5-(3-Phenylpropoxy)psoralen and 5-(4-phenylbutoxy)psoralen are blockers of the lymphocyte potassium channel Kv1.3 with EC50 values < 10 nM and may be used as new potential immunosuppressive drugs. As reported for many furocoumarins, these compounds show phototoxic properties and react under UV radiation with different targets in the cell, e.g. DNA or proteins and lipids in membranes. The photoreactions with these targets were investigated and compared to the well-known derivatives psoralen, 5-methoxypsoralen, 8-methoxypsoralen and 4,5',8-trimethylpsoralen. Moreover, absorption properties and t

The objective of our studies under the National Toxicology Program on psoralens was to evaluate a new furocoumarin derivative that would be highly efficacious and yet possess little or no systemic toxicity while also having a short effective biologic half-life. In addition, this work allowed for the development of a test system for compound evaluation of various psoralen derivatives. A guinea pig model was used first, followed by definitive studies in hairless mice for evaluation of the phototoxic potentialities of 32 furocoumarins and 4 benzofuran derivatives. Rank order depends on whether th

Excision repair of UV damage in human cells was measured by the incorporation of new bases into DNA after exposure to UV light and variety of other carcinogens including X-rays, furocoumarins plus 360 nm light (8-methoxypsoralen (8-MOP), 4'-aminomethyl 4,5',8-trimethylpsoralen hydrochloride (AMT) and angelicin), methyl methanesulfonate (MMS), dimethyl sulfate (DMS) and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). Repair of UV damage was unaffected by concomitant exposure to X-rays. Furocoumarin adducts and alkylating agents, however, interacted with UV repair and reduced the amount of repair r

Photoinduced binding of drugs to endogenous biomacromolecules may cause both toxic and therapeutic effects. For example, photobinding of certain phenothiazines to biomolecules possibly underlies their phototoxic and photoallergic potential, whereas photobinding of furocoumarins to epidermal DNA is held responsible for their advantageous effects in the photochemotherapy of psoriasis. Usually, the in vitro photobinding of drugs is investigated. However, under in vivo conditions, the metabolism and distribution of the drug and the light absorption by endogenous compounds will significantly affect

The presence and levels of furocoumarins in several parts of Ficus carica including the milky sap, were investigated. The results show that psoralen and bergapten are the only significant photoactive compounds, and are present in appreciable quantities in the leaf and shoot sap but are not detected in the fruit or its sap. These compounds are more concentrated in the leaf sap compared to the shoot sap. The psoralen levels are several times higher than those of bergapten. Lower concentrations of both compounds are present in autumn compared to spring and summer. These findings suggest that the

Pain is underdiagnosed and often not adequately treated, contributing to behavioral and psychological symptoms of dementia (BPSD). BPSD are treated with atypical antipsychotics that are associated with severe cerebrocardiovascular effects. Interestingly, treatment of pain may reduce agitation. Research is focusing on nonpharmacological treatment, such as aromatherapy, for pain and BPSD in dementia. This clinical study will assess the effect on agitation in severely demented elderly of BEO loaded in a nanotechnological odorless cream indistinguishable from placebo. This is a protocol for a rand

An efficient protocol for synthesizing furo[3,2-c]coumarin derivatives is described. The novel reaction could afford the desired furocoumarins with good to excellent yields in a mild and rapid manner. Large substrate scope screening and scale-up preparation have also been accomplished, and selected compounds were evaluated for their photophysical properties.

Furocoumarins (psoralen and its derivatives) are used to photoinactivate a variety of viruses and cell types. In the presence of long-wavelength ultraviolet light (UVA), furocoumarins bind covalently with pyrimidine residues via a cyclobutane ring. A second photoevent allows pyrimidines located on the opposite DNA strand in an adjacent base pair to react, forming a cross-link. In the experiments in this report, psoralen photoinactivation is employed to investigate human DNA repair pathways by analyzing the ability of xeroderma pigmentosum (XP) and Fanconi's anemia (FA) cells to rescue psoralen

The genus Ficus belongs to the Moraceae (the mulberry family). Figs can cause irritant or phototoxic reactions. Phytophotodermatitis is a common cutaneous phototoxic reaction. Contact with plant-derived phototoxic substances (furocoumarins or psoralens) followed by sunlight exposure produces the clinical lesions. Here, we present a case of phytophotodermatitis after contact with fig fruits and leaves. The vesicular dermatitis was primarily located in areas of vitiligo.

The most well-known cause of phytophotodermatitis in Denmark is accidental contact with plants belonging to the Apiaceae (Umbelliferae), such as hodgweed (Heracleum spondylium). However, many other plants with furocoumarins (psoralens) may induce the same kind of reaction. We describe a case of phytophotodermatitis caused by Dictamnus alba, gas plant or burning bush also known from the Bible as the "burning bush of Moses".

A series of angelicin heteroanalogues, in which the furan was replaced by thiophene or a 1-substituted pyrazole moiety, was synthesised in order to obtain potential therapeutic agents with antiproliferative and/or other biological activities. In general, the antiproliferative activity of the new thioangelicin, tested in different biological substrates, appeared to be higher than that of the angelicin, the natural parent compound, but lower than that of 8-MOP, the furocoumarin ordinarily used in PUVA therapy and photopheresis. Thioangelicin 6 induced strong inhibition of T2 bacteriophage infect

Citrus products are rich sources of furocoumarins, a class of photoactive compounds. Certain furocoumarins combined with ultraviolet radiation can induce skin cancer. We examined the relationship between citrus consumption and cutaneous melanoma risk among 56,205 Caucasian postmenopausal women in the Women's Health Initiative. Cox proportional hazards models were used to estimate hazard ratios (HRs) and 95% confidence intervals (CIs) of melanoma by citrus intake level. During a mean follow-up of 15.7 years, 956 incident melanoma cases were documented. In multivariable adjusted models, the HR (

The geographical pathology of hepatocellular carcinoma (HCC) was essentially anecdotal until systematic cancer registration was introduced, but it is now clear that sub-Saharan Africa is a high-incidence area. The disease is multifactorial in etiology, the possible etiological agents including hepatitis B virus and a number of chemical carcinogens, among which the most important appear to be the aflatoxins and N-nitroso compounds. Medicinal plants and herbal teas used in the tropics also contain compounds such as furocoumarins that are mutagenic and/or carcinogenic. A modified form of the Ames

Phytophotodermatitis is a phototoxic dermatologic reaction that occurs with exposure to ultraviolet light after contact with certain plant chemicals. Recognition is vital to proper management and avoidance of unnecessary distress for patients. Sun-sensitizing compounds, known as furocoumarins, are found in many plants (limes, celery, and natural grasses) and are excited by ultraviolet A (UVA) irradiation. UVA irradiation induces covalent linkages of nuclear DNA resulting in cutaneous photosensitivity and vesicular skin damage while increasing melanin production. Delayed erythema, hyperpigmenta

Khellin, a furochromone structurally related to furocoumarins, is a phototherapeutic agent used against psoriasis and vitiligo. This paper reports that pretreatment of HeLa cells with pertussis toxin reverses both the inhibition of NaF-stimulated adenylyl cyclase activity and the stimulation of GTPase by khellin alone and plus UVA light, as previously reported. Our results demonstrate, for the first time, that khellin is able to elicit an intracellular response by Gi alpha protein.

Babchi (Psorylea corylifolia) is occasionally used by patients as a herbal treatment in conditions such as psoriasis and vitiligo, due to its anti-inflammatory, anti-oxidant and immune-modulatory properties (Shrestha et al., 2018; Chopra et al., 2013). Due to its psoralen containing furocoumarins, it has the potential to induce photosensitive reactions. We present the first reported case of Babchi-induced phytophotodermatitis in the UK, presenting as sunburn.

Phytophotodermatitis is a skin eruption that occurs after contact with photosensitizing compounds in plants and exposure to UV light. There are two common presentations of phytophotodermatitis. Acutely, erythema and vesiculation similar to a severe sunburn are noted. After resolution of the inflammation, the involved skin has marked hyperpigmentation. Many plants have been identified that contain furocoumarins (psoralens), including limes, lemons, and celery. We present a patient with an acute phototoxic eruption and hyperpigmentation after contact with limes during a beach vacation.

Plants of the family Umbelliferae are known to cause a phytophotodermatitis due to a phototoxic reaction caused by furocoumarin (psoralens) and simultaneous exposure to sunlight. We report four patients with partial skin thickness burns, induced by this phototoxic reaction. One occurred after contact with parsley (Apium petroselinum) and three others after contact with giant hogweed (Heracleum mantegazzianum) and simultaneous exposure to sunlight.
