BACKGROUND: Tannerella forsythia (Tf) is a Gram-negative anaerobe implicated in the development of periodontal disease. Bacterial surface protein A (BspA) is a surface-expressed and -secreted protein that is recognized as an important virulence factor of Tf. This study was undertaken to determine whether Tf BspA induces an antibody response in periodontal disease. We hypothesized that serum immunoglobulin (Ig)G antibody levels against BspA correlate with the disease of patients.
METHODS: Sera were obtained from 100 patients with cardiac disorders and periodontal disease and 73 patients who ex

CONTEXT: Periodontitis, a chronic inflammatory disease affecting the periodontium, is primarily caused by gram-negative anaerobic bacteria. Conventional periodontal therapy aims to eradicate pathogenic microflora through mechanical debridement, often supplemented with chemical agents. However, the use of these chemical adjuvants is frequently associated with adverse drug reactions and the development of antimicrobial resistance. Consequently, researchers are increasingly exploring herbal antibacterial agents, such as Ocimum sanctum (Tulsi), as viable alternatives due to their broad-spectrum an

BACKGROUND: Accumulating evidence suggests that acute and chronic infections with periodontopathogens are associated with an increased risk of cardiovascular disease. The objective of this study was to assess whether Tanerella forsythia and Porphyromonas gingivalis are associated with increased levels of atherogenic low-density lipoprotein (LDL), high-density lipoprotein, total cholesterol (TC), triglycerides and body mass index (BMI) in chronic periodontitis patients.
MATERIAL AND METHODS: Medical history and clinical and radiographic examination were conducted in 80 chronic periodontitis pa

BACKGROUND: Periodontitis is a chronic inflammatory disease affecting more than 50% of the global population, resulting in irreversible gum damage, alveolar bone loss and ultimately tooth loss. Periodontitis arises from stable, multi-species biofilms whose pathogenicity is driven by emergent ecological interactions persistent to antimicrobial perturbations. Fusobacterium spp. are considered key microbial components that provide scaffolding for this ecological niche, which includes the pathobionts Tannerella forsythia, Prevotella intermedia and Porphyromonas gingivalis.
OBJECTIVES: To examined

Liver fibrosis, a cardinal feature of chronic liver injury, involves hepatic stellate cells (HSCs) activation and excessive extracellular matrix (ECM) deposition. Upon liver fibrosis, TLR4 is highly expressed on HSCs and Kupffer cells. TLR4 activation induces liver inflammation followed by exacerbated fibrosis. Inhibition of TLR4 signaling represents a validated anti-liver fibrosis therapeutic approach. In the realm of liver fibrosis treatment, traditional Chinese medicine (TCM) has demonstrated certain anti-fibrotic effects in clinical practice after long-term application. However, due to the

Porphyromonas gingivalis is considered the principal etiologic agent and keystone pathogen of chronic periodontitis. As an auxotrophic bacterium, it must acquire heme to survive and multiply at the infection site. P. gingivalis HmuY is the first member of a novel family of hemophore-like proteins. Bacterial heme-binding proteins usually use histidine-methionine or histidine-tyrosine residues to ligate heme-iron, whereas P. gingivalis HmuY uses two histidine residues. We hypothesized that other 'red complex' members, i.e. Tannerella forsythia and Treponema denticola might utilize similar heme u

BACKGROUND: T. forsythia a gram negative, anaerobe inhabits the mature biofilm present at sites expressing progressive periodontitis. It is a part of "red complex" group which contributes to the pathogenesis of periodontitis. The BspA protein and prtH gene encoded cysteine protease play a vital role in the virulence of T. forsythia. The present study aims to detect the two genotypes (bspA and prtH) in periodontitis and healthy subjects.
MATERIALS & METHOD: Subgingival plaque samples were collected from periodontitis patients and healthy subjects (Chronic Periodontitis n = 128, Aggressive Peri

Forsythia suspensa (Thunb.) Vahl. has been used widely in traditional medicines to treat gonorrhea, erysipelas, inflammation, pyrexia and ulcer. It has also shown antioxidant activity, as well as antibacterial, antiviral, choleretic and antiemetic effects. A high-speed counter-current chromatography (HSCCC) method was developed for the preparative separation and purification of the bioactive molecule phillyrin from F. suspensa (Thunb.) Vahl. The crude phillyrin was obtained by extraction with 50% ethanol from the dried fruits of F. suspensa (Thunb.) Vahl. under sonication. Preparative HSCCC wi

This study systematically investigated the effects and intrinsic mechanisms of inoculating SeNPs-enriched Rhodopseudomonas palustris (SR) on the physiological and biochemical traits, Cd accumulation, and root metabolome of Forsythia suspensa under Cd stress. A pot experiment was designed with a full factorial combination of three Cd concentrations (3.75, 7.5, and 15.0 mg kg-1 soil) and four SR inoculation levels (0.5 × 109, 1.0 × 109, 1.5 × 109, and 2.0 × 109 CFU mL-1). The results demonstrated that SR inoculation significantly reduced shoot Cd content by up to 54.14 % and dramatically enhance

The discovery of high-quality natural product-based enzyme inhibitors remains a major challenge due to the chemical complexity of herbal extracts. Although affinity-based ligand fishing and ultrafiltration are rapid methods for screening active compounds, non-specific interactions often interfere with the binding of high-quality active compounds to target enzymes, resulting in the fished components with weak or no biological activity. To overcome this limitation, we developed a novel hierarchical enrichment-driven ultrafiltration strategy by integrating affinity ultrafiltration (AUF) with soli

In this study, a homogenous polysaccharide (FSP), with an average molecular weight of 9.08 × 104 Da, was isolated from Forsythia suspense and its antibacterial potential against Enterobacter cloacae producing SHV-12 ESBL was investigated. Growth kinetics, in vitro competition and biofilm formation experiments demonstrated that SHV-12 ESBL contributed to a fitness benefit to E cloacae strain. The antibacterial activity of FSP (2.5, 5.0 and 10.0 μg/mL) was tested against E cloacae bearing SHV-12 ESBL gene using bacterial sensitivity, agar bioassay and agar well diffusion assays. It was found th

BACKGROUND: Pulmonary infections resulting from respiratory syncytial virus (RSV) continue to pose a significant threat to the well-being of infants and the elderly, but there is no safe, effective and specific treatment except symptomatic treatment. Forsythia Suspensa Leaf (FSL) is cold in nature and bitter in taste, and has the efficacy of clearing away heat and toxic materials. Previous research by our research group showed that the active components in FSL have the pharmacological effect of anti-RSV. Based on that, this study aims further to clarify the anti-RSV active components and mecha

Tannerella forsythia, a Gram-negative oral bacterium closely associated with chronic periodontitis, naturally produces outer membrane vesicles (OMVs). In this study, OMVs were purified by gradient centrifugation, and the proteome was investigated together with cellular fractions using LC-MS/MS analyses of SDS-PAGE fractions, resulting in the identification of 872 proteins including 297 OMV proteins. Comparison of the OMV proteome with the subcellular proteomes led to the localization of 173 proteins to the vesicle membrane and 61 proteins to the vesicle lumen, while 27 substrates of the type I

The oral pathogen Tannerella forsythia possesses a unique surface (S-) layer with a complex O-glycan containing a bacterial sialic acid mimic in the form of either pseudaminic acid or legionaminic acid at its terminal position. We hypothesize that different T. forsythia strains employ these stereoisomeric sugar acids for interacting with the immune system and resident host tissues in the periodontium. Here, we show how T. forsythia strains ATCC 43037 and UB4 displaying pseudaminic acid and legionaminic acid, respectively, and selected cell surface mutants of these strains modulate the immune r

INTRODUCTION: Porphyromonas gingivalis and Porphyromonas endodontalis belong to the Bacteroidota phylum. Both species inhabit the oral cavity and can be associated with periodontal diseases. To survive, they must uptake heme from the host as an iron and protoporphyrin IX source. Among the best-characterized heme acquisition systems identified in members of the Bacteroidota phylum is the P. gingivalis Hmu system, with a leading role played by the hemophore-like HmuY (HmuYPg) protein.
METHODS: Theoretical analysis of selected HmuY proteins and spectrophotometric methods were employed to determi
