Evolution of putative barrier loci at an intermediate stage of speciation with gene flow in campions (Silene).

Understanding the origin of new species is a central goal in evolutionary biology. Diverging lineages often evolve highly heterogeneous patterns of genetic differentiation; however, the underlying mechanisms are not well understood. We investigated evolutionary processes governing genetic differentiation between the hybridizing campions Silene dioica (L.) Clairv. and S. latifolia Poiret. Demographic modelling indicated that the two species diverged with gene flow. The best-supported scenario with heterogeneity in both migration rate and effective population size suggested that a small proporti

Divergent sorting of a balanced ancestral polymorphism underlies the establishment of gene-flow barriers in Capsella.

In the Bateson-Dobzhansky-Muller model of genetic incompatibilities post-zygotic gene-flow barriers arise by fixation of novel alleles at interacting loci in separated populations. Many such incompatibilities are polymorphic in plants, implying an important role for genetic drift or balancing selection in their origin and evolution. Here we show that NPR1 and RPP5 loci cause a genetic incompatibility between the incipient species Capsella grandiflora and C. rubella, and the more distantly related C. rubella and C. orientalis. The incompatible RPP5 allele results from a mutation in C. rubella,

Flow cytometry and gene expression profiling of immune cells of the carotid plaque and peripheral blood.

OBJECTIVES: The relative contribution of the local vs. peripheral inflammation to the atherothrombotic processes is unknown. We compared the inflammatory status of the immune cells of the carotid plaque with similar cells in peripheral circulation of patients with advanced carotid disease (PCDs). METHODS: Mononuclear cells (MNCs) were extracted from carotid endarterectomy (CEA) samples by enzymatic digestion and subsequent magnetic cell sorting. The cell surface antigenic expressions, and mRNA expression levels were compared between CEA MNCs and peripheral MNCs, using flow cytometry and RT-PC

Genome-wide barriers to gene flow reveal the genetic basis of viviparity evolution in a lizard.

Viviparity (live-bearing) is a major evolutionary transition repeatedly linked with ecological and evolutionary diversification throughout vertebrates. Live-bearing reproduction entails a novel suite of phenotypes and life history traits, but the genetic processes by which such a reproductive innovation evolves are unknown. Remarkable among amniotes, the common lizard (Zootoca vivipara) has extant oviparous (egg-laying) and viviparous lineages and a to-date unresolved history of parity mode emergence. This species represents an ideal model to reconstruct the evolutionary and genetic mechanisms

Cessation of Gene Flow Associated With the Reduction of a Sexually Selected Phenotype in the Island Stag Beetle.

Whether gene flow acts as a creative or constraining force on local adaptation is a fundamental question in evolutionary biology, yet its role in the evolution of sexually selected traits remains poorly understood. Here we combine whole-genome and functional analyses to investigate how geological isolation is associated with the evolution of sexually selected traits. We focused on the stag beetle genus Prosopocoilus of the Izu Islands, Japan. In the mainland population (MLP), males possess sexually selected enlarged mandibles. In contrast, across the Izu Islands, relative mandible size decreas

Effect of the Central American Isthmus on gene flow and divergence of the American crocodile (Crocodylus acutus).

The final formation of the Central American Isthmus (CAI) about 3.5 million years ago altered global ocean circulation, connected North and South America terrestrial biotas, and established the Caribbean Sea. The nature of this event creates a natural scenario to test vicariance, divergence, and speciation by allopatry. Studies have shown the effect of the CAI on marine and terrestrial species, but none have examined a large-bodied amphibious taxon. We used RAD sequencing on populations of the American crocodile (Crocodylus acutus) to study the genomic variation of C. acutus on both sides of t

Iron stress genes in marine Synechococcus and the development of a flow cytometric iron stress assay.

Marine Synechococcus are frequently found in environments where iron (Fe) is a limiting nutrient. To understand their capacity to respond to Fe stress, we screened picoplankton genomes and the Global Ocean Survey metagenome for known Fe stress genes. Many open ocean strains of Synechococcus lack most known genes for Fe stress, while coastal and upwelling strains contain many, suggesting that maintaining multiple Fe limitation compensation strategies is not a selective advantage in the open ocean. All genomes contained iron deficiency-induced protein A (IdiA) and its complementary Fe(3+) transp

Genomic Connectivity and Adaptive Gene Flow of Anopheles gambiae s. s. Across East and Central Africa.

Population connectivity influences the adaptive capacity of disease vectors by mediating the spread of insecticide resistance alleles. High gene flow poses challenges for tailored intervention strategies targeting localised populations, but connectivity is often poorly characterised for particular geographical regions. This study uses whole-genome data to investigate the population connectivity of the Anopheles gambiae malaria vector across East-Central Africa, including understudied populations from Ethiopia. We also investigated adaptive gene flow using Diplotype clustering at known insectic

Population genomics reveals gene flow and positive selection patterns in the wine-related yeast Hanseniaspora uvarum.

Hanseniaspora uvarum is a representative non-Saccharomyces species that plays a significant role in fermentation processes such as winemaking. In recent years, this species has gained attention in food engineering and evolutionary biology. However, the population genomic signatures in this species remain poorly understood. In this study, a population genomics analysis was conducted on 151 H. uvarum strains (45 from Ningxia, China; 21 from other regions of China; 67 from Australia; and 18 from other regions or of unspecified origin), and a pangenome analysis was performed on 159 strains, incorp

Fine-Scale Landscape Genomics Show Asymmetric Patterns of Gene Flow for the Invasive Mosquito Aedes albopictus.

Mosquito-borne viruses like dengue, Zika, and chikungunya pose increasing health risks in the United States due to the expanding range of Aedes albopictus, a highly invasive mosquito species that now has a global distribution. Aedes albopictus thrive in artificial containers associated with anthropogenic land use, allowing populations to reach high numbers in urban and suburban environments. While the global spread of Ae. albopictus has been well characterized, the effects of heterogeneous urban landscapes on dispersal and gene flow at fine spatial scales remain unclear. This study analyzed th

Effects of luteal blood flow on endometrial progesterone concentrations and gene expression in Japanese Black cows.

Luteal blood flow (LBF) is essential for progesterone (P4) biosynthesis in the corpus luteum (CL) and affects bovine fertility. However, the mechanism by which LBF affects fertility remains unclear. This study was conducted to investigate the effects of LBF on endometrial P4 concentrations and gene expression. Endometrial biopsies and blood samples were collected from 13 Japanese Black cows after ultrasound examination on Day 7 (Day 0 = day of estrus). Based on LBF, the cows were divided into low- (LV; n = 5), medium- (MV; n = 2), and high-vascularity (HV; n = 6) groups. Plasma and endometrial

Interploidy gene flow involving the sexual-asexual cycle facilitates the diversification of gynogenetic triploid Carassius fish.

Asexual vertebrates are rare and at risk of extinction due to their restricted adaptability through the loss of genetic recombination. We explore the mechanisms behind the generation and maintenance of genetic diversity in triploid asexual (gynogenetic) Carassius auratus fish, which is widespread in East Asian fresh waters and exhibits one of the most extensive distribution among asexual vertebrates despite its dependence on host sperm. Our analyses of genetic composition using dozens of genetic markers and genome-wide transcriptome sequencing uncover admixed genetic composition of Japanese as

Spatial gradients of introgressed ancestry reveal cryptic connectivity patterns in a high gene flow marine fish.

Assessing genetic connectivity among populations in high gene flow species is sometimes insufficient to evaluate demographic connectivity. Genetic differentiation quickly becomes zero as soon as a few dozen migrants are exchanged per generation. This provides little information to determine whether migration can ensure demographic coupling. The resulting difficulties in delineating conservation units for the management of commercially exploited marine fish species are well illustrated in the case of the European sea bass (Dicentrarchus labrax). Previous attempts to assess connectivity patterns

β-catenin promotes endothelial survival by regulating eNOS activity and flow-dependent anti-apoptotic gene expression.

Increased endothelial cell (EC) apoptosis is associated with the development of atherosclerotic plaques that develop predominantly at sites exposed to disturbed flow (DF). Strategies to promote EC survival may therefore represent a novel therapeutic approach in cardiovascular disease. Nitric oxide (NO) and β-catenin have both been shown to promote cell survival and they interact in ECs as we previously demonstrated. Here we investigated the physiological role of β-catenin as a mediator of NO-induced cell survival in ECs. We found that β-catenin depleted human umbilical vein ECs (HUVEC) stimula

MYH9 gene polymorphisms may be associated with cerebrovascular blood flow in patients with type 2 diabetes.

Genetic factors play an important role in type 2 diabetes (T2D) complications. Alteration of cerebrovascular blood flow (CBF) is a direct result of cerebrovascular diseases. However, few studies have reported the role of genetics on CBF in patients with T2D. We investigated whether single-nucleotide polymorphisms (SNPs) in metabolic disease genes are associated with CBF in patients with T2D. CBF velocities of CBF were measured in 337 Han Chinese patients with T2D using transcranial Doppler sonography, with 54 cerebrovascular blood flow parameters documented. Fifty-two SNPs from 31 metabolic di

Gene transcriptional expression of cerebral blood flow alterations in Parkinson's disease: a transcription-neuroimaging association study.

AIM: Despite the intimate link between cerebral blood flow (CBF) alterations and Parkinson's disease (PD) pathogenesis and cognitive decline, the precise pathophysiological mechanisms driving this relationship remain elusive. This study seeks to correlate changes in CBF with regional gene expression to advance mechanistic understanding of the disease. METHODS: CBF group differences were first determined from ASL data in 53 PD and 40 healthy controls (HC) and subsequently correlated with cognitive scores (Dataset 1). A coordinate-based meta-analysis of published literature provided a second se

Cline Widths, Barriers to Gene Flow and Reproductive Isolation: A Comment on Gippner et al. 2026.

Hybrid zones provide opportunities to study the operation of barriers to gene flow as they play out in natural environments. However, comparative analysis is difficult because both hybrid zones and the methods used to analyse them vary widely. Gippner et al. (2026) brought together data on 108 animal hybrid zones. They argued that cline width in these zones provides a proxy for reproductive isolation (RI). Here, we comment on what cline widths do and don't tell us about the evolutionary progression towards the completion of speciation.

Simultaneous Dual-Gene Diagnosis of SARS-CoV-2 Based on CRISPR/Cas9-Mediated Lateral Flow Assay.

Few methods for the detection of SARS-CoV-2 currently have the capability to simultaneously detect two genes in a single test, which is a key measure to improve detection accuracy, as adopted by the gold standard RT-qPCR method. Developed here is a CRISPR/Cas9-mediated triple-line lateral flow assay (TL-LFA) combined with multiplex reverse transcription-recombinase polymerase amplification (RT-RPA) for rapid and simultaneous dual-gene detection of SARS-CoV-2 in a single strip test. This assay is characterized by the detection of envelope (E) and open reading frame 1ab (Orf1ab) genes from cell-
