Boronate Affinity Fluorescent Nanoparticles for Förster Resonance Energy Transfer Inhibition Assay of cis-Diol Biomolecules.

Förster resonance energy transfer (FRET) has been essential for many applications, in which an appropriate donor-acceptor pair is the key. Traditional dye-to-dye combinations remain the working horses but are rather nonspecifically susceptive to environmental factors (such as ionic strength, pH, oxygen, etc.). Besides, to obtain desired selectivity, functionalization of the donor or acceptor is essential but usually tedious. Herein, we present fluorescent poly(m-aminophenylboronic acid) nanoparticles (poly(mAPBA) NPs) synthesized via a simple procedure and demonstrate a FRET scheme with suppre

A new spectrofluorimetric method for the determination of some tetracyclines based on their interfering effect on resonance fluorescence energy transfer.

A rapid, simple, inexpensive and highly sensitive spectrofluorimetric method was developed for the determination of trace amounts of some tetracyclines (TCs), namely tetracycline hydrochloride (TCH), oxytetracycline hydrochloride (OTCH) and minocycline hydrochloride (MCH). Binding rhodamine B (RhB) to gold nanoparticles (Au NPs) resulted in quenching of the fluorescence of RhB by a resonance energy transfer (FRET) mechanism, with Au NPs as the energy acceptors. The presence of TCs caused the release of RhB molecules and recovered their fluorescence, and this was used as a basis for the quantit

Evaluating the potential for iodinated radiocontrast agents to interfere with ADAMTS13 activity testing via fluorescence resonance energy transfer methodology.

OBJECTIVES: Fluorescence resonance energy transfer (FRET)-based ADAMTS13 activity assays are critical for the diagnosis of thrombotic thrombocytopenic purpura. However, these assays are susceptible to interference. As iodide has been suggested to interfere in laboratory testing via fluorophore quenching or promotion, we aimed to determine whether iodinated contrast (Omnipaque) interferes with the ATS-13 ADAMTS13 Activity Assay 2.0. METHODS: We evaluated the excitation, emission, and absorbance spectrum of Omnipaque alone and spiked in patient plasma with known ADAMTS13 activity and ADAMTS13 a

Ratiometric fluorescence resonance energy transfer for reliable and sensitive detection of intracellular telomerase RNA via strand displacement reaction amplification.

Human telomerase RNA (hTR) is one essential component of telomerase and is overexpressed in tumor cells. Therefore, the reliable and sensitive detection of hTR is essential for the early cancer diagnosis. Herein, to avoid the false positive signals caused by co-existing components in the cell, a ratiometric fluorescence resonance energy transfer (FRET) strategy was developed to achieve reliable detection of intracellular hTR. Manganese dioxide nanosheets (MnO2NS) with good biocompatibility carry two fluorophore-labelled hairpin DNA probes into the cancer cell and then release the probes via de

Characterization of the adsorption of oligonucleotides on mercaptopropionic acid-coated CdSe/ZnS quantum dots using fluorescence resonance energy transfer.

Semiconductor quantum dots (QDs) coated with thioalkyl acid ligands are often used as probes and reporters for nucleic acid sensing, or protein sensing using aptamers, and are also potential vectors for gene delivery. In such applications, the interactions that potentially lead to the adsorption of oligonucleotides onto the surface of colloidal QDs are an important consideration. To explore such interactions, fluorescence resonance energy transfer (FRET) between QDs and oligonucleotides labeled with a fluorescent dye was used to identify and characterize a set of conditions that favor strong a

Using fibre photometry with a fluorescence resonance energy transfer-based biosensor to test efficacy of calpain inhibitors in vivo.

Calcium-activated proteases, known as calpains, cleave proteins into smaller protein fragments, altering protein structure and function. Calpain overactivity is a pathological hallmark of many human diseases, including neurodegenerative diseases, which are characterized by excessive proteolytic cleavage and formation of protein aggregates. Current methods for determining calpain activity, such as examination of the presence or absence of calpain substrates in post-mortem tissue, are low-throughput, costly and retrospective, hampering the search for potentially therapeutically efficacious calpa

Encapsulation of anticancer drug curcumin and co-loading with photosensitizer hypericin into lipoproteins investigated by fluorescence resonance energy transfer.

Low-density lipoproteins (LDL) and high-density lipoproteins (HDL) are natural occurring vehicles attractive for drug delivery and targeting tumor cells. Here we have investigated the encapsulation and interaction of a well-known anticancer agent curcumin with LDL and HDL. LDL particles have been found to accumulate more curcumin molecules inside their structure than HDL. The chemical stability of curcumin is enhanced and its photo-physical properties are altered due to encapsulation inside both lipoproteins. Combining photodynamic therapy with chemotherapy can improve anticancer treatment by

Bioluminescence Imaging of Inflammation in Vivo Based on Bioluminescence and Fluorescence Resonance Energy Transfer Using Nanobubble Ultrasound Contrast Agent.

Inflammation is an immunological response involved in various inflammatory disorders ranging from neurodegenerative diseases to cancers. Luminol has been reported to detect myeloperoxidase (MPO) activity in an inflamed area through a light-emitting reaction. However, this method is limited by low tissue penetration and poor spatial resolution. Here, we fabricated a nanobubble (NB) doped with two tandem lipophilic dyes, red-shifting luminol-emitted blue light to near-infrared region through a process integrating bioluminescence resonance energy transfer (BRET) and fluorescence resonance energy

Sensitive detection of polycyclic aromatic hydrocarbons using CdTe quantum dot-modified TiO₂ nanotube array through fluorescence resonance energy transfer.

CdTe quantum dots (QDs) are prepared on TiO(2) nanotubes (TiO(2) NTs), for the first time, with pulse electrodeposition. A novel single-drop optical sensor is prepared with the CdTe QDs-modified TiO(2) NTs, and applied for the detection of polycyclic aromatic hydrocarbons (PAHs) based on fluorescence resonance energy transfer (FRET). Excited at 270 nm, the sensor shows fluorescence emission at around 370 nm. As PAHs are with absorption/fluorescence emission at around 364/410 nm, FRET happens between the CdTe QDs and PAHs with the CdTe QDs as donors and PAHs as receptors. The sensitivity is dep

Spatially pathogenic forms of tau detected in Alzheimer's disease brain tissue by fluorescence lifetime-based Förster resonance energy transfer.

In tauopathies including Alzheimer's disease (AD) tau molecules have lost their normal spatial distance to each other and appear in oligomeric or aggregated forms. Conventional immunostaining methods allow detection of abnormally phosphorylated or conformationally altered aggregated tau proteins, but fail to visualize oligomeric forms of tau. Here we show that tau molecules that lost their normal spatial localization can be detected on a subcellular level in postmortem central nervous system (CNS) tissue sections of AD patients by fluorescence lifetime-based Förster resonance energy transfer (

Conformational substates of calmodulin revealed by single-pair fluorescence resonance energy transfer: influence of solution conditions and oxidative modification.

A calmodulin (CaM) mutant (T34,110C-CaM) doubly labeled with fluorescence probes AlexaFluor 488 and Texas Red in opposing domains (CaM-DA) has been used to examine conformational heterogeneity in CaM by single-pair fluorescence resonance energy transfer (spFRET). Burst-integrated FRET efficiencies of freely diffusing CaM-DA single molecules yielded distributions of distance between domains of CaM-DA. We recently reported distinct conformational substates of Ca(2+)-CaM-DA and apoCaM-DA, with peaks in the distance distributions centered at approximately 28 A, 34-38 A, and 55 A [Slaughter et al.

Use of fluorescence resonance energy transfer hybridization probes to evaluate quantitative real-time PCR for diagnosis of ocular toxoplasmosis.

Toxoplasma gondii infection is an important cause of chorioretinitis in Europe and the United States. Ophthalmological examination and a good clinical response to adequate therapy mainly support ocular toxoplasmosis diagnosis. However, clinical diagnostic may be difficult in some atypical cases. In these cases, laboratory confirmation, based on detection of local specific antibodies and parasite DNA by conventional PCR, is therefore important to confirm the disease etiology. More recently, real-time PCR has been developed to improve prenatal congenital toxoplasmosis diagnosis. We therefore exa

Rapid detection and differentiation of human pathogenic orthopox viruses by a fluorescence resonance energy transfer real-time PCR assay.

BACKGROUND: The orthopox viruses that are pathogenic for humans include variola major virus (VAR), monkeypox virus (MPV), cowpox virus (CPV), and to a lesser extent, camelpox virus (CML) and vaccinia virus (VAC). PCR is a powerful tool to detect and differentiate orthopox viruses, and real-time PCR has the further advantages of rapid turnaround time, low risk of contamination, capability of strain differentiation, and use of multiplexed probes. METHODS: We used real-time PCR with fluorescence resonance energy transfer technology to simultaneously detect and differentiate VAR, MPV, CPV/VAC, an

Detection, differentiation, and quantitation of pathogenic leishmania organisms by a fluorescence resonance energy transfer-based real-time PCR assay.

Real-time technology eliminates many of the pitfalls of diagnostic PCR, but this method has not been applied to differentiation of Leishmania organisms so far. We have developed a real-time PCR that simultaneously detects, quantitates, and categorizes Leishmania organisms into three relevant groups causing distinct clinical pictures. The analytical sensitivity (detection rate of >or=95% at 94.1 parasites/ml of blood) was within a range that has been determined previously to facilitate the confirmation of visceral leishmaniasis from peripheral blood. Parasites were successfully detected in 12 d

A fluorescence aptasensor for high sensitivity detection of Golgi protein 73 based on N, S-GQDs and MoS2@RGO via fluorescence resonance energy transfer mechanism.

Quantitative detection of Golgi protein 73 (GP73), a promising biomarker for hepatocellular carcinoma (HCC), is crucial for early diagnosis. In this study, we developed a fluorescence aptasensor for sensitive and specific detection of GP73 based on fluorescence resonance energy transfer (FRET) between GP73 aptamer-functionalized nitrogen/sulfur co-doped graphene quantum dots (N, S-GQDs-GP73Apt) as the fluorescence donor and molybdenum disulfide@reduced graphene oxide (MoS2@RGO) as the fluorescence acceptor. In the absence of GP73, the fluorescence of N, S-GQDs-GP73Apt was effectively quenched

A Förster Resonance Energy Transfer Switchable Fluorescent Probe With H2S-Activated Second Near-Infrared Emission for Bioimaging.

Real-time and accurate detection of endogenous hydrogen sulfide is of great biomedical significance. Here, a FRET-based fluorescent probe for ratiometric detection of H2S was designed to comprise an AIE luminophore TPE as an energy donor and a monochlorinated BODIPY dye as an energy acceptor and H2S-responsive site. Such a designed probe showed H2S-dependent ratiometric and light-up NIR-II emission, enabling accurate imaging of H2S-rich cancer cells and identification of H2S-rich tumors with high resolution.

Self-Catalyzed Surface Reaction-Induced Fluorescence Resonance Energy Transfer on Cysteine-Stabilized MnO2 Quantum Dots for Selective Detection of Dopamine.

A simple one-step ultrasonic method was developed for the synthesis of luminescent MnO2 quantum dots (MnO2 QDs) in the presence of cysteine, in which cysteine acted as the exfoliating agent and stabilization ligand. The cysteine-stabilized MnO2 QDs (Cys-MnO2 QDs) possess a fluorescence quantum yield of 4.7%, and the fluorescence intensity of Cys-MnO2 QDs is sensitive to dopamine (DA). The mechanism by which the Cys-MnO2 QDs catalyzed the self-polymerization of DA to form polydopamine nanoparticles (PDA NPs) and caused the fluorescence resonance energy transfer (FRET) between MnO2 QDs and PDA N

Synthesis and characterization of fluorescence resonance energy transfer-based nanoprobes by coating CdTe QDs with rhodamine B in gelatin nanoparticles.

A FRET-based nanoprobe consisting of CdTe quantum dot as donor and rhodamine B as acceptor embedded in gelatin nanoparticles was constructed and the photophysics of the system was characterized. The FRET-based sensors were led to at fixed donor/acceptor distance by synthesizing via a two-step desolvation way. This approach allowed for short acceptor-donor separation and thus for high energy transfer efficiencies. The morphology of the nanoprobes was presented spherically and relatively uniform with a diameter of about 100 nm, which was characterized by transmission electron microscope. The lum
