Molecular characterization of three gonadotropin subunits and their expression patterns during ovarian maturation in Cynoglossus semilaevis.

The endocrine regulation of reproduction in a multiple spawning flatfish with an ovary of asynchronous development remains largely unknown. The objectives of this study were to monitor changes in mRNA expression patterns of three gonadotropin hormone (GTH) subunits (FSHβ, LHβ and CGα) and plasma GTH levels during ovarian maturation of half-smooth tongue sole Cynoglossus semilaevis. Cloning and sequence analysis revealed that the cDNAs of FSHβ, LHβ and CGα were 541, 670 and 685 bp in length, and encode for peptides of 130, 158 and 127 amino acids, respectively. The number of cysteine residues a

Regulation of troponin T expression during muscle development in sea bream Sparus auratus Linnaeus: the potential role of thyroid hormones.

In the sea bream Sparus auratus three stage-specific fast troponin T (fTnT) isoforms have been cloned and correspond to embryonic-, larval- and adult-specific isoforms. Characterisation, using database searches, of the putative genomic organisation of Fugu rubripes and Tetraodon nigroviridis fTnT indicates that alternative exon splicing in the 5 region of the gene generates the different isoforms. Moreover, comparison of teleost fTnTs suggests that alternative splicing of fTnT appears to be common in teleosts. A different temporal expression pattern for each fTnT splice varotnt is found during

Chromosomal manipulation in Senegalese sole (Solea senegalensis Kaup, 1858): induction of triploidy and gynogenesis.

In this study we have developed protocols for induced triploidy and gynogenesis of Senegalese sole (Solea senegalensis), a promising flatfish species for marine aquaculture, in order to: 1) identify the sex-determination mechanism; and 2) to improve its production by generating a) sterile fish, avoiding problems related with sexual maturation, and b) all-female stocks, of higher growth rate. Triploidy was induced by means of a cold shock. Gynogenesis was induced by activating eggs with UV-irradiated sperm, and to prompt diploid gynogenesis, a cold-shock step was also used. Ploidy of putative t

Size-, sex- and site-dependent morphological variation in the stone flounder (Kareius bicoloratus) from Korean coastal waters.

Kareius bicoloratus is a commercially important flatfish in Korean coastal fisheries and an ecologically relevant benthic species in nearshore ecosystems. To evaluate how multiple biological and environmental factors jointly shape flatfish morphology, we analysed 81 morphometric indices and two-dimensional geometric morphometrics (GMM) in wild K. bicoloratus collected from two Korean coastal regions, incorporating size, sex and sampling site. Classical morphometrics based on a three-way ANOVA framework identified body size and sex as the primary drivers of shape variation, particularly influen

Asymmetrical development of bones and soft tissues during eye migration of metamorphosing Japanese flounder, Paralichthys olivaceus.

The symmetrical body of flatfish larvae dramatically changes into an asymmetrical form after metamorphosis. Eye migration results in the most significant asymmetrical development seen in any vertebrate. To understand the mechanisms involved in eye migration, bone and cartilage formation was observed during metamorphosis in laboratory-reared Japanese flounder, Paralichthys olivaceus, by using whole-body samples and histological sections. Most of the hard tissues of the cranium (parasphenoid, trabecular cartilage, supraorbital canal, and supraorbital bar) exist symmetrically in the larval period

The life cycle of Pygidiopsis ardeae Køie, 1990 (Digenea, Heterophyidae).

Ocellate cercariae, consistent with the description of Cercaria angularis Honer, 1961, developing in rediae in Hydrobia ulvae (Pennant) and Hydrobia ventrosa (Montagu) (Prosobranchia, Hydrobiidae) from Denmark, were shown to be larvae of Pygidiopsis ardeae Køie, 1990 (Heterophyidae, Pygidiopsinae). Euryhaline fishes such as the sticklebacks Gasterosteus aculeatus Linnaeus and Pungitius pungitius (Linnaeus), and the common goby Pomatoschistus microps (Krøyer) are second intermediate hosts in Danish inlets. These species and the flatfishes Platichthys flesus (Linnaeus) and Limanda limanda (Linna

Comparative persistence of opportunistic Vibrio alginolyticus and major foodborne Vibrio species (Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus) in ready-to-eat seafood under different storage conditions.

Vibrio alginolyticus is an opportunistic marine bacterium capable of infecting both humans and aquatic animals, in addition to major foodborne pathogenic Vibrio species (Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus). Although these species are increasingly detected in seafood, differences in their persistence under conditions simulating real food systems remain poorly understood. This study investigated the growth, survival, and recovery of V. alginolyticus and compared its behavior with those of the three pathogenic species in ready-to-eat seafood (flatfish, smoked salmon,

RNA-seq and microRNA-seq analysis of Japanese flounder (Paralichthys olivaceus) larvae treated by thyroid hormones.

Flatfish pigmentation is a complex process, affected by environmental factors including light, nutrients, and hormones. Of those, the thyroid hormone has been reported to increase the albinism rate of Japanese flounder (Paralichthys olivaceus). However, the underlying mechanism remains unclear. In the present study, triiodothyronine (T3), thyroxine, and thiourea were introduced into P. olivaceus larvae from 16 to 57 days after hatching (DAH). By comparison of albinism rate, T3 treatment and control larvae of 42 DAH were chosen for mRNA and miRNA high-throughput sequencing analyses. A total of

Morphological and immunohistochemical characterisation of the thymus in juvenile turbot (Psetta maxima, L.).

We performed structural and immunohistochemical studies on the thymus of juvenile turbot (Psetta maxima L.) in order to define its cellular composition. The thymic stroma was mainly composed of two subpopulations of reticulo-epithelial cells (RECs). RECs immunoreactive to anti-actin antibody were distributed through the organ, while RECs that were cytokeratin-immunopositive were located in the outer zone of the thymus. The parenchyma of the thymus was composed of several cell types such as lymphocytes/thymocytes, lymphoblasts, melano-macrophages and to a lesser extent of nurse-like cells, immu

Asymptomatic gastric anisakiasis detected in gastric cancer screening: A case report.

Anisakiasis is a parasitic disease caused by Anisakis simplex and has become an emerging zoonosis as preferences for eating raw or undercooked seafood have become more common. Few case reports of asymptomatic anisakiasis have been published to date. A 79-year-old asymptomatic man underwent esophagogastroduodenoscopy (EGD) for gastric cancer screening. The gastroenterologist diagnosed superficial gastritis without any malignant lesions but found an Anisakis larva while reviewing EGD images. The physician performed a second EGD and removed the larva. The patient reported that he ate the flatfish

Histopathology reveals environmental stress in dusky flounder Syacium papillosum of the Yucatan Peninsula continental shelf.

The histological changes in the liver, kidney, spleen, and gills of Syacium papillosum from the continental shelf of the Yucatan Peninsula, Gulf of Mexico, and their statistical associations with environmental conditions and pollutants were assessed in 2010, 2011, and 2012. We evaluated the extension and severity of the lesions through a degree of tissue change (DTC), and, with the sum of the number of lesion types within each of their DTC stages, we determined the histological alteration index (HAI). The liver and kidney were the most affected organs, with HAI values > 100. Fish with the most

Pardaxin produces postjunctional muscle contraction in guinea-pig intestinal smooth muscle.

The action of pardaxin (PX), a toxin isolated from the secretion of the Red Sea flatfish, Pardachirus marmoratus, was studied on longitudinal muscle of guinea-pig ileum. Pardaxin contracted the ileum and subsequently abolished muscle contraction to 5-hydroxytryptamine (5-HT), but did not affect the responses to acetylcholine (ACh) and substance P(SP). Pardaxin-induced contraction was only partially suppressed by atropine and not affected by tetrodotoxin or morphine. Preparations desensitized to 5-HT or SP responded normally to pardaxin. Pardaxin-induced contractions were normal in K+-depolariz

Effects of acute hypoxia on nutrient metabolism and physiological function in turbot, Scophthalmus maximus.

Acute hypoxia is a common stress in aquaculture, and causes energy deficiency, oxidative damage and death in fish. Many studies have confirmed that acute hypoxia activated hif1α expression, anaerobic glycolysis and antioxidant system in fish, but the effects of acute hypoxia on lipid and protein metabolism, organelle damage, and the functions of hif2α and hif3α in economic fishes have not been well evaluated. In the present study, turbot was exposed to acute hypoxia (2.0 ± 0.5 mg/L) for 6 h, 12 h, and 24 h, respectively. Then, the contents of hemoglobin (HB), metabolite, gene expressions of hi

Horizontal transmission of Endolimax piscium, causative agent of systemic amoebiasis in Senegalese sole Solea senegalensis.

Systemic amoebiasis of Senegalese sole Solea senegalensis is caused by Endolimax piscium Constenla, Padrós & Palenzuela, 2014 a cryptic parasitic member of the Archamoebae whose epidemiology is yet unknown. To test whether the parasite can be transmitted horizontally, an experimental trial by cohabitation between non-infected and infected fish was designed. Transmission of the parasite from naturally infected to healthy fish was confirmed in the experiment, with the water as the most likely route of infection. Under the conditions of the study, the infection process was remarkably slow, as par

Morphological and histological study of larval development of the Senegal sole Solea senegalensis: an integrative study.

This study provides a comprehensive description of the main morphological and histological events that take place during larval and post-larval development of Senegal sole Solea senegalensis in order to establish a reference for its normal developmental organogenesis. Five stages have been described. Before gill development at the onset of metamorphosis (eye migration process, stage 4c), the skin was the main site of gas and ion exchange, whereas during stages 3 and 4, the skin begins differentiating into the definitive juvenile structure. The timing of development of the endocrine system depe

Histopathology of the skin of UV-B irradiated sole (Solea solea) and turbot (Scophthalmus maximus) larvae.

Larval stages of two economically important flatfish, the sole (Solea solea) and turbot (Scophthalmus maximus) were exposed to ambient and elevated levels of UV-B. Sole larvae, which naturally occur in the plankton in early spring, demonstrated skin lesions at elevated levels of UV-B. Histopathology of the sole revealed cellular changes in the integument, characteristic of sunburn damage, with a reduction in the size of mucus-secreting cells and an increased epidermal thickening, especially at the highest doses of UV-B (2.15 KJ bio eff/m2). Pigmentation in the sole is restricted to a few isola

The galaninergic enteric nervous system of pleuronectiformes (Pisces, Osteichthyes): an immunohistochemical and confocal laser scanning immunofluorescence study.

The galaninergic enteric nervous system of three flatfishes was studied using immunohistochemical and immunofluorescence methods. Galanin-like immunoreactivity was mainly detected within the enteric intramural neurons of the stomachs and the proximal intestines. The sole, Solea solea L. and the flounder, Platichthys flesus L. showed a similar occurrence and relative distribution of galaninergic intramural neurons. Rare nervous fibre immunoreactive to the anti-galanin serum were observed in the muscular layers of the oesophagus of the turbot, Psetta maxima L. The presence and relative abundance

Genotype and host range analysis of infectious spleen and kidney necrosis virus (ISKNV).

Infectious spleen and kidney necrosis virus (ISKNV) is the causative agent of a disease causing high mortality in mandarin fish, Siniperca chuatsi. In this study, complete major capsid protein (MCP) genes of nine ISKNV isolates were sequenced and compared with other known megalocytiviruses to evaluate genetic variation and host range of the viruses. Comparison of nucleotide sequences of MCP gene revealed 92.6-100% identity among nine ISKNV isolates. A phylogenetic tree revealed that 33 megalocytiviruses were divided into three genotypes, and there was a strong host species signal in three geno

Identification and functional characterization of annexin A2 in half-smooth tongue sole (Cynoglossus semilaevis).

Annexin A2 (AnxA2), belonging to the annexin family, plays a crucial role in immune responses. In this study, the cDNA of the AnxA2 gene was identified in half-smooth tongue sole, Cynoglossus semilaevis. The transcript of AnxA2 gene in C. semilaevis (CsAnxA2) showed broad tissue distribution, with the highest expression level observed in the gut. CsAnxA2 expression was significantly up-regulated in the intestine, spleen, and kidney tissues following exposure to Shewanella algae. Immunohistochemical staining revealed that CsAnxA2 was predominantly expressed in epithelial cells and significantly
