Five novel sulfur-containing benzyl metabolites, designated as gastrabenzylsulfoxides A and B (1 and 2), gastrabenzylsulfinate A (3) and gastrabenzylsulfides A and B (4 and 5), along with four known compounds (6-9), were isolated from the aqueous extracts of Gastrodia elata. Compounds 1 and 4 are 4-hydroxy-3-(4'-hydroxybenzyl)benzyl-substituted sulfoxide and sulfide, respectively, which are unprecedented in natural products. Compound 3 represents a rare sulfinate. Several isolates and their sulfone and disulfide analogs (10-13) were synthesized to evaluate their anti-inflammatory activity. Not

Acetylcholinesterase (AChE) inhibitors are crucial for the symptomatic management of Alzheimer's disease (AD), with natural products-particularly botanical sources like Yellow Gastrodia elata (YGE)-serving as promising reservoirs of such inhibitors. Nevertheless, comprehensive screening and mechanistic characterization of their inhibitory potential remain limited. This study sought to identify potent AChE inhibitors from YGE, investigate their mechanisms of action, and assess their therapeutic prospects for AD. Methodologically, an integrated approach was employed, combining ultrafiltration-li

Hypertension has become a major contributor to the morbidity and mortality of cardiovascular diseases worldwide. Despite the evidence of the anti-hypertensive effect of gastrodia-uncaria granules (GUG) in hypertensive patients, little is known about its potential therapeutic targets as well as the underlying mechanism. GUG components were sourced from TCMSP and HERB, with bioactive ingredients screened. Hypertension-related targets were gathered from DisGeNET, OMIM, GeneCards, CTD, and GEO. The STRING database constructed a protein-protein interaction network, visualized by Cytoscape 3.7.1. Co

Gastrodia elata Bl. is a widely used traditional Chinese medicine known for its medicinal properties. However, during the drying process, G. elata is often fumigated with sulfur to prevent corrosion and improve its appearance. Sulfur-fumigation can result in a reduction in the effective components of the herb and can also be hazardous to human health due to the remaining sulfur dioxide. Sulfur-fumigation of G. elata poses a significant challenge to both end-users and researchers. The detection of p-hydroxybenzyl hydrogen sulfite (p-HS) is a useful tool in determining whether G. elata has been

Gastrodia elata Blume is a vital herb requiring precise subtype discrimination for market authentication and traceability. However, the current analytical focus on small molecules limits the extraction of highly discriminative polysaccharide features, making chemometric authentication under class-imbalanced conditions highly challenging. To address these limitations, we established a robust multi-detector size-exclusion chromatography platform utilizing a 5 mM NH₄Ac mobile phase to induce a mixed-mode separation, effectively resolving the cryptic structural heterogeneities and multidimensional

Microgravity during long-duration spaceflights promotes oxidative damage, impairs mitochondrial efficiency, and induces neurodegeneration, highlighting the need for safe countermeasures. Gastrodia elata (Tianma) is a traditional medicinal herb with known antioxidant and neuroprotective properties that has not yet been examined under microgravity-analog conditions. The main aim of this study was to evaluate whether Tianma mitigates oxidative, mitochondrial, and neurobehavioral deficits induced by simulated microgravity (SM) in Caenorhabditis elegans (C. elegans). Plant-derived extracellular vesicles (PDEVs) are nanoscale lipid bilayer structures that facilitate cross-kingdom communication by transporting bioactive molecules, including proteins and small RNAs. While their roles in plant immunity against fungal pathogens are well-established, their functions in beneficial or symbiotic interactions, particularly with mycorrhizal fungi, remain largely unexplored. Here, we investigated the function of PDEVs in the symbiosis between the fully mycoheterotrophic orchid Gastrodia elata and its fungal partner Armillaria gallica. We isolated and characterized G.

BACKGROUND: According to Compendium of Materia Medica, Gastrodia elata (GE) Blume as a top grade and frequently prescribed herbal medicine has been used in treating dizziness, headaches, and epilepsy, indicating a neuroprotective effect. Because GE is capable of suppressing a hyperactive liver and thus calming endogenous wind, and because Huntington's disease (HD) can be classified as a phenomenon of disturbed liver wind, it is suggested that GE might be beneficial in treating HD. However, although current studies support GE for the prevention of diverse neurodegenerations such as HD, its deta

BACKGROUND: Insomnia and depressive disorder are two common symptoms with a reciprocal causal relationship in clinical practice, which are usually manifested in comorbid form. Several medications have been widely used in the treatment of insomnia and depression, but most of these drugs show non-negligible side effects. Currently, many treatments are indicated for insomnia and depressive symptom, including Chinese herbal medicine such as Gastrodia elata Blume (G. elata), which has excellent sedative-hypnotic and antidepressant effects in clinical and animal studies.

BACKGROUND: Fully mycoheterotrophic orchids depend entirely on fungal partners for carbon and mineral nutrition, yet the structural mechanisms that regulate fungal accommodation within host tissues remain poorly understood. This question is particularly relevant in the Gastrodia elata-Armillaria symbiosis, where the fungal partner is a wood-decaying basidiomycete capable of aggressive host colonization and responsible for root rot disease in many tree species. Here, we examined whether host cell wall remodeling and extensin deposition contribute to the establishment and regulation of the host-

BACKGROUND: Excessive consumption of high-fat diet (HFD) can easily cause obesity, hyperlipidemia, and fatty liver disease. Persistent elevation of blood lipid levels increases the risk of cardiovascular disease, with hyperlipidemia being a well-established risk factor. Gastrodia elata Blume (GEB) is a perennial orchid plant that is known to have beneficial effects on obesity and blood circulation. This study aimed to evaluate the effects of GEB extract on improving blood lipids in the hyperlipidemia model induced by HFD in rats. Wistar rats (five-week-old) were divided into 3 groups: Control

This study aimed to fabricate a novel codelivery system to simultaneously load β-carotene and curcumin in a controlled and synergistic manner. We hypothesized that the aggregates of octenylsuccinated Gastrodia elata starch (OSGES) could efficiently load and control the release of β-carotene and curcumin in combination. Mechanisms underlying the self-assembly of OSGES, coloading, and corelease of β-carotene and curcumin by relevant aggregates were studied. The OSGES could form aggregates with a size of 120.2 nm containing hydrophobic domains surrounded by hydrophilic domains. For coloading, the
