Pathological findings in a case of hypoxic myoclonus treated with 5-hydroxytryptophan and a decarboxylase inhibitor.

A 72-year-old woman suffered a respiratory arrest following intoxication with barbiturates. Her examination 27 months after the anoxic incident revealed involuntary jerks of trunk and limb muscles triggered by willed movements. On a regimen of 1 g L-5-HTP and 100 mg l-alpha-methyldopa hydrazine (carbidopa), action myoclonus disappeared completely. This medication had to be discontinued because of a regressive hysterical reaction. Two months later, she was found unconscious; resuscitation efforts were unsuccessful. Autopsy showed death was caused by choking on food. Coronal slices of the cerebr

Increased 5-HT2 receptor number in brain as a probable explanation for the enhanced 5-hydroxytryptamine-mediated behaviour following repeated electroconvulsive shock administration to rats.

Following electroconvulsive shock (ECS) administration daily for 10 days there was an increase (35%) in 5-hydroxytryptamine2 (5-HT2) receptor number in rat frontal cortex 24 h after the last ECS, compared with handled controls. A similar increase was seen after intermittent ECS administration (5 ECS over 10 days) given during halothane anaesthesia, compared with halothane-treated controls. The dissociation constant was also increased at this time. A single ECS had no effect. Treatment of rats with pentylenetetrazol, p-chlorophenylalanine or alpha-methyl p-tyrosine during the intermittent ECS a

Plasma amino acids in patients with the carcinoid syndrome.

Plasma tryptophan (Trp) is reported to be decreased in some patients with the carcinoid syndrome. To determine if the plasma levels of other amino acids are also altered in the carcinoid syndrome, we used a fas-liquid chromatographic method to determine the plasma amino acid concentration of nine patients with the carcinoid syndrome and nine age-matched healthy control subjects. In comparison to the control subjects, the patients with the carcinoid syndrome had decreased plasma concentration of valine (Val), isoleucine (Ile), lysine (Lys), and ornithine (Orn), and an increased plasma concentra

The regulation of phenylalanine hydroxylase in rat tissues in vivo. The maintenance of high plasma phenylalanine concentrations in suckling rats: a model for phenylketonuria.

Maximum inhibition of phenylalanine hydroxylase activity in the liver (85%) and in the kidney (50%) of suckling rats required the administration of over 9 mumol of p-chlorophenylalanine/10g body weight. Despite the decrease in the total activity from 184 to 34 units per 10g body weight, the injection of as much as 26 mumol of phenylalanine was required for its concentration in plasma to be still considerably elevated 12h later. In rats injected with p-chlorophenylalanine every 48h and with phenylalanine every 24h from 3 to 18 days of age, the hepatic and renal phenylalanine hydroxylase remaine

Serotonin disruption at gestation alters expression of genes associated with serotonin synthesis and reuptake at weaning.

RATIONALE: Serotonin (5-HT) is a monoamine neuromodulator that plays a key role in the organization of the central nervous system. 5-HT alterations may be associated to the emergence of social deficits and psychiatric disorders, including anxiety, depression, and substance abuse disorders. Notably, disruption of the 5-HT system during sensitive periods of development seems to exert long-term consequences, including altered anxiety responses and problematic use of alcohol. OBJECTIVE: We analyzed, in mice, the effects of transient 5-HT depletion at gestation (a developmental stage when medial p

Brain serotonin depletion attenuates diabetogenic effects of streptozotocin.

The diabetogenic effects of streptozotocin (STZ) were studied on blood glucose, plasma insulin, feeding and drinking, body weight, islet morphology, and hypothalamic serotonin (5-HT) release in vehicle-pretreated rats and in rats pretreated with either intracerebroventricular injection of 5,7-dihydroxytryptamine (5,7-DHT; a 5-HT nerve fiber depletor), intraperitoneal injection of p-chlorophenylalanine (PCPA; a tryptophan hydroxylase inhibitor), or intraperitoneal injection of p-chloroamphetamine (PCA; a neurotoxin for 5-HT nerve fiber). At four days after STZ administration, vehicle-treated ra

Serotonin is necessary for short-term modulation of the exercise ventilatory response.

The exercise ventilatory response is augmented during conditions of increased respiratory dead space (delta Vd), a phenomenon that we refer to as short term modulation (STM). To test the hypothesis that serotonin is necessary in the mechanism underlying STM, experiments were conducted on ten awake goats. Ventilation, CO2 production and PaCO2 were measured at rest and during treadmill exercise (4 km/h, 5% grade), with and without delta Vd (0.25 L), before and after systemic administration of the serotonin receptor antagonist, methysergide maleate (n = 6; 1 mg/kg, i.v.), or the tryptophan hydrox

Female sexual behaviors in male rats with dorsal raphe nucleus lesions: treatment with p-chlorophenylalanine.

The effects of the serotonin-synthesis inhibitor, p-chlorophenylalanine (PCPA) on female sexual behaviors were examined in male rats with or without lesions (DRL) of the dorsal raphe nucleus, which contains a large number of serotonergic cell bodies. Estrogen-primed castrated males without brain surgery (control) showed extremely low levels of lordosis compared with females. On the other hand, DRL males displayed lordosis response more frequently than control males, but the lordosis quotient (LQ) in this group was lower than that in females. As well as DRL males, all PCPA-treated males showed

Roles of the pontine dorsomedial tegmentum and midbrain central gray in regulating female rat sexual behaviors: effects of p-chlorophenylalanine.

Lesions in the midbrain dorsal central gray (MCGL) markedly suppressed both lordosis and soliciting behavior in estradiol benzoate-progesterone (EB-P)-primed castrated female rats. Similarly, the pontine dorsomedial tegmental lesions (PDMTL) caused a severe loss of lordosis behavior. However, the PDMTL females while demonstrating no lordotic response displayed ear-wiggling. These results suggest that the midbrain dorsal central gray plays an important role in regulation of both lordosis and soliciting behaviors. In contrast, the pontine dorsomedial tegmentum appears to participate in regulatin

[Mechanism of action of the analgesic flupirtine].

To answer the questions of mode and site of action partly supplementary, partly new investigations with flupirtine (Katadolon) were carried out which are described below. The investigation for opiate receptor affinity of flupirtine in rat brain homogenate did not show any reduction in 3He-etorphine binding up to the highest concentration of flupirtine of 10(-5) mol/1. This result suggests that flupirtine either has a very low opiate receptor affinity or lacks it fully. Therefore the analgesic activity of flupirtine is not based on opiate mechanism. The intracerebroventricular and intrathecal a

Differential analgetic actions of amphetamine enantiomers in the mouse: a drug-drug interaction study.

A series of agents were tested for their ability to interact with the analgetic actions of either d-amphetamine (d-AMP) or l-amphetamine (l-AMP), or morphine in rats using the hot plate procedure. The analgetic action of l-AMP was potentiated by morphine and slightly antagonized by naloxone; it was antagonized by clonidine, alpha-MT, yohimbine, fluoxetine and metergoline, but enhanced by PCPA. The analgetic action of morphine was antagonized by both naloxone and PCPA. The analgetic actions of l-AMP and morphine appear to involve different biogenic amine systems.

Modification of rat brain 5-hydroxytryptamine metabolism by sublethal doses of organophosphate agents.

The early and late effects of sublethal doses of two organophosphate agents (paraoxon and soman) on 5-hydroxytryptamine (5-HT) metabolism were investigated in several rat brain areas. Parallel determinations of acetylcholinesterase (AcChE) inhibition were performed. An increase in 5-HT level was observed during the first phase of soman intoxication and a rise in 5-hydroxyindol acetic acid (5-HIAA) appeared in the early and late effects of both anticholinesterase agents with a predominant action in the striatum. These data suggested that paraoxon and soman induce a long-lasting increase in 5-HT

Role of serotonin (5-HT) in tolerance to ethanol and barbiturates.

Previously it has been shown that chronic administration of p-chlorophenylalanine (p-CPA), slowed the development of tolerance to ethanol, pentobarbital and cross-tolerance development to ethanol in rats chronically treated with pentobarbital. These findings have been extended by the following observations: (1) p-CPA slowed the development of tolerance to barbital as measured by motor impairment on the moving belt test, without altering the acute response. (2) p-CPA also reduced the tolerance to barbital as measured by sleeping time, in animals chronically treated with pentobarbital. (3) Admin

Effect of humoral modulators of morphine-induced increase in locomotor activity of mice.

The effect of humoral modulators on the morphine-induced increase in locomotor activity of mice was studied. The subcutaneous administration of 10 mg/kg of morphine-HC1 produced a marked increase in locomotor activity in mice. The morphine-induced hyperactivity was potentiated by scopolamine and attenuated by physostigmine. In contrast, both methscopolamine and neostigmine, which do not penetrate the blood-brain barrier, had no effect on the hyperactivity produced by morphine. Pretreatment of mice with alpha-methyltyrosine (20 mg/kg i.p., one hour), an inhibitor of tyrosine hydroxylase, signif

Interspecies aggressiveness of rats towards mice after the application of p-chlorophenylalanine.

The administration of pCPA (400 mg/kg) induces in 30% of Wistar rats the potentiation of interspecies aggressiveness and develops in them the instinct to kill mice. It can be supposed that 5-HT system inhibits aggressive behaviour in this type of aggressiveness. Applying mouse killing test it has been stated that cholinergic receptor blockers (atropine, scopolamine) suppress the predatory instinct towards mice in killer rats and the pilocarpine, the cholinomimetic, develops such instinct in rats which previously did not kill mice. The results of the mouse killing test indicate that cholinergic

Subcellular storage organelles for 5-hydroxytryptamine in parafollicular cells of drugs which deplete the amine.

A study was made of the effect of the administration of reserpine and parachlorophenylalanine, an inhibitor of 5-hydroxytryptamine (serotonin; 5-HT), on the capacity of thyroid parafollicular cells to synthesize and store 5-HT. The two drugs were given to nonhibernating bats in doses which produced an equivalent degree of depletion of 5-HT from the thyroid. Tritiated 5-hydroxytryptophan, the precursor of 5-HT, was then given intravenously to assess the ability of parafollicular cell granules to take up and retain newly synthesized 5-HT. Reserpine, but not parachlorophenylalanine, decreased the

Neonatal maternal deprivation facilitates the expression of a panic-like escape behavior in adult rats.

A wealth of evidence associates disruptions of the parent-infant relationship (e.g. childhood parental loss or parental neglect) with the later appearance of panic disorder. In rodents, neonatal maternal separation and maternal deprivation (MD) are reported to increase the expression of anxiety-related defensive responses in adult animals. However, little is known about the long-term consequences of these early-life stressors in animal models of panic. We here investigated the effects of a single 24 h-episode of MD on post-natal day 11 (PND 11) in adult male Wistar rats submitted to two animal
