Advances in the research of fetal DNA in maternal plasma for noninvasive prenatal diagnostics.

Molecular analysis of fetal DNA present in the maternal circulation allows noninvasive, early, and precise determination of fetal genetic status in prenatal diagnostics. The most common clinical applications, i.e. prenatal gender determination and fetal RhD genotyping, are possible already in the first trimester using specialized protocols for DNA isolation from plasma and subsequent real-time PCR detection. Recent advances in molecular techniques enable other applications of fetal DNA purified from maternal plasma samples. Chromosomal abnormalities (e.g. trisomy 21) can be diagnosed by digita

The Polkinghorne Report on Fetal Research: nice recommendations, shame about the reasoning.

In 1989, in the wake of the first operations to transplant fetal tissue into the brains of sufferers from Parkinson's Disease, the UK Code of Practice governing the use of the fetus for research was overhauled by an eminent committee under the chairmanship of the Reverend Dr John Polkinghorne. The Polkinghorne Report has, however, attracted remarkably little comment or analysis. This paper is believed to be the first to subject it to sustained ethical and legal scrutiny. The author concludes that, although the committee's recommendations meet the major objections to the Code of Practice, the r

Fetal intervention for critical aortic stenosis: advances, research and postnatal follow-up.

PURPOSE OF REVIEW: Fetal aortic valvuloplasty is intended to alter the natural history of aortic stenosis evolving to hypoplastic left heart syndrome. The most recently reported data and advances on this procedure were reviewed. RECENT FINDINGS: The highlights of the latest experience are the advances in further understanding of the prenatal and postnatal natural history of this disease, and the way in which fetal aortic valvuloplasty impacts on it, the identification of new predictors of biventricular outcome, and the report of postnatal survival of intervened patients. Recently reported sho

Congenitally Corrected Transposition of the Great Arteries: Fetal Diagnosis, Associations, and Postnatal Outcome: A Fetal Heart Society Research Collaborative Study.

Background Fetal diagnosis of congenitally corrected transposition of the great arteries (ccTGA) has been increasingly reported; however, predictors of clinical outcomes remain underexplored. We undertook a multicenter, retrospective study to investigate natural history, associated anomalies, and outcomes of fetal ccTGA. Methods and Results Fetuses with ccTGA diagnosed from January 2004 to July 2020 within 20 North American programs were included. Fetuses with severe ventricular hypoplasia thought to definitively preclude biventricular repair were excluded. We included 205 fetuses diagnosed wi

Ethical, legal, and social issues associated with human fetal tissue research in Japan.

The Informed Consent Standards for Human Fetal Tissue Donation for Research, published by the International Society for Stem Cell Research, provide important guidance for ethical research. They are particularly relevant in countries such as Japan, where clear legal frameworks for the use of human fetal and associated tissues obtained through induced abortion are lacking. However, because such guidelines do not fully account for national regulatory and legal frameworks, additional considerations are required to support their effective global implementation. Here, we describe three ethical, lega

Large-Pore Channels at the Maternal-Fetal Interface: Progress and Open Research Avenues.

The maternal-fetal interface functions as an integrated physiological unit whose homeostatic balance determines pregnancy success. Large-pore channels, composed of connexins (Cxs), pannexins (Panxs), calcium homeostasis modulators (CALHMs) and leucine-rich repeat-containing 8 (LRRC8) proteins, mediate direct intercellular communication, autocrine and paracrine release of ATP and other signaling molecules, and scaffold-based signal integration across this interface. In this review, we synthesize current knowledge on large-pore channel expression and their physiological and pathophysiological ro

Research status of fetal hydrocephalus from 2003 to 2022 based on bibliometric analysis.

Hydrocephalus is the most common and devastating condition affecting the fetus. The aim of this study was to provide a comprehensive overview of the relevant literature through bibliometric analysis. The survey covers the articles related to congenital hydrocephalus published in the Web of Science Core Collection (WoSCC) database from January 1, 2003 to December 31, 2022. In addition to repeated literature, reviews and articles are included. We visualized the annual publication number, citation frequency, country/region, institution, author, periodical, and keywords with a range of software su

Maternal weight and fetal injury at birth: data deriving from medico-legal research.

The relationships among maternal weight, gestational weight gain, fetal birth weight and birth injuries have been investigated in connection with 62 cases of shoulder dystocia that involved permanent impairment of the newborn or neonatal demise. A high prevalence of > 13.5 kgs. gestational weight increase between the first and last office visits (62%) obesity expressed as > or = 87 kg. body weight at term (64.1%) and > or = 4000 gm. birth weight (80%) was found in this group of patients. The results indicate that the relationship between excessive maternal and fetal body weights and shoulder d

Essential variables for reporting research studies on fetal growth restriction: a Delphi consensus.

OBJECTIVE: To determine, by expert consensus using a Delphi procedure, a minimum reporting set of study variables for fetal growth restriction (FGR) research studies. METHODS: A panel of experts, identified based on their publication record as lead or senior author of studies on FGR, was asked to select a set of essential reporting study parameters from a literature-based list of variables, utilizing the Delphi consensus methodology. Responses were collected in four consecutive rounds by online questionnaires presented to the panelists through a unique token-secured link for each round. The e

In vitro models of fetal lung development to enhance research into congenital lung diseases.

PURPOSE: This paper aims to build upon previous work to definitively establish in vitro models of murine pseudoglandular stage lung development. These can be easily translated to human fetal lung samples to allow the investigation of lung development in physiologic and pathologic conditions. METHODS: Lungs were harvested from mouse embryos at E12.5 and cultured in three different settings, i.e., whole lung culture, mesenchyme-free epithelium culture, and organoid culture. For the whole lung culture, extracted lungs were embedded in Matrigel and incubated on permeable filters. Separately, dist

Advancements in medical research using fetal sheep: Implications for human health and treatment methods.

Sheep are typically considered as industrial animals that provide wool and meals. However, they play a significant role in medical research in addition to their conventional use. Notably, sheep fetuses are resistant to surgical invasions and can endure numerous manipulations, such as needle puncture and cell transplantation, and surgical operations requiring exposure beyond the uterus. Based on these distinguishing characteristics, we established a chimeric sheep model capable of producing human/monkey pluripotent cell-derived blood cells via the fetal liver. Furthermore, sheep have become cru

Applications for Research Concerning Fetal or Placental Tissue and Expected Institutional Review Board Responses.

Proposals for research concerning fetal and/or placental tissue may be refused institutional review board (IRB) review, effectively preventing the research from occurring. We conducted an anonymous electronic survey of IRB chairs to determine their assessment of the likely response to research projects using fetal/placental tissue obtained from various procedures. We found that proposals concerning tissue obtained from diagnostic procedures or miscarriage were anticipated to be considered at most institutions. Tissue obtained after abortion was likely to be refused consideration by more than 2

Sir Joseph Barcroft, Cambridge placental and fetal research (1933-1966) and inter-generational Science.

The nature of Cambridge (UK) placental and fetal research in the middle third of the twentieth century is reviewed on the basis of published literature and personal recollection. Joseph Barcroft is a central figure who came to fetal research late in an extremely productive career which is briefly sketched. Contemporaneous Cambridge academics in the field included J.D. Boyd (the authors father), J. Hammond, F.H.A. Marshall, R.A. McCance, J. Needham, A.S. Parkes and Elsie Widdowson. The then current Cambridge academic geography is explained and features of its scientific life such as funding, in
