A Kinetic Origin of Vps34-IN-3 Selectivity for VPS34 over PI3Kα Revealed by Supervised Molecular Dynamics and Potential of Mean Force Analysis.

The rational design of selective kinase inhibitors is hampered by the conserved ATP-binding pockets. Vps34-IN-3, a potent VPS34 inhibitor developed by Genentech, Inc., showed ∼95-fold selectivity over the closely related PI3Kα, yet the atomic-level mechanism underlying this selectivity remains unclear. Here, we combined supervised molecular dynamics (SuMD) and potential of mean force (PMF) analysis to reconstruct the complete binding and unbinding pathways of Vps34-IN-3 to both targets. Our calculations revealed that selectivity arose primarily from a dramatically higher dissociation barrier f

Application of Pan-Viral Metagenomic Sequencing on United States Air Force Academy Wastewater to Uncover Potential Causes of Acute Gastroenteritis.

INTRODUCTION: Wastewater surveillance is an important technique to monitor public health and is being studied extensively for pandemic prevention, force health protection and readiness, and as a potential early warning system for chem-bio defense. Wastewater surveillance has traditionally relied on techniques such as quantitative PCR or targeted sequencing, both of which are microbe- or disease-specific tools. Newer pan-viral metagenomics strategies may provide higher specificity for pathogens of interest, resulting in a lower false negative rate and reduced sequencing of undesired background

A bradykinin potentiating peptide from Egyptian cobra venom strongly affects rat atrium contractile force and cellular calcium regulation.

Peptide fractions were isolated from venoms of the Egyptian snake Naja haje haje (cobra BPP) and the scorpions Buthus occitanus (BPP(B)) and Leirus quenquestriatus (BPP(L)). The pharmacological effects of these peptides were bioassayed and showed bradykinin potentiating activities. Amino acid analysis revealed that 14 amino acids contribute to the structure of BPP(B) and 16 for BPP(L), while cobra BPP was composed of 15 amino acids. Treatment of rat atrial preparations with 50 microg/ml of cobra BPP caused a significant reduction (P<0.001) in myocardial force. Elevation of extracellular calciu

18AA Reprograms the Pyruvate Cycle and Enhances Proton Motive Force to Potentiate Ceftazidime-Avibactam against Drug-Resistant Escherichia coli.

The rising threat of carbapenem-resistant Enterobacteriaceae demands novel strategies to restore antibiotic efficacy. Here we show that compound amino acid injection (18AA), a clinically approved parenteral nutrition formulation, markedly potentiates ceftazidime-avibactam (CZA) against multidrug-resistant and carbapenem-resistant Escherichia coli. Checkerboard and Bliss independence analyses demonstrated strong synergy. Time-kill kinetics confirmed dose-dependent bactericidal enhancement across susceptible, multidrug-resistant, and carbapenem-resistant strains. In a panel of 113 clinical isola

Sprint Acceleration Mechanics in Fatigue Conditions: Compensatory Role of Gluteal Muscles in Horizontal Force Production and Potential Protection of Hamstring Muscles.

Aim: Hamstring muscle injury is the main injury related to sports requiring sprint acceleration. In addition, hamstring muscles have been reported to play a role in horizontal force production during sprint acceleration performance. The aim of the present study was to analyze (i) the determinants of horizontal force production and (ii) the role of hip extensors, and hamstring muscles in particular, for horizontal force production during repeated sprint-induced fatigue conditions. Method: In this experimental laboratory setting study including 14 sprint-trained male athletes, we analyzed (i) th

Simultaneous mechanical stiffness and electrical potential measurements of living vascular endothelial cells using combined atomic force and epifluorescence microscopy.

The degree of mechanical stiffness of vascular endothelial cells determines the endogenous production of the vasodilating gas nitric oxide (NO). However, the underlying mechanisms are not yet understood. Experiments on vascular endothelial cells suggest that the electrical plasma membrane potential is involved in this regulatory process. To test this hypothesis we developed a technique that simultaneously measures the electrical membrane potential and stiffness of vascular endothelial cells (GM7373 cell line derived from bovine aortic endothelium) under continuous perfusion with physiological

Membrane knobs of unfixed Plasmodium falciparum infected erythrocytes: new findings as revealed by atomic force microscopy and surface potential spectroscopy.

Cerebral malaria, a severe complication of malaria, is caused by the obstruction of cerebral microvessels by Plasmodium falciparum-infected erythrocytes. Such cells adhere to endothelial cells by means of "knobs" induced on the red cell membrane by the parasites. When atomic force microscopy was used to investigate the structure of the knobs of unfixed infected red cells, each knob was found to consist of two distinct subunits, knob components that have never been seen in chemically fixed knobs examined by conventional transmission electron microscopy. Surface potential spectroscopy revealed t

There Are No Differences in Startle Conditioned Cervicomedullary Motor Evoked Potentials Across Isometric, Concentric, and Eccentric Muscle Actions at the Same Absolute Force Output.

Human movement involves a dynamic interplay of isometric, concentric, and eccentric muscle actions. There is a need to understand the contribution of the reticulospinal tract (RST) to human movement control during different muscle actions. This research aimed to determine the excitability of the RST during isometric, concentric, and eccentric muscle actions. Fourteen neurologically intact participants (age: 26 ± 7 years; sex: 3 female, 11 male; stature: 176 ± 8 cm; mass: 78.5 ± 10.9 kg) performed isometric, concentric, and eccentric muscle actions with the right biceps brachii. Riding (High) into the danger zone: a review of potential differences in chemical exposures in fighter pilots resulting from high altitude and G-forces.

When in flight, pilots of high performance aircraft experience conditions unique to their profession. Training flights, performed as often as several times a week, can expose these pilots to altitudes in excess of 15 km (~50,000 ft, with a cabin pressurized to an altitude of ~20,000 ft), and the maneuvers performed in flight can exacerbate the G-forces felt by the pilot. While the pilots specifically train to withstand these extreme conditions, the physiologic stress could very likely lead to differences in the disposition of chemicals in the body, and consequently, dangerously high exposures.

Nanoscale biophysical properties of small extracellular vesicles from senescent cells using atomic force microscopy, surface potential microscopy, and Raman spectroscopy.

Cells secrete extracellular vesicles (EVs) carrying cell-of-origin markers to communicate with surrounding cells. EVs regulate physiological processes ranging from intercellular signaling to waste management. However, when senescent cells (SnCs) secrete EVs, the EVs, which are newly regarded as senescence-associated secretory phenotype (SASP) factors, can evoke inflammation, senescence induction, and metabolic disorders in neighboring cells. Unlike other soluble SASP factors, the biophysical properties of EVs, including small EVs (sEVs), derived from SnCs have not yet been investigated. In thi

Simultaneous quantification of lower limb skeletal posture and ground reaction forces using upright computed tomography: Potential implications for osteoarthritis assessment and treatment.

Understanding the transmission of forces through the joints of the lower limb under weight-bearing conditions is critical for assessing the progression of knee and ankle osteoarthritis, as well as for developing effective surgical and preventive strategies. This study presents the first simultaneous quantification of three-dimensional skeletal posture and ground reaction force vectors during quiet standing, utilizing upright computed tomography and a custom-made force plate. We constructed the force plate using two six-axis force sensors, and the coordinate system of the CT was aligned with th

Cueing Dancers to "Externally Rotate From the Hips" Improves Potentially Injurious Ankle Joint Angles and Contact Forces During a Demipointe Ballet Position.

Introduction: The demipointe dance position puts the ankle at high risk of overuse injury and posteromedial ankle pain due to increased ankle valgus forces. Previous work has shown that creating lower limb external rotation intrinsic to demipointe with hip external rotation reduces foot pronation that causes ankle valgus stress. Therefore, the purpose of this work was to examine long axis rotation kinematics of the hip, knee, and ankle as well as the ankle joint contact forces in demipointe to better understand the biomechanical impact(s) of the specific cue to increase hip external rotation i

Sustained release of bone morphogenetic protein-4 in adult rabbit extraocular muscle results in decreased force and muscle size: potential for strabismus treatment.

Purpose. To assess the effect of a sustained-release preparation of bone morphogenetic protein-4 (BMP-4) on EOM force generation and muscle size. Methods. Sustained-release pellets, releasing 500 nanograms/day of BMP-4 for a maximum of 3 months, were implanted beneath the superior rectus muscle (SR) belly in anesthetized adult rabbits. The contralateral side received a placebo pellet as a control. After 1, 3, and 6 months, SRs were removed, and force generation at twitch and tetanic frequencies as well as fatigue resistance were determined in vitro. Beyond spinal stabilization: Engaging the functional core: The potential of non-invasive force sensor for clinical AWT measurement in adaptive LBP rehabilitation: A critical review.

BACKGROUND AND PURPOSE: LBP remains a pain point for millions worldwide. The interplay between the opoid crisis and LBP underscores the urgent need for effective, non-pharmacological solutions. Current approaches lack precision, particularly due to the limited understanding of core muscle function and the complex anatomical, biomechanical, and physiological factors involved. This study aims to explore the potential of Non-invasive force sensing (NIFS) technologies to quantify abdominal wall tension (AWT), intra-abdominal pressure (IAP), and anterolateral abdominal wall (ALAW) activation, colle

Development and validation of dual-cardiotoxicity evaluation method based on analysis of field potential and contractile force of human iPSC-derived cardiomyocytes / multielectrode assay platform.

A recent in vitro cardiovascular safety pharmacology test uses cardiomyocytes derived from human induced pluripotent stem cells (hiPSCs) to overcome the limitations of the classical test systems, such as species differences and local channel analysis. The Comprehensive in vitro Proarrhythmia Assay (CiPA) is a new proarrhythmia screening paradigm proposed by a CiPA steering expert group, which essentially requires iPSCs derived cardiomyocyte-based electrophysiological evaluation technology. Moreover, the measurement of the contractile force is also emerging as an important parameter to recapitu
