Hepatic fibrosis (HF) can be reversed via a self-repair mechanism targeting the activation of hepatic stellate cells (HSCs), which are considered the central driving force behind its progression. Currently, many inhibitors or antagonists of HF are in the development phase. However, research is often halted due to adverse reactions triggered by drug interactions or because the drugs fail to demonstrate significant efficacy, with many trials being discontinued at phase II. Natural polysaccharides have shown notable advantages in treating HF, making them promising candidates for therapy. This rev

BACKGROUND: The incidence of autoimmune diseases is increasing in developed countries, possibly due to the modern Western diet and lifestyle. We showed earlier that polysaccharides derived from the medicinal fungus Hirsutella sinensis produced anti-inflammatory, anti-diabetic and anti-obesity effects by modulating the gut microbiota and increasing the abundance of the commensal Parabacteroides goldsteinii in mice fed with a high-fat diet.
METHODS: We examined the effects of the prebiotics, H. sinensis polysaccharides, and probiotic, P. goldsteinii, in a mouse model of imiquimod-induced system

Sporothrix schenckii, Sporothrix brasiliensis, and Sporothrix globosa are etiological agents of sporotrichosis, a human subcutaneous mycosis. Although the protocols to evaluate Sporothrix virulence in animal models are well described, the cell preparation before inoculation is not standardized, and several culturing media are used to grow yeast-like cells. Here, we found that carbon or nitrogen limitation during fungal cell preparation negatively impacted the ability of S. schenckii and S. brasiliensis to kill Galleria mellonella larvae, but not S. globosa. The fungal growth conditions associa

Cryptococcus neoformans is an opportunistic fungal pathogen with a global distribution and is the causative agent of cryptococcosis, a disease that primarily affects immunocompromised individuals. The infection typically begins in the lungs and, in severe cases, can progress to meningoencephalitis and even death. Biochemical studies have shown that the fungal capsule is predominantly composed of glucuronoxylomannan (GXM), which accounts for approximately 88% of the total composition, followed by glucuronoxylomannogalactan (GXMGal, ∼10%) and mannoproteins (∼2%). Purified capsular components hav

Galactosaminogalactan (GAG) is an extracellular polysaccharide produced by the mycelium of the opportunistic human fungal pathogen Aspergillus fumigatus GAG is the first polysaccharide described as a virulence factor in medical mycology. This review presents our current knowledge of the structural organization and biosynthesis of this polymer. The function of this molecule as an adhesin that also masks Aspergillus PAMPs and the impact of GAG on the modulation of the host immune response by inducing neutropenia and blocking the IL-1 signaling pathway also will be emphasised.

Extracellular polymeric substances (EPSs) extracted from fungal mycelium by four chemical methods (NaOH, H2SO4, formaldehyde-NaOH, glutaraldehyde-NaOH), three physical methods (heating, ultrasound, vibration), and a control method (centrifugation alone) were investigated. Results indicated formaldehyde-NaOH outperformed other methods with 186.6 ± 8.0 mg/g of polysaccharides and 23.2 ± 4.6 mg/g of protein extracted and ensured little contamination by intracellular substances. Thereafter, this method was applied in extracting EPS from a mixed fungal culture in the adaptation process with 0.5% (w

The differential recognition of fungal cell wall polysaccharides that program innate and adaptive immunity to the human opportunistic fungal pathogen Aspergillus fumigatus has been a focus of considerable interest. In a mouse model of fungal conidia aspiration, decreased relative levels of cell wall core carbohydrates β-1,3-glucan to chitin in A. fumigatus isolates and mutant strains were correlated with increased airway eosinophil recruitment. In addition, an increase in fungal surface chitin exposure induced by the β-1,3-glucan synthesis-targeting drug caspofungin was associated with increas

Hepatocellular carcinoma (HCC) is a major cause of cancer mortality worldwide, and its limited response to conventional chemotherapeutics underscores the need for safer, multifunctional approaches. Natural polysaccharides, complex carbohydrates derived from plants, fungi, algae, animals, and bacteria, represent a structurally diverse class of bioactive macromolecules that combine therapeutic and material properties. Their monosaccharide composition, linkage types, branching patterns, molecular weights, and charge densities determine receptor recognition, intracellular trafficking, and signalin

The polysaccharide capsule of the fungus Cryptococcus neoformans is its main virulence factor. In this study, we determined the effects of mannitol and glucose on the capsule and exopolysaccharide production. Growth in mannitol significantly increased capsular volume compared with cultivation in glucose. However, cells grown in glucose concentrations higher than 62.5 mM produced more exopolysaccharide than cells grown in mannitol. The fibre lengths and glycosyl composition of capsular polysaccharide from yeast grown in mannitol was structurally different from that of yeast grown in glucose. Fu

Recent findings indicate that fungi use vesicular transport to deliver substances across their cell walls. Fungal vesicles are similar to mammalian exosomes and could originate from cytoplasmic multivesicular bodies. Vesicular transport enables the export of large molecules across the cell wall, and vesicles contain lipids, proteins and polysaccharides, many of which are associated with virulence. Concentration of fungal products in vesicles could increase their efficiency in food acquisition and/or delivering potentially noxious substances to other cells, such as amoebae or phagocytes. The di

Fungal extracellular vesicles (FEVs) have emerged as sophisticated emissaries in cross-kingdom communication, redefining our understanding of fungal secretome in pathogens like Cryptococcus neoformans and Candida species. Rather than mere cellular debris, these bilayered structures are actively exported across the fungal cell wall via complex biogenesis mechanisms, carrying a potent cargo of virulence factors, genetic material, proteins, lipids, and polysaccharides. In human hosts, FEVs facilitate myco-pathogenesis by interacting directly with immune cells to modulate the immune response, eith

Paracoccidioides brasiliensis is the causative agent of paracoccidioidomycosis or South American blastomycosis. Many aspects of the disease and its agent are unknown. One of the most important factors regarding the infection and the host-parasite relationships seems to be the fungal cell wall whose biochemical aspects are reviewed here. Biochemical studies, done mainly by Kanetsuna et al., (21,22), have demonstrated that the yeastlike (Y) and the mycelial (M) forms have chitin as a common polysaccharide, with alpha-1, 3-glucan in the Y form and beta-1, 3-glucan in the M form. These polysacchar

Cryptococcus neoformans is an opportunistic fungus that can cause lethal brain infections in immunosuppressed individuals. Infection usually occurs via the inhalation of a spore or desiccated yeast which can then disseminate from the lung to the brain and other tissues. Dissemination and disease is largely influence by the production of copious amounts of cryptococcal polysaccharides, both which are secreted to the extracellular environment or assembled into a thick capsule surrounding the cell body. There are two important polysaccharides: glucuronoxylomannan (GXM) and galactoxylomannan, also

Ganoderma lucidum spores are widely consumed natural functional materials; however, the influence of sporoderm disruption on the physicochemical characteristics and biological performance of spore polysaccharides remains insufficiently understood. In this study, polysaccharides extracted from intact spores (GLSP-I) and sporoderm-broken spores (GLSP-SB) were systematically compared to clarify how industrial processing affects their physicochemical properties and cytoprotective activity. Chemical characterization demonstrated that sporoderm disruption increased extraction yield and modified mole

Skin cancers, including basal cell carcinoma, squamous cell carcinoma, and melanoma, remain a major therapeutic challenge due to their prevalence, invasive potential, and the limited efficacy of conventional treatments. Fungal-derived secondary metabolites have emerged as promising sources of anticancer agents, acting through pro-apoptotic, anti-angiogenic, immunomodulatory, and anti-metastatic mechanisms. This review summarizes current evidence on representative fungal compounds - including cordycepin, ganoderic acid D, and immunomodulatory polysaccharides such as lentinan - with particular f

Invasive fungal infections (IFI) are an increasing threat to the developing world, with fungal spores being ubiquitous and inhaled every day. Some fungal species are commensal organisms that are part of the normal human microbiota, and, as such, do not pose a threat to the immune system. However, when the natural balance of this association is disturbed or the host's immune system is compromised, these fungal pathogens overtake the organism, and cause IFI. To understand the invasiveness of these pathogens and to address the growing problem of IFI, it is essential to identify the cellular proce

The Candida albicans high-affinity phosphate transporter Pho84 is required for normal Target of Rapamycin (TOR) signaling, oxidative stress resistance, and virulence of this fungal pathogen. It also contributes to C. albicans' tolerance of two antifungal drug classes, polyenes and echinocandins. Echinocandins inhibit biosynthesis of a major cell wall component, beta-1,3-glucan. Cells lacking Pho84 were hypersensitive to other forms of cell wall stress beyond echinocandin exposure, while their cell wall integrity signaling response was weak. Metabolomics experiments showed that levels of phosph
