Lighting up forensic science by aggregation-induced emission: A review.

Forensic science requires a fast, sensitive, and anti-interfering imaging tool for on-site investigation and bio-analysis. The aggregation-induced emission (AIE) phenomenon exhibits remarkable luminescence properties (large Stokes shift, diverse molecular structures, and high photo-stability), which can provide a viable solution for on-site analysis, while at the same time overcoming the problem of aggregation-caused quenching (ACQ). Based on the outstanding performance in chemical analysis and bio-sensing, AIE materials have great prospects in the field of forensic science. Therefore, the app

Ancestral Information Analysis of Chinese Korean Ethnic Group via a Novel Multiplex DIP System.

Deletion/insertion polymorphism (DIP) is one of the more promising genetic markers in the field of forensic genetics for personal identification and biogeographic ancestry inference. In this research, we used an in-house developed ancestry-informative marker-DIP system, including 56 autosomal diallelic DIPs, three Y-chromosomal DIPs, and an Amelogenin gene, to analyze the genetic polymorphism and ancestral composition of the Chinese Korean group, as well as to explore its genetic relationships with the 26 reference populations. The results showed that this novel panel exhibited high genetic po

Mass spectrometry-based cDNA profiling as a potential tool for human body fluid identification.

Several mRNA markers have been exhaustively evaluated for the identification of human venous blood, saliva, and semen in forensic genetics. As new candidate human body fluid specific markers are discovered, evaluated, and reported in the scientific literature, there is an increasing trend toward determining the ideal markers for cDNA profiling of body fluids of forensic interest. However, it has not been determined which molecular genetics-based technique(s) should be utilized to assess the performance of these markers. In recent years, only a few confirmatory, mRNA/cDNA-based methods have bee

External visible characteristics prediction through SNPs analysis in the forensic setting: a review.

Numerous major advances have been made in forensic genetics over the past decade. One recent field of research has been focused on the analysis of External Visible Characteristics (EVC) such as eye colour, hair colour (including hair greying), hair morphology, skin colour, freckles, facial morphology, high myopia, obesity, and adult height, with important repercussions in the forensic field. Its use could be especially useful in investigative cases where there are no potential suspects and no match between the evidence DNA sample under investigation and any genetic profiles entered into crimin

Research progress and potential application of microRNA and other non-coding RNAs in forensic medicine.

At present, epigenetic markers have been extensively studied in various fields and have a high value in forensic medicine due to their unique mode of inheritance, which does not involve DNA sequence alterations. As an epigenetic phenomenon that plays an important role in gene expression, non-coding RNAs (ncRNAs) act as key factors mediating gene silencing, participating in cell division, and regulating immune response and other important biological processes. With the development of molecular biology, genetics, bioinformatics, and next-generation sequencing (NGS) technology, ncRNAs such as mic

The neurobiology of antisocial behavior in adolescence; current knowledge and relevance for youth forensic clinical practice.

Antisocial behavior in adolescents is generally seen as a neurodevelopmental problem; however, in spite of increasing knowledge on the neurobiology of persistent antisocial behavior, conduct disorders, and psychopathic traits, this knowledge is hardly used in clinical practice. The aim of this review is to give an overview of current research on the neurobiology of antisocial behavior in adolescents and to discuss how this knowledge can be translated to youth forensic clinical practice. First, an overview of recent literature on genetics, neuroimaging, neuropsychology, neurophysiology/neuroend

Allelic frequencies of 20 visible phenotype variants in the korean population.

The prediction of externally visible characteristics from DNA has been studied for forensic genetics over the last few years. Externally visible characteristics include hair, skin, and eye color, height, and facial morphology, which have high heritability. Recent studies using genome-wide association analysis have identified genes and variations that correlate with human visible phenotypes and developed phenotype prediction programs. However, most prediction models were constructed and validated based on genotype and phenotype information on Europeans. Therefore, we need to validate prediction

Population genetics of 30 insertion-deletion polymorphisms in two Chinese populations using Qiagen Investigator® DIPplex kit.

Insertion-deletion polymorphisms (INDELs) are short length diallelic polymorphisms caused by the insertion or deletion of several bases. INDEL markers can serve as useful supplementary or stand-alone assays for human identification. The Qiagen Investigator(®) DIPplex kit multiplexes 30 autosomal INDELs plus amelogenin for forensic use. The objective of this study was to estimate genetic diversity of 30 INDEL markers in the Han (the largest ethnic group of China, n=565) and She population (almost the smallest ethnic group of China, n=119), and to evaluate their usefulness in forensic genetics.

Population Genetic Data for 23 STR Loci of the Pech Ethnic Group in Honduras.

Background: The Pech ethnic group, comprising approximately 6024 individuals in northeastern Honduras, represents one of the country's smallest indigenous communities with a rich cultural heritage extending to pre-Columbian times. Despite their historical significance, no population genetic studies have been conducted on this group, and population-specific databases are essential for accurate forensic applications. Methods: Allele frequencies for 23 autosomal short tandem repeat (STR) loci were determined in 100 unrelated Pech individuals (58 females, 42 males) from communities in the departme

Human airborne eDNA as forensic evidence in simulated residential burglaries.

Airborne environmental DNA (eDNA) is emerging as a valuable tool in forensic genetics to support individual identification in cases where other biological traces are limited or absent. Here, we investigate the potential of human airborne eDNA as a source of forensic evidence by evaluating its recovery following simulated residential burglaries. A total of 391 eDNA samples were collected from air, dust, and surfaces in 22 households by two laboratories. Samples were recovered under two conditions: immediately after residents vacated the premises and following a 15-minute simulated burglary. Res

What do the X and Y chromosomes tell us about sex and gender in forensic case analysis?

Sex determination can be particularly crucial in forensic casework such as rape cases or cases of missing persons. Biological traces have to be genetically typed and the classification of the sex is of great importance for further investigations. Lately, several papers were published on reliability of sex determination by genetic typing of amelogenin gene-specific fragments. Problems may arise not only from false detection (or non-detection) of amelogenin-specific fragments, but also in cases of chimerism (bone marrow transplants) or micro chimerism (pregnant women carrying male fetuses), and

Pharmacogenetics and Forensic Toxicology: A New Step towards a Multidisciplinary Approach.

Pharmacogenetics analyzes the individual behavior of DNA genes after the administration of a drug. Pharmacogenetic research has been implemented in recent years thanks to the improvement in genome sequencing techniques and molecular genetics. In addition to medical purposes, pharmacogenetics can constitute an important tool for clarifying the interpretation of toxicological data in post-mortem examinations, sometimes crucial for determining the cause and modality of death. The purpose of this systematic literature review is not only to raise awareness among the forensic community concerning ph

Comparison of the Allelic Alterations between InDel and STR Markers in Tumoral Tissues Used for Forensic Purposes.

Background and objectives: Over the last two decades, human DNA identification and kinship tests have been conducted mainly through the analysis of short tandem repeats (STRs). However, other types of markers, such as insertion/deletion polymorphisms (InDels), may be required when DNA is highly degraded. In forensic genetics, tumor samples may sometimes be used in some cases of human DNA identification and in paternity tests. Nevertheless, tumor genomic instability related to forensic DNA markers should be considered in forensic analyses since it can compromise genotype attribution. Therefore,

Population Genetic Diversity and Clustering Analysis for Chinese Dongxiang Group With 30 Autosomal InDel Loci Simultaneously Analyzed.

In comparison with the most preferred genetic marker utilized in forensic science (STR), insertion/deletion analysis possesses further benefits, like absence of stutter peak, low mutation rate, and enabling mixed stain analysis. At present, a total of 169 unrelated healthy Dongxiang individuals dwelling in Dongxiang Autonomous county of Gansu province were recruited in our study to appraise the forensic usefulness of the panel including 30 autosomal diallelic genetic markers. The insertion allele frequencies were in the range of 0.1598 at HLD 111 to 0.8550 at HLD 118. The cumulative match of p

Sudden death of a young male with previously undiagnosed autosomal dominant polycystic kidney disease (ADPKD).

Autosomal dominant polycystic kidney disease (ADPKD) is one of the best known genetic diseases. However, in only very rare cases does it present as an abnormal death followed by clarification of its genetic background. We experienced a case in which ADPKD first became evident from the results of forensic autopsy, on the basis of which all potentially affected family members were offered genetic and other medical examinations. In this way, forensic medicine was able not only to determine the cause of death but to contribute to preventive medicine as well.
