Tests for the presence of pathogen DNA or antibodies are routinely used to survey for current or past infections. In diseases that emerge following a host jump estimates of infection rate might be under- or overestimated. We here examine whether observed rates of infection are biased for a non-focal host species in a model system. The bacterium Mycoplasma gallisepticum is a widespread pathogen in house finches (Haemorhous mexicanus), a fringillid finch, but an unknown proportion of individuals of other songbird species are also infected. Our goal is to determine the extent to which detection o

BACKGROUND: Due to its high polymorphism and importance for disease resistance, the major histocompatibility complex (MHC) has been an important focus of many vertebrate genome projects. Avian MHC organization is of particular interest because the chicken Gallus gallus, the avian species with the best characterized MHC, possesses a highly streamlined minimal essential MHC, which is linked to resistance against specific pathogens. It remains unclear the extent to which this organization describes the situation in other birds and whether it represents a derived or ancestral condition. The sequen

To investigate whether changes in testosterone alter the frequency range in which a zebra finch produces song, we assigned male zebra finches to two groups, one of which received testosterone implants and the other empty silastic capsule implants. We then recorded songs up to 52 weeks after the surgery and measured frequency changes in the fundamental frequencies of arbitrarily chosen harmonic stacks in the songs of birds in either group. We found statistically significant decreases after 5 weeks in the songs of the testosterone-treated birds. No changes were found in the fundamental frequenci

Juvenile male zebra finches (Taeniopygia guttata) learn a stereotyped song by imitating sounds from adult male tutors. Their song is composed of a series of syllables, which are separated by silent periods. How acoustic units of song are translated into respiratory and syringeal motor gestures during the song learning process is not well understood. To learn about the respiratory contribution to the imitation process, we recorded air sac pressure in 38 male zebra finches and compared the acoustic structures and air sac pressure patterns of similar syllables qualitatively and quantitatively. RATIONALE: Rodent models dominate autism spectrum disorder (ASD) research, yet their translational validity is limited for a heterogeneous condition with complex social-vocal phenotypes.
OBJECTIVES: To evaluate the zebra finch (Taeniopygia guttata) - a highly social, vocal-learning songbird - as an ASD model by adapting embryonic valproic acid (VPA) exposure and identifying dose-timing conditions that preserve viability while perturbing social behavior.
METHODS: We used a 3 × 3 design crossing VPA dose (0, 0.3, 0.6 µmol/egg) with manipulation day (incubation days 8-10). Hatching success, pos

Mycobacterium avium subsp. hominissuis (MAH) is a zoonotic pathogen with a broad host range and diverse clinical manifestations. We report here the first documented case of MAH-induced fatal vasculitis in zebra finch (Taeniopygia guttata). Histopathological examination revealed acid-fast bacilli within macrophages and endothelial cells, primarily affecting the heart and aorta. Mycobacterial DNA was detected in cloacal swabs from affected finches and environmental samples from their housing facility. PCR targeting the rpoB gene and insertion elements IS1245 and IS901, followed by sequencing, co

Predicting the nature of behavioral plasticity can provide insight into mechanisms of behavioral expression and control. Songbirds like the zebra finch rely on vocal signals for communication, and the performance of these signals demonstrate considerable plasticity over development. Traditionally, these signals were thought to be fixed in adulthood, but recent studies have revealed significant age-dependent changes to spectral and temporal features of song in adult songbirds. A number of age-dependent changes to song resemble acute changes to adult song performance across social contexts (e.g.

At a cellular level, oxidative stress is known to increase telomere attrition, and hence cellular senescence and risk of disease. It has been proposed that dietary micronutrients play an important role in telomere protection due to their antioxidant properties. We experimentally manipulated dietary micronutrients during early life in zebra finches (Taeniopygia guttata). We found no effects of micronutrient intake on telomere loss during chick growth. However, females given a diet high in micronutrients during sexual maturation showed reduced telomere loss; there was no such effect in males. Th

Forty-one outbreaks of mortality in wild finches were reported in southern Norway, Sweden, and Finland in the second half of 2008 (n = 40) and in February 2009 (n = 1). Greenfinches (Carduelis chloris) and occasional chaffinches (Fringilla coelebs) primarily were affected. Forty-eight greenfinches, eight chaffinches, one hawfinch (Coccothraustes coccothraustes), and one blue tit (Parus caeruleus) from 22 incidents were examined postmortem. Birds were in poor nutritional condition and had necrotizing ingluvitis, esophagitis, and/or oropharyngitis. Viable trichomonads with morphology consistent

Individual differences in animal behavior can be attributed to genetic as well as non-genetic influences. One mechanism by which the behavioral phenotype of an individual can be shaped is via transmission of maternal sex steroids. In this study, we examined the role of yolk testosterone (T) in controlling neophobia in 9-month-old, sexually mature zebra finches (Taeniopygia guttata). Offspring hatched from either T-treated or control eggs were subjected to a sequential series of behavioral tests in which we measured the neophobic response and its persistence towards two unfamiliar stimuli. Bird

Zebra finches (Taeniopygia guttata) learn a specific song pattern during a sensitive period of development, after which song changes little or not at all. However, recent studies have demonstrated substantial behavioral plasticity in song behavior during adulthood under a range of conditions. The current experiment examined song behavior of adult zebra finches temporarily deprived of auditory feedback by chronic exposure to loud white noise (WN). Long-term exposure to continuous WN resulted in disruption of song similar to that observed after deafening. When auditory feedback was restored by d

Antibody titers against St. Louis encephalitis virus (SLE) measured by a plaque reduction neutralization test (PRNT) decreased rapidly in house finches (Capodacus mexicanus) after initial infection, whereas antibodies measured by enzyme immunoassay (EIA) remained detectable in all birds for the length of the experiment, indicating long-term persistence and greater assay sensitivity of the EIA. After 52 wk, birds were challenged by subcutaneous inoculation with the same strain of SLE virus. Virus was not detected for 1-4 d postchallenge in blood samples tested by plaque assay and RT-PCR or by x

By 2030, 1.4 billion people worldwide will be 60 years or older. A significant proportion will experience changes in voice, which can indicate early signs of neurodegenerative disease. Most research and treatment studies have focused on the larynx because of its accessibility and known role in moderating pitch, loudness, and voice quality. Preclinical animal models offer important insight into brain mechanisms involved in aging vocalizations. Previously, we conducted a cross-sectional analysis in adult male zebra finches, discovering that song changes occur at transitional time points, between

Food-handling time, or the time elapsed between capturing a food item and consuming it, is a key factor in foraging decisions. For example, quickly consuming more items that are easier to handle, but of lower nutritional value, can be more beneficial than attempting to consume items that are harder to handle but more nutritious. Infection likely alters such foraging decisions, as sick animals often express sickness behaviours (lethargy and anorexia), which could affect both motivation and foraging proficiency. Although such changes could alter interindividual contact rates at food patches, and

Ever since 1994, when the bacterial pathogen Mycoplasma gallisepticum jumped from poultry to wild birds, it has been assumed that the primary host species of this pathogen in wild North American birds was the house finch (Haemorhous mexicanus), in which disease prevalence was higher than in any other bird species. Here we tested two hypotheses to explain a recent increase in disease prevalence in purple finches (Haemorhous purpureus) around Ithaca, New York. Hypothesis 1 is that, as M. gallisepticum evolved and became more virulent, it has also become better adapted to other finches. If this i

The capacity to adequately respond to (physiological) perturbations is a fundamental aspect of physiology, and may affect health and thereby Darwinian fitness. However, little is known of the degree of individual variation in this capacity in non-model organisms. The glucose tolerance test evaluates the individual's ability to regulate circulating glucose levels, and is a widely used tool in medicine and biomedical research, because glucose regulation is thought to play a role in the ageing process, among other reasons. Here, we developed an application of the intraperitoneal glucose tolerance

Parasites co-infecting hosts can interact directly and indirectly to affect parasite growth and disease manifestation. We examined potential interactions between two common parasites of house finches: the bacterium Mycoplasma gallisepticum that causes conjunctivitis and the intestinal coccidian parasite Isospora sp. We quantified coccidia burdens prior to and following experimental infection with M. gallisepticum, exploiting the birds' range of natural coccidia burdens. Birds with greater baseline coccidia burdens developed higher M. gallisepticum loads and longer lasting conjunctivitis follow

Mycoplasma gallisepticum (MG) has become a common cause of conjunctivitis in free-living house finches (Carpodacus mexicanus) since its emergence in the early 1990s. To date, temporal and spatial genotypic variation in MG has been documented, but phenotypic variation in pathogenicity and immunogenicity has not been examined. House finches were inoculated with MG isolates Virginia (VA)1994, California (CA)2006, or North Carolina (NC)2006, which were cultured from free-living house finches with conjunctivitis in 1994, 2006, and 2006, respectively. Infection with NC2006 resulted in the most sever

Severe tracheitis and bronchitis were identified in two fatal cases of respiratory disease affecting a flock of Gouldian finches (Erythrura [Chloebia] gouldiae). Intranuclear inclusion bodies in epithelial cells of the upper respiratory tract were identified in samples from two birds. Electron microscopic examination showed that the inclusions consisted of viral particles consistent in appearance with Herpesviridae. Degenerate PCR primers targeting a conserved region of the herpesviral-DNA-dependent DNA polymerase were used to amplify a region of DNA isolated from tissues with lesions from eac

Since 1995, the epidemic of mycoplasmal conjunctivitis in eastern house finches has affected the Auburn, AL, house finch population. To better characterize the current status of this host-parasite interaction, we established a captive flock of 38 seronegative, healthy finches in fall 1998. After a minimum quarantine period of 4 wk, two Mycoplasma gallisepticum (MG)-infected house finches were introduced into this flock. Over a 12-wk period, the flock was captured every 2 wk and each bird was observed for conjunctivitis. Blood and choanal swabs were collected from each bird for serologic analys
