Phage-derived depolymerases represent a promising antibiotic alternative for treating Klebsiella pneumoniae infections. Depolymerases can increase the sensitivity of bacteria to the host immune system and complement-mediated killing by specifically degrading capsular polysaccharides (CPS). However, the impact of depolymerases on bacterial biofilm formation remains unclear. This study found that depolymerase treatment significantly enhances the biofilm formation capability of hypervirulent K. pneumoniae (hvKp) strains, which inherently exhibit relatively weak biofilm formation due to their thic

Biofilm formation by Klebsiella pneumoniae is mediated by the type 3 fimbriae Mrk, and regulated by MrkH and 3',5'-cyclic diguanylic acid (c-di-GMP). We sought to identify specific chemical inhibitors of K. pneumoniae biofilm formation that reduced the activity of MrkH. A compound N-(3-cyano-5,6,7,8-tetrahydro-4H-cyclohepta[b]thien-2-yl)-2-methoxybenzamide, JT71, reduced K. pneumoniae mrkA promoter activity and biofilm formation by 50% without affecting cell viability. Western blot analysis, hemagglutination assays, electron microscopy and qPCR showed that JT71 reduced type 3 fimbriae producti

We investigated the ability of meconium, feces from human milk-fed (HMF) newborns, and feces from formula-fed (FF) newborns to inhibit adhesion of S-fimbriated E. coli to human buccal epithelial cells. S-fimbriae are a common property of E. coli strains causing sepsis and meningitis in neonates. Meconium had the highest content of neuraminic acid and the strongest inhibitory effect on bacterial adhesion. HMF also exerted high inhibitory activity while FF was markedly less active: To achieve inhibitory effects comparable to HMF a sixfold amount of FF was required. Glycoproteins from excretions

Neisseria gonorrhoeae, the causative agent of gonorrhoea, is a major burden on global healthcare systems, with an estimated ~80-90 million new global cases annually. This burden is exacerbated by increasing levels of antimicrobial resistance, which has greatly limited viable antimicrobial therapies. Decreasing gonococcal drug susceptibility has been driven largely by accumulation of chromosomal resistance determinants, which can be acquired through natural transformation, whereby DNA in the extracellular milieu is imported into cells and incorporated into the genome by homologous recombination

Uropathogenic Escherichia coli strains rely on specialized surface appendages to colonize the urinary tract, including Dr fimbriae, which are strongly associated with recurrent infection and long-term persistence. Although traditionally described as receptor-binding fibers, their broader contribution to bacterial surface architecture and physicochemical behavior has remained unclear. Here, combining quantitative proteomics, computational modeling, rheology, surface physicochemistry, and flow-based adhesion assays, we show that Dr fimbriae collectively form a dense, highly hydrated amphipathic

Klebsiella pneumoniae is a Gram-negative pathogen that has become a worldwide concern due to the emergence of multidrug-resistant isolates responsible for various invasive infectious diseases. Biofilm formation constitutes a major virulence factor for K. pneumoniae and relies on the expression of fimbrial adhesins and aggregation of bacterial cells on biotic or abiotic surfaces in a coordinated manner. During biofilm aggregation, bacterial cells communicate with each other through inter- or intra-species interactions mediated by signallng molecules, called autoinducers, in a mechanism known as

Using crystal violet binding assay, we examined the potential for biofilm formation in 194, 76 and 107 isolates from urine of patients with uncomplicated acute cystitis, pyelonephritis and prostatitis, respectively. The prostatitis isolates showed significantly higher optical density (OD) values compared with cystitis and pyelonephritis isolates (OD(540): 0.82, 0.29 and 0.43, respectively, P<0.0001). Similarly, strains of serotypes O4 and O22, which were commonly isolated from prostatitis, exhibited significantly higher OD values than did other strains. Furthermore, when the 21 prostatitis iso

Fluid flow is thought to prevent bacterial adhesion, but some bacteria use adhesins with catch bond properties to enhance adhesion under high shear forces. However, many studies on bacterial adhesion either neglect the influence of shear force or use shear forces that are not typically found in natural systems. In this study, we use microfluidics and single-cell imaging to examine how the human pathogen Pseudomonas aeruginosa interacts with surfaces when exposed to shear forces typically found in the human body (0.1 pN to 10 pN). Through cell tracking, we demonstrate that the angle between the

In Pseudomonas aeruginosa, type IV pili (TFP)-dependent twitching motility is required for development of surface-attached biofilm (SABF), yet excessive twitching motility is detrimental once SABF is established. In this study, we show that mucin significantly enhanced twitching motility and decreased SABF formation in strain PAO1 and other P. aeruginosa strains in a concentration-dependent manner. Mucin also disrupted partially established SABF. Our analyses revealed that mucin increased the amount of surface pilin and enhanced transcription of the pilin structural gene pilA. Mucin failed to

The social motility (S-motility) of Myxococcus xanthus is driven by the intricate interplay between type IVa pili (T4aP) and exopolysaccharide (EPS). Although T4aP retraction furnishes the mechanical force necessary for cellular movement, EPS serves as a matrix for T4aP anchoring and instigates pilus retraction. Despite this interdependence, the molecular determinants governing specific T4aP-EPS recognition and interaction have yet to be fully elucidated. Utilizing a comprehensive approach, our study identifies glucosamine as the key monosaccharide within EPS that interacts with the major pili

Porphyromonas gingivalis plays a key role in periodontal disease and has been associated with several serious systemic diseases. Its fimbriae are a major virulence factor. We recently demonstrated the formation of bundles of long FimA fimbriae in strain ATCC 33277. Transmission (TEM) and scanning electron microscopy (SEM) were used to examine a collection of P. gingivalis strains representing all seven known FimA types (I, Ib, IIa, IIb, III-V) and a P. gulae strain (type A). Additionally, two P. gingivalis strains (ATCC 49417 and OMI 1127) were investigated in dual-species approaches together

Interleukin-33 (IL-33) is an IL-1 cytokine family member that is involved in the development of chronic inflammatory diseases and the initiation of allergic inflammation in response to pathogens. Porphyromonas gingivalis is a primary pathogen that is involved in chronic periodontitis and its bacterial components induce inflammatory responses. Dendritic cells (DCs) recognize pathogen- associated molecular patterns by expression of pattern-recognition receptors, such as Toll-like receptors (TLRs). DCs play an essential role in resistance to infection and maintenance of mucosal immune system. In

Experiments were performed to determine whether immunization of mice with fimbrial adhesins isolated from Actinomyces viscosus T14V could modulate infection of tooth surfaces in animals challenged with the homologous strain. Saliva and sera from animals immunized in the submandibular gland region contained elevated levels of fimbria-specific immunoglobulin A (IgA) and IgG, whereas saliva and sera from sham-immunized animals did not. There was a statistically significant inverse correlation between the presence of fimbria-specific antibodies in saliva and serum and the levels of bacterial colon

Antibodies reactive with type 1 and type 2 fimbriae from Actinomyces viscosus T14V specifically inhibit the adherence of A. viscosus T14V to salivary pellicle-coated tooth surfaces and other bacteria, and these antibodies are thought to modulate colonization by this microorganism. These studies were done to determine whether previously noted differences in the antibody responses of inbred mice to type 1 and type 2 fimbriae might be under genetic control. The serum immunoglobulin G (IgG) and IgM antibody responses of inbred, F1 hybrid, and H-2 congenic mice, immunized with A. viscosus T14V cell

Urinary tract infections (UTIs) are one of the most common bacterial infections in humans, causing cystitis, pyelonephritis, and renal failure. Uropathogenic Escherichia coli (UPEC) is the leading cause of UTIs. Accurate and rapid discrimination of UPEC lineages is useful for epidemiological surveillance. Fimbriae are necessary for the adherence of UPEC strains to host uroepithelia, and seem to be abundant and diverse in UPEC strains. By analyzing all the possible fimbrial operons in UPEC strains, we found that closely related strains had similar types of chaperone-usher fimbriae, and the dive

Enterotoxigenic Escherichia coli possessing K99 fimbriae (E. coli K99) causes diarrhea in piglets of less than 1 week old. The first stage of the bacterial infection is adhesion by the fimbriae on the small intestinal mucosa and the adhesion is followed by colony formation. K99 fimbriae bind specifically to N-glycolylneuraminyl-lactosyl-ceramide, GM3(NeuGc) [Ono, E. et al. (1989) Infect. Immun. 57,907-911]. We examined the postnatal change of the content and the molecular species of GM3(NeuGc) in the small intestinal mucosa of 0- to 14-day-old piglets and adult pigs. GM3(NeuGc) was a major gan

An experimental pyelonephritis model was developed in monkeys (Macaca fascicularis) using P-fimbriated Escherichia coli as the infecting organism. The relevant receptor molecules for P-fimbriae were also shown to be present in Macaca fascicularis. Atraumatic administration of P-fimbriated E. coli into the ureter induced a ureteritis followed by acute and chronic pyelonephritis. The decisive role of P-fimbriae as an adhesive virulence factor was proven by the receptor blockade of P-fimbriae-mediated bacterial adhesion by a synthetic receptor analogue (alpha-D-Galp-(1-4)-beta-D-Galp-1-OMe), whic

BACKGROUND: The Neisseria gonorrhoeae pilE gene encodes the PilE protein, the major subunit of the Type IV pilus and a primary colonization and virulence factor. The pilE gene undergoes high-frequency diversification mainly through gene conversion from one of many pilS copies. These unique molecular processes contribute to gonococcal population diversity, facilitating immune evasion. While the process of pilin variation is understood, the diversity of pilE and pilS genes from clinical isolates is understudied.
METHODS: We analyzed 15 186 N. gonorrhoeae genomes, including finished (n = 65) and

Hypervirulent (hvKP) and classical (cKP) strains of Klebsiella pneumoniae are major contributors to serious hospital-acquired infections, and the growing issue of antibiotic resistance highlights the urgent need for new antimicrobial approaches. In this study, we investigated the inhibitory effects of Sarcotoxin II 50, an insect-derived antimicrobial peptide, on K. pneumoniae. Our findings show that Sarcotoxin II 50 exerts potent antibacterial activity by impairing capsular polysaccharide (CPS) production and fimbrial formation, which compromises bacterial adhesion and colonization capacity. Escherichia coli is the major causative agent of human cystitis. In this study, a preliminary molecular analysis carried out by PCR (polymerase chain reaction) demonstrated that 100% of 31 E. coli strains isolated from patients with recurrent UTIs (urinary tract infections) showed the presence of the curli fimbria gene (csgA). Curli fimbria is known to be associated with bacterial biofilm formation but not with the adhesion of human cystitis-associated E. coli. Therefore, this work aimed to study how curli fimbria is associated with uropathogenic E. coli (UPEC) as an adhesion factor. For this
