Silver lampreys (Ichthyomyzon unicuspis) lack a gonadotropin-releasing hormone- and FMRFamide-immunoreactive terminal nerve.

The terminal nerve is a ganglionated cranial nerve with peripheral processes that enter the nasal cavity and centrally directed processes that enter the forebrain. Members of all classes of gnathostomes have been found to possess a terminal nerve, some components of which demonstrate immunoreactivity to the peptides Phe-Met-Arg-Phe-NH2 (FMRFamide) and gonadotropin-releasing hormone (GnRH). To explore the possibility that lampreys possess a terminal nerve, we examined the distribution of these peptides in the silver lamprey, Ichthyomyzon unicuspis, by using antisera to FMRFamide and to four for

Pattern of distribution and cycling of SLOB, Slowpoke channel binding protein, in Drosophila.

BACKGROUND: SLOB binds to and modulates the activity of the Drosophila Slowpoke (dSlo) calcium activated potassium channel. Recent microarray analyses demonstrated circadian cycling of slob mRNA. RESULTS: We report the mRNA and protein expression pattern of slob in Drosophila heads. slob transcript is present in the photoreceptors, optic lobe, pars intercerebralis (PI) neurons and surrounding brain cortex. SLOB protein exhibits a similar distribution pattern, and we show that it cycles in Drosophila heads, in photoreceptor cells and in neurosecretory cells of the PI. The cycling of SLOB is al

Netrin-3 protein is localized to the axons of motor, sensory, and sympathetic neurons.

The netrin family of axon guidance cues has been shown to play a pivotal role in the guidance of a variety of axon projections during embryonic development, both in the vertebrate and invertebrate. While the guidance potential of netrin-1 has been examined in depth in many regions of the developing mouse brain very little information is available on the expression and activity of netrin-3. Here we show that the netrin-3 protein is present on motor neurons and subpopulations of neurons within sensory and sympathetic ganglia. Moreover, significant levels of netrin-3 protein were found to be asso

Distribution of PACAP-like immunoreactivity in the nervous system of oligochaeta.

The marked similarity between the primary structures of human, other vertebrate, and the invertebrate tunicate PACAP suggests that PACAP is one of the most highly conserved peptides during the phylogeny of the metazoans. We investigated the distribution of PACAP-like immunoreactivity in the nervous system of three oligochaete (Annelida) worms with immunocytochemistry. The distribution pattern of immunoreactivity was similar in all three species (Lumbricus terrestris, Eisenia fetida, and Lumbricus polyphemus). The cerebral ganglion contains numerous immunoreactive cells and fibers. A few cells

Component placement optimization in the brain.

This computational neuroanatomy study evaluates how well some formalisms derived from combinatorial network optimization theory fit as models for brain structure. At multiple hierarchical levels--brain, ganglion, individual cell--physical placement of neural components appears consistent with a single, simple goal: minimize cost of connections among the components. The most dramatic instance of this "save wire" organizing principle is reported for adjacencies among ganglia in the nematode nervous system; among about 40,000,000 alternative layout orderings, the actual ganglion placement in fact

Neuronal processing of complex sounds in the prothoracic ganglion of a bushcricket.

Male bushcrickets produce complex species-specific songs to attract mating partners. Since the neuronal processing underlying song-pattern detection is not fully understood, we analysed extracellularly recorded song-induced spike responses and local field potentials across the first stage of auditory processing in the prothoracic ganglion of Mecopoda elongata. We analysed the influence of the temporal structure and of any change in amplitude of spiking and local field potentials. We presented the complete chirp and compared the neuronal response pattern with those responses evoked individually

Neuropeptide modulation of bidirectional internetwork synapses.

Oscillatory networks underlying rhythmic motor behaviors, and sensory and complex neural processing, are flexible, even in their neuronal composition. Neuromodulatory inputs enable neurons to switch participation between networks or participate in multiple networks simultaneously. Neuromodulation of internetwork synapses can both recruit and coordinate a switching neuron in a second network. We previously identified an example in which a neuron is recruited into dual-network activity via peptidergic modulation of intrinsic properties. We now ask whether the same neuropeptide also modulates int

Motor neurons within a network use cell-type specific feedback mechanisms to constrain relationships among ion channel mRNAs.

Recently, activity has been proposed as a primary feedback mechanism used by continuously bursting neurons to coordinate ion channel mRNA relationships that underlie stable output. However, some neuron types only have intermittent periods of activity and so may require alternative mechanisms that induce and constrain the appropriate ion channel profile in different states of activity. To address this, we used the pyloric dilator (PD; constitutively active) and the lateral gastric (LG; periodically active) neurons of the stomatogastric ganglion (STG) of the crustacean Cancer borealis. We experi

Similarities and differences in circuit responses to applied Gly1-SIFamide and peptidergic (Gly1-SIFamide) neuron stimulation.

Microcircuit modulation by peptides is well established, but the cellular/synaptic mechanisms whereby identified neurons with identified peptide transmitters modulate microcircuits remain unknown for most systems. Here, we describe the distribution of GYRKPPFNGSIFamide (Gly1-SIFamide) immunoreactivity (Gly1-SIFamide-IR) in the stomatogastric nervous system (STNS) of the crab Cancer borealis and the Gly1-SIFamide actions on the two feeding-related circuits in the stomatogastric ganglion (STG). Gly1-SIFamide-IR localized to somata in the paired commissural ganglia (CoGs), two axons in the nerves

Maintaining phase of the crustacean tri-phasic pyloric rhythm.

We construct and analyze a model network of the pyloric rhythm of the crustacean stomatogastric ganglion consisting of an oscillator neuron that inhibits two reciprocally inhibitory follower neurons. We derive analytic expressions that determine the phase of firing of the follower neurons with respect to the oscillator. An important aspect of the model is the inclusion of synapses that exhibit short-term synaptic depression. We show that these type of synapses allow there to be a complicated relationship between the intrinsic properties of the neurons and the synapses between them in determini

Neurotoxicity of paraquat and triphenyltin in the earthworm, Eisenia fetida Sav. A histo- and cytopathological study.

Neuropathological effects of the pesticides paraquat (PQ, a herbicide) and triphenyltin (TPT, a fungicide) were studied on the postclitellar segmental ganglia of juvenile E. fetida specimens using light and electron microscopic methods. Growth retardation of the worms in standard culture medium was used as quantitative marker of sublethal effect. There is a distinct neuron group in the segmental ganglia characterised by high sensitivity against PQ- and another one against TPT-toxication. However the marked histo- and neurocytopathological alterations were similar in PQ and TPT treated worms. B

Analysis of inter-leg coordination mechanisms in cricket locomotion: insights from thoracic Ganglion network transection.

Adaptive insect locomotion depends on interactions between the nervous system and body dynamics, yet how these components contribute to inter-leg coordination remains unclear. We investigated the role of direct neural coupling by unilaterally transecting the posterior intermediate connective linking the mesothoracic and metathoracic ganglia in the cricketGryllus bimaculatus. We analyzed kinematic changes and compared them with predictions from a purely neural phase oscillator network model featuring hierarchical asymmetric coupling. The experiments showed a clear dissociation: transection disr

Transcriptome analysis of eyestalk ganglion provides new insights into the immune response of Eriocheir sinensis.

Generally, eyestalk ganglion is regarded as a basic neuroendocrine tissue in decapods. In the past decades, increasing evidence has implied that the nervous system plays important roles in modulating the immune defense against pathogenic microorganisms. However, the molecular mechanisms of nervous system on the innate immunity of decapods remain largely unclear. In this study, we investigated the transcriptomic responses of eyestalk ganglion in Eriocheir sinensis challenged by Vibrio parahaemolyticus. A total of 77 differentially expressed genes (DEGs) were identified, and these DEGs were pred

Neuromodulation Reduces Interindividual Variability of Neuronal Output.

In similar states, neural circuits produce similar outputs across individuals despite substantial interindividual variability in neuronal ionic conductances and synapses. Circuit states are largely shaped by neuromodulators that tune ionic conductances. It is therefore possible that, in addition to producing flexible circuit output, neuromodulators also contribute to output similarity despite varying ion channel expression. We studied whether neuromodulation at saturating concentrations can increase the output similarity of a single identified neuron across individual animals. Using the latera

Synaptic modulation of the interspike interval signatures of bursting pyloric neurons.

The pyloric network of the lobster stomatogastric nervous system is one of the best described assemblies of oscillatory neurons producing bursts of action potentials. While the temporal patterns of bursts have been investigated in detail, those of spikes have received less attention. Here we analyze the intraburst firing patterns of pyloric neurons and the synaptic interactions shaping their dynamics in millisecond time scales not performed before. We find that different pyloric neurons express characteristic, cell-specific firing patterns in their bursts. Nonlinear analysis of the interspike

Neuropeptides are ubiquitous chemical mediators: Using the stomatogastric nervous system as a model system.

The stomatogastric nervous system (STNS) controls the movements of the foregut and the oesophagus of decapod crustaceans and is a good example for demonstrating that peptides are ubiquitously distributed chemical mediators in the nervous system. The stomatogastric ganglion (STG), one of the four ganglia of the STNS, contains the most intensively investigated neuronal circuits. The other ganglia, including the two commissural ganglia (CoGs) and the oesophageal ganglion (OG), are thought to be modulatory control centres. Peptides reach the STNS either as neurohormones or are released as transmit

Outgrowth patterns and directed growth of identified neurons induced by native substrates in culture.

In this study, we have tested how various identified leech neurons in culture grow on surfaces that they normally contact in situ. Neurons were cultured either on ganglion capsules from which neurons had been removed or on skin. On these substrates, outgrowth patterns were characteristic for each cell type. Retzius cells plated on capsules extended bundles of thick, fasciculated processes with few branching points and in the opposite direction a tangle of fine neurites. Anterior pagoda (AP) neurons plated on capsules extended two single processes in opposite directions but failed to grow on sk

A hormone-activated central pattern generator for courtship.

BACKGROUND: Medicinal leeches (Hirudo spp.) are simultaneous hermaphrodites. Mating occurs after a stereotyped twisting and oral exploration that result in the alignment of the male and/or female gonopores of one leech with the complementary gonopores of a partner. The neural basis of this behavior is presently unknown and currently impossible to study directly because electrophysiological recording techniques disrupt the behavior. RESULTS: Here we report that (Arg(8))-conopressin G and two other members of the oxytocin/vasopressin family of peptide hormones induce in Hirudo verbana a sequenc

Alcohol-, nicotine-, and cocaine-evoked release of morphine from human white blood cells: substances of abuse actions converge on endogenous morphine release.

BACKGROUND: Normal human white blood cells (WBC) have the ability to synthesize morphine as do invertebrate ganglia. Furthermore, invertebrate neural tissues incubated with ethanol, cocaine, or nicotine results in a statistically significant enhancement of labeled morphine release. We now demonstrate that this also occurs with human WBC. MATERIAL/METHODS: Human blood was obtained from the Long Island Blood Services (Melville, NY). Polymorphonuclear cells (PMN) or mononuclear cells (MN) (10 million/ml) were bathed in phosphate buffered saline (PBS) medium containing purified RIA grade 125I-lab

TENS stimulates constitutive nitric oxide release via opiate signaling in invertebrate neural tissues.

BACKGROUND: There is a major societal concern relating to the addictive properties of analgesic drugs such as morphine with regard to alleviating pain. Because of this, alternative methods of pain relief are, and have been, actively pursued. An extremely promising method for treatment of low to moderate levels of chronic pain in humans is transcutaneous electrical nerve stimulation (TENS). MATERIAL/METHODS: All experiments utilized the invertebrate marine bivalve mollusc Mytilus edulis pedal ganglia. TENS was achieved using a stimulation apparatus developed by Professor Han of Peking Universi
