Triple helix formation of a collagen model peptide induces cell aggregation formation.

Biofabrication refers to the creation of tissues and organs from biological materials such as cells and proteins, and is often applied in transplantation therapy and drug efficacy evaluation. For such a construct to function as a tissue, it is important for individual cells to interact closely with each other, so tailor-made approaches for the fabrication of cell aggregates to facilitate intercellular interactions are needed. Some researchers are using the DNA chain as a crosslinking site to make cell-cell assemblies, but DNA degradation and the potential risk of an immune response are concern

IMPERFECTIVE EXINE FORMATION (IEF) is required for exine formation and male fertility in Arabidopsis.

IEF, a novel plasma plasma membrane protein, is important for exine formation in Arabidopsis. Exine, an important part of pollen wall, is crucial for male fertility. The major component of exine is sporopollenin which are synthesized and secreted by tapetum. Although sporopollenin synthesis has been well studied, the transportation of it remains elusive. To understand it, we analyzed the gene expression pattern in tapetal microdissection data, and investigated the potential transporter genes that are putatively regulated by ABORTED MICROSPORES (AMS). Among these genes, we identified IMPERFECTI

Simultaneous Formate and Syngas Conversion Boosts Growth and Product Formation by Clostridium ragsdalei.

Electrocatalytic CO2 reduction to CO and formate can be coupled to gas fermentation with anaerobic microorganisms. In combination with a competing hydrogen evolution reaction in the cathode in aqueous medium, the in situ, electrocatalytic produced syngas components can be converted by an acetogenic bacterium, such as Clostridium ragsdalei, into acetate, ethanol, and 2,3-butanediol. In order to study the simultaneous conversion of CO, CO2, and formate together with H2 with C. ragsdalei, fed-batch processes were conducted with continuous gassing using a fully controlled stirred tank bioreactor.

Pyruvate-formate lyase-derived formate regulates the response of Salmonella Typhimurium to meropenem and ciprofloxacin.

Salmonella enterica serovar Typhimurium (STM) employs various strategies to endure antibiotic stress, but the contribution of central metabolites to resistance remains insufficiently characterized. In this work, we identify intracellular formate, produced by pyruvate-formate lyase (PflB), as a key determinant of STM susceptibility to meropenem and ciprofloxacin. Deletion of pflB disrupts cellular pH homeostasis and is associated with impaired efflux pump function, increased reactive oxygen species, and membrane depolarization, collectively correlating with heightened antibiotic sensitivity. Formation of hydroxyl radicals in the presence of ferritin and haemosiderin. Is haemosiderin formation a biological protective mechanism?

Horse spleen and human spleen ferritins increase the formation of hydroxyl radicals (OH) at both pH 4.5 and pH 7.4 in reaction mixtures containing ascorbic acid and H2O2. The generation of OH is inhibited by the chelator desferrioxamine. Human spleen haemosiderin also accelerates OH generation in identical reaction mixtures, but is far less effective (on a unit iron basis) than ferritin under all reaction conditions. It is proposed that conversion of ferritin into haemosiderin in iron overload is biologically advantageous in that it decreases the ability of iron to promote oxygen-radical react

Exploring Ectoine Production From Methanol, Formate, and Electrochemically Produced Formate by Methyloligella halotolerans.

Microbial synthesis using renewable C1-carbon sources like electrochemically produced formate (e-formate) represents a promising approach for climate-neutral chemical production. This study investigates formate utilization for ectoine biosynthesis by the halophilic methylotroph Methyloligella halotolerans. Preliminary growth assays confirmed formate utilization using 15-20 mM formate as the sole energy source substrate, when supplemented with yeast extract or vitamin solutions in a mineral salt medium. In a systematic study for ectoine production, formate utilization reached 0.305 ± 0.020 mmol

Biofilm Formation in Mycobacterial Genus; Mechanism of Biofilm Formation and Anti-mycobacterial Biofilm Agents.

Mycobacterium tuberculosis, Mycobacterium leprae, and non-tuberculous mycobacteria (NTM) are among the most significant human pathogens within the Mycobacterium genus. These pathogens can infect people who come into contact with biomaterials or have chronic illnesses. A characteristic pathogenic trait of mycobacteria is the development of biofilms, which involves several molecules, such as the GroEL1 chaperone, glycopeptidolipids, and shorter-chain mycolic acids. Bacterial behavior is influenced by nutrients, ions, and carbon sources, which also play a regulatory role in biofilm development. C

From formate oxidation to CO₂ reduction: The role of formate dehydrogenase in sustainable carbon utilization.

The escalation of global climate change and environmental degradation has made it imperative to develop innovative strategies to mitigate carbon dioxide (CO₂) emissions and enhance its utilization. Formate dehydrogenase (FDH) is a key enzyme capable of catalyzing the reversible conversion between CO₂ and formate. Due to its critical role in sustainable carbon recycling processes, FDH has garnered significant attention in recent times. This review offers a thorough analysis of FDH, emphasizing its dual function of converting one carbon (C1) substrates and providing reducing power. Recent advanc

Intracellular Eimeria bovis macromeront formation induces bystander cell accumulation and TNT formation.

INTRODUCTION: Eimeria bovis first merogony is an intracellular process (~ 3 weeks) resulting in the formation of large macromeronts (≤ 400 μm) containing up to 140,000 merozoites I, each. The production of merozoites I poses critical metabolic stress on bovine endothelial host cells, leading to mitochondrial dysregulation and premature senescence. In this context, an accumulation of non-infected bystander cells (BCs) around E. bovis macromeront-carrying host cells (MCHCs), eventually supporting MCHCs, was observed. METHODS: BC accumulation was quantified by 3D confocal microscopy. A meront-tr

In Situ Vivianite Formation in Intertidal Sediments: Ferrihydrite-Adsorbed P Triggers Vivianite Formation.

Coastal sediments are a key contributor to oceanic phosphorus (P) removal, impacting P bioavailability and primary productivity. Vivianite, an Fe(II)-phosphate mineral, can be a major P sink in nonsulfidic, reducing coastal sediments. Despite its importance, vivianite formation processes in sediments remain poorly understood. Here, we applied a novel approach to detect and quantify in situ vivianite formation in three intertidal flats. We conducted 7-week long incubations of mesh-bags filled with sediments mixed with (1) 57Fe-ferrihydrite, (2) 57Fe-ferrihydrite with adsorbed phosphate, and (3)

Photocatalytic treatment of IGCC power station effluents in a UV-pilot plant.

The aim of this work is to improve the quality of water effluents coming from an Integrated Gasification Combined Cycle (IGCC) power station to meet with future environmental legislation. This study has been made using an homogeneous photocatalytic oxidation process (UV/Fe(II)/H(2)O(2)) in a pilot plant. The efficiency of the process was determined from the analysis of the following parameters: cyanides, formates and TOC content. In the first stage, a factorial experimental design allowed to determine the influence of operation variables (initial concentration of H(2)O(2) and Fe(II), pH and te

Hydroxamate formation in Thermochelin Biosynthesis.

Nitrogen-hydroxylating monooxygenases (NMOs) are key enzymes in siderophore biosynthesis that rely on coordinated protein and cofactor conformational changes for catalysis. Here, we characterize TheA, an NMO from Thermocrispumagreste. Steady-state kinetics of TheA reveal a substrate preference for L-ornithine over L-lysine, with L-lysine primarily supporting uncoupled hydrogen peroxide formation rather than productive hydroxylation. TheA also shows a preference for NADPH over NADH and remains stable up to 50 °C. Pre-steady-state kinetics reveal that L-ornithine binding leads to a slower rate o

The formation, dynamics, and disassembly of caveolae.

Caveolae are a highly abundant surface feature of many mammalian cells, including skeletal muscle, smooth muscle, adipocytes, and fibroblasts. Dysregulation of caveolae has been implicated in a range of diseases, including muscular dystrophies, lipodystrophies, pulmonary hypertension, and various cancers. Despite their well-established links to human disease, our molecular understanding of caveolae function at the cellular level is only now emerging. Recent advances have begun to clarify how changes in caveolar components contribute to the diverse cellular processes attributed to these special

Kinetics of Amyloid Oligomer Formation.

Low-molecular-weight oligomers formed from amyloidogenic peptides and proteins have been identified as key cytotoxins across a range of neurodegenerative disorders, including Alzheimer's disease and Parkinson's disease. Developing therapeutic strategies that target oligomers is therefore emerging as a promising approach for combating protein misfolding diseases. As such, there is a great need to understand the fundamental properties, dynamics, and mechanisms associated with oligomer formation. In this review, we discuss how chemical kinetics provides a powerful tool for studying these systems.
