Self-Assembling Nanoclay Diffusion Gels for Bioactive Osteogenic Microenvironments.

Laponite nanoparticles have attracted attention in the tissue engineering field for their protein interactions, gel-forming properties, and, more recently, osteogenic bioactivity. Despite growing interest in the osteogenic properties of Laponite, the application of Laponite colloidal gels to host the osteogenic differentiation of responsive stem cell populations remains unexplored. Here, the potential to harness the gel-forming properties of Laponite to generate injectable bioactive microenvironments for osteogenesis is demonstrated. A diffusion/dialysis gelation method allows the rapid format

Generation of 3D Collagen Gels with Controlled Diverse Architectures.

Rat tail collagen solutions have been used as polymerizable in vitro three dimensional (3D) extracellular matrix (ECM) gels for single and collective cell migration assays as well as spheroid formation. Factors such as ECM concentration, pH, ionic concentration, and temperature can alter collagen polymerization and ECM architecture. This unit describes how to generate 3D collagen gels that have distinct architectures ranging from a highly reticular meshwork of short thin fibrils with small pores to a loose matrix consisting of stiff, parallel-bundled long fibrils by changing collagen polymeriz

Physicochemical properties of surimi gels fortified with dietary fiber.

Although dietary fiber provides health benefits, most Western populations have insufficient intake. Surimi seafood is not currently fortified with dietary fiber, nor have the effects of fiber fortification on physicochemical properties of surimi been thoroughly studied. In the present study, Alaska pollock surimi was fortified with 0-8 g/100 g of long-chain powdered cellulose as a source of dietary fiber. The protein/water concentrations in surimi were kept constant by adding an inert filler, silicon dioxide in inverse concentrations to the fiber fortification. Fiber-fortified surimi gels were

Arginine-rich self-assembling peptides as potent antibacterial gels.

Hydrogel materials that display inherent activity against bacteria can be used to directly treat accessible wounds to prevent or kill existing infection. Hydrogels composed of self-assembling β-hairpin peptides, having a high content of arginine, were found to be extremely effective at killing both gram-positive and gram-negative bacteria, including multi-drug resistant Pseudomonas aeruginosa. No added antibacterial agents are necessary to realize activity. Using self-assembling peptides for material construction allows facile structure-activity relationships to be determined since changes in

Characterization and release kinetics of nicotinamide microemulsion-based gels.

The aim of this study was to investigate physicochemical characteristics and to determine in vitro release kinetics of prepared nicotinamide microemulsion-based gels (MBGs). Nicotinamide microemulsions (ME) were composed of 3% w/w nicotinamide, 7% w/w water, 25% w/w soybean oil, and 65% w/w of 9:1 oleth-10:isopropyl alcohol mixture. A water-in-oil (w/o) type ME was converted to three MBGs. ME was combined with 5% w/w of colloidal silica to obtain MBG-1, with 5% w/w of 0.5% w/w carbomer solution to obtain MBG-2, or with a mixture of 3% w/w of 0.5% w/w carbomer solution and 2% w/w of PEG-40 hydr

Shear wave propagation in anisotropic soft tissues and gels.

The propagation of shear waves in soft tissue can be visualized by magnetic resonance elastography (MRE) to characterize tissue mechanical properties. Dynamic deformation of brain tissue arising from shear wave propagation may underlie the pathology of blast-induced traumatic brain injury. White matter in the brain, like other biological materials, exhibits a transversely isotropic structure, due to the arrangement of parallel fibers. Appropriate mathematical models and well-characterized experimental systems are needed to understand wave propagation in these structures. In this paper we revie

Development of the ambroxol gels for enhanced transdermal delivery.

Ambroxol is an expectoration improver and mucolytic agent that has been used to treat acute and chronic disorders. However, ambroxol needs to be administered percutaneously in order to avoid systemic adverse effects, such as headache, drowsiness, dizziness, and insomnia, which can occur after oral administration. The aim of this study was to develop a gel preparation containing a permeation enhancer to enhance the delivery of ambroxol. The ambroxol gels were prepared using hydroxypropyl methylcellulose (HPMC) and poloxamer 407. The release characteristics of the drug from the gels were examine

Complement-binding anti-Jka not detectable by DiaMed gels.

BACKGROUND: An anti-Jka was not detectable by the DiaMed-ID Micro Typing System but was strongly reactive when tested using other indirect antiglobulin test (IAT) methods. However, it was detectable in the DiaMed-ID system when the recommended diluent ID-Diluent 2, was replaced by other low ionic Strength saline (LISS) solutions. This led us to believe that the problem lay with the ID-Diluent 2. METHODS: The anti-Jka was investigated using various low ionic strength diluents in the IAT against both polyspecific and monospecific antiglobulin reagents. We tested the ID-Diluent 2 for its ability

Controlled trials of aluminum hydroxide gels for peptic ulcer.

Relief of duodenal ulcer pain by aluminum hydroxide gel (AG) was compared with that obtained by a dummy gel (DG) in randomized trials. In 44 individual pain episodes, complete relief was obtained by 15-ml doses of AG in 79% and by DG in 45% (P less than 0.05). In 48 identical blind trials replicated at another hospital the difference was not significant. The gels were also tested against ulcer pain induced by intragastric acid instillation (Palmer test) in 35 patients; pain was relieved by AG in 63% and by DG in 62%. Presumed effectiveness in terminating ulcer episodes was studied in 65 patien

Drug Delivery Investigation of Trans-Ferulic Acid Loaded Proniosomal Gels.

BACKGROUND: Proniosomes are anhydrous, free-flowing formulations that, upon hydration, yield niosomes-lipid-based vesicles. These biocompatible vesicles represent a promising vehicle for drug delivery in melanoma treatment. METHODS: This study aimed to develop a melanoma therapy by combining the benefits of proniosomes with the reported anticancer activity of the natural phenolic compound Trans-ferulic acid (T-FA). Six formulations of T-FA-loaded-proniosomes (PrN-1 to PrN-6) were prepared and compared in terms of their physicochemical parameters, entrapment efficiency, and antioxidant activit

Spontaneous Phase Separation Enables Rapid, Polymerization-Free Fabrication of Gels.

Hydrogels are cross-linked polymeric networks with wide applications in drug delivery, tissue engineering, biosensing, and environmental remediation. These hydrogels additionally host living cells, small molecules, and biological propagules, which further expand the applications of these materials. However, most, if not all, fabrication methods require covalent modifications. In this work, by deliberately selecting polymers with a known propensity to phase separate and formulating compositions far from the binodal boundary, we demonstrate the propensity of the system to transition directly int

Platinum(II) terpyridine-based supramolecular polymer gels with induced chirality.

Metal-ligand binding plays a crucial role in regulating the photophysical properties of supramolecular gels. In this study, we designed 1-Pt complexes comprising a central benzene-1,3,5-tricarboxamide unit functionalized with three terpyridines, which can form supramolecular gels with Pt(II). The resulting supramolecular gel of 1-Pt exhibited strong orange emission, which was attributed to the metal-to-metal ligand charge transfer during gel formation. Furthermore, the temperature-dependent absorption spectrum changes of the supramolecular polymer 1-Pt exhibited a nonsigmoidal transition, foll

Current applications of platelet gels in wound healing-A review.

Human platelets play important roles in several physiologic and pathologic processes. Platelet concentrates are activated with thrombin or calcium, resulting in a viscous coagulum (platelet gel [PG]), composed of 95% platelets at least. PG is increasingly used for the treatment of a variety of soft and hard tissue defects, most notably in the management of chronic non-healing wounds. During wound healing, platelets not only play a critical role in primary hemostasis and thrombosis, but also release growth factors and cytokines to promote tissue regeneration, enhance collagen synthesis, and tri

Formulation and evaluation of secnidazole or doxycycline dento-oral gels.

Local delivery of antibiotics has been shown to be effective in reducing periodontopathic microorganisms. The purpose of this study is to formulate gels containing secnidazole or doxycycline hydrochloride that could be used in the treatment of periodontitis by direct periodontal intrapocket administration. Different mucoadhesive polymers were used as cellulose derivatives, carbopol and eudragit. The prepared gels were evaluated for their in vitro drug release, rheological behavior, and mucoadhesive force. Increasing the concentration of each polymer increased the viscosity, mucoadhesion, and t

Induction of plasmacytomas in genetically susceptible mice with silicone gels.

Silicone gels injected intraperitoneally into strains of mice related to BALB/c develop plasmacytomas in approximately the same numbers and with similar phenotypes as previously obtained with pristane. Silicone gels produce few side effects and are well tolerated for long periods. Silicone gels contain several components that are potentially biologically active: residual vinyl groups and platinum. Microscopic and histological evidence suggests the silicone gel is degraded over a long period of time. Artificial Life Approach for Controlling Fibrillation in Self-Oscillating Gels.

Biological rhythms are fundamental to life but are highly complex and difficult to study directly in living systems. Understanding how these rhythms can be controlled is crucial for addressing rhythm disorders. This challenge has increased interest in software-based Artificial Life (ALife) approaches, which allow systematic investigation of rhythmic phenomena under controlled conditions. Here, we numerically examined the response of self-oscillating gels to external mechanical forces, focusing on fibrillation control and resonance. A coupled model of the Belousov-Zhabotinsky (BZ) reaction and

Lanthanide-based metal-organic gels for flexible X-ray imaging.

Flexible X-ray imaging technology requires scintillators that combine excellent flexibility and high stability. Although traditional inorganic scintillators offer good performance, their brittleness and demanding synthesis conditions limit their applications. In this work, a series of lanthanide-based metal-organic gels (Ln-MOGs, Ln = Tb3+, Eu3+), named Ln-CPTPY, was prepared through a facile one-pot method at room temperature. Through adjusting molar ratio of Tb3+ and Eu3+ ions, we achieved tunable emission color from green to red in both photoluminescence and X-ray excited luminescence modes

Thermoresponsive Gels Based on Cross-Linked Polymer-Grafted Cellulose Nanocrystals.

Stimuli-responsive nanocomposite hydrogels have garnered significant interest as alternatives to conventional hydrogels, enabling the engineering of stimuli-responsive behavior and network connectivity through composition and architecture. Here, we report thermoresponsive, "one-component" nanocomposite hydrogels composed of copolymer-grafted cellulose nanocrystals (CNCs). Thermoresponsive polyacrylamides or poly(oligoethylene glycol acrylate) copolymers bearing terminal olefin side chains were grafted from the CNC surfaces using atom-transfer radical polymerization, yielding densely grafted ha
