Fusobacterium nucleatum is a Gram-negative anaerobic oral commensal associated with periodontal disease. F. nucleatum has been implicated in a wide spectrum of systemic diseases, including oral, gastro-intestinal, rheumatologic, and vascular pathologies. As pregnancy risk has been linked to periodontal disease, there has also been significant research into the effects of periodontal disease on adverse pregnancy outcomes. This article reviews the epidemiological and mechanistic evidence of the role of F. nucleatum in adverse pregnancy outcomes.

An abscess in a gum pocket, resulting from bacterial infection, is a common source of chronic halitosis. Although antibiotics are generally prescribed for abscesses, they require multiple treatments with risks of creating resistant bacterial strains. Here we develop a novel vaccine using ultraviolet-inactivated Fusobacterium nucleatum (F. nucleatum), a representative oral bacterium for halitosis. A gum pocket model, established by continuous inoculation of F. nucleatum, was employed to validate the vaccine potency. Mice immunized with inactivated F. nucleatum effectively minimized the progress

We report the first description of portal and mesenteric vein thrombosis associated with suppurative mesenteric adenitis in a 71-year-old woman. The bacterium detected in mesenteric lymph nodes was Fusobacterium nucleatum, an anaerobic Gram-negative bacillus. Our patient had a clinical syndrome of pharyngitis and fever preceding portal vein thrombosis. Abdominal symptoms improved with antibiotics and anticoagulant therapy. This location of F. nucleatum in mesenteric lymph nodes provides an interesting insight into the occurrence of septic thrombosis in the portal vein following pharyngo-tonsil

Endometriosis is a chronic, inflammatory gynecological condition characterized by the ectopic growth of endometrial-like tissue, with an unclear etiology and limited treatment efficacy. Recent studies implicate the oral and gut commensal bacterium Fusobacterium nucleatum in the pathogenesis of endometriosis, with uterine colonization reported in up to 64% of affected women. This review highlights the potential role of F. nucleatum in disease progression, particularly through its metabolic activation within the endometrial microenvironment. We explore the contribution of key bacterial metabolit

Fusobacterium nucleatum is an anaerobic Gram-negative bacterium associated with periodontal disease and colorectal cancer. Outer membrane vesicles (OMVs) produced by F. nucleatum are nanoscale lipid vesicles enriched with immunogenic and virulence-associated proteins, making them promising candidates for vaccine development. This protocol describes a comprehensive workflow for culturing F. nucleatum, isolating and purifying OMVs via ultracentrifugation and density gradient ultracentrifugation, and characterizing their biophysical properties using transmission electron microscopy (TEM) and dyna

Fusobacterium nucleatum is implicated in esophageal cancer; however, its distribution in esophageal cancer tissues remains unknown. This study aimed to clarify the presence and distribution of F. nucleatum in esophageal cancer tissues using fluorescence in situ hybridization (FISH). Tissues collected from 70 patients with esophageal squamous cell carcinoma were examined using FISH. Corresponding normal epithelium and metastatic lymph nodes were assessed. F. nucleatum was identified more frequently in esophageal cancer tissues than in the normal epithelium. F. nucleatum also showed significant

Fusobacterium nucleatum and Helicobacter pylori are two microbial species increasingly recognized for their roles in gastrointestinal (GI) carcinogenesis, particularly in colorectal cancer (CRC) and gastric cancer (GC), respectively. While H. pylori has been long classified as a Group 1 carcinogen due to its well-characterized pathogenic mechanisms, F. nucleatum has more recently emerged as a key microbial contributor to CRC, with growing evidence linking it to tumor progression, immune evasion, and poor clinical outcomes. Despite occupying anatomically distinct niches within the GI tract, bot

Fusobacterium nucleatum, a gram-negative anaerobic bacterium abundantly found in the human oral cavity, is widely recognized as a key pathobiont responsible for the initiation and progression of periodontal diseases due to its remarkable aggregative capabilities. Numerous clinical studies have linked F. nucleatum with unfavorable prognostic outcomes in various malignancies. In further research, scholars have partially elucidated the mechanisms underlying F. nucleatum's impact on various types of cancer, thus gaining a certain comprehension of the role played by F. nucleatum in cancer. In this

The Gram-negative anaerobic species Fusobacterium nucleatum was originally described as a commensal organism from the human oral microbiome. However, it is now widely recognized as a key inflammophilic pathobiont associated with a wide variety of oral and extraoral diseases. Historically, F. nucleatum has been classified into four subspecies that have been generally considered as functionally interchangeable in their pathogenic potential. Recent studies have challenged this notion, as clinical data reveal a highly biased distribution of F. nucleatum subspecies within disease sites of both infl

BACKGROUND: Fusobacterium nucleatum (F. nucleatum) is a resident anaerobic bacterium, which in rare cases may invade blood from the head and neck or the digestive tract to cause bacteremia and induce venous thrombosis. F. nucleatum is closely related to abdominal tumors, but it has not been reported in relation to renal tumors. We report herein a possible case.
CASE PRESENTATION: This patient had kidney cancer with thrombosis in the right renal vein but had no sign of infection. After radical nephrectomy, thrombi formed in his left renal vein, and when removed, severe sepsis occurred. He did

Hydrogen sulfide and methyl mercaptan are the two major compounds associated with oral malodor. These compounds are highly toxic, and are thought to play an important role in periodontal disease. Fusobacterium nucleatum, an oral bacterium, produces large amounts of hydrogen sulfide from L-cysteine by the enzymatic action of L-cysteine desulfhydrase. We cloned and sequenced the cdl gene encoding L-cysteine desulfhydrase from F. nucleatum ATCC 10953, and revealed that the structural cdl gene consists of 921 bp and encodes a 33.4-kDa protein. The cloned gene was inserted into an expression vector

Understanding gene expression in Fusobacterium nucleatum and related species is essential for uncovering the molecular mechanisms that underlie their adaption to various environments. However, the isolation of high-quality RNA from these Gram-negative anaerobes is challenging because of their cell wall structure and the limitations of conventional column-based RNA purification kits, particularly their transcript size bias. Here, we present a robust, phenol-based RNA extraction protocol optimized for F. nucleatum that consistently yields high-integrity, DNA-free RNA suitable for transcriptomic

Fusobacterium is a genus of anaerobic Gram-negative bacteria that has been increasingly implicated in a range of diseases, including periodontitis and cancer. This review critically evaluates the reported role of Fusobacterium nucleatum in cancer, highlighting new findings that suggest a more nuanced cross-talk with the disease. We contextualise current evidence on the interactions with the wider tumour micro-environment, including the roles of polymicrobial communities, microbial metabolites and taxonomic heterogeneity. Collectively, the evidence suggests that Fusobacterium nucleatum should n

Fusobacterium nucleatum is frequently found in the colon microbiome of colorectal cancer (CRC) patients. The bacterium is not only a passive bystander of CRC, but is actively involved in disease progression through mediating tumor growth stimulation, metastasis, and immune evasion. These outcomes are achieved through the action of several adhesins that are located on the outer membrane surface of F. nucleatum, which bind to different receptors on tumor or immune cells. Adhesin-receptor interaction as the initial step of host-pathogen interaction then triggers signal transduction pathways respo

UNLABELLED: Many bacteria metabolize ethanolamine as a nutrient source through cytoplasmic organelles named bacterial microcompartments (BMCs). Here we investigated the molecular assembly, regulation, and function of BMCs in Fusobacterium nucleatum-a Gram-negative oral pathobiont that is associated with adverse pregnancy outcomes. The F. nucleatum genome harbors a conserved ethanolamine utilization (eut) locus with 21 genes that encode several putative BMC shell proteins and a two-component signal transduction system (TCS), in addition to the enzymes for ethanolamine transport and catabolism.

As evidence has been linking the oral bacterium Fusobacterium nucleatum (F. nucleatum) to colorectal tumorigenesis, we aimed to produce preliminary data on the expression of F. nucleatum in both oral and colorectal body sites in cases diagnosed with colorectal neoplasms (CRN) and CRN-free controls. We conducted a pilot hospital-based case-control study among patients who underwent colonoscopy examination. Saliva samples and biopsies from healthy colon mucosa from CRN cases and CRN-free controls, and from tumors in cases, were collected, as well as data on periodontal condition and potential CR

Fusobacterium nucleatum plays a key role in creating the pathogenic subgingival biofilm that initiates destructive periodontitis. It is also a common resident of the human gastrointestinal tract and has been associated with inflammatory bowel disease. The aim of the present study was to investigate the effects of green and black tea extracts as well as two of their bioactive components, EGCG and theaflavins, on the growth and virulence properties of F. nucleatum. The tea extracts and components displayed various degrees of antibacterial activity that may involve damage to the bacterial cell me

Porphyromonas gingivalis is associated with chronic periodontitis and forms multi-species biofilms. They can communicate within species as well as with other species found in the subgingiva, which may induce changes in the growth ratio and virulence of periodontopathogens. The lipopolysaccharide (LPS) of P. gingivalis shows different virulence by growth condition. The purpose of this study was to investigate the characteristics of P. gingivalis LPS when co-cultured with Fusobacterium nucleatum. After culture of P. gingivalis in the presence or absence of F. nucleatum, P. gingivalis LPS was ext

Endothelial cell senescence plays a critical role in the development of atherosclerosis. Fusobacterium nucleatum (Fn), a periodontal pathogen, has demonstrated pro-atherosclerotic effects, yet its role in endothelial senescence remains underexplored. This study aimed to elucidate whether and how Fn drives endothelial senescence and contributes to atherosclerosis. In vivo, chronic Fn infection accelerated atherosclerotic plaque progression, increased oxidative stress and vascular senescence, and impaired endothelial function. In vitro studies revealed that Fn induces dose- and time-dependent RO

The identification and quantification of Fusobacterium nucleatum subspecies is of paramount importance to researchers investigating fusobacterial composition in clinical specimens of human health and disease. Here, we present two validated methods to identify and/or quantify oral fusobacteria present within clinical specimens. Importantly, both approaches can unambiguously distinguish the four F. nucleatum subspecies and Fusobacterium periodonticum. The first method is a convenient PCR-based approach used for rapid genotyping of individual fusobacterial colonies and mixed cultures. The second
