Frozen section diagnosis in ophthalmic pathology.

Frozen section diagnosis is extensively used in various branches of pathology, but its application in ophthalmic pathology was recognised only in the 1970s. We studied 10 sections of ocular and adenexal lesions by frozen section diagnosis, which included orbital lesions (4 cases), lid lesions (3 cases), and intraocular tissue (1 case). The time taken for processing ranged between 10 to 15 minutes. Diagnoses based on frozen section evaluation included lymphoma, mesenchymal chondrosarcoma, solar keratosis, compound naevus, silicone oil globules in cataractous lens, neurofibromatosis, pseudotumou

Translation of frozen sections into FFPE images for skin cancer resection margins using generative AI.

Frozen section (Frozen) analysis is essential for intraoperative margin assessment in skin cancer surgery, but assessment is limited by freezing artifacts that obscure cellular detail. Formalin-fixed paraffin-embedded (FFPE) histopathology, the gold standard, provides higher quality but requires >24 h for processing. Herein, we developed and validated generative AI models to translate Frozen images into AI-generated FFPE images (GenFFPE) using 2594 slides from 283 cases across five major skin cancer types. Four unpaired image-to-image models (CycleGAN, CUT, AIFFPE, and SANTA) were trained and

Optimization of electron microscopy for human brains with long-term fixation and fixed-frozen sections.

BACKGROUND: Abnormal connectivity across brain regions underlies many neurological disorders including multiple sclerosis, schizophrenia and autism, possibly due to atypical axonal organization within white matter. Attempts at investigating axonal organization on post-mortem human brains have been hindered by the availability of high-quality, morphologically preserved tissue, particularly for neurodevelopmental disorders such as autism. Brains are generally stored in a fixative for long periods of time (often greater than 10 years) and in many cases, already frozen and sectioned on a microtome

Immunophenotyping of cutaneous lymphoid infiltrates in frozen and paraffin-embedded tissue sections: a comparative study.

A panel of antibodies reactive in routinely fixed, paraffin-embedded tissue sections was compared with a panel of antibodies reactive in frozen sections for the immunophenotyping of cutaneous lymphoproliferative disorders. Three T cell-associated markers (UCHL1, MT-1, MT-2, six B cell-associated markers (MB-1, MB-2, LN-1, LN-2, L-26, 4KB5), immunoglobulin heavy and light chains, anti-LCA antibody, two markers for Reed-Sternberg cells (Ber-H2, Leu-M1), one marker for macrophages (Mac-387) and anti-S-100 protein antibody were tested on normal skin, inflammatory skin diseases, and cutaneous lymph

Calcium uptake in frozen muscle biopsy sections compared with other predictors of malignant hyperthermia susceptibility.

The frozen-section Ca45 uptake procedure of Mabuchi and Sreter has been applied to 51 frozen human muscle biopsies previously tested for halothane and for caffeine contracture responses. In preliminary tests with cat muscle, uptake varied with skeletal muscle fiber type, was absent from smooth muscle, and was blocked by agents which damage or extract sarcoplasmic reticulum. In the human biopsies, however, the calcium uptake values showed no significant correlation with any of the following parameters: time of frozen-biopsy storage up to 5 yr; fiber type distribution; positive versus negative r

Frozen-section-guided breast-conserving surgery: implications of diagnosis by frozen section as a guide to determining the extent of resection.

This study was conducted to analyze retrospectively the results of performing sector resection on 56 breasts in 54 patients with breast cancer. The glands were resected with a 2-cm tumor-free margin on both lateral sides and the distal side, and with more than a 3-cm tumor-free margin on the nipple side. The frequency of positive resection margins for the cancer cells was 7/56 (12.5%) on the nipple side and 12/46 (26.1%) on the lateral sides, with an overall frequency of 15/56 (26.8%). There were positive resected margins for cancer cells on both the nipple and lateral sides in 4/46 patients (

Mohs micrographic surgery for dermatofibrosarcoma protuberans: our experience using frozen sections and a literature review.

INTRODUCTION AND OBJECTIVES: Mohs micrographic surgery (MMS) is considered the gold standard for the treatment of dermatofibrosarcoma protuberans (DFSP). Although paraffin-based MMS is the most widely accepted technique, no study has demonstrated its superiority over frozen-section MMS. We present our experience with frozen-section MMS for DFSP and review the literature. MATERIALS AND METHODS: We conducted a retrospective, observational, single-center study of all patients with DFSP treated with frozen-section MMS between July 2014 and July 2023. Demographic and tumor characteristics, surgica

Comparing Whole Slide Imaging and Light Microscopy to Identify Tumor in en face Frozen Sections.

OBJECTIVE: To assess accuracy of whole slide imaging (WSI) in the interpretation of permanent and frozen sections in surgical pathology and the identification of tumors in cutaneous en face frozen sections. METHODS: Twenty glass slides containing cutaneous en face frozen sections were selected from twenty cases of keratinocyte carcinomas treated with Mohs micrographic surgery. Ten slides contained tumor and ten did not. A blinded dermatologic surgeon used traditional light microscopy (LM) to assess physical slides for tumor presence and type, while noting the confidence (scale 1-10) and time

Accuracy of frozen sections in oral cancer resections, an experience of a tertiary care hospital.

Reactive gliosis and inflammation are risk factors for white matter injury (WMI) development, which are correlated with the development of many neurodevelopmental deficits with no treatment. This study aimed to understand the mechanisms correlated with WMI, with a particular focus on the role of nuclear factor‑kappa B (NF‑kB) and p38 mitogen‑activated protein kinases (MAPKs) pathways. Seven‑day‑old Wistar rats were used to generate cerebellar tissue slices. Slices were cultured and randomly allocated to one of 3 groups and treated as follows: group‑I (control); group‑II (WMI), slices were subj

Identification of IgE-positive cells and mast cells in frozen sections of multiple sclerosis brains.

There is evidence that nervous system mast cells may be involved in the pathogenesis of experimental demyelinating diseases, and that their involvement may be mediated by IgE. In order to examine the possible relevance of these observations to human demyelinating diseases, we have histologically and immunocytochemically examined multiple sclerosis (MS) and control brains for the presence of mast cells and IgE. Using a highly specific antiserum, we found IgE-positive cells in MS brains, but not in controls. IgE-positive cells could be detected in all regions of MS lesions, but were far more fre

[Comparative studies of simultaneous intraoperative frozen sections and imprint cytology of the mamma (author's transl)].

250 tissue biopsies of the mamma were examined simultaneously by intraoperative histology (frozen sections) and imprint cytology. In 95% of the cases there was a good agreement between the histologic and cytologic diagnoses. The reliability of the imprint cytology was tested in some complicated cases such as proliferating fibroadenomas, sclerosing adenosis and carcinomas and the significance of the imprint cytology for the diagnosis of difficult cases was pointed out in detail. The results have demonstrated, that such a cytologic examination can not be a substitute for intraoperative frozen se

Concordance in Distinguishing Actinic Keratosis from Squamous Cell Carcinoma in Situ on Mohs Histological Frozen Sections.

BACKGROUND: Mohs surgeons routinely encounter squamous cell carcinoma at surgical margins and often base the decision to take another layer on the severity of atypia observed. Currently, no criteria exists for distinguishing borderline histological patterns that could be interpreted differently as actinic keratosis (AK), squamous cell carcinoma in situ (SCCIS), or AK with focal SCCIS. OBJECTIVE: To assess interrater concordance amongst Mohs surgeons in distinguishing AK from SCCIS when evaluating Mohs frozen sections. METHODS: Seventeen slides were selected and converted into digitally forma

Immunohistochemistry (IHC): Chromogenic Detection of 3-Phosphohistidine Proteins in Formaldehyde-Fixed, Frozen Mouse Liver Tissue Sections.

The development of antibodies that specifically detect histidine-phosphorylated proteins is a recent achievement and allows potential roles of histidine phosphorylated proteins in pathological and physiological conditions to be characterized. Immunohistochemical analyses enable the detection of proteins in tissues and can reveal alterations to the quantity and/or localization of these proteins through comparisons of normal and diseased specimens. However, the sensitivity of phosphohistidine modifications to phosphatases, acidic pH, and elevated temperatures poses unique challenges to the detec

Ureteral and urethral frozen sections during radical cystectomy or cystoprostatectomy: an analysis of denudation and atypia.

OBJECTIVE: To evaluate denudation (no urothelium present for evaluation) and atypia (urothelial carcinoma in situ [CIS] cannot be ruled out) on frozen sections (FSs) of ureteral and urethral margins in radical cystectomies and cystoprostatectomies. MATERIALS AND METHODS: In radical cystectomies from 2000-2012, we compared FS diagnoses with the corresponding permanent section of the same tissue (frozen section control [FSC]). We also compared FS to "any CIS," which assessed whether there was any CIS in a given ureter or urethra, combining the diagnoses on "frozen section control" and on all su

Label-free histopathological diagnosis of frozen sections based on multi-excitation and broad-emission autofluorescence imaging.

Tumor surgery requires rapid intraoperative histopathological diagnosis to guide treatment and improve patient prognosis. Autofluorescence (AF) imaging offers a label-free approach with strong potential for rapid intraoperative use; however, most studies to date have been performed on deparaffinized sections based on bandpass emission settings. Its compatibility with frozen sections-which is essential for time-constrained intraoperative settings-remains unclear. Here, we systematically compared AF imaging of frozen sections with conventional deparaffinized sections, finding that the bandpass-f
