRS) in Asia. To date, however, progress in defining mechanisms of host-pathogen interaction has been limited by the absence of an appropriate animal model. In the present study, the tree shrew ( Tupaia belangeri) was shown to be permissive to HTNV infection and to recapitulate key renal pathology and immunological features observed in patients with HFRS. Following intranasal challenge, viral replication occurred predominantly in the kidney, spleen, and lung, accompanied by tubular dilation, glom

Mitochondrial dysfunction and lipid homeostasis.

This review is aimed at illustrating that mitochondrial dysfunction and altered lipid homeostasis may concur in a variety of pathogenesis states, being either contributive or consecutive to primary disease events. Underlying mechanisms for this concurrence are far from being the exhaustive elements taking place in disease development. They may however complicate, contribute or cause the disease. In the first part of the review, physiological roles of mitochondria in coordinating lipid metabolism and in controlling reactive oxygen species (ROS), ATP and calcium levels are briefly presented. In

FASN-mediated fatty acid biosynthesis remodels immune environment in Clonorchis sinensis infection-related intrahepatic cholangiocarcinoma.

BACKGROUND & AIMS: Intrahepatic cholangiocarcinoma (iCCA) is the second most common primary liver cancer and is highly lethal. Clonorchis sinensis (C. sinensis) infection is an important risk factor for iCCA. Here we investigated the clinical impact and underlying molecular characteristics of C. sinensis infection-related iCCA. METHODS: We performed single-cell RNA sequencing, whole-exome sequencing, RNA sequencing, metabolomics and spatial transcriptomics in 251 patients with iCCA from three medical centers. Alterations in metabolism and the immune microenvironment of C. sinensis-related iCC

CCT2 Promotes Prostate Cancer Progression Through EIF3F-Dependent Stabilization of FASN.

Prostate cancer (PCa) is increasingly recognized to be driven by dysregulated lipid metabolism. Although fatty acid synthase (FASN) is highly expressed in PCa, the mechanisms governing FASN protein stability and its functional integration into oncogenic lipid metabolism remain poorly defined. In this study, we identified chaperonin-containing TCP1 subunit 2 (CCT2) as a key oncogenic regulator that promotes lipid synthesis and enhances malignant phenotypes both in vitro and in vivo. Mechanistically, CCT2 transcription is upregulated by the transcription factor Forkhead Box A1 (FOXA1); the CCT2

Elevated hydrostatic pressure dysregulates lipid metabolism of hepatocytes.

Elevation of interstitial fluid hydrostatic pressure is observed in the early stages of fatty liver disease. We herein examined the hypothesis that elevated hydrostatic pressure dysregulates lipid metabolism in hepatocytes and thereby contributes to the progression of fatty liver disease. Human hepatocytes were exposed to 20 and 50 mmHg hydrostatic pressure for 1 and 24 h. RT-qPCR data on the expression of fatty acid metabolism-associated genes suggested the alteration of lipid metabolism. Although we did not observe an obvious change of cell growth, the accumulation of lipid droplets increase

Inhibition of Fatty Acid Synthase (FASN) synergistically enhances the efficacy of 5-fluorouracil in breast carcinoma cells.

The lipogenic enzyme fatty acid synthase (FASN) is differentially overexpressed and hyperactivated in a biologically aggressive subset of breast carcinomas and minimally in most normal adult tissues, rendering it an interesting target for anti-neoplastic therapy development. We previously reported that the FASN blockade can induce a synergistic chemosensitization of breast cancer cells to microtubule interfering agents (MIAs) such as docetaxel, paclitaxel and vinorelbine. Upon pharmacological inhibition of FASN activity using the natural antibiotic cerulenin [(2S,3R)-2,3-epoxy-4-oxo-7E,10E-dod

Anthraquinone-Based Electron Reservoir-Pump Photosensitizer Generating Radical Ion Pairs for Metabolism-Targeted Photodynamic Therapy.

Type I photodynamic therapy (PDT) overcomes oxygen (O2) dependence of type II PDT, but developing hypoxia-efficient electron-transfer photosensitizers remains challenging. Herein, we proposed an anthraquinone (AQ)-based single-component "electron reservoir-pump" strategy to enhance the electron transfer ability of type I photosensitizers. In this molecular design, the AQ scaffold functioned as an intrinsic electron reservoir, while the electron-rich tetraphenylethylene (TPE) served as an electron pump that actively donated electrons. The synergistic reservoir-pump interaction enabled the photo

FASN-mediated metabolic reprogramming drives CD4+ T cell hyperactivation in Sjögren's syndrome via fatty acid oxidation-dependent oxidative phosphorylation.

BACKGROUND: Sjögren's syndrome (SS) is a chronic systemic autoimmune disease in which CD4+ T cells play a critical role. Recent advances in immunometabolism suggest that metabolic reprogramming contributes to autoimmune pathogenesis. This study investigates the role of fatty acid synthase (FASN) in CD4+ T cell dysfunction in SS. METHODS: Peripheral blood CD4+ T cells were isolated from SS patients and healthy controls. FASN expression was assessed via PCR, Western blot, and immunofluorescence. Functional and metabolic assays, including flow cytometry, Seahorse analysis, transcriptomic profili

Cigarette smoke induces FASN-dependent fatty acid metabolic rewiring to drive bladder cancer progression.

Cigarette smoke promotes bladder tumor growth by enhancing cancer cell survival and proliferation through smoke mediated carcinogens. FASN, a key enzyme in fatty acid synthesis, is dysregulated in many cancers and correlates with aggressive phenotypes. In this study, we demonstrate elevated fatty acid levels and FASN specifically in smokers with bladder cancer. Elevated FASN under smoke exposure imparted epigenetic alterations, particularly histone acetylation, impacts DNA repair and DNA-binding transcription factors which regulate metabolic pathways. Under cigarette smoke, bladder cancer cell

Creating New Access Points for Strategic Synthase Engineering: Ethane Thioester Analogs as Truncated Coenzyme A Building Blocks Unlock Expanded Acyl Carrier Protein Loading Capabilities.

Strategic engineering of natural product biosynthetic pathways through the incorporation of alternative, tunable carbon-based building blocks represents a promising approach for accessing medicinally relevant molecules. However, efforts toward this goal have been hindered by the substrate specificity of component enzymes. In type I and type II fatty acid synthases (FASs) and polyketide synthases (PKSs), the acyltransferase (AT) selects a specific malonyl-based coenzyme A (CoA) building block and transfers it onto the acyl carrier protein (ACP) for subsequent processing. Inspired by the observa

METTL3 inhibits hepatic insulin sensitivity via N6-methyladenosine modification of Fasn mRNA and promoting fatty acid metabolism.

Type 2 diabetes (T2D) is characterized by lack of insulin, insulin resistance and high blood sugar. However, the underlying mechanisms of insulin resistance during T2D development remains unclear. As the most common mRNAs modification, N6-Methyladenosine (m6A) is involved in many of pathological processes in aging disease. However, it remains unclear whether m6A is involved in T2D development and what is the regulatory mechanism. This study is aimed to illustrate the roles of m6A and its methyltransferase METTL3 in the regulation of blood glucose homeostasis and insulin sensitivity. The result
