Aluminum and ABC transporter activity.

Aluminum is the third most common element on Earth´s crust and despite its wide use in our workaday life it has been associated with several health risks after overexposure. In the present study the impact of aluminum salts upon ABC transporter activity was studied in the P-GP-expressing human blood-brain barrier cell line hCMEC/D3, in MDCKII cells overexpressing BCRP and MRP2, respectively, and in freshly isolated, functionally intact kidney tubules from Atlantic killifish (Fundulus heteroclitus), which express the analog ABC transporters, P-gp, Bcrp and Mrp2. In contrast to previous findings

Fin duplications and deletions induced by disruption of retinoic acid signaling.

Retinoic acid (RA), a derivative of vitamin A, plays a critical role as a signaling molecule in axial patterning of vertebrates. Here we report that RA exposure of zebrafish (Danio rerio) and mummichog (Fundulus heteroclitus) embryos during gastrulation results in homeotic duplications of the pectoral fins in up to 94% of fish. We have observed three to four pairs of fins in an individual fish. Although some duplications are partial, many represent complete axial duplications of the pectoral girdle and fin and include coracoscapulae, proximal radials, and dermal fin elements. Fin duplications

Ultrastructural and histochemical aspects of zinc accumulation by fish scales.

The effect of zinc exposure on the ultrastructure of the scales and scale associated cells of the estuarine teleost, Fundulus heteroclitus was investigated in laboratory experiments. The Timm sulfide silver stain indicated that in the calcified region of the scales, Zn was colocalized with the calcium phosphate mineral crystals. X-ray diffraction analysis showed that Zn did not have an effect on crystal structure. The scale osteoblasts of Zn-exposed fish showed an increase in the number of lysosome-like structures contained by the cytoplasm. In Zn-exposed animals, X-ray microanalysis revealed

Low-conductance anion channel activated by cAMP in teleost Cl- -secreting cells.

We studied characteristics and modulation of ion channels in primary cultures of opercular epithelium from the euryhaline marine killifish Fundulus heteroclitus. Primary cultures, 17-28 h old, retain mitochondria-rich Cl- cells identifiable by fluorescence microscopy. Cell-attached patches revealed frequent low-conductance 8.1 +/- 0.35 pS channels that usually became inactive on excision; high-conductance anion channels were not apparent. Ion substitution experiments demonstrated selectivity for Cl- over gluconate of 1:0.07. With addition of 1-isobutyl-3-methylxanthine (0.1 mM) and dibutyrylad

Mummichog gill and operculum exhibit functionally consistent claudin-10 paralog profiles and Claudin-10c hypersaline response.

Claudin (Cldn)-10 tight junction (TJ) proteins are hypothesized to form the paracellular Na+ secretion pathway of hyposmoregulating mummichog (Fundulus heteroclitus) branchial epithelia. Organ-specific expression profiles showed that only branchial organs [the gill and opercular epithelium (OE)] exhibited abundant cldn-10 paralog transcripts, which typically increased following seawater (SW) to hypersaline (2SW) challenge. Post-translational properties, protein abundance, and ionocyte localization of Cldn-10c, were then examined in gill and OE. Western blot analysis revealed two Cldn-10c immun

Converging adrenergic and cholinergic mechanisms in the inhibition of Cl secretion in fish opercular epithelium.

The rate of Cl secretion (Isc) by the opercular epithelium of Fundulus heteroclitus is stimulated by elevations in cyclic AMP (cAMP) levels elicited via beta 1-adrenergic receptor activation, and inhibited by both alpha 2-adrenergic and muscarinic cholinergic receptor activation via mechanisms presently unknown. A comparison of these two inhibitory responses was made using clonidine, an alpha 2-adrenergic agonist, and acetylcholine (ACh), a cholinergic agonist. The dose required for maximum inhibition was 100 times greater for ACh, but in all other respects the responses elicited by both agoni

Effects of spironolactone and RU486 on gene expression and cell proliferation after freshwater transfer in the euryhaline killifish.

We have explored the possible mechanisms by which mineralocorticoid (MR) and glucocorticoid (GR) receptors regulate the response to freshwater transfer in the gills of the euryhaline killifish Fundulus heteroclitus. Killifish were implanted with RU486 (GR antagonist) or spironolactone (MR antagonist) at doses of 0.1-1.0 mg g(-1), and subsequently transferred from 10 per thousand brackish water to freshwater. Compared to brackish water sham fish, mRNA expression of CFTR and NKCC1 decreased in the gills of sham fish transferred to freshwater, whereas Na(+), K(+)-ATPase alpha(1a) mRNA expression

Experimental and natural warming elevates mercury concentrations in estuarine fish.

Marine food webs are the most important link between the global contaminant, methylmercury (MeHg), and human exposure through consumption of seafood. Warming temperatures may increase human exposure to MeHg, a potent neurotoxin, by increasing MeHg production as well as bioaccumulation and trophic transfer through marine food webs. Studies of the effects of temperature on MeHg bioaccumulation are rare and no study has specifically related temperature to MeHg fate by linking laboratory experiments with natural field manipulations in coastal ecosystems. We performed laboratory and field experimen

How Model Organisms and Model Uncertainty Impact Our Understanding of the Risk of Sublethal Impacts of Toxicants to Survival and Growth of Ecologically Relevant Species.

Understanding how sublethal impacts of toxicants affect population-relevant outcomes for organisms is challenging. We tested the hypotheses that the well-known sublethal impacts of methylmercury (MeHg) and a polychlorinated biphenyl (PCB126) would have meaningful impacts on cohort growth and survival in yellow perch (Perca flavescens) and Atlantic killifish (Fundulus heteroclitus) populations, that inclusion of model uncertainty is important for understanding the sublethal impacts of toxicants, and that a model organism (zebrafish Danio rerio) is an appropriate substitute for ecologically rele

The Elizabeth River Story: A Case Study in Evolutionary Toxicology.

The Elizabeth River system is an estuary in southeastern Virginia, surrounded by the towns of Chesapeake, Norfolk, Portsmouth, and Virginia Beach. The river has played important roles in U.S. history and has been the location of various military and industrial activities. These activities have been the source of chemical contamination in this aquatic system. Important industries, until the 1990s, included wood treatment plants that used creosote, an oil-derived product that is rich in polycyclic aromatic hydrocarbons (PAH). These plants left a legacy of PAH pollution in the river, and in parti

Type II iodothyronine deiodinase is preferentially expressed in rainbow trout (Oncorhynchus mykiss) liver and gonads.

It is well admitted that thyroid hormones (TH) play a role in the development of vertebrates. The major secretory product of the thyroid is a pro-hormone, T(4), which is activated in peripheral tissues by outer ring deiodination to T(3). We have isolated from rainbow trout testis, a full length cDNA encoding type II iodothyronine deiodinase (rtD2). The cDNA was 2410 nucleotides long and coded for a polypeptide of 264 amino acids including a selenocysteine residue. The predicted molecular weight of rtD2 was 29.3 kDa and the isoelectric point 8.71. The deduced amino acids sequence showed 80% ide

New insights into SREB orphan receptor roles in the ovary, using comparative transcriptomics across 3 fishes.

Superconserved receptors expressed in brain (SREB) are a family of orphan G protein-coupled receptors with 3 members in most vertebrates (GPR27 or SREB1, GPR85 or SREB2, and GPR173 or SREB3). They are associated with diverse physiological processes, ranging from glucose homeostasis to ovarian development. Despite a lack of confirmed ligands, our understanding of these receptors has increased based on interactions with putative SREB3 ligands such as phoenixin (PNX) and the development of synthetic SREB1 agonists (A8535 and PT-91). However, to date no studies have used these agonists in any comp

An examination of the factors influencing the bioaccumulation of methylmercury at the base of the estuarine food web.

Estuarine systems have received ongoing mercury (Hg) inputs from both point sources and regional contamination and have high legacy Hg in sediments. This is an environmental concern given that coastal seafood is an important vector for human exposure to methylmercury (MeHg). The base of the food chain represents the most important trophic steps for MeHg bioaccumulation. The magnitude of the uptake by phytoplankton, and their consumers, is influenced by many factors, in addition to sediment and water MeHg concentrations, that impact MeHg assimilation into phytoplankton and the trophic transfer

No adverse effects of three size classes of UV-aged and fragmented polyethylene microplastics on life-history traits of two marine teleost fish species in standardized toxicity tests.

Microplastics (MPs) detected in marine environments exhibit substantial variability in shape, size, polymer type, and other physicochemical properties. Although numerous laboratory studies have investigated the effects of MPs on marine organisms, studies specifically examining the effects of environmentally abundant aged polyethylene (PE) MPs on the early life stages of marine fishes, using officially recognized standardized test protocols remain limited. In this study, we evaluated the effects of additive-free, ultraviolet (UV)-aged, fragmented PE MPs (afPE-MPs) in three size classes (10, 30,

Molecular characterization of the medaka (Oryzias latipes) Hsp90ab1.

Nervous necrosis virus (NNV) can infect many species of fish and has caused severe economic losses to the aquaculture industry worldwide. Recently, there is a wealth of research indicating that heat shock protein 90ab1 (Hsp90ab1) is a receptor for red-spotted grouper nervous necrosis virus (RGNNV), however the potential function has not been examined so far. In this study, medaka (Oryzias latipes) hsp90ab1 gene (OlHsp90ab1) was identified, which contained 13 exons with a genome length of 5191 bp and coding sequences of 2175 bp. OlHsp90ab1 had 724 amino acids, containing three regions including

Paracellular pathway remodeling enhances sodium secretion by teleost fish in hypersaline environments.

In vertebrate salt-secreting epithelia, Na(+) moves passively down an electrochemical gradient via a paracellular pathway. We assessed how this pathway is modified to allow Na(+) secretion in hypersaline environments. Mummichogs (Fundulus heteroclitus) acclimated to hypersaline [2× seawater (2SW), 64‰] for 30 days developed invasive projections of accessory cells with an increased area of tight junctions, detected by punctate distribution of CFTR (cystic fibrosis transmembrane conductance regulator) immunofluorescence and transmission electron miscroscopy of the opercular epithelia, which form

Wood preservative leachates from docks in an estuarine environment.

Environmental concentrations and biological effects of certain metals and organic compounds found in wood preservatives were examined. The study focused on leachates from private residential docks in South Carolina tidal creeks. Copper, chromium, arsenic, and polynuclear aromatic hydrocarbons (PAHs) were measured in composite samples of surficial sediments and naturally occurring oyster populations (Crassostrea virginica) from creeks with high densities of docks, and from nearby reference creeks with no docks. In some cases, metal concentrations in sediments and oysters were higher immediately

The influence of nutrient loading on methylmercury availability in Long Island estuaries.

Estuaries provide critical habitat for food webs supporting fish and shellfish consumed by humans, but estuarine ecosystem health has been threatened by increases in nitrogen loading as well as inputs of the neurotoxin, mercury (Hg), which biomagnifies in food webs and poses risk to humans and wildlife. In this study, the effects of nutrient loading on the fate of Hg in shallow coastal estuaries were examined to evaluate if their interaction enhances or reduces Hg bioavailability in sediments, the water column, and concentrations in lower trophic level fish (Fundulus heteroclitus and Menidia m
