DIMBOA and melatonin mediate a recruitment feedback loop between wheat and Bacillus pseudomycoides to alleviate herbicide stress.

Herbicide residues disrupt soil-plant-microbe interactions and threaten agricultural sustainability, yet how plants actively recruit beneficial microbes to mitigate stress remains unclear. Here, we uncover a metabolite-mediated recruitment model in which wheat secretes DIMBOA and melatonin in response to bensulfuron-methyl (BM) stress to recruit Bacillus pseudomycoides A3, with cross-sectional evidence supporting a self-reinforcing feedback architecture. Chemotaxis assays confirmed that DIMBOA and melatonin act as potent chemoattractant for strain A3 at environmentally relevant concentrations.

Synthetic gene network restoring endogenous pituitary-thyroid feedback control in experimental Graves' disease.

Graves' disease is an autoimmune disorder that causes hyperthyroidism because of autoantibodies that bind to the thyroid-stimulating hormone receptor (TSHR) on the thyroid gland, triggering thyroid hormone release. The physiological control of thyroid hormone homeostasis by the feedback loops involving the hypothalamus-pituitary-thyroid axis is disrupted by these stimulating autoantibodies. To reset the endogenous thyrotrophic feedback control, we designed a synthetic mammalian gene circuit that maintains thyroid hormone homeostasis by monitoring thyroid hormone levels and coordinating the exp

The vitamin rheostat: A unified model of epigenetic feedback in vitamin homeostasis.

Recent evidence implicates epigenetic mechanisms in vitamin metabolism, yet most studies remain focused on individual vitamins. In this mini-review, we extend beyond single-vitamin paradigms to propose the Vitamin Rheostat: a unifying model in which vitamins influence regulatory signaling networks that shape their own uptake, utilization, and degradation. By integrating fat-soluble vitamins (nuclear receptor-mediated signaling) and water-soluble vitamins (methyl-donor and enzymatic cofactor systems), this model provides a predictive and experimentally testable framework that may help explain i

The Mechano-Immune-Vascular Activation Loop: An Integrative Conceptual Framework for Positive Feedback in Hypertension.

Hypertension is increasingly recognized as involving chronic inflammation beyond its hemodynamic origins. This review proposes the Mechano-Immune-Vascular Activation Loop as an integrative conceptual framework-a positive feedback circuit in which aberrant mechanical forces in the hypertensive vascular wall may activate immune cells through mechanosensitive sensors, potentially triggering inflammation that drives vascular stiffening and amplifies mechanical stress. We examine molecular force sensors including piezo-type mechanosensitive ion channel component 1 (Piezo1), transient receptor poten

Integrated Feedforward and Feedback Mechanisms in Neurovascular Coupling.

Neurovascular coupling (NVC) is the mechanism that drives the neurovascular response to neural activation, and NVC dysfunction has been implicated in various neurologic diseases. NVC is driven by (1) nonmetabolic feedforward mechanisms that are mediated by various signaling pathways and (2) metabolic feedback mechanisms that involve metabolic factors. However, the interplay between these feedback and feedforward mechanisms remains unresolved. We propose that feedforward mechanisms normally drive a swift, neural activation-induced regional cerebral blood flow (rCBF) overshoot, which floods the

Unilateral transfemoral amputees exhibit altered strength and dynamics of muscular co-activation modulated by visual feedback.

Objective.Somatosensory perception is disrupted in patients with a lower limb amputation. This increases the difficulty to maintain balance and leads to the development of neuromuscular adjustments. We investigated how these adjustments are reflected in the co-activation of lower body muscles and are modulated by visual feedback.Approach.We measured electromyography (EMG) signals of muscles from the trunk (erector spinae and obliquus external), and the lower intact/dominant leg (tibialis anterior and medial gastrocnemius) in 11 unilateral transfemoral amputees and 11 age-matched able-bodied co

Regulation of energy metabolism by inflammation: a feedback response in obesity and calorie restriction.

Caloric restriction (CR), in the absence of malnutrition, delays aging and prevents aging-related diseases through multiple mechanisms. A reduction in chronic inflammation is widely observed in experimental models of caloric restriction. The low inflammation status may contribute to the reduced incidence of osteoporosis, Alzheimer's disease, cardiovascular diseases and cancer in the aging subjects. The association of caloric restriction with low inflammation suggests a role of energy accumulation in the origin of the chronic inflammation. This point is enforced by recent advances in obesity re

From synaptic guardian to neurodegenerative culprit: rewiring the amyloid-β feedback loop in Alzheimer's disease.

Studies of amyloid-β (Aβ) in Alzheimer's disease pathology have revealed the peptide's complex roles in synaptic function. The study by Siddu et al. in this issue clarifies the contexts in which Aβ peptides may be synaptogenic or synaptotoxic. This commentary integrates the study's major findings with the salient findings of others that, over recent years, have redefined Aβ from a troublesome waste product into a physiological agent of the innate immune response and a modulator of synaptic homeostasis. Convergent evidence demonstrates how free, nonaggregated Aβ supports synaptic structure and

The role of the bone myoregulation reflex in impact-loading activities: a new view of feedback control mechanisms.

BACKGROUND: The short-latency reflex (SLR), which occurs immediately after ground contact during jumping, is traditionally attributed to a muscle spindle-mediated stretch reflex, with a longer latency explained by slow muscle stretching. However, emerging evidence suggests that the bone myoregulation reflex (BMR) may provide a more physiologically parsimonious and biomechanically integrated explanation for this response. OBJECTIVES: This study compared the latencies of these reflexes and assessed the mechanical stimulus transmission delay to the muscle during impact. METHODS: Two experiments

Intrinsic regulation of microvascular tone by myoendothelial feedback circuits.

The endothelium is an important regulator of arterial vascular tone, acting to release nitric oxide (NO) and open Ca2+-activated K+ (KCa) channels to relax vascular smooth muscle cells (VSMCs). While agonists acting at endothelial cell (EC) receptors are widely used to assess the ability of the endothelium to reduce vascular tone, the intrinsic EC-dependent mechanisms are less well characterized. In small resistance arteries and arterioles, the presence of heterocellular gap junctions termed myoendothelial gap junctions (MEGJs) allows the passage of not only current, but small molecules includ

Interface between metabolic balance and reproduction in ruminants: focus on the hypothalamus and pituitary.

This article is part of a Special Issue "Energy Balance". The interface between metabolic regulators and the reproductive system is reviewed with special reference to the sheep. Even though sheep are ruminants with particular metabolic characteristics, there is a broad consensus across species in the way that the reproductive system is influenced by metabolic state. An update on the neuroendocrinology of reproduction indicates the need to account for the way that kisspeptin provides major drive to gonadotropin releasing hormone (GnRH) neurons and also mediates the feedback effects of gonadal s

Quantitative analyses of principal and secondary compound parieto-occipital feedback pathways in cat.

The purpose of our study was to quantify the magnitude of principal and secondary pathways emanating from the middle suprasylvian (MS) region of visuoparietal cortex and terminating in area 18 of primary visual cortex. These pathways transmit feedback signals from visuoparietal cortex to primary visual cortex. (1) WGA-HRP was injected into area 18 to identify inputs from visual structures. In terms of numbers of neurons, feedback projections to area 18 from MS sulcal cortex (areas PMLS, AMLS and PLLS) comprise 26% of inputs from all visual structures. Of these neurons, between 21% and 34.9% ar

Non-invasive somatotopic proprioceptive feedback for closed-loop robotic hand aperture control.

Objective.Current robotic prostheses developed for individuals with transradial amputation often lack physiological feedback, particularly proprioceptive information, which limits control precision and increases reliance on vision.Approach.This study investigates the effect of introducing non-invasive somatotopic feedback using transcutaneous electrical nerve stimulation (TENS) to convey hand aperture information, as an artificial form of proprioceptive feedback. Twenty healthy participants with intact limbs were divided into two groups: one receiving TENS-based feedback and one without feedba

Development of a biofeedback tilt-table for investigating orthostatic syncope in patients with spinal cord injury.

The purpose of this study was to develop a biofeedback tilt-table for automatic tilt-table training, helping patients with spinal cord injury (SCI) to recover more rapidly from orthostatic hypotension, and increasing safety to avoid syncope during training. This biofeedback tilt-table implemented automatic training maneuvers and included three closed feedback loops to monitor the acquisition of physiological signals from patients and the feedback of presyncope symptoms (PS) to regulate the angle of tilt. The results of clinical testing revealed that the mean blood pressure and oxygen saturatio

Effects of Wearable Continuous Normal-Force Tactile Feedback Applied to the Fingertip on Standing Balance in Healthy Young Adults.

Wearable balance-assistance systems predominantly rely on vibrotactile cues that are well suited for transient event detection but may provide a physiologically mismatched representation for the slow, quasi-static postural sway during quiet standing. To address this gap, this study investigates the effects of wearable continuous normal-force feedback on standing balance. We developed a real-time closed-loop system that maps mediolateral center-of-pressure deviation ( $\Delta $ COP ${}_{x}\text {)}$ to a continuous normal-force cue on fingertip using a dead zone-based piecewise linear algorithm

A physiologically inspired hybrid CPG/Reflex controller for cycling simulations that generalizes to walking.

Predictive simulations based on explicit, physiologically inspired, control policies, can be used to test theories on motor control and to evaluate the effect of interventions on the different components of control. Several control architectures have been proposed for simulating locomotor tasks, based on fully feedback, reflex-based, controllers, or on feedforward architectures mimicking the Central Pattern Generators. Hybrid architectures integrating both feedback and feedforward components represent a viable alternative to fully feedback or feedforward controllers. Current literature on cont

Effects of Visual Feedback Availability on Aerobic Performance and Pacing Strategy During a 5 km Running Time Trial.

Running is a widely practiced exercise modality in which central and peripheral fatigue can influence performance and pacing strategy. This study investigated the influence of cognitive-emotional factors, based on the psychobiological model of fatigue, on 5 km time trial performance using a randomized crossover design. Twenty-two recreational male runners (23.0 ± 3.05 years) completed four laboratory visits. During the first visit, participants underwent body composition assessment and an incremental test to determine maximal oxygen uptake (VO2max) and the velocity associated with VO2max. In t
