Molecular systematics of five Onchocerca species (Nematoda: Filarioidea) including the human parasite, O. volvulus, suggest sympatric speciation.

The genus Onchocerca (Nematoda: Filarioidea) consists of parasites of ungulate mammals with the exception of O. volvulus, which is a human parasite. The relationship between O. volvulus, O. ochengi and O. gibsoni remains unresolved. Based on morphology of the microfilariae and infective larvae, vector transmission and geographical distribution, O. ochengi and O. volvulus have been placed as sister species. Nevertheless, the cuticle morphology and chromosomal data (O. volvulus and O. gibsoni have n=4 while O. ochengi is n=5) suggest that O. gibsoni could be more closely related to O. volvulus t

Regional genetic variation in the major sperm protein genes of Onchocerca volvulus and Mansonella ozzardi (Nematoda: Filarioidea).

Onchocerca volvulus and Mansonella ozzardi are two human filarial parasites present in South and Central America. In the Brazilian Amazonia they are found in sympatry, and the lack of clear morphological diagnostic characters in the microfilariae hinders their identification. The major sperm protein (MSP) gene of both species has been sequenced and characterised to determine its potential as a molecular diagnostic character. The length of the MSP gene is different in each species, and this could be used to detect and differentiate them by running the polymerase chain reaction (PCR) product in

Isoenzymatic diagnosis of filariae: a method for separation of lactate dehydrogenase isoenzymes from Molinema dessetae (Nematoda: Filarioidea).

Lactate dehydrogenase (LDH) is highly active in filariae and could be a valuable tool for phyllogeny studies. Unfortunately, the isoenzymatic diagnosis of filariae is often difficult for LDH because of a poor mobility of the enzymes in starch gels which are the most commonly used in such studies. We propose here a method to separate filarial LDH isoenzymes using disc electrophoresis. The experiments were carried out on male and female Molinema dessetae in order to compare their respective isoenzymes. The study of several parameters such as buffer systems, percentage of bisacrylamide and progre

Intracellular melanization in the mosquito Anopheles quadrimaculatus (Diptera: Culicidae) against the filarial nematodes, Brugia spp. (Nematoda: Filarioidea).

Intracellular melanization responses to developing larvae of Brugia species (B. malayi (Buckley), B. pahangi (Buckley and Edeson), and B. patei (Buckley, Nelson, and Heisch] in the thoracic muscle fibers of eight strains of Anopheles quadrimaculatus Say were first observed 48 to 72 h after an infective blood meal. Three to 4 d later, large numbers of melanized first-stage larvae were found within the thoracic muscle fibers. These intracellular responses were in addition to the extracellular responses to microfilariae and microfilarial sheaths of B. pahangi in the abdominal hemocoel of An. quad

The host-parasite relationships in normal and protein-malnourished cotton rats infected with Litomosoides carinii (Nematoda: Filarioidea).

A total of 120 young cotton rats were fed ad libitum for 10 weeks on either synthetic diets containing 2.5, 5, 10 or 15% casein or on a stock breeding diet containing 20% crude protein. Half of the animals in each group were infected with the filarial nematode Litomosoides carinii and the food intake and body weight of each animal was measured. There was a trend towards increased food consumption in protein-deficient cotton rats and this was increased further in infected animals; however, differences between groups were not significant. Low protein diets reduced the rate of growth of uninfecte

Litomosoides (Nemata: Filarioidea) of bats from Bolivia with records for three known species and the description of a new species.

Herein, we describe Litomosoides salazari n. sp. collected from the body cavity of the yellow bat, Lasiurus ega , from Bolivia. This new species of filarioid nematode is most closely related to the carinii group and is characterized by its relatively small size with the vulva located slightly posterior to the esophago-intestinal junction; an arrow-shaped buccal capsule; and a short, rounded tail. New host and locality records for both Litomosoides hamletti and L. chandleri in phyllostomid bats, and L. brasiliensis occurring in both phyllostomid and vespertilionid bats, are provided. Density and distribution of Dirofilaria immitis (Nematoda: Filarioidea) third-stage larvae in Aedes sollicitans and Aedes taeniorhynchus (Diptera: Culicidae).

Numbers and the distribution of third-stage larvae (L3) were investigated in Aedes sollicitans (Walker) and Aedes taeniorhynchus (Wiedemann) female mosquitoes fed Dirofilaria immitis (Leidy) infectious-blood with densities of microfilariae (mf) ranging from approximately 7,100-43,400 mf/ml. At each microfilarial density, a maximum of 63-66 infective larvae were recovered from an Ae. sollicitans alive on day 15 after infection. In comparison with Ae. taeniorhynchus, Ae. sollicitans averaged greater numbers of L3 and from 1.4 to 2.4 times more L3 in the head and labium per infected female. The t

Development of Onchocerca volvulus (Filarioidea: Onchocercidae) in the West African black fly Simulium yahense (Diptera: Simuliidae) in Liberia.

Simulium yahense black flies infected with microfilaria of Onchocerca volvulus were kept in a defined insectary environment in Liberia, West Africa. A daily sample of infected flies was dissected for larvae developing in the thoracic muscles and examined for growth in stadial development. Microfilariae ingested by black flies transformed to the L1 larval stage without molting. Successive larval development included molting to the L2 stage and, finally, to the L3 stage, which was infective in humans. The cephalic cap, consisting of a laterally located hook and central stoma, occurs in the first

Homologous and crossreacting immune response of the jird and cotton rat against microfilariae of Dipetalonema viteae and Litomosoides carinii (Nematoda: Filarioidea).

Consecutive injections of microfilariae of L. carinii and D. viteae were carried out in the homologous or heterologous sequence into the corresponding natural or into the experimental hosts. Microfilariae of D. viteae are imunogenic in Sigmodon specifically and crossreacting as well. However, in the natural host, Meriones, they are doubtlessly tolerogenic. The sojourn and level of microfilaraemia are prolonged resp. higher when living D. viteae-Mf are injected s.c. followed by an i.v. injection. Elaeophora elaphi n. sp. (Filarioidea: Onchocercidae) parasite of the red deer (Cervus elaphus). With a key of species of the genus Elaeophora.

We described a new species of nematode filarioid (Onchocercidae) parasiting the hepatic vessels of the red deer Cervus elaphus. This new species is characterized by the number and disposition of the papillae on the genital area of the male, and the presence and characteristics of an "area rugosa" situated just in front of the ventral impair precloacal papilla. We discussed this new species, giving a key to the identification of all the known species of the genus Elaeophora.

Scanning electron microscopy of Onchocerca fasciata (Filarioidea: Onchocercidae) adults, microfilariae and eggs with notes on histopathological findings in camels.

BACKGROUND: Onchocerca fasciata is a prevalent filarial species in camelids of Asia and Africa forming nodules in the skin of dromedary and Bactrian camels. In spite of recent advances in the biology and epidemiology of this nematode species, a relatively scant number of studies have focussed on the morphology of this parasite. The main objective of this study was to describe morphological characteristics of adults, microfilariae and eggs of O. fasciata by scanning electron microscopy (SEM), staining and histology. METHODS: From April 2016 to March 2017 dromedary camels (n = 456) were inspect

Fine structure and cytochemical analysis of the intestinal wall along the body of adult female of Litomosoides chagasfilhoi (Nematoda: Filarioidea).

Litomosoides chagasfilhoi is a filariid nematode parasite of the abdominal cavity of the wild rodent Akodon cursor (Winge, 1887), that has been described and used in Brazil as a new model for human filariasis. The fine structure of the intestine of this nematode was analyzed based on observations made by light and transmission electron microscopies of serial sections along the body. Cytochemical analysis was carried out to investigate the composition of the intestinal wall. This structure consisted of a basal lamina and an epithelium of variable thickness, composed of cells that have an irregu

Zoonotic infection caused by Onchocerca japonica (Nematoda: Filarioidea) in a 69-year-old woman in Kanto Region, Eastern Honshu, Japan.

Reports of zoonotic infections caused by the filarial nematode Onchocerca japonica have recently increased in Japan. A 69-year-old woman living in Sosa City, Chiba Prefecture, Kanto Region, Honshu, developed a painful nodule at the metacarpophalangeal joint of the index finger of her right hand. The causative agent was identified as a female O. japonica based on the histopathological characteristics (i.e., cuticle with transverse triangular ridges but without inner striae) of the biopsy specimens of the nodule. The species identification was corroborated by cox1 gene sequencing of the worm tis

Onchocerca takaokai n. sp. (Nematoda: Filarioidea) in Japanese wild boars (Sus scrofa leucomystax): Description and molecular identification of intradermal females.

Human zoonotic onchocercosis is caused by Onchocerca dewittei japonica, parasitic in wild boars (Sus scrofa leucomystax) in Japan. Previously, microfilariae longer than those of Onchocerca dewittei japonica were observed in skin snips from wild boars during the study of O. dewittei japonica. Moreover, the third-stage larvae (L3) of these longer microfilariae were obtained from the blackfly Simulium bidentatum after experimental injections. Based on morphometric and molecular studies, similar L3 were found in blackflies during fieldwork in Oita, Japan. However, except for O. dewittei japonica,

Heartworms in Halichoerus grypus: first records of Acanthocheilonema spirocauda (Onchocercidae; Filarioidea) in 2 grey seals from the North Sea.

The assumed definitive host of the heartworm Acanthocheilonema spirocauda (Onchocerdidae; Filarioidea) is the harbour seal (Phoca vitulina). This filaroid nematode parasitizing in cardiac ventricles and blood vessel lumina of harbour seals (P. vitulina) has a low prevalence and seldom causes severe health impacts. The seal louse (Echinophthirius horridus) is the assumed intermediate host for transmission of A. spirocauda filariae between seals, comprising a unique parasite assembly conveyed from the terrestrial ancestors of pinnipeds. Although grey seals (Halichoerus grypus) are infected by se

The complete mitochondrial genome of Setaria digitata (Nematoda: Filarioidea): Mitochondrial gene content, arrangement and composition compared with other nematodes.

In the present study, we determined the complete mitochondrial (mt) genome sequence (13,839bp) of parasitic nematode Setaria digitata and its structure and organization compared with Onchocerca volvulus, Dirofilaria immitis and Brugia malayi. The mt genome of S. digitata is slightly larger than the mt genomes of other filarial nematodes. S. digitata mt genome contains 36 genes (12 protein-coding genes, 22 transfer RNAs and 2 ribosomal RNAs) that are typically found in metazoans. This genome contains a high A+T (75.1%) content and low G+C content (24.9%). The mt gene order for S. digitata is th

Identification and phylogenetic analysis of Dirofilaria ursi (Nematoda: Filarioidea) from Wisconsin black bears (Ursus americanus) and its Wolbachia endosymbiont.

Dirofilaria ursi is a filarial nematode of American black bears (Ursus americanus Pallas, 1780) that is vectored by black flies (Simuliidae) in many parts of the United States. In northwestern Wisconsin, the prevalence of microfilaremic bears during the fall hunting season was 21% (n = 47). Unsheathed blood microfilariae from Wisconsin bears possess characters consistent with the original description of D. ursi, as do adult worms observed histologically and grossly. Immunohistochemistry was used to identify the Wolbachia endosymbiont in the hypodermis and lateral cords of an adult female D. ur
